    Abstract:

    The White-Eared-Opossum (Didelphis albiventris) is a marsupial that occupies a large variety of habitats, spreading through several Brazilian biomes and some urban areas [1], with an important role in seed dissemination. As a mammalian, its Tongue evolved to specialized functions and, in the neonate, in addition to milk suckling, it also serves to keep them attached to the female nipple, as a support for protection within the marsupium, reflecting the success of organogenesis. Furthermore, the lingual papillae provides an indicative of the animal habits, mechanical uses, diet and taxonomic relationships [2]. The organs were collected from individuals found dead by the roads in the state of Rio Grande do Sul, southern of Brazil. This material was previously fixed in modified Karnovsky aqueous solution and, once identified the tissue quality, it was submitted to processing for scanning (SEM) and transmission (TEM) electron microscopy. Through TEM the epithelial layers that forms the lingual Mucosa were easily distinguished and identified, both in adults and fetuses. The stratum corneum at both stages demonstrate the epithelial renewal process through the release of the most superficial cells; the granular layer is more tenuous in fetuses than in adults, where many tonofilaments and several keratohyaline granules were observed; the spinous layer was similar in both ages and marked by the large number of desmossomes between the cells. Two adult Tongues were used in the SEM study. One was used for the Mucosa analysis, and revealed filiform papillae with different morphologies according to the region. The apex is filled with “major” papillae, like acute columns. The space among them is filled with tuft of “minor” thin and flat filiform papillae. This “minor” filiform papillae covers the entire top of the body surface but is much denser than in the apex, giving a velvet appearance to the organ. The fungiform papillae are scattered in the apex and body, and partially covered by an elevation of Mucosa, looking like circumvallate papillae, since it present a moat-like trough. Circumvallate papillae are 3 in number and form a triangle on the lingual root, with the apex oriented caudally. Foliate papillae are small and thin elevations in the caudolateral border of the Tongue, ranging from 12 to 16. The other SEM study used corrosion with Sodium Hydroxide to remove the epithelial layer, reaching the connective tissue that supports the lingual Mucosa. It was observed that the filiform papillae subepithelial tissue is similar to what is seen with the conventional technique. The other types of papillae present an irregular surface with some dorsal depressions. The subepithelial tissue of the two circumvallate papillae, located more rostrally, are oval like the epithelium, while the third is rounded.     With 4 figures Summary The presence of lingual papillae and the nerve endings in the middle region of the Tongue Mucosa of collared peccary (Tayassu tajacu) were studied using scanning electron microscopy and light microscopy, based upon the silver impregnation method. The middle region of Tongue Mucosa revealed numerous filiform and fungiform papillae. The thick epithelial layer showed epithelial cells and a dense connective tissue layer containing nerve fibre bundles and capillaries. The sensory nerve endings, intensely stained by silver impregnation, were usually non-encapsulated and extended into the connective tissue of the filiform and fungiform papillae very close to the epithelial cells. In some regions, the sensory nerves fibres formed a dense and complex network of fine fibrils. The presence of these nerve fibrils may characterize the mechanisms of transmission of sensitive impulses to the Tongue Mucosa.

    Abstract:

    Togue Mucosa surface of 3-day postnatal rats was examined under transmission electron microscopy (TEM) and high-resolution scanning electron microscopy (HRSEM). For HRSEM analysis, the specimens were fixed in the same solution for 24 h, postfixed in 2% osmiun tetroxide, critical-point dried and coated with platinum-palladium. For TEM analysis, the specimens were fixed using modified Karnovsky solution and embedded in Spurr resin. The results revealed the presence of numerous microplicae in the membrane surface of keratinized epithelial cells to which groups of bacteria were attached. These bacteria were staphylococcus and coccus organized either in rows or at random, which were visualized in three-dimensional HRSEM images. At high magnification, the TEM images revealed the adhesion of bacteria to the cell membrane through numerous filamentous structures comprising the glycocalyx. The fine fibrillar structures rising from each bacterium and from cell membrane were clearly seen. These characteristics on bacteria structure may be used for future control or prevention of bacterial diseases and for installation of the oral native flora.

    El objetivo de esta investigacion fue estudiar las fibras sensitivas mielinizadas y amielinicas localizadas en la lamina propia subepitelial de la Mucosa lingual de ratas. Se uso el metodo de impregnacion argentica, microscopia MET y mediciones morfometricas. Los resultados revelaron que los fasciculos de fibras subepiteliales de las regiones anterior, media y posterior de la lengua provenian de su capa muscular profunda. Los grupos de fibras fueron localizados dentro del tejido conectivo de la lamina propia. Estas fibras nerviosas se ramificaron varias veces y en el tejido conectivo de las papilas formaron terminaciones simples o ramificados. Las papilas fungiformes y valadas contenian numerosas terminaciones nerviosas. La ultraestructura demostro en el axoplasma la presencia de neurofilamentos, mitocondrias y microtubulos; aunque los registros morfometricos de las fibras mielinizadas mostraron que alrededor del 44% tenian un diametro entre 3 y 4 µm, el valor promedio fue de 4.5 µm. El diametro mayor fue de 12 µm y el menor de 1.4 um. Los rangos de menores diametros fueron de 1 a 3 µm, siendo el promedio de 2.33 µm. En relacion al espesor de las vainas de mielina, los valores obtenidos fueron de 0.2 a 0.8 um siendo el valor promedio de alrededor de 0,5 µm, en el 90% de ellas. En las fibras amielinicas los mayores diametros (62%) variaron entre 0.25 y 0.75 µm. El valor promedio fue de 0.6 µm, siendo el valor maximo 3.17 µm y el minimo 0.2 µm. El menor diametro fue obtenido en 44% de las fibras amielinicas y los rangos variaron entre 0.2 y 0.4 µm. El valor maximo obtenido fue de 1 µm y el minimo 0.12 µm

    A traves de miscroscopia electronica de transmision se examinaron las celulas del epitelio de la Mucosa lingual de ratas Wistar recien nacidas. Los tejidos fueron fijados en solucion de Karnovksy modificada e incluidos en resina Spurr. Se obtuvieron delgados cortes de 90 nm y se observaron al microscopio electronico de transmision Jeol 1010. Se encontro que las celulas epiteliales queratinizadas escamosas de la Mucosa lingual expuestas al cadmio, presentaron grupos de numerosas bacterias las cuales estaban fijadas a las superficies de las membranas celulares. Los cocos y bacilos estaban fijados a las membranas celulares organizadas en pilas o columnas o al azar, como se aprecia claramente en las imagenes obtenidas a traves de la microscopia electronica de transmision. Las imagenes mostraron que las bacterias estaban fijadas por estructuras fibrilares desde una a otra y desde las memebranas de las celulas epiteliales a las bacterias. El grupo control demostro que en las membranas celulares no era evidente la presencia de cadmio

    Summary The quantitative differences in consistency of normal Mucosa, leukoplakia and squamous cell carcinoma of the Tongue were studied. Forty-five patients with squamous cell carcinoma of the Tongue, 26 with leukoplakia and 80 with normal Tongues were examined using an instrument capable of measuring consistency as a function of the load (in grams) needed to displace Tongue Mucosa by 2.0 mm using a probe 2.0 mm in diameter. The mean value for consistency in Tongue carcinoma, leukoplakia and normal Tongue was 22.1±8.0 g, 8.3±2.0 g and 4.8±0.7 g, respectively.     Lipid metabolic reprogramming is one hallmark of cancer. Lipid metabolism is regulated by numerous enzymes, many of which are targeted by several drugs on the market. We aimed to characterize the lipid alterations in oral squamous cell carcinoma (OSCC) as a basis for understanding its lipid metabolism, thus identifying potential therapeutic targets. We compared lipid species, classes, and glycerophospholipid (GPL) fatty acid species between paired tumor tissue and healthy oral Tongue Mucosa samples from 10 OSCC patients using a QExactive mass spectrometer. After filtering the 1370 lipid species identified, we analyzed 349 species: 71 were significantly increased in OSCC. The GPL metabolism pathway was most represented by the lipids differing in OSCC (P = .005). Cholesterol and the GPLs phosphatidylcholines, phosphatidylethanolamines, and phosphatidylinositols were most significantly increased in OSCC tissue (FC 1.8, 2.0, 2.1, and 2.3 and, P = .003, P = .005, P = .002, P = .007). In conclusion, we have demonstrated a shift in the lipid metabolism in these OSCC samples by characterizing the detailed landscape. Predominantly, cholesterol and GPL metabolism were altered, suggesting that interactions with sterol regulatory binding proteins may be involved. The FA composition changes of the GPLs suggest increased de novo lipogenesis.

    Abstract The application of the carcinogen 7,12-dimethylbenz(a)anthracene (DMBA) can initiate and promote the development of oral squamous cell carcinoma of the Tongue and buccal Mucosa. In this study the level of expression of various markers related to the development of programmed cell death (PCD) in the respective oral carcinomas was analyzed. Sixteen male and female Syrian hamsters ( Mesocrietus auratus ) were treated with 0.05% DMBA for 16 weeks. Immunohistochemistry was used to observe the expression of p53, proliferating cell nuclear antigen (PCNA), Bcl-2, and nucleosome formation. Single-strand conformational polymorphism (SSCP) for exons 2–9 and sequence analysis of exon 9 of the p53 gene from normal buccal or Tongue Mucosa as well as the squamous cell carcinomas from the buccal Mucosa or the Tongue were determined. p53 (wild type) expression was significantly reduced in the Tongue dysplastic Mucosa or squamous cell carcinoma. The SSCP disclosed banding shifts or new bands in exons 2/3, 4, 8, and 9 for the Tongue or buccal oral carcinomas (five of each). In exon 9 the mutation in codon 307 (ala)GCC–GTC(val) was present in the Tongue but not in the buccal carcinoma. Other markers included the level of PCNA. PCNA was initially lower in the premalignant Tongue lesions but increased in oral squamous cell carcinoma at both sites. In contrast, the amount of nucleosome formation in the Tongue carcinomas was less than the level noted for buccal cancers but premalignant dysplasias in the Tongue Mucosa exhibited higher levels. The inhibitor of PCD, Bcl-2 was lower for dysplasias and carcinomas of the Tongue compared to similar lesions of the buccal Mucosa. These results indicate that oral carcinomas of different anatomical sites can exhibit differences in growth, oncogene mutation expression, and the development of PCD. The differences in Bcl-2 and nucleosome formation may signify their influence on oncogene expression and growth potential for developing transformed clones and established oral carcinomas.

    Background: Mucopolysaccharidosis type I (MPS I) is caused by a deficiency of alfa‑iduronidase (IDUA), which leads to intralysosomal accumulation of glysosaminoglycans. Evidences point secondary events like oxidative stress on lysosomal storage diseases including MPS I. Patients with MPS I present a wide range of oral clinical manifestations, including Tongue hypertrophy, hypertrophyc alveolar process, and carious teeth. However, the mechanisms by which these alterations occur are still not fully understood. The aim of this study was to analyze the proliferative activity as well as apoptosis in Tongue Mucosa cells from murine model of MPS I. Materials and Methods: Protein expression of apoptotic markers such as p53, bcl‑2 and bax were evaluated in this setting. Ki‑67 was used as a proliferative marker. All analyses were made by immunohistochemistry in Tongue cells. Statistical analysis was perfomed by Kruskal-Wallis nonparametric test followed by the Dunn’s test. P < 0.05 was considered for statistic significance. Results: Histopathological analysis revealed no remarkable differences in Tongue Mucosa on MPS I mice when compared to control. By contrast, our results demonstrated that bcl‑2 immunoexpression was decreased in mice Tongue Mucosa cells of MPS I mice. p53, bax and ki‑67 immunoexpresssion did not show significant differences among controls and MPS I mice. Conclusion: Taken together, our results suggest that IDUA deficiency, which characterizes MPS I, may induce apoptosis in mice Tongue cells as a result of bcl‑2 down regulation.     Beedi smokers (2.65%) and chewers of pan with tobacco (2.3%) had more prevalence of leukoplakia compared to other form of smoking and chewing habits. Present study showed that tobacco users having the habit for the past 2–5 years of duration had higher prevalence of oral Mucosal conditions. Discussion: The present study showed that tobacco use in any form caused oral Mucosal conditions. The different forms of tobacco use along with duration of use had its significant effect on severity of oral Mucosal and periodontal condition.

    While it has been clearly demonstrated that smoking is the most significant exogenous factor involved in oral carcinogenesis, little is known about the global molecular and cellular changes that occur prior to the appearance of clinically detectable symptoms. Thus, the aim of this study was to investigate the expressivity of bcl-2, bax and PCNA in the rat Tongue Mucosa exposed to cigarette smoke by means of immunohistochemistry. A total of twelve male Wistar rats were distributed into 2 groups: negative control and experimental group exposed to cigarette smoke during 75 days. After experimental period, no histopathological changes in the Tongue Mucosa were detected in the negative control and the experimental group. On the other hand, an overexpression of bcl-2 was detected (p 0.05). Also, the labeling index for bcl-2 and bax showed an increase 75 days after cigarette exposure (p < 0.01). PCNA-labeling index did not show remarkable changes between groups. Taken together, our results show that bcl-2 is overexpressed in the rat Tongue keratinocytes after cigarette smoke exposure.
