    Increased muscle loading results in the phosphorylation of the 70 kDa ribosomal S6 kinase (p70S6k), and this event is strongly correlated with the degree of muscle adaptation following resistance exercise. Whether insulin resistance or the comorbidities associated with this disorder may affect the ability of skeletal muscle to activate p70S6k signaling following an exercise stimulus remains unclear. Here, we compare the contraction-induced activation of p70S6k signaling in the plantaris muscles of lean and insulin resistant obese Zucker Rats following a single bout of increased contractile loading. Compared to lean animals, the basal phosphorylation of p70S6k (Thr389; % and Thr421/Ser424; %), Akt (Thr308; %), and mTOR (Ser2448; %) was higher in obese animals. Contraction increased the phosphorylation of p70S6k (Thr389), Akt (Ser473), and mTOR (Ser2448) in both models however the magnitude and kinetics of activation differed between models. These results suggest that contraction-induced activation of p70S6k signaling is altered in the muscle of the insulin resistant obese Zucker Rat.

    Abstract The metabolic syndrome (MetS) is a major public health problem in the United States. Chronic inflammation is a critical component of the MetS, leading to dramatically increased risk of type II diabetes and cardiovascular disease. This study investigates the ability of a wild-blueberry-enriched diet to improve the proinflammatory status associated with MetS in the obese Zucker Rat (OZR). Circulating levels of key inflammatory markers and their expression in the liver and abdominal adipose tissue were examined in OZR and its genetic control, the lean Zucker Rat (LZR), after feeding a control or an 8% wild blueberry diet (WB) for 8 weeks from age 8 to 16 weeks. In the OZR, WB consumption resulted in decreased plasma concentRations of tumor necrosis factor (TNF)-α (−25.6%, P P P P Results of this study suggest that wild blueberry consumption exerts an overall anti-inflammatory effect in the OZR, a model of the metabolic syndrome.

    This study evaluates the effect of wild blueberry (WB) consumption on the biomechanical properties of the aorta in the obese Zucker Rat (OZR), a model of the metabolic syndrome. Thirty-six OZRs and 36 lean controls (lean Zucker Rats) were placed either on a WB-enriched or a control (C) diet for 8 weeks. Phenylephrine (Phe)-mediated vasoconstriction and acetylcholine (Ach)-mediated vasorelaxation in the aortic vessel were investigated, as well as the contribution of the nitric oxide synthase and cyclooxygenase (COX) pathways in each of the above responses by using specific inhibitors. Obese Zucker Rats exhibited a reduced vasocontstrictor response to Phe and an exaggeRated vasorelaxant response to Ach. The WB diet partially restored Phe-induced constrictor responses and attenuated Ach-induced relaxant responses in OZR.     To evaluate the ontogeny of adipose tissue dynamics in obese and lean Zucker Rat pups, from suckling to puberty. The trial had a two-group parallel design. Sixty-two male Zucker Rat pups shared within 15 litters received deuteRated water for 5 days, prior killing at different age. Adipose tissues were collected for 2H-enrichment analyses using mass spectrometry to determine fat cell prolifeRation and lipid synthesis Rates. Rats were assigned to obese and lean Rat groups by genotyping. The time course (from days 13 to 55) of all adipose tissue growth showed that the highest fractional Rates of fat cell prolifeRation, triacylglycerol (TG) synthesis and de novo lipogenesis (DNL) took place during early suckling in all Rat pups. The appearance of excessive fat mass growth in the obese Rats, as compared with lean Rats, was first shown through a significant increase in DNL at the end of suckling (P<0.05). The TG synthesis Rate was enhanced (P<0.05) from the end of suckling and early postweaning until day 55 (from 122±10 to 498±78 in obese pups and from 25±6 to 75±26 mg new TG per day in lean pups (median±s.e.m., P<0.01)). In contrast, only by day 55 did the fractional prolifeRation Rate of fat cells in retroperitoneal and epididymal depots in the obese Rats supersede that of the lean Rats (P<0.05). The early suckling period constitutes the most active period for adipose tissue development in normal Rats. In the obese Zucker Rat model, adipose hypertrophy primarily contributes to the early onset of obesity, while hyperplasia increases after puberty. Early onset of adipose tissue growth may play a determinant role in the development of obesity later in life.
