New insights into morphological features of Hadjelia truncata (Spirurida: Habronematidae), as revealed by SEM.

Houston, Gabriel Sentíes-cuéAbstract:Hadjelia truncata (Spirurida: Habronematidae) is a nematode found in the gizzard of several avian species. However, it has been reported pathogenic only in pigeons (Columba livia), in which it causes severe ventriculitis. Scanning electron microscopy (SEM) was used to study the morphological aspects of adult worms of this nematode in detail. Males and females measured 6.5-9 and 12-16.5 mm, respectively. The bodies were straight with moderate uniform thickness in both males and females, narrowing at the anterior end. The cuticle was striated transversely. The mouth was surrounded by two lateral trilobed lips, and the dorsal and ventral interlabia were present. The two cephalic papillae were present on each side lodged just behind the upper and lower bases of each lip. Around the mouth, a large amphid was seen on each lip. At the anterior end of both male and female worms, an excretory Pore on the ventral side and a pair of lateral cervical papillae were observed. In the female, the vulva was located at the anterior end of the body. The lips of the vulva protruded above the body surface, and the bluntly rounded posterior end showed an Anal Pore and two subterminal phasmids. The posterior end of the males curved ventrally, with large caudal alae supported by four pairs of stalked precloacal papillae, a single medial precloacal papilla, two pairs of postcloacal papillae, and a cluster of small papillae on the caudal extremity. Spicules were unequal and dissimilar.

Fine structure of body surface of Thelazia skrjabini (Nematoda: Spirurida, Thelaziidae).
Parasitology research, 2006Co-Authors: Soraya NaemAbstract:Thelazia skrjabini (Spirurida, Thelaziidae) is a parasite of cattle that causes ocular infections. Scanning electron microscopy was used to study the surface ultrastructure of adult worms of this nematode. At the anterior end of both sexes, the buccal opening was orbicular. Around the mouth, two circles of cephalic papillae were seen: the inner circle with six papillae and the outer circle with four submedian papillae. At both lateral sides of the head, two amphids were observed. A pair of lateral cervical papillae was present. In the female, the vulva was located at the anterior part of the body and the tail carried an Anal Pore and two phasmids near the tip. In the male, the tail was blunt without caudal alae and curved ventrally. There were 31 to 38 unpaired preAnal papillae, two paired postAnal papillae, and two phasmids at the posterior end. The cuticle had fine, scarcely visible transversally striations. At the anterior end of the two cases, one abnormal mass was seen.

Ultrastructural observations on the surface of Thelazia lacrymalis (Nematoda: Spirurida, Thelaziidae).

At the anterior end, the buccal capsule was cup shaped. The mouth was without lips and was surrounded by four cephalic papillae and two amphids. There were two lateral cervical papillae. Also, a single small papilla was seen on the ventral side, close to the vulvar opening. The vulva was located posterior to the end of the oesophagus and the tail carried an Anal Pore and a pair of phasmids near the tip. The cuticle was transversally annulated with fine striations.

Abomasa of 42 Egyptian camels Camelus dromedarius (Artiodactyla: Camelidae) were collected monthly from September 2013 to April 2014 from the main slaughter house of Cairo, Egypt. Adult male and female nematode worms were recovered from 26 (62 %) specimens of the examined abomasa. The parasites were of yellow color; the body was filiform (slender) tapered towards the anterior end in male and towards both ends in female. Buccal capsules absent, the buccal cavity was small with a conspicuous dorsal lancet extended from dorsal wall. The cervical papillae were prominent and spine-like. The body length of the female worm was 16.6–20.5 (18.5 ± 0.3) mm. The anterior end to the cervical papillae was 3.19–4.30 (4.12 ± 0.5) mm. The vulva of the female had a linguiform process or flap, the tail is without a spine, and the Anal Pore at the posterior end of the body had a simple dorsal rim. The body of male was 10.4–14.7 (13.9 ± 2.0) mm in length. The male bursa had elongated lobes supported by long, slender rays. The small dorsal lobe was asymmetrical with Y-shaped dorsal rays. The spicules were long with a length of 0.52–0.54 (0.53 ± 0.05) mm, each provided with a small barb and Pore near its extremity. Synlophe was bilaterally and dorsoventrally symmetrical; it extended from cephalic expansion over anterior 50 % of prebursal or prevulvar body and consisted of a maximum of 42 ridges. The described species herein was compared with the three morphologically similar species Haemonchus mitchelli, Haemonchus okapiae, and H. longistipes with their synlophes consist of 42 ridges distributed over the anterior half of the body. These species can be separated by unique structural characteristics of their synlophes, spicules, and copulatory bursa. The most morphologically similar species to the recovered worm was H. longistipes. Also, some of the parameters with regard to morphology and morphometry of this parasite were described for the first time.

Taxonomic Justification of the Pathogenic Strongylid Infecting the Arabian Camel Camelus Dromedarius as Haemonchus Longistipes by Morphological and Molecular Phylogeny.

Al-kahtani, Rewaida Abdel-gaberAbstract:Introduction: There is still lack of morphological and phylogenetic information on the pathogenic nematode of the camel Haemonchus longistipes. In the present study, this parasite was isolated in Saudi Arabia and described. Material and Methods: The abomasa of two Arabian camels were collected from a slaughterhouse in Abha province and examined for nematode infection. Worms were described morphologically and morphometrically by electron microscopy. Multiple sequence alignment and the phylogenetic tree of the parasite were constructed from maximum likelihood Analysis of its ITS-2 rDNA sequences. Results: These nematodes had a slender body terminating anteriorly at a conspicuous dorsal lancet. A pair of lateral cervical papillae distant from the anterior end was observed. The buccal aperture was hexagonal and surrounded by two amphids, six externo-labial papillae, and four cephalic papillae. Males terminated posteriorly at a bursa supported by spicules and lateral and dorsal rays. Females were linguiform and knobbed morphotypes with distinct ovijectors and a dorsal rim covering the Anal Pore. The taxonomy was confirmed by the morphology and number of the longitudinal cuticular ridges in a 43–46 range. The sequence alignment and phylogeny revealed 92% homology with H. longistipes (AJ577461.1), and the sequence was deposited into GenBank. Conclusion: The present study describes H. longistipes morphologically and molecularly which facilitates further discrimination of this species worldwide.

Parascaris equorum and Habronema microstoma are of the most endemic nematodes of the world which are currently the major cause of PGE of the domestic horses in Egypt. The present investigation introduced the first morphological description of these nematodes recovered from the domestic horse, Equus ferus caballus (Equidae), in Egypt by light and scanning electron microscopy. Seven P. equorum (fifth stage) and 18 adults of H. microstoma were recovered from the gastrointestinal tracts of four young domestic horses collected during the year of 2015. Microscopic examination of the isolated fifth stage P. equorum revealed that it possessed a long body with a broad anterior end equipped by large shamrock-like lips with deep transverse groove on medial surface set off from the rest of the body by a deep post-labial constriction giving the body a shouldered appearance. The total body length was 12–15 (14 ± 2) cm for males and 13–18 (16 ± 2) cm for females. Lips were three in number in the form of one dorsal and two sub-ventral surrounding the central stoma. The isolated adult worms of H. microstoma were whitish in color narrowed slightly at the anterior end. Single lateral ala in the cephalic region in both sexes was observed. The buccal vestibule was markedly thickened and equipped by two tridentate teeth. The adult worms had two bilobed lateral lips surrounding the mouth with four sub-median cephalic papillae and two amphids. The males were 14.5–18.0 (17.2 ± 0.3) mm long and 1.23–1.57 (1.42 ± 0.3) mm wide. The posterior end was spirally coiled and had wide caudal alae. The spicules were unequal. The females were 13.5–21.0 (16.2 ± 0.3) mm long and 1.55–1.75 (1.69 ± 0.3) mm wide. The Anal Pore had a thin upper rim and was located 177.0 μm from the posterior end.

Vector competence of Haemaphysalis longicornis ticks for a Japanese isolate of the Thogoto virus

In this study, we investigated the vector competence of H. longicornis ticks for a Japanese isolate of the Thogoto virus using Anal Pore microinjection and experimental virus acquisition. Our results showed that Anal Pore microinjection can readily infect adult ticks, and THOV-infected ticks can successfully transmit the virus to mice. Blood feeding was also critical in the distribution of the virus in tick organs, most especially in the salivary glands. Furthermore, co-feeding between an infected adult and naïve nymphs can also produce infected molted adults that can horizontally transmit THOV to mice. Altogether, our results suggest that H. longicornis is a competent vector for the Japanese THOV isolate and could be the primary tick vector of the virus in Japan.

Synchronous Langat Virus Infection of Haemaphysalis longicornis Using Anal Pore Microinjection

The transmission of this group of viruses is commonly associated with tick species such as Ixodes spp., Dermacentor spp., and Hyalomma spp. In the case of Haemaphysalis longicornis, the detection and isolation of flaviviruses have been previously reported. However, studies showing survival dynamics of any tick-borne flavivirus in H. longicornis are still lacking. In this study, an Anal Pore microinjection method was used to infect adult H. longicornis with Langat virus (LGTV), a naturally attenuated member of the TBEV serocomplex. LGTV detection in ticks was done by real-time PCR, virus isolation, and indirect immunofluorescent antibody test. The maximum viral titer was recorded at 28 days post-inoculation, and midgut cells were shown to be the primary replication site. The tick can also harbor the virus for at least 120 days and can successfully transmit LGTV to susceptible mice as confirmed by detection of LGTV antibodies. However, no transovarial transmission was observed from the egg and larval samples. Taken together, our results highly suggest that Anal Pore microinjection can be an effective method in infecting adult H. longicornis, which can greatly assist in our efforts to study tick and virus interactions.

Buccal capsules absent, the buccal cavity was small with a conspicuous dorsal lancet extended from dorsal wall. The cervical papillae were prominent and spine-like. The body length of the female worm was 16.6–20.5 (18.5 ± 0.3) mm. The anterior end to the cervical papillae was 3.19–4.30 (4.12 ± 0.5) mm. The vulva of the female had a linguiform process or flap, the tail is without a spine, and the Anal Pore at the posterior end of the body had a simple dorsal rim. The body of male was 10.4–14.7 (13.9 ± 2.0) mm in length. The male bursa had elongated lobes supported by long, slender rays. The small dorsal lobe was asymmetrical with Y-shaped dorsal rays. The spicules were long with a length of 0.52–0.54 (0.53 ± 0.05) mm, each provided with a small barb and Pore near its extremity. Synlophe was bilaterally and dorsoventrally symmetrical; it extended from cephalic expansion over anterior 50 % of prebursal or prevulvar body and consisted of a maximum of 42 ridges. The described species herein was compared with the three morphologically similar species Haemonchus mitchelli, Haemonchus okapiae, and H. longistipes with their synlophes consist of 42 ridges distributed over the anterior half of the body. These species can be separated by unique structural characteristics of their synlophes, spicules, and copulatory bursa. The most morphologically similar species to the recovered worm was H. longistipes. Also, some of the parameters with regard to morphology and morphometry of this parasite were described for the first time.
