While vertical transmission of L. infantum from pregnant bitches to their offspring has been documented, thus far no studies have explored the possibility of vertical transmission of A. platys in dogs. This study investigated the occurrence of vertical transmission of L. infantum and A. platys in sheltered dogs during the first half of gestation, in an area of southern Italy characterised by a high incidence of infection by both pathogens.

Both patients were exposed to Rhipicephalus sanguineus, the presumed tick vector, and experienced chronic, nonspecific clinical signs including headaches and muscle pains. Intra-platelet inclusion bodies resembling A. platys were observed in buffy coat smears and A. platys DNA was amplified and sequenced from whole blood; however, treatment with doxycycline did not alleviate their symptoms. These cases provide further support for A. platys as a zoonotic tick-borne pathogen, most likely of low pathogenicity; nonetheless, the cause of illness in humans by A. platys is yet to be confirmed.

During elective surgeries, substantial intraoperative hemorrhage occurs in some seroreactive dogs. Objectives To assess hemostatic parameters and bleeding tendencies as well as prevalence of PCR positivity in apparently healthy A. platys and E. canis seroreactive and seronegative free-roaming dogs from Grenada. Animals Forty-seven elective surgery dogs allocated to 4 groups: Seronegative control (n = 12), A. platys (n = 10), E. canis (n = 14) and A. platys, and E. canis (n = 11) seroreactive. Methods Preoperatively, hemostasis was assessed by platelet count, prothrombin time, activated partial thromboplastin time, and buccal mucosal bleeding time. Intra- and postoperative bleeding scores were subjectively assigned. Blood, spleen, bone marrow, and lymph node aspirates were tested by PCR. Results Bleeding scores in dogs coseroreactive for A. platys and E. canis were higher (P = .015) than those of seronegative dogs. A. platys DNA was amplified from 7/21 (33%) A. platys seroreactive dogs and from 1 E. canis seroreactive dog; E. canis DNA was amplified from 21/25 (84%) E. canis seroreactive dogs. E. canis DNA was amplified most often from blood, whereas A. platys DNA was amplified most often from bone marrow. Conclusions and Clinical Importance Apparently healthy, free-roaming dogs coseropositive for A. platys and E. canis may have increased intraoperative bleeding tendencies despite normal hemostatic parameters. Future investigations should explore the potential for vascular injury as a cause for bleeding in these dogs. Improved tick control is needed for dogs in Grenada.

Persistent intravascular infection with these bacteria is well documented in dogs, but is less well documented in human beings. Methods: Serology and PCR targeting multiple microbial genes, followed by DNA sequencing, was used to test sequential blood samples. Tissue culture isolation was attempted in two laboratories. Results: A. platys, E. chaffeensis, and E. ewingii DNA was amplified from two Anaplasma and Ehrlichia seronegative family members and their dog, all lacking typical symptoms of anaplasmosis or ehrlichiosis. Following treatment with doxycycline, the dog and mother were Anaplasma and Ehrlichia spp. PCR negative. Conclusions: Sequential PCR testing provided molecular evidence supporting intravascular persistence of A. platys and Ehrlichia spp. in two humans and their dog. Diagnosticians and clinicians should consider the potential for co-infections due to these tick borne organisms.

Alguns recentes relatos tem descrito uma doenca severa principalmente associada a infeccao por Ehrlichia canis. Este relato descreve os principais achados clinicos, hematologicos e bioquimicos da coinfeccao por A. platys e E. canis diagnosticada atraves da PCR e nested PCR, usando oligonucleotideos especificos. Os caes apresentavam sinais inespecificos, sem disturbios hemostaticos como trombocitopenia, considerado principal achado da infeccao por A. platys e comum na E. canis . Anemia foi observada nos dois casos, e em nenhum houve alteracao nos niveis sericos de ureia, creatinina, alanino aminotransferase e fosfatase alcalina. Em um cao, observou-se hiperglobulinemia, decorrente da resposta imune, concordando com a literatura que cita exacerbacao clinica associada a este tipo de coinfeccao.

Studies show a high risk of haemoparasite transmission during blood transfusion in canines. These parasites include Leishmania chagasi, Anaplasma platys, and Ehrlichia canis, which are endemic to the city of Cuiaba. This study aimed to evaluate the occurrence of L. chagasi, Trypanosoma cruzi, Babesia (canis) vogeli, A. platys, and E. canis in canine blood donor candidates, and identify possible factors associated with the infection of these agents. Sixty-six canines were evaluated using serologic and molecular tests, for the presence of the Leishmania species. While one canine sample showed a positive result for L. chagasi with indirect fluorescent antibody test, with titer of 1:40, and seven canine samples were positive using DPP, all other samples were negative when using PCR and ELISA. All canines were negative for T. cruzi when using PCR. The B. (c.) vogeli infection was identified in one canine and A. platys was identified in six canines. E. canis was identified in 17 canines, with a prevalence of 25.7%. There were no significant factors associated with the infection of the pathogens investigated. Given the observation of infection, even in the absence of clinical symptoms, emphasis must be placed on the need for the use of more sensitive and specific diagnostic methods for the screening of donor canines.

Dentre estas, estao a erliquiose monocitica canina e trombocitopenia ciclica com distribuicao cosmopolita. Com o objetivo de verificar a presenca de DNA de Anaplasma platys e Ehrlichia canis em carrapatos R. sanguineus coletados no periodo de 2008 a 2009 de 380 caes infestados. Os carrapatos, apos a maceracao, foram submetidos a extracao de DNA e, em seguida, foi realizada a Nested PCR para a amplificacao da especie A. platys e E. canis . Destes, 81 (29.7%) amplificaram o DNA dos agentes ehrlichiais, onde 38 (17.9%) amplificaram para E. canis e 32 (15.7%) para A. platys . A observacao dos dois patogenos, combinado com distribuicao mundial do carrapato R. sanguineus , demonstra o elevado risco de infeccao por esses patogenos de caes na cidade de Cuiaba.

O diagnostico pode ser feito pela deteccao de morulas em esfregaco sanguineo, sorologicamente, pelo isolamento, com excecao de A. platys e pelos metodos moleculares, como a PCR. O objetivo desta pesquisa foi avaliar a presenca de infeccao por E. canis e A. platys em caes neonatos e suas respectivas maes. Durante o periodo de junho a dezembro de 2006, foram avaliados clinicamente 21 femeas higidas e 40 filhotes de suas respectivas ninhadas com idade entre cinco a 29 dias de idade. Para a avaliacao da infeccao por E. canis , realizaram-se analise sorologica com o ensaio SNAP 3DX, observacao de morulas em esfregaco sanguineo e nested PCR e para A. platys, a nested PCR. Dos 61 animais avaliados, 36 (59%) apresentaram-se positivos para E. canis e 16 (26,2%) para A. platys . Infeccao isolada por A. platys e co-infeccao com E. canis foi observada em cinco (13,8%) e 11 (30,5%) caes, respectivamente, nao havendo diferenca estatisticamente significativa (P=0,63). Com os dados obtidos pela tecnica da n PCR conseguiu-se detectar infeccao por E. canis e A. platys em neonatos e suas respectivas maes, levantando a possibilidade de transmissao transplacentaria, entretanto estudos mais aprofundados devem ser realizados para se confirmar tal aspecto, principalmente em areas endemicas para a doenca.

2006Co-Authors: Valeria Regia Franco SousaAbstract:Este trabalho teve por objetivo investigar a infeccao por Ehrlichia canis e Anaplasma platys nos caes atendidos no Hospital Veterinario da Universidade Federal de Mato Grosso, atraves das tecnicas de esfregaco sanguineo e de PCR, analisando os achados clinicos e laboratoriais. Durante o periodo de maio de 2004 a julho de 2005 foram examinados 195 caes com sinais clinicos sugestivos de erlichiose ou com contactantes com a infeccao. A trombocitopenia canina ciclica, causada pelo A. platys, provoca diminuicao das plaquetas a cada 14 dias, sem provocar sinais severos, mas quando associada a infeccao por E. canis a gravidade clinica aumenta. Nos treze caes atendidos com PCR positiva para E. canis foi possivel verificar a diversidade de sinais, com predominância estatisticamente significativa dos disturbios hemorragicos. Ja nos 195 caes testados pela tecnica de esfregaco sanguineo nao houve predominio de nenhum sinal clinico. Os achados hematologicos tambem foram inespecificos, ocorrendo tanto anemia, leucopenia e trombocitopenia, quanto normalidade ou aumento das celulas. As alteracoes observadas na analise bioquimica nao foram exclusivas dos grupos com infeccao. Nestes grupos ocorreu aumento das proteinas plasmaticas, com hiperglobulinemia, sem, no entanto haver diferenca significativa, apesar deste achado ser frequente na erliquiose. A partir da analise da PCR confirmou-se que a infeccao por E. canis e A. platys ocorre nos caes atendidos no Hospital Veterinario da UFMT, devendo ser tomadas medidas preventivas, principalmente no controle do vetor, ja que todos os carrapatos encontrados nos caes examinados foram da especie Rhipicephalus sanguineus.

Se recolectaron 144 muestras de sangre entre enero y diciembre de 2012. De estas, el 29.2% (42/144) fue positiva (individuos trombocitopenicos con presencia de corpusculos de inclusion en plaquetas) y el 12.5% (18/144) resulto sospechoso (individuos no trombocitopenicos y presencia de corpusculos de inclusion en plaquetas) a la identificacion hematologica. Comparative Immunology Microbiology and Infectious Diseases, 2013Co-Authors: Diego Fernando Eiras, Maria Belen Craviotto, Dario Vezzani, Osnat Eyal, Gad BanethAbstract:Bacteria belonging to the Anaplasmataceae family are vector transmitted agents that affect a variety of vertebrate hosts including the tick-borne pathogens Ehrlichia canis and Anaplasma platys, which cause canine monocytic ehrlichiosis and cyclic thrombocytopenia, respectively. These two infections, typically reported from tropical and sub-tropical regions, have not been previously reported in dogs from Argentina. A total of 86 blood samples from dogs with suspected rickettsial disease and 28 non-suspected dogs were studied. Analysis included evaluation of hematological findings, PCR for Ehrlichia and Anaplasma species and sequencing of the positive PCR products. E. canis was detected in the blood of six dogs and A. platys in eighteen. All the dogs categorized as non-suspected were negative by PCR. Co-infection with Hepatozoon canis and Babesia vogeli was documented. This first report of E. canis and A. platys infections in dogs from Argentina indicates that these tick-borne infections have a considerably broader range than previously recognized in South America.

Therefore, their microbial carriage capacity is constantly being investigated. The aim of this study was to characterize the diversity of domestic animal pathogens in ticks collected from vegetation and the ground, from different parts of Israel. Non-engorged questing adult ticks were collected from 13 localities. A total of 1196 ticks in 131 pools-83 pools of Rhipicephalus turanicus and 48 of Rhipicephalus sanguineus (with two to ten ticks per pool)-were included in this study. In addition, 13 single free-roaming Hyalomma spp. ticks were collected. Screening by molecular techniques revealed the presence of Ehrlichia canis, Anaplasma platys, Anaplasma bovis and Babesia canis vogeli DNA in R. turanicus ticks. E. canis, A. bovis, B. canis vogeli and Candidatus Midichloria mitochondrii DNA sequences were detected in R. sanguineus ticks. Candidatus Midichloria mitochondrii DNA was also detected in Hyalomma spp. ticks. Neither Hepatozoon spp. nor Bartonella spp. DNA was detected in any of the ticks examined. This study describes the first detection of E. canis in the tick R. turanicus, which may serve as a vector of this canine pathogen; E. canis was the most common pathogen detected in the collected questing ticks. It also describes the first detection of A. bovis and Candidatus Midichloria mitochondrii in Israel. To the best of the author's knowledge, this is the first report describing the detection of DNA of the latter two pathogens in R. sanguineus, and of A. bovis in R. turanicus.

In this study, four dogs from the North of Portugal, in which an ehrlichial disease was suspected clinically, were tested by molecular methods. After DNA extraction from blood on filter paper, a 345 bp fragment of the Ehrlichia/Anaplasma 16S rRNA gene was amplified by the polymerase chain reaction (PCR). Sequence analysis of PCR products revealed one dog infected with Ehrlichia canis and three with Anaplasma platys. One of these latter animals was co-infected with Babesia canis subspecies vogeli. This is the first report of the genetic characterisation of both A. platys and E. canis in naturally infected dogs from the North of Portugal.

Samples were tested for Ehrlichia canis infection by PCR assays using 16S rRNA, dsb and p28 gene, while detection of A. platys was performed with 16S rRNA and groESL gene. The assignment of the ticks to each lineage was corroborated with 16S rDNA sequences. All ticks infected with E. canis and A. platys belonged to the tropical lineage. These results constitute the first record of E. canis infection in R. sanguineus s.l ticks from Argentina. No ticks from the temperate lineage were found to be infected with E. canis, coinciding with previous studies performed in Argentina and Uruguay where E. canis infection was not detected in R. sanguineus s.l from the temperate lineage. Because the presence of the tropical lineage of R. sanguineus s.l has been documented in tropical areas of northern Argentina between 22° and 24° of south latitude, the findings of this work indicate that transmission of E. canis and A. platys to dogs by R. sanguineus s.l probably occurs along this region.

Objetivo: Detectar y caracterizar mo-lecularmente estos patogenos en garrapatas y caninos domesticos del municipio de Bahia Blanca (provincia de Buenos Aires, Argentina). Material y Metodos : Se estudiaron 56 muestras sanguineas de caninos, 75 garrapatas Rhipicephalus sanguineus y 7 Amblyomma tigrinum . Las muestras fueron analizadas por RPC para Rickettsia (espacio intergenico 23S-5S ARNr), Ehrlichia y Anaplasma (16S ARNr), y Anaplasma platys (16S ARNr). Resultados : Se detecto positividad a Rickettsia en 12% de R. sanguineus , identificandose por secuenciacion a Rickettsia massiliae. Las muestras sanguineas de los caninos resultaron en 37,5% positivas a A. platys . The aim of the present study was to use molecular techniques to detect and characterize those pathogens in dogs and ticks from Buenos Aires city. We studied 207 Rhipicephalus sanguineus ticks and 52 canine blood samples from poor neighborhoods of Buenos Aires city. The samples were molecularly screened for the genera Rickettsia, Ehrlichia, and Anaplasma by PCR and sequencing. DNA of Rickettsia massiliae (3.4%) and Anaplasma platys (13.5%) was detected in ticks and blood samples, respectively. For characterization, the positive samples were subjected to amplification of a fragment of the 190-kDa outer membrane protein gene (spotted fever group rickettsiae) and a fragment of the groESL gene (specific for A. platys). A phylogenetic tree was constructed using the neighbor-joining method, revealing that the sequences were closely related to those of strains from other geographic regions. The results indicate that human and animal pathogens are abundant in dogs and their ticks in Buenos Aires city and portray the potentially high risk of human exposure to infection with these agents, especially in poor neighborhoods, where there is close contact with animals in an environment of poor health conditions.
