    Abstract Background NSCLC is the major type of lung cancer and the survival rates of NSCLC patients remain low. AZD9291 is a third-generation EGFR-TKI and approved to treat NSCLC patients harboring EGFR T790M mutation and common targetable activating EGFR mutations, but it has a limited effect for wtEGFR NSCLC. Purpose The current study investigated whether shikonin could enhance the antitumor effect of AZD9291 in wtEGFR NSCLC cells. Methods SRB and colony formation assay were used to detect the proliferation of NSCLC cells, propidium iodide staining was performed to detect the apoptosis, ROS was analyzed using DCFH-DA staining, and western blot was used to detect the expression of indicated proteins. Results We demonstrated that shikonin, a natural ROS inducer, could enhance the antitumor effect of AZD9291 in wtEGFR NSCLC cells. In addition, shikonin increased AZD9291-induced apoptosis accompanying with the generation of ROS and activation of ER stress. Furthermore, ROS inhibition by NAC or GSH reversed the apoptosis induced by shikonin plus AZD9291, and recovered the ER stress activated by combination treatment, indicating that ROS mediated ER stress played a vital role in this combination therapy. Moreover, shikonin increased the anticancer activity of AZD9291 in primary wtEGFR NSCLC cells through ROS-mediated ER stress. Conclusion Our study suggests that combining shikonin with AZD9291 is a promising therapeutic strategy for treating wtEGFR NSCLC patients.

    Abstract:

    Abstract Emodin (1, 3, 8-trihydroxy-6-methyl-Anthraquinone) is an Anthraquinone Derivative from the roots of Rheum officinale Baill, a Chinese herb widely and traditionally used for wound healing. Our objective was to determine whether topically applied emodin enhanced repair of rats' excisional wounds and its possible mechanism. Wounds were treated with either topical emodin (100, 200 and 400 μg/ml), recombinant human epidermal growth factor (rhEGF, 10 μg/ml), or vehicle for 7 or 14 days consecutively. At day 5 postinjury, wounds receiving emodin (400 μg/ml) were significantly smaller than those treated with vehicle. Emodin treatments had markedly more hydroxyproline content in day 7 wounds and tensile strength in day14 wounds than that of vehicle control. The level of transforming growth factor- β 1 (TGF-β 1 ) in wound tissues assessed by immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR), showed a dose-dependent increase in emodin-treated wounds compared with vehicle. Western immunoblotting analysis of wound tissues for Smad 2, 3, 4, 7 protein expression showed increase in Smad 2, 3 in the emodin-treated wounds compared with vehicle. In contrast, a reduction of Smad 7 was observed in emodin-treated wounds compared with vehicle and no change of Smad 4. In summary, our results showed that emodin promoted repair of rats' excisional wounds via a complex mechanism involving stimulation of tissue regeneration and regulating Smads-mediated TGF-β 1 signaling pathway.

    A water-soluble Anthraquinone Derivative (2-[methylamino-N-(1′-methylethyl)]-9,10-Anthraquinone monophosphate), previously found to be selectively toxic towards Planktothrix perornata at submicromolar concentrations, was studied to determine its toxic mode of action towards this cyanobacterium. Chlorophyll fluorescence was monitored as an indicator of photosynthetic efficiency, and measurement of reactive oxygen species (ROS) was performed using the ROS-sensitive probe 2′,7′-dichlorodihydrofluorescein diacetate. The effects of the herbicide paraquat (a ROS generator) as well as ascorbate and α-tocopherol (ROS scavengers) on ROS formation by P. perornata were studied. Also, the effects of different concentrations of ascorbate, α-tocopherol, and the herbicide diuron on reducing the toxicity of the water-soluble Anthraquinone Derivative towards P. perornata were determined. Our results indicate that the water-soluble Anthraquinone Derivative does not inhibit photosynthetic electron transport directly, but does generate ROS at levels that may cause toxicity towards P. perornata.

    Abstract:

    A highly selective membrane electrode for the determination of ultratrace amounts of lead was prepared. The PVC membrane electrode based on 2-(2-ethanoloxymethyl)-1-hydroxy-9,10-Anthraquinone (AQ), directly coated on graphite, exhibits a good Nernstian response for Pb(II) ions over a very wide concentration range (1.0×10−7–1.0×10−2 M) with a limit of detection of 8.0×10−8 M. It has a fast response time of ca. 10 s and can be used over a period 2 months with good reproducibility (SD=±0.2 mV). The electrode revealed a very good selectivity respect to common alkali, alkaline earth, transition and heavy metal ions and could be used in the pH range of 3.5–6.8. It was used as an indicator electrode in potentiometric titration of lead ions with chromate and oxalate, and in indirect determination of lead in spring water samples.

    Abstract A simple and fast method for extraction and determination of trace amounts of lead(II) ions using octadecyl-bonded silica membrane disks modified with 1,8-dihydroxy-2,7-bis(prop-1′-enyl)-9,10-Anthraquinone and atomic absorption spectrometry (AAS) is introduced. Extraction efficiency and the influence of flow rates, pH, and type and smallest amount of stripping acid were investigated. Maximum capacity of the membrane disks modified with 2 mg of the Anthraquinone Derivative used was found to be 255 μg Pb 2+ . The limit of detection of the proposed method is 5 ng/ml. The method is applied to the recovery of Pb 2+ from different synthetic samples and a spring water sample.
