Objectives To investigate the association of maternal urinary bisphenol concentrations during pregnancy with maternal, newborn and early childhood thyroid function. Methods This study was embedded in Generation R, a prospective, population-based birth cohort (Rotterdam, the Netherlands). Maternal urine samples were analyzed for eight Bisphenols at early ( 25 weeks) pregnancy. Maternal serum thyroid stimulating hormone (TSH), free thyroxine (FT4) and total thyroxine (TT4) were measured in early pregnancy and child TSH and FT4 were measured in cord blood and childhood. Results The final study population comprised 1,267 mothers, 853 newborns and 882 children. Of the eight Bisphenols measured, only bisphenol A (BPA) was detected in >50% of samples at all three time-points and bisphenol S (BPS) at the first time-point. There was no association of BPA or the bisphenol molar sum with maternal thyroid function. Higher BPS concentrations were associated with a higher maternal TT4 (β [95% CI] per 1 (natural-log) unit increase: 0.97 [0.03 to 1.91]) but there was no association with TSH or FT4. Furthermore, higher BPS was associated with an attenuation of the association between maternal FT4 and TSH (Pinteraction = 0.001). There was no association of early or mid-pregnancy BPA or early pregnancy BPS with cord blood or childhood TSH and FT4. A higher late pregnancy maternal BPA exposure was associated with a higher TSH in female newborns (Pinteraction = 0.06) and a higher FT4 during childhood in males (Pinteraction = 0.08). Discussion Our findings show that exposure to Bisphenols may interfere with the thyroid system during pregnancy. Furthermore, the potential developmental toxicity of exposure to Bisphenols during pregnancy could affect the thyroid system in the offspring in a sex-specific manner.

We examined associations of maternal phthalate and bisphenol urine concentrations during pregnancy with lipids, insulin, and glucose concentrations at school age. Methods In a population-based, prospective cohort study among 757 mother–child pairs, we measured maternal phthalate and bisphenol urine concentrations in first, second and third trimester of pregnancy. We measured non-fasting lipids, glucose and insulin blood concentrations of their children at a mean age of 9.7 (standard deviation 0.2) years. Analyses were performed for boys and girls separately. Results An interquartile range (IQR) higher natural log transformed third trimester maternal urine phthalic acid concentration was associated with a 0.20 (95% confidence interval (CI) 0.07–0.34) standard deviation score (SDS) higher triglycerides concentration among boys. Maternal bisphenol urine concentrations were not associated with non-fasting lipid concentrations during childhood. An IQR higher natural log transformed second trimester maternal high molecular weight phthalates (HMWP) and di-2-ethylhexylphthalate (DEHP) urine concentration were associated with a 0.19 (95% CI 0.31–0.07) respectively 0.18 (95% CI 0.31–0.06) SDS lower glucose concentration among boys. An IQR higher natural log transformed third trimester maternal bisphenol F urine concentration was associated with a 0.22 (95% CI 0.35–0.09) SDS lower non-fasting insulin concentration among boys. Conclusions Our results suggest potential persisting sex specific effects of fetal exposure to phthalates and Bisphenols on childhood lipid concentrations and glucose metabolism. Future studies are needed for replication and exploring underlying mechanisms.

We hypothesized that exposure to Bisphenols and phthalates during fetal life has persistent effects on bone development. Objectives: To analyze the associations of fetal exposure to Bisphenols and phthalates with bone health in school-aged children. Methods: Among 1,362 mother-child pairs participating in a population-based cohort study, we measured maternal urinary concentrations of Bisphenols and phthalates at first, second and third trimester with high performance liquid chromatography electrospray ionization-tandem mass spectrometry. Total body bone mineral density (BMD) and bone area (BA) were measured using dual-energy X-ray absorptiometry (DXA) at 6 and 10 years, and were both used to calculate bone mineral content (BMC) and area-adjusted BMC (aBMC, a measure of volumetric BMD). Results: Maternal bisphenol concentrations were not associated with childhood bone measures at 6 years. After adjustment for covariates and multiple testing correction, an interquartile range increase in maternal first trimester bisphenol S (BPS) concentrations was associated with lower BMD and aBMC at 10 years (−6.08 (95% confidence interval (CI), −9.97 to −2.19) mg/cm2 and -0.12 (95% CI, −0.20 to −0.04) g). Maternal third trimester low molecular weight (LMW) phthalate concentrations were associated with higher aBMC at 6 years whereas, maternal third trimester di-n-octylphthalate (DNOP) concentrations were associated with lower aBMC at 10 years. However, these associations did not remain statistically significant after multiple testing correction. Discussion: Maternal first trimester BPS concentrations are associated with lower BMD and aBMC in school-aged children. These findings should be considered as hypothesis generating and need further replication and exploration of potential underlying mechanisms.

In this study, we determined the concentrations of several parabens and their metabolites as well as bisphenol analogues in sediment collected from coastal areas of northern China. All sediment samples, including surface sediment and sediment cores, contained at least one of the parabens analyzed, and the total concentrations of parabens (ΣPBs; sum of six parabens) ranged from 1.37 to 24.2 ng/g dw (geometric mean: 3.30–6.09 g/g dw), which was comparable to or slightly higher than those found for the total concentrations of five detectable Bisphenols (ΣBPAs; geometric mean: 2.18–4.61 ng/g dw). 4-hydroxybenzoic acid, a common metabolite of parabens, was found in all samples at concentrations in the range of 6.85–437 ng/g dw, which was one order of magnitude lower than those found for benzoic acid. Methyl-, ethyl-, and propyl-parabens were the predominant paraben analogues, collectively accounting for >88% of ΣPBs. Bisphenol A and bisphenol F were the two major Bisphenols, collectively accounting for >86% of ΣBPAs. We also examined vertical profiles in concentrations of target analytes in sediment cores. The sediment core from the Shandong Peninsula showed a gradual increase in the concentrations of several parent and metabolic parabens as well as Bisphenols during the past decade. Relatively higher concentrations of parabens and Bisphenols were found in sediment cores collected from industrialized areas. Significant positive correlations were found among the concentrations of parabens in sediment, which suggested the existence of similar sources for these compounds. Overall, our findings suggest that the Bohai Sea coast is moderately contaminated with parabens and Bisphenols in comparison to other coastal areas in China or elsewhere.

Because gestational BPS can disrupt placental function and result in reproductive and metabolic disorders in the progeny, the aim of the study was to investigate BPS and BPF toxicokinetics during pregnancy using an in vivo approach. Fetal catheterizations were conducted in pregnant sheep (n = 6) at mid-pregnancy and injected with either a single dose of BPS (n = 3, 0.5 mg/kg, s.c.), or a combination of BPS, BPF, and BPA (n = 3, 0.5 mg/kg for each chemical, s.c.). Maternal and fetal blood and urine and amniotic fluid were collected over 72 h and analyzed for Bisphenols by HPLC-MS/MS. We observed significant differences in half-life, maximum concentration, and total body clearance in maternal circulation among Bisphenols. Longer half-lives were observed in fetal vs. maternal circulation for all Bisphenols. Fetal toxicokinetics differed among Bisphenols with BPS having the longest fetal half-life. All Bisphenols reached basal levels at 48 h in maternal plasma, but were still detectable in amniotic fluid, fetal urine, and fetal plasma at 72 h. In this first pregnancy toxicokinetic study of BPS and BPF we have demonstrated maternal and fetal toxicokinetic differences among all three Bisphenols. Higher BPS persistence in the fetal compartment warrants studies into progeny adverse outcomes following gestational exposure. Additionally, toxicokinetic differences among Bisphenols call for a more careful approach when extrapolating kinetic information from one bisphenol chemical to another.

Science of The Total Environment, 2019Co-Authors: Chunyang Liao, Kurunthachalam KannanAbstract:Abstract Information on the occurrence and bioaccumulation of Bisphenols and benzophenones in coastal marine invertebrates is limited. In this study, concentrations of eight bisphenol and five benzophenone analogues were determined in 186 mollusks collected from coastal areas of the Chinese Bohai Sea during the period 2006–2015. Total concentrations of Bisphenols and benzophenones were in range of not detected (nd)–58 (geometric mean: 13.0 ng/g) and nd–59.1 (2.06 ng/g) on a dry-weight basis, respectively. BPA and BPF were the major Bisphenols, collectively accounting for >90% of the total concentrations. BP-3 was the predominant benzophenone, representing 74% of the total concentrations. The mollusk species, Meretrix meretrix, accumulated high concentrations of both Bisphenols and benzophenones. A steady increase in bisphenol concentrations in mollusks was observed in recent years, whereas no obvious temporal trend was found for benzophenones. The estimated daily intakes of Bisphenols through consumption of mollusks were lower than the current reference dose. This study establishes a baseline concentration for Bisphenols and benzophenones in mollusks in Chinese coastal waters.

A survey of bisphenol A and other bisphenol analogues in foodstuffs from nine cities in China
2014Co-Authors: Chunyang Liao, Kurunthachalam KannanAbstract:Bisphenol A (BPA) is a high-production-volume chemical that is widely used in polycarbonate plastics and epoxy food-can coatings. Following several studies that have reported adverse effects of BPA over the past decade, other bisphenol analogues, such as bisphenol F (BPF), bisphenol S (BPS), bisphenol AF (BPAF), and bisphenol B (BPB), have been gradually developed as substitutes for BPA in several applications. Nevertheless, few studies have reported on the occurrence of compounds other than BPA in foodstuffs. In this study, 289 food samples (13 categories: cereals and cereal products, meat and meat products, fish and seafood, eggs, milk and milk products, bean products, fruits, vegetables, cookies/snacks, beverages, cooking oils, condiments, and others), collected from nine cities in China, were analysed for eight bisphenol analogues using high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). BPA and BPF were found widely in foodstuffs at concentrations ranging from below the limit of quantitation (LOQ) to 299 ng g–1 (mean = 4.94 ng g–1) and from below the LOQ to 623 ng g–1 (mean = 2.50 ng g–1), fresh weight, respectively. The highest total concentrations of Bisphenols (∑BPs: sum of eight Bisphenols) were found in the category of vegetables that included canned products (mean = 27.0 ng g–1), followed by fish and seafood (16.5 ng g–1) and beverages (15.6 ng g–1). ∑BP concentrations (mean = 2–3 ng g–1) in milk and milk products, cooking oils, and eggs were low. Food samples sold in metallic cans contained higher mean ∑BP concentrations (56.9 ng g–1) in comparison with those packaged in glass (0.43 ng g–1), paper (11.9 ng g–1), or plastic (6.40 ng g–1). The daily dietary intakes of Bisphenols were estimated, based on the mean concentrations measured and daily consumption rates of foods, to be 646 and 664 ng kg–1 bw day–1 for men and women, respectively.

The highest overall mean concentration of ΣBPs was found in the "others" category, which included condiments (preserved, ready-to-serve foods). A sample of mustard (dressing) and ginger, placed in the category of vegetables, contained the highest concentrations of 1130 ng/g for bisphenol F (BPF) and 237 ng/g for bisphenol P (BPP). Concentrations of BPs in beverages (mean = 0.341 ng/g) and fruits (0.698 ng/g) were low. The predominant bisphenol analogues found in foodstuffs were BPA and BPF, which accounted for 42 and 17% of the total BP concentrations, respectively. Canned foods contained higher concentrations of individual and total Bisphenols in comparison to foods sold in glass, paper, or plastic containers. On the basis of measured concentrations and daily ingestion rates of foods, the daily dietary intakes of Bisphenols (calculated from the mean concentration) were estimated to be 243, 142, 117, 63.6, and 58.6 ng/kg body weight (bw)/day for toddlers, infants, children, teenagers, and adults, respectively.

Food Chemistry, 2018Co-Authors: Hao Dong, Xiaofang Zeng, Weidong BaiAbstract:Abstract The present study reports an ultra high-performance liquid chromatography tandem mass spectrometry method for the simultaneous determination of six Bisphenols (bisphenol A, bisphenol B and bisphenol F) and alkylphenols (4-nonylphenol, 4-n-nonylphenol and octylphenol) in hotpot seasoning. Samples were dispersed in n-hexane after addition of internal standards bisphenol A-d4 and 4-n-nonylphenol-d4. Sample solutions were then centrifuged, and the supernatants purified using solid phase extraction with high polarity Carb/PSA composite fillers. Six target analytes were separated on a Waters ACQUITY BEH C18 column by gradient elution with methanol and 0.05% ammonium hydroxide in water as the mobile phase, and determined under multiple reactions monitoring mode. The limits of detection and quantitation, matrix effect, recovery and precision of the method were investigated. Results were linear in the concentration range 0.1–250 µg/L for all compounds of interest, with R2 > 0.9950. Limits of detection were in the range 0.1–0.4 μg/kg, and limits of quantitation were between 0.5 μg/kg and 1.0 μg/kg. The mean recoveries for negative samples at three spiked concentrations were in the range 87.9%–102.4%, and the intra-day precision and inter-day precision were in the ranges 2.1–8.2% and 4.8–11.2%, respectively. This method is accurate and sensitive, and had good clean-up characteristics, which might apply to screening and quantitation of target Bisphenols and alkylphenols in hotpot seasoning.

The edible oil sample was dispersed by n-hexane and extracted with ammoniated methanol-water solution. Then the target analytes were dispersedly absorbed using the polymer anion exchange (PAX) as the sorbent and eluted by acidic methanol. After that, four Bisphenols were separated on a C18 column by gradient elution with methanol and 0.05% ammonium hydroxide in water as mobile phase, detected by MS/MS under multiple reactions monitoring (MRM) mode and quantified by internal standard method. The PAX amounts, adsorption time, concentrations of formic acid in the elution solvent and volume of elution solvent for the DMSPE technique were optimized. The limit of detection and quantitation (LOD and LOQ), matrix effect, recovery and precision of the developed method were investigated. Results indicated that BPS and the rest three Bisphenols displayed excellent linearity in the concentration ranges of 0.1–50 μg/L and 0.5–250 μg/L, respectively, with correlation coefficients (R2) all larger than 0.998. Achieved MLODs (S/N = 3) varied between 0.1–0.4 μg/kg for all Bisphenols. The mean recoveries at three spiked levels in edible oil were in the range of 87.3–108%. The MP-CDP was characterized by Fourier transform infrared spectroscopy, solid-phase 13C nuclear magnetic resonance, thermo-gravimetric analysis, scanning electron microscopy, and nitrogen adsorption and desorption analysis. Results indicated that the MP-CDP had micron-level particle size, microporous structure, a high BET surface area, and a high product yield. Adsorption tests showed that the MP-CDP sorbent had high adsorption ability and fast binding kinetics for Bisphenols. Moreover, the MP-CDP presented high extraction efficiencies, high enrichment factor, good reusability and good batch-to-batch reproducibility for SPE of Bisphenols. Based on the MP-CDP sorbent, a SPE-HPLC-UV method was developed and successfully applied to simultaneously detect three Bisphenols in water samples and orange juice with the recoveries of 95.7–106.3% (RSD = 2.0–5.2%) for BPF, 92.9–107.0% (RSD = 1.5–5.1%) for BPA, and 96.0–103.5% (RSD = 1.7–5.0%) for BPAF, respectively. The limit of detection (S/N = 3) and the limit of quantification (S/N = 10) for all Bisphenols in these samples were 0.15 ng/mL and 0.5 ng/mL, respectively. The new MP-CDP can be potentially utilized as a good sorbent for simultaneous SPE of trace Bisphenols in environmental water samples and beverages.

However, there is a big knowledge gap on bioaccumulation and biomagnification of these emerging Bisphenols in aquatic organisms. The bioaccumulation and magnification of nine bisphenol analogues in aquatic organisms at different trophic levels collected from Taihu Lake, China, were evaluated. The total concentrations of the nine Bisphenols in the lake waters were in the range of 49.7-3480ng/L (mean, 389ng/L). BPA, bisphenol AF (BPAF) and bisphenol S (BPS) were the most predominant analogues in the water. The mean natural logarithm bioaccumulation factor (log BAFs) of BPAF, bisphenol C (BPC), bisphenol Z (BPZ) and bisphenol E (BPE) were greater than BPA, and there was a significantly positive correlation between log BAFs of the biphenols and their octanol-water partition coefficients (log Kow). The trophic magnification factors of BPAF, BPC and BPZ were 2.52, 2.69 and 1.71, respectively, suggesting that they had the potential to biomagnify in the food web. The results of this study call for further investigations on risk assessment of these emerging pollutants in the environment.

However, there is a big knowledge gap on bioaccumulation and biomagnification of these emerging Bisphenols in aquatic organisms. The bioaccumulation and magnification of nine bisphenol analogues in aquatic organisms at different trophic levels collected from Taihu Lake, China, were evaluated. The total concentrations of the nine Bisphenols in the lake waters were in the range of 49.7-3480ng/L (mean, 389ng/L). BPA, bisphenol AF (BPAF) and bisphenol S (BPS) were the most predominant analogues in the water. A highly class-selective dummy molecularly imprinted polymer was used for solid phase extraction (SPE) and clean-up of the samples. Bisphenols was quantified by high-performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS). The developed method had acceptable recoveries (43.6–101%), precision (RSDs: 1.5–15%) and matrix effects (−6.7 to 28%). The method limits of quantitation (MLOQs) for nine Bisphenols in sewage and sludge samples were 0.0007–16.3 ng L−1 and 0.0004–8.28 ng g−1 dry weight (dw), respectively. The method was applied to a survey of a municipal wastewater treatment plant (WWTP) in Dalian. All of the tested Bisphenols, except BPB and BPZ, were presented in the analyzed samples. BPA, BPS, and BPF with the concentrations 412, 109 and 66.4 ng L−1 in the WWTP influent, respectively, were the predominant Bisphenols. The results demonstrated that BPS and BPF have become the most frequently used substitutes of BPA.

The microsphere particles were characterized by scanning electron microscopy and nitrogen adsorption-desorption measurements, demonstrating that the regular-shaped and medium-sized particles (40-70 μm) were obtained with a specific surface area of 355.759 m(2) /g and a total pore volume of 0.561 cm(3) /g. The molecular imprinting properties of the particles were evaluated by static adsorption and chromatographic evaluation experiments. The association constant and maximum adsorption amount of bisphenol A were 0.115 mmol/L and 3.327 μmol/g using Scatchard analysis. The microsphere particles were then used as a solid-phase extraction sorbent for selective extraction of seven Bisphenols. The method of dummy molecularly imprinted solid-phase extraction coupled with high-performance liquid chromatography and diode array detection was successfully established for the extraction and determination of seven Bisphenols from environmental sediment samples with method detection limits of 0.6-1.1 ng/g. Good recoveries (75.5-105.2%) for sediment samples at two spiking levels (500 and 250 ng/g) and reproducibility (RSDs < 7.7%, n = 3) were obtained.
