purification of proctolin binding proteins from the foregut of the insect Blaberus Craniifer

Proctolin-binding sites were efficiently solubilized with either the nonionic detergent digitonin or the zwitterionic detergent Chaps, as indicated by the specific binding of 3H-proctolin to solubilized samples. A solubilized sample obtained from 1600 foregut membranes was subjected to a five-step chromatographic purification including chromatofocusing, anion-exchange and size-exclusion chromatographies. The final size-exclusion separation resulted in the isolation of approximately 100 pmol of purified proctolin-binding proteins, eluting as a single peak at approximately 74 kDa. Analysis of the purified sample using SDS/PAGE and silver staining showed two bands at 80 kDa and 76 kDa. Densitometric analysis of the gel indicated that each band contained approximately 7-8 microg of protein, suggesting that one band corresponds to the proctolin-binding activity. Proctolin-binding proteins were thus purified 1800-fold using standard chromatography.

In order to investigate lindane effects on protein release by the fat body, we used antibodies raised against egg proteins to quantify protein levels in fat body, hemolymph and ovaries of treated-fed or -decapitated females 3- or 5-days -old. In vitro assays used fat body in Grace's medium to quantify the protein amount released in the medium. Individual data for each treatment were related to their corresponding control in paired series. In vivo, ovarian enhanced protein content was linked to an enhanced protein secretion by the fat body. This was ascertained in vitro by high levels of released protein in the medium containing lindane (1 microM) by fat body from females, but not from males. This effect was inhibited by EDTA, a calcium chelator. The present results confirmed that low doses of lindane (about 3 microg/g of body mass) acted as a juvenile hormone analogue, at the level of the ovaries, by enhancing protein uptake, and also at the level of the fat body, by triggering protein release. This property is calcium-dependent.

Until now, assays were performed using juvenile stages. The aim of the present paper was to investigate the W-328 effects both on vitellogenesis (vitellogenin synthesis by the fat body and uptake by the oocytes) and reproductive behavior in the ovoviviparous cockroach Blaberus Craniifer. Imaginal molt decapitated or fed females were used in vivo for this experiment. Vitellogenesis onset and ovarian growth were demonstrated using biometrical, immunocytochemical, ELISA and electrophoretical investigations. In vitro, biosynthesis and secretory capabilities of the fat body were ascertained by antivitellin antibodies. In headless females, the analogue restores the onset of vitellogenesis. An increase in all the ovarian parameters was observed as compared to controls. The ovarian proteinogram in decapitated females resembled that of fed females of the same age. The proteins sequestered by oocytes, which originated from the fat body, were released into the haemolymph. In intact fed females, a significant dose-dependent increase in the ovarian growth was observed by comparison to controls. Up to 50 μg/female, the analogue induced a positive effect on the process, whereas it became inhibitory for higher doses. W-328 induced behavioural disturbances leading to altered mating and oviposition. 10 μg W-328 reduced mating success, and oothecae production. Sterilisation was achieved with 20 μg. These findings showed that the carbamate compound W-328 acts as a juvenile hormone analogue in adult females and causes a physiological disturbance of the reproductive physiology. They suggest that W-328 could be a useful insecticide against adult populations.

In headless females, lindane (4–20 μg/female) induced vitellogenesis onset, as detected by immunocytochemical location of sites binding anti-egg antibodies in the basal oocyte, and the electrophoregram looked like an ovary from an intact 4-day old female, but the increased ovarian parameters (oocyte volume, ovarian weight, and protein content) remained below those observed in fed virgin and isolated females 48-h old. In lindane-treated (4–20 μg/female) fed females, mating occured at D7 as in untreated females. In treated females, in which a spermatophore was present in the bursa copulatrix, the oviposition was not inhibited and vitellogenesis was not impaired by sublethal doses (4 or 10 μg/female) of this insecticide. These results suggest that vitellogenesis onset, which can be induced by low doses of lindane, a compound known to be neurotoxic, given to headless females on D2, is assumed by extracephalic factors. With regard to oviposition in fed females, we questioned the existence of a dose-dependent stimulation.

Enhancement of oocyte growth in the cockroach Blaberus Craniifer by a scorpion toxin, charybdotoxin

These results suggest that low doses charybdotoxin enhance vitellogenesis, possibly via the nervous system by means of a substance conditioning both protein synthesis by the fat body and ovarian uptake.

Proctolin promotes vitellogenesis onset in the imaginal molt decapitated cockroach Blaberus Craniifer.

Perriere, Paulette Brousse-gauryAbstract:The ovarian response of the imaginal molt decapitated cockroach, Blaberus Craniifer, to nanomolar quantities of synthetic proctolin was evaluated. Sites binding anti-egg proteins were detected in the basal oocyte in 67% (18 out of 27) of the treated females, indicating that vitellogenin uptake is mediated by proctolin. The injected concentration of proctolin was consistent with a physiological role. The results suggest that proctolin, a putative neurotransmitter or neuromodulator, possibly acts on a non-excitable cell, the oocyte. At this level, the precise mechanism is discussed with respect to the calcium-dependent processes involved in both the proctolin myogenic effect and vitellogenin uptake in other species of cockroach.

Ovarian response to trauma in imaginal molt decapitated Blaberus Craniifer (Dictyoptera, Blaberidae). Severance of gonapophyses

Sites binding anti‐egg protein antibodies were detected in the periphery of basal oocytes in 68% operated females. In headless Blaberus Craniifer, the onset of vitellogenesis which is triggered by extraphysiological doses of juvenile hormone or indolamines, was induced by this surgical procedure. It is suggested that amputation of gonapophyses affects the delivery of natural messengers inducing vitellogenesis at physiological concentration.
