    Abstract:

SEARCH FOR QUALITY: METHODOLOGICAL VALIDATION APPLIED TO THE IDENTIFICATION AND QUANTIFICATION OF CARBARYL BY DERIVATIVE SPECTROPHOTOMETRIC Abstract Agriculture is the main Parana State’s economic activity and it is the major national producer of maize, beans, soya beans, wheat and oat. It comes second on the production of manioc and potatoes. In order to guarantee the production of all these agricultural commodities it is necessary to use large quantities of pesticides. Carbaryl is one of the pesticides largely used all over the State’s agricultural land and the overexposure toCarbaryl or itsmisuse results in important cases of intoxication. Thus, it was suggested the development of a simple, reliable and reproducible analytical method for determining the presence of Carbaryl in biological material originated from those cases. In order to guarantee a high level of quality to this study, it was decided to apply to it the principles of the methodological validation. This paper presents the first part of the validation protocol, which comprehends: the experimental draft, the method adjustment to the laboratory’s work conditions, the study of Carbaryl’s physicochemical properties and its toxicokinetic data. The method suggested is the derivative spectrophotometric.

  • exposure to insecticides reduces populations of rhynchophorus palmarum in oil palm plantations with bud rot disease
    Insects, 2019
Co-Authors: Luis Carlos Martinez, Angelica Platarueda, Francisco Andres Rodriguezdimate, Juliana Mendonca Campos, Valdeir Celestino Dos Santos, Gabriela Da Silva Rolim, Flavio Lemes Fernandes, Wiane Meloni Silva, Carlos Frederico Wilcken, José Cola Zanuncio
    Abstract:

The South American palm weevil (SAPW), Rhynchophorus palmarum Linnaeus (Coleoptera: Curculionidae) is the main pest of Elaeis guineensis and damages palm trees with bud rot disease in the Americas. The effects of six neurotoxic insecticides (abamectin, carbaryl, deltamethrin, fipronil, imidacloprid and spinosad) were evaluated against SAPW for toxicity, survival, reproduction, and mortality. Abamectin (LC50 = 0.33 mg mL−1), Carbaryl (LC50 = 0.24 mg mL−1), deltamethrin (LC50 = 0.17 mg mL−1), and fipronil (LC50 = 0.42 mg mL−1) were the most toxic to SAPW. Adult survival was 95% without exposure to insecticides, decreasing to 78–65% in insects treated with the LC25 and 49–35% in insects exposed to LC50. Sublethal doses of carbaryl, fipronil and imidacloprid showed significant effect on the reproduction of this insect. Mortality of SAPW populations caused by insecticides had similar effects in the laboratory and field conditions. The results suggest that carbaryl, deltamethrin, fipronil, and imidacloprid caused significantly higher mortality as compared to the control in SAPW and may be used to control its populations in oil palm trees where bud rot appears as the key disease for SAPW attraction and infestation.

  • Insecticide selectivity to Supputius cincticeps (Stal) (Het., Pentatomidae) and its prey Spodoptera frugiperda (J. E. Smith) (Lep., Noctuidae)
    Journal of Applied Entomology, 1998
Co-Authors: José Cola Zanuncio, V. C. Batalha, Raul Narciso C. Guedes, Marcelo Coutinho Picanço
    Abstract:

: The selectivity of malathion, carbaryl, permethrin, and deltamethrin to Supputius cincticeps (Stal) (Het., Pentatomidae) and its prey Spodoptera frugiperda (J. E. Smith) (Lep., Noctuidae) (fall armyworm) was evaluated using leaves dipped in insecticide dilutions. Based on LC50 values, malathion was the least toxic insecticide to the fall armyworm followed by carbaryl, permethrin and deltamethrin which was the most toxic compound to this insect pest. Malathion was also the least toxic insecticide to the predaceous bug S. cincticeps, while carbaryl and permethrin showed intermediate toxicity and deltamethrin was the most toxic insecticide to this species. Malathion and carbaryl were more toxic to the predator than to the fall armyworm showing low selectivity in favour of the first. However, the pyrethroids showed the opposite effect with deltamethrin being the most selective insecticide in favour of S. cincticeps. Nymphs of the fifth instar and adults of the predator were more tolerant to the pyrethroids than third-instar nymphs. The other insecticides showed similar toxicity to all three developmental stages of the predator.

  • in vitro metabolism of methiocarb and carbaryl in rats and its effect on their estrogenic and antiandrogenic activities
    Environmental Toxicology and Pharmacology, 2016
Co-Authors: Satoko Tange, Nariaki Fujimoto, Naoto Uramaru, Fung Fuh Wong, Kazumi Sugihara, Shigeru Ohta, Shigeyuki Kitamura
    Abstract:

Abstract In this work, we examined the metabolism of the carbamate insecticides methiocarb and carbaryl by rat liver microsomes and plasma, and its effect on their endocrine-disrupting activities. Methiocarb and carbaryl were not enzymatically hydrolyzed by rat liver microsomes, but were hydrolyzed by rat plasma, mainly to methylthio-3,5-xylenol (MX) and 1-naphthol, respectively. When methiocarb was incubated with rat liver microsomes in the presence of NADPH, methiocarb sulfoxide was formed. The hydrolysis product, MX, was also oxidized to the sulfoxide, 3,5-dimethyl-4-(methylsulfinyl)phenol (SP), by rat liver microsomes in the presence of NADPH. These oxidase activities were catalyzed by cytochrome P450 and flavin-containing monooxygenase. Methiocarb and carbaryl both exhibited estrogen receptor α (ERα) and ERβ agonistic activity. MX and 1-naphthol showed similar activities, but methiocarb sulfoxide and SP showed markedly decreased activities. On the other hand, methiocarb and carbaryl exhibited potent antiandrogenic activity in the concentration range of 1 × 10 −6 –3 × 10 −5  M. Their hydrolysis products, MX, and 1-naphthol also showed high activity, equivalent to that of flutamide. However, methiocarb sulfoxide and SP showed relatively low activity. Thus, hydrolysis of methiocarb and carbaryl and oxidation of methiocarb to the sulfoxide markedly modified the estrogenic and antiandrogenic activities of methiocarb and carbaryl.
