Expression, purification and identification of CtCVNH, a novel anti-HIV (Human Immunodeficiency Virus) protein from Ceratopteris thalictroides.

In a previous study, we first reported the full-length sequences of the CVNH gene cloned from Ceratopteris thalictroides. Based on the finding, the coding sequence of CtCVNH was optimized in the study, and then a pET prokaryotic expression vector was constructed. The purification and identification of CtCVNH protein were investigated, as well. SDS-PAGE analysis indicated that a 31 kDa protein was overexpressed and mainly accumulated in the soluble fraction. Only a single protein was obtained after the Ni- nitrilotriacetic acid (NTA) affinity chromatography. The purified protein was identified to be the recombinant CtCVNH by both Western blot and peptide mass fingerprinting analysis.

It is 1993 bp long, contains a 723 bp open reading frame (ORF) that encodes a deduced protein (named CtCVNH) with 150 amino acid residues. CtCVNH has a predicted isoelectric point (PI) of 4.47 and a calculated molecular mass 15.9556 kDa. It possesses the conserved anti-HIV (human immunodeficiency virus) CV-N domain, which is the same as the cyanovirin-N homology (CVNH) members that were isolated from filamentous ascomycetes and C. richardii. Modeling of the tertiary structure indicated that CtCVNH is an elongated, largely β-sheet protein that displays internal two-fold pseudosymmetry. As espécies encontradas nos ambientes amostrados foram Salvinia auriculata, Salvinia molesta (Salviniaceae) e Ceratopteris thalictroides (Pteridaceae). As três espécies apresentaram caráter de ocorrência anual e as Salviniaceae estiveram amplamente distribuídas nos ambientes aquáticos observados. Dentre as monilófitas amostradas, apenas o gênero Salvinia apresenta heterosporia, com a presença de megasporângios e microsporângios. O gênero Ceratopteris, por sua vez, é homosporado, apresentando apenas esporos masculinos, que possuem um potencial para desenvolver gametófitos que futuramente poderão ser masculinos ou hermafroditas. A superfície foliar (tricomas), no caso de Salvinia, e o caule, no caso de Ceratopteris, são importantes estruturas na flutuabilidade das plantas. The species found in the study sites were Salvinia auriculata, Salvinia molesta (Salviniaceae) and Ceratopteris thalictroides (Pteridaceae), all presenting an annual occurrence in nature and Salviniaceae were widely distributed in the ponds observed. Among the monilophytes sampled, only the Salviniagenus shows heterosporia, with the presence of megasporangia and microsporangia. The genus Ceratopteris in turn is homosporate, presenting only male spores, which have a potential to develop gametophytes that in the future may be male or hermaphrodite. The leaf surface (trichomes), in the case of Salvinia, and the stem, in the case of Ceratopteris, are important structures in the buoyancy of plants.
