Osbeck) is reported to have antidiabetic, anti-hypercholesterolemia, anti-inflammatory and antioxidant activities. This study aims to find out the effectiveness of Sunkist orange peel ethanol extract (Citrus Sinensis (L.) Osbeck) on blood sugar levels at doses of 500, 750 and 1000 mg/kgBB. This study used the True Experimental Pre-Test and Post-Test Control Group as research design where sunkist orange peel was extracted by maceration method and was given orally for 14 days to streptozotocin induced wistar rat with hypercholesterolemia. Statistical test results showed that ethanol extract of Sunkist orange peel was able to lower blood sugar levels and cholesterol levels of h streptozotocin induced wistar rat with hypercholesterolemia (p<0.05). The most effective dose of Sunkist orange peel ethanol extract in lowering blood sugar levels and cholesterol levels of streptozotocin induced wistar rat with hypercholesterolemia is 1000mg/kgBB.Kondisi kadar gula darah yang tinggi pada penderita diabetes ternyata memicu peningkatan LDL dan penurunan HDL dimana pada orang dengan diabetes”energi yang didapatkan tubuh berasal dari penguraian lemak dan metabolisme protein. Kulit jeruk Sunkist (Citrus Sinensis (L.) Osbeck) dilaporkan memiliki aktivitas antidiabetik, antihiperkolesterolemia, antiinflamasi serta antioksidan . Penelitian ini bertujuan untuk mengetahui efektivitas dari ekstrak etanol kulit jeruk Sunkist (Citrus Sinensis (L.) Osbeck) terhadap kadar gula darah pada dosis 500, 750 dan 1000 mg/kgBB. V.cholerae dapat menginfeksi manusia melalui rute pencernaan (fecal-oral). Manifestasi klinik berupa penyakit kolera akan timbul apabila jumlah bakteri yang masuk mencapai jumlah tertentu. Indonesia merupakan negara megabiodiversity yang kaya akan tanaman obat, dan sangat potensial untuk dikembangkan, tapi belum dikelola secara maksimal. Tumbuhan jeruk manis merupakan salah satu jenis tanaman yang dapat dimanfaatkan untuk pengobatan. Penelitian ini bertujuan untuk mengetahui efektifitas antibakteri ekstrak kulit jeruk manis (Citrus Sinensis L) terhadap pertumbuhan bakteri Vibrio cholerae. Jenis penelitian ini adalah eksperimen laboratorik dengan menggunakan difusi cakram dengan rancangan Posttest Only Control Group Design untuk melihat aktivitas ekstrak kulit jeruk manis (Citrus Sinensis L) terhadap bakteri Vibrio cholerae. Penelitian dilakukan dengan tahapan sebagai berikut (1) Sterilisasi Alat, (2) Persiapan dan Pembuatan Ekstrak Kulit Jeruk Manis (Citrus Sinensis), (3) Pembuatan Konsentrasi Ekstrak Kulit Jeruk Manis (Citrus Sinensis), (4) Pembuatan Biakan Bakteri (5) Pembuatan Media Biakan, (6) Uji Sensitivitas. Hepatoprotective Effect of Citrus Sinensis (L.) Osbeck Ethanol Extract in Paracetamol-Induced Rats

Citrus Sinensis (L.) Osbeck (Sunkist) is a fruit that has been widely studied for its various therapeutic effects. This study aimed to explore the potential of Sunkist peels as a hepatoprotection agent against the toxicity of paracetamol. This was an experimental study conducted in May 2019 at the Pharmacy Biology Laboratory of Pharmacy Faculty, University of North Sumatera on 15 rats that were divided into 5 groups of treatment (negative, positive, and 300, 450, and 600 mg/kgBW Sunkist ethanol extract) and received treatment for 14 days. The AST and ALT was assessed using the dyasis manual assay. Data were then analyzed using one-way ANOVA, followed by Post-hoc test. Histology Study of Liver Changes Paracetamol-Induced Wistar Rats Treated with Sunkist (Citrus Sinensis L. Osbeck) Extract

This study is aimed to investigae the protection effect of ethanol extract from Citrus Sinensis by histologic study of liver tissues in paracetamol-induced wistar rats. This study was using 15 rats which were divided into 5 groups included negative control, positive control, and ethanol extract groups (300 mg/kg b.w; 450 mg/kg b.w; 600 mg/kg b.w), after 14 days the rats was terminated and the liver was process to histology view. The result of this study showed that there was reducing in severity of degeneration following increasing of ethanol extract dosage. Here we present a comprehensive analysis of the draft genome of sweet orange (Citrus Sinensis). The assembled sequence covers 87.3% of the estimated orange genome, which is relatively compact, as 20% is composed of repetitive elements. We predicted 29,445 protein-coding genes, half of which are in the heterozygous state. With additional sequencing of two more Citrus species and comparative analyses of seven Citrus genomes, we present evidence to suggest that sweet orange originated from a backcross hybrid between pummelo and mandarin. Focused analysis on genes involved in vitamin C metabolism showed that GalUR, encoding the rate-limiting enzyme of the galacturonate pathway, is significantly upregulated in orange fruit, and the recent expansion of this gene family may provide a genomic basis. This draft genome represents a valuable resource for understanding and improving many important Citrus traits in the future.

Plant Cell Reports, 2009Co-Authors: Zhiyong Pan, Rui Guan, Shiping Zhu, Xiuxin DengAbstract:Two dimensional gel electrophoresis combined with matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) was employed to study the somatic embryogenesis (SE) in Valencia sweet orange (Citrus Sinensis Osbeck). Twenty-four differentially expressed proteins were identified at five time points of Citrus SE (0, 1, 2, 3, 4 weeks after embryo initiation) covering globular, heart/torpedo and cotyledon-shaped embryo stages. The general expression patterns for these proteins were consistent with those appeared at 4 weeks of Citrus SE. The most striking feature of our study was that five proteins were predicted to be involved in glutathione (GSH) metabolism and anti-oxidative stress, and they exhibited different expression patterns during SE. Based on that oxidative stress has been validated to enhance SE, the preferential representation for anti-oxidative proteins suggests that they could have a developmental role in Citrus SE. Some proteins involved in cell division, photosynthesis and detoxification were also identified, and their possible roles in Citrus SE were discussed.

Euphytica, 2000Co-Authors: Wenwu Guo, Xiuxin DengAbstract:Protoplasts from cell suspension cultures of ‘Bonnaza’ navel orange (Citrus Sinensis (L.) Osbeck) were electrically fused with mesophyll protoplasts isolated from seedless ‘Red Blush’ grapefruit (Citrusparadisi). After 6 months of culture, a total of 20 plants were regenerated. Root tip chromosome counting revealed that 4 of them were tetraploids (2n = 4x = 36)and the rest were diploids (2n = 2x = 18) morphologically resembling the mesophyll parent. After 6 months of transplantation into the greenhouse, 4 of the diploidmesophyll regenerants unexpectedly flowered, but this phenomenon disappeared in the next year. This is the first report of precocious flowering in Citrus via protoplast fusion. RAPD analysis further confirmed that the tetraploid regenerants were somatic hybrids while the diploid regenerants were mesophyll parent type. This somatic hybrid will be utilized as a possible pollen parent for improving the seedy pummelo cultivars in China by producing triploid seedless pummelo hybrid. The mechanism of early flowering was also discussed.

GmitterAbstract:Abstract Leaf-derived protoplasts isolated from in vitro grown seedlings of Fortunella crassifolia Swing cultivar ‘Meiwa’ were fused chemically with protoplasts isolated from embryogenic suspension cultures of Citrus Sinensis (L.) Osbeck cultivar ‘Valencia’. Protoplasts were cultured following fusion, and mitosis was first observed after 10 days incubation in the dark at 28°C. Embryoids were recovered from regenerated calli 2 weeks after transfer to agar-solidified medium. Cytological evaluation of 27 embryoids revealed seven tetraploids and 20 diploids. Embryos were germinated and shoots dissected from germinated embryos were rooted in vitro. Counts of chromosomes in cells of young leaf meristems indicated that most regenerated plants were tetraploid (2n = 4x = 36). Peroxidase (PER) leaf isozyme banding patterns confirmed the somatic hybridity of recovered tetraploids. More than 50 somatic hybrid plants have been recovered and successfully acclimated.
