Jurnal Fitopatologi Indonesia, 2018Co-Authors: Dian Syahfitri, Nisa Rachmania Mubarik, Lisdar A ManafAbstract:Use of Chitinolytic Bacteria as Biological Control of Colletotrichum capsici on Chili Plants Colletotrichum capsici is known as the causal agent of anthracnose disease in chili plant and may cause reduction of crop yield. Chitinolytic bacteria, namely Serratia marcescens KAHN 15.12, Bacillus thuringiensis SAHA 12.12, and BAE 36 were reported to have antagonistic activity against C. capsici . Therefore, a study was conducted to determine the potential of chitinolytic bacteria on controlling C. capsici on chili plants in greenhouse experiment. Three bacterial isolates used as biocontrol agent was formulated by using talcum as carrier materials. The methodologies consisted of characterization of bacterial isolates, formulation of biocontrol agent, viability test of bacterial isolate, efficacy of biocontrol agents in the laboratory and in the greenhouse . Disease severity in the laboratory reached 64% when chili treated with isolate formulation of BAE 36. In the greenhouse, BAE 36 isolate formulation and consortium formulation were able to suppress infection of C. capsici; each was indicated by disease incidence of 25% and 50%, respectively . These results indicated that chitinolytic bacterial formulations could be potencial as biocontrol agents of C. capsici.

'The Indonesian Phytopathological Society', 2018Co-Authors: Dian Syahfitri, Nisa Rachmania Mubarik, Lisdar A ManafAbstract:Cendawan patogen Colletotrichum capsici merupakan salah satu penyebab penyakit antraknosa pada tanaman cabai dan menyebabkan penurunan produktivitas cabai di daerah tropis dan subtropis. Bakteri kitinolitik dan enzim kitinase mempunyai potensi sebagai pengendali biologis cendawan patogen tanaman. Penelitian sebelumnya telah berhasil mengisolasi bakteri kitinolitik yang bersifat antagonis terhadap Colletotrichum capsici, yaitu: KAHN 15.12, SAHA 12.12, dan BAE 36. Penelitian ini dilakukan untuk mengetahui potensi bakteri kitinolitik dalam mengendalikan Colletotrichum capsici pada tanaman cabai dalam skala rumah kaca. Ketiga isolat bakteri digunakan sebagai agens biokontrol dengan menggunakan talek sebagai bahan pembawa. Metode penelitian ini terdiri atas karakterisasi isolat, formulasi agens biokontrol, uji viabilitas isolat, uji in vitro agens biokontrol pada buah cabai, dan uji in vivo pada cabai dalam percobaan rumah kaca. Hasil penelitian menunjukkan bahwa formulasi isolat bakteri dapat menghambat serangan Colletotrichum capsici 50%-75%. Secara keseluruhan, penelitian ini menunjukkan bahwa formulasi bakteri kitinolitik dapat digunakan sebagai agens biokontrol dari C. capsici.

Plant Disease, 2009Co-Authors: P Montri, Paul J Taylor, Orarat MongkolpornAbstract:Eleven isolates of Colletotrichum capsici were screened on nine chili genotypes derived from four cultivated species of Capsicum: Capsicum annuum, C. baccatum, C. chinense, and C. frutescens. Host reactions were assessed 9 days after inoculation by microinjection of spores into the pericarp of red fruit. A set of disease scales, with 0 to 9 scores, were developed for anthracnose infection of each Capsicum sp. based on percent lesion size in relation to fruit size, appearance of necrotic or water-soaked tissue, and presence of acervuli. Three pathotypes, PCc1, PCc2, and PCc3, were identified according to differential qualitative infection of fruit of C. chinense genotypes PBC932 and C04714. PCc1 was the most virulent pathotype, infecting all genotypes of C. annuum, C. chinense, and C. frutescens, whereas PCc3 was the least virulent pathotype, infecting only the genotypes C. annuum and C. frutescens. Quantitative infection occurred in all chili genotypes except for genotypes of C. baccatum, where no infection occurred, demonstrating various levels of aggressiveness of isolates within pathotypes.

Butler & Bisby was studied in interspecific Capsicum populations derived from a cross between a Thai elite cultivar Capsicum annuum L. 'Bangchang' and a resistant line C. chinense Jacq. 'PBC932'. The resistance was assessed by measuring lesion area per fruit area (LFA) on detached chili fruits, using a laboratory-based injection inoculation. Nil symptoms resembling the resistant parent 'PBC932' were also identified in the progeny F2 and BC1 populations. Segregation of resistance (nil LFA) and susceptibility in the F2 fitted a 1:3 Mendelian ratio, indicating that resistance was responsible by a single recessive gene. The segregation of the trait in the test crosses in both BC1s also confirmed the 1:3 gene segregating model as found in the F2

Salah satu kendala utama dalam produksi tanaman cabai adalah serangan penyakit antraknosa yang disebabkan oleh infeksi jamur Colletotrichum sp. dan Gleosporium sp.Pengendalian penyakit antraknosa menggunakan fungisida memiliki dampak negatif seperti menyebabkan pencemaran lingkungan dan kekebalan penyebab penyakit terhadap fungisida. Salah satu alternatif yang dapat digunakan dalam pengendalian penyakit antraknosa adalah penggunaan mikroba endofit. Mikroba endofit merupakan mikroba yang tumbuh dalam jaringan tanaman pada periode tertentu dan mampu hidup dengan membentuk koloni dalam jaringan tanaman tanpa membahayakan inangnya Penelitian ini dilakukan untuk mengetahui kemampuan antagonistis mikroba endofit dari daun sirih (Piper betle L.) dan daun papaya (Carica papaya L.) terhadap jamur Colletotrichum capsici penyebab penyakit antraknosa pada cabai. Kemampuan antagonistik mikroba endofit terhadap jamur patogen ditunjukkan dengan tebentuknya zona bening pada media uji. Mikroba endofit yang berhasil diisolasi dari dari daun sirih (Piper bettle L.) sebanyak 2 isolat jamur endofit dan 6 isolat bakteri endofit. Hasil uji antagonis memperlihatkan bahwa 1 isolat bakteri endofit yakni isolat S6 dapat menghambat pertumbuhan jamur Colletotrichum capsici. Mikroba endofit yang berhasil diisolasi dari dari daun pepaya (Carica papaya L.) sebanyak 2 isolat jamur endofit dan 3 isolat bekteri endofit. Hasil uji antagonis memperlihatkan bahwa 1 isolat bakteri endofit yakni isolat P2 dan 1 isolat jamur endofit yakni isolat P42 dapat menghambat pertumbuhan jamur Colletotrichum capsici

2017Co-Authors: Rurry DahliaAbstract:Air rebusan tumbuhan kecombrang merupakan salah satu alternatif untuk mengendalikan penyakit antraknosa pada tanaman cabai. Tujuan penelitian ini untuk mengetahui aktivitas air rebusan bagian tumbuhan kecombrang (Nicolaia speciosa) yang paling berpotensi untuk mengendalikan Colletotrichum capsici penyebab penyakit antraknosa pada tanaman cabai. Penelitian telah dilaksanakan di Laboratorium Fitopatologi Jurusan Hama dan Penyakit Tumbuhan Fakultas Pertanian Universitas Andalas dari bulan Juli sampai September 2016. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 5 ulangan. Perlakuan adalah air rebusan tumbuhan kecombrang (daun, batang, akar, rimpang dan bunga) dan kontrol (aquades) dengan konsentrasi 5%. Data hasil pengamatan dianalisis dengan Uji F dan diuji lanjut dengan Least Significance Different (LSD) pada taraf 5%. Parameter yang diamati adalah pertumbuhan koloni jamur, luas koloni, jumlah konidia/ml suspensi, berat basah, berat kering dan daya kecambah konidia. Hasil uji secara in vitro didapatkan perlakuan air rebusan bunga kecombrang yang terbaik dengan efektivitas rata-rata sebesar 36,64%. A Colletotrichumine A-pathogen combination was more toxic to the host than individual inoculations of Colletotrichumine A or the pathogen. The Colletotrichumine A-pathogen combination led to an increased activity of defense related enzymes viz. PPO and SOD, while levels of the lignification enzymes PAL and PO decreased. Higher levels of phenols including catechin and ferulic acid were also observed with the Colletotrichumine A-pathogen combination compared to individual treatment. The likely role of Colletotrichumine A during chilli anthracnose infection is supported by histochemical analysis of infected plants that showed increased cell death after infection.

Journal of Plant Growth Regulation, 2016Co-Authors: Amrita Saxena, Richa Raghuwanshi, H B SinghAbstract:Biocontrol strategies have been mainly focused on proposing the use of biocontrol agents (BCAs) isolated from the rhizospheric region of the plant for protection against phytopathogens. The present study evaluates the effectiveness of phyllospheric Trichoderma isolates in elevating the defense responses in chilli against Colletotrichum capsici infection and comparing its efficiency to the conventionally recommended rhizospheric Trichoderma strains. The elicitation of the defense network in the plants was analyzed using biochemical assays for important enzymes, that is, PAL, PO, PPO, TPC, SOD along with the total protein level in challenged plants over untreated and unchallenged control plants. The results recorded 2.1, 5.18, 3, 0.67, and 0.5-fold increases in TPC, PAL, PO, PPO, and total protein content in BHUF4 (phyllopsheric Trichoderma isolate)-treated plants when compared to control plants under C. capsici challenge. This was at par with the increment recorded in T16A (rhizospheric Trichoderma isolate)-treated chilli plants. The increment in growth parameters was also recorded after treatment with the isolated Trichoderma strains. Interestingly, the phyllospheric isolate (BHUF4) treatment recorded comparable growth promotion in chilli plants recording 36, 62, and 60 % increases in one of the major parameters of plant growth, that is, root length, no. of leaves, and dry weight, respectively. This study proposes the use of combined application of both rhizospheric as well as phyllospheric Trichoderma isolates for better and all around protection of plants against foliar as well as soil phytopathogens. This would be a novel approach in biological control strategy for better management of anthracnose disease of chilli.
