Culex quinquefasciatus (Diptera: Culicidae) From Florida Transmitted Zika Virus.

ZIKV RNA was detected in bodies and in saliva deposited on filter paper cards with subsequent studies demonstrating the presence of live ZIKV in saliva.

Culex quinquefasciatus storage proteins.

CardosoAbstract:Insect storage proteins accumulate at high levels during larval development of holometabolous insects. During metamorphosis they are degraded, supplying energy and amino acids for the completion of adult development. The genome of Culex quinquefasciatus contains eleven storage protein-coding genes. Their transcripts are more abundant in larvae than in pupae and in adults. In fact, only four of these genes are transcribed in adults, two of which in blood-fed adult females but not in adult males. Transcripts corresponding to all Cx. quinquefasciatus storage proteins were detected by RT-PCR, while mass spectrometric analysis of larval and pupal proteins identified all storage proteins with the exception of one encoded by Cq LSP1.8. Our results indicate that the identified Cx. quinquefasciatus storage protein-coding genes are candidates for identifying regulatory sequences for the development of molecular tools for vector control.

Salivary gland proteins of the mosquito Culex quinquefasciatus.

The amount of protein in female salivary glands increased from 0.26 microg on day one after emergence to about 1.4 microg on day seven. The major polypeptides found in the female salivary glands had molecular weights of 35.7, 28.3, and 20.5 kDa. Antibodies produced by mice immunized by bites of Culex quinquefasciatus female mosquitoes reacted with the 35.7 and 28.3 kDa polypeptides, showing that these molecules were secreted by mosquitoes during blood feeding. The salivary glands of C. quinquefasciatus females displayed the same morphological and biochemical organization as that of Aedes aegypti mosquitoes, accumulating apyrase in the distal portions and alpha-glucosidase in the proximal portions of the gland. Bionomics of Culex quinquefasciatus within urban areas of Rio de Janeiro, Southeastern Brazil Bionomia de Culex quinquefasciatus em áreas urbanas no Rio de Janeiro, RJ
2012Co-Authors: Mariana Rocha David, Gabriel Sylvestre RibeiroAbstract:OBJECTIVE: To evaluate density, parity rates, daily survival and longevity of natural populations of Culex quinquefasciatus in three neighborhoods with distinct socio-economic and infrastructure profi les. METHODS: Mosquito collections of the Culex quinquefasciatus species were performed weekly during two four month periods, from August to November 2008 (spring) and March to June 2009 (fall), in a favela (slum), a suburban area and a middle class area of Rio de Janeiro, Southeastern Brazil. Collections were performed with backpack aspirators, in 20 randomly selected houses in each area per week, during 15-20 minutes per house. Ovaries were removed from captured females and classifi ed as initial, intermediary or fi nal stage. Furthermore, females were dissected for determination of parity based on the condition of the tracheal system. Mosquito survival rate and longevity were estimated on a per month basis for each neighborhood. RESULTS: We collected a total of 2,062 Culex quinquefasciatus, but monthly vector density was not correlated with temperature and rainfall. We dissected the ovaries of 625 Culex quinquefasciatus, and overall, there was a higher proportion of nulliparous females during the dryer months, while gravid females were more frequent in rainy months. In the middle class neighborhood, the parity rate reached up to 93.75% with survivorship of 0.979. Lower parity and survival rates were obtained in the suburban area (as low as 36.4% parity and 0.711 daily survival). Up to 84.7% of Culex quinquefasciatus females could survive the eight day period needed to complete West Nile Virus incubation. CONCLUSIONS: The survival rate of Culex quinquefasciatus varied signifi cantly between the neighborhoods. This suggests that vectorial capacity and disease transmission risk may vary greatly between different urban areas, which is potentially useful information for vector control programs.

Toxicity of Chitinase to Culex quinquefasciatus and Its Synergetic Activity with Binary Toxin
Journal of Tropical Medicine, 2010Co-Authors: Cai Yajun, Yuan Zhiming, Cai Quanxin, Yang Yankun, Wei SuzhenAbstract:Objective To study the toxicity of chitinase and it′s synergistic activity with binary toxin on mosquito larvae.Methods 2nd and 3rd stages of Culex quinquefasciatus larvae were treated with chitinase from Serretia marcescens S3 strain and binary toxin from Bacillus sphaericus C3-41 strain for 48 h.Results Chitinase had a lower level of toxicity on the susceptible Culex quinquefasciatus larvae,and the LC50 and LC90 was 8.99 μg/ml and 28.5 μg/ml,respectively.Toxicity of chitinase to the B.sphaericus-resistant Culex quinquefasciatus larvae was similar to the susceptible larvae,with the value of LC50 and LC90 of 11.9 and 168 μg/ml,respectively.In addition,chitinase not only had a delayed toxic effect on the susceptible C.quinquefasciatus larvae after the treatment,but also enhanced the larvicidal activity of binary toxin against the susceptible mosquito.When used in combination with binary toxin,1.59 μg/ml(LC2),2.84 μg/ml(LC10) and 5.61 μg/ml(LC30) of chitinase could enhance 1.98,3.53 and 8.11-fold of the toxicity of binary toxin.Conclusion Chitinase is less toxic and has a delayed effect against Culex quinquefasciatus larvae.The toxicity can be enhanced by the binary toxin.This result might provide information for further application of chitinase for mosquito control.
