First report of Culex flavivirus infection from Culex coronator (Diptera: Culicidae), Colombia

Culex flavivirus (CxFV) is an insect-specific virus of the genus Flavivirus, detected in a wide variety of mosquito species. Objective To detect Flavivirus in mosquitoes of a tropical region of the Colombian Caribbean. Methods In 2014, an entomological surveillance of arboviruses was conducted in the department of Cordoba area of the Caribbean, Colombia. A total of 8270 mosquitoes were captured as follow: Mansonia ( n = 3271/39.5%), Culex ( n = 2668/32.26%), Anopheles ( n = 840/10.15%), Aedeomyia ( n = 411/4.9%), Psorophora ( n = 397/4.8%), Coquilletidia ( n = 369/4.46%), Uranotaenia ( n = 261/3.15%) and Aedes ( n = 53/0.6%). All mosquito species were collected in dry tropical forest of the Caribbean area. Universal primers for NS5 gene (958 pb), RT-PCR for flavivirus and sequencing were used for molecular identification of viruses detected. Results Two pools belonging to Culex coronator were positive for flavivirus RNA sequence by RT-PCR. The sequences of the PCR amplicons, matched that of the Culex flaviviruses, CxFv COL PM_149 (GenBank: KR014201) and CxFv COL PM_212 (GenBank: KT307717). Phylogenetic analysis of the NS5 protein sequences of the Culex flaviviruses sequences with those of reference sequences available in GenBank indicated viruses of Genotype II, closely related to the Brazilian strain, BR_SJRP_01_ (GenBank: KT726939), from Culex sp. The alignment of Culex flavivirus sequences CxFv COL_ PM 212 and CxFv COL_ PM 149 with sequences of strains detected in different geographical regions grouped the strains in a Latin American clade reported in Brazil, Argentina and Mexico. Conclusions The present work illustrated that CxFV was circulating among vectors of human pathogenic arboviruses in Colombia, but the impact of CxFV on other flaviviruses which are endemic in the study area still remains to be explored.

Culex saltanensis and Culex interfor diptera culicidae are susceptible and competent to transmit st louis encephalitis virus flavivirus flaviviridae in central argentina

St. Louis encephalitis virus (SLEV) is endemic and autochthonous in the American continent. Culex pipiens quinquefasciatus is the primary urban vector of SLEV; however, Culex interfor and Culex saltanensis have also been found naturally infected with the virus, suggesting their potential role as vectors. OBJECTIVE: The aim of this study was to determine the vector competence of Cx. interfor and Cx. saltanensis for SLEV from central Argentina in comparison to a Cx. p. quinquefasciatus. METHODS: Adult female mosquitoes of the three Culex species were orally infected by feeding on viremic chicks that had been inoculated with SLEV CbaAr-4005, it is the same strain detected in the US and Argentina, and responsible for human disease. Then, abdomens, legs and saliva blood-fed mosquitoes were analyzed by viral plaque assay and the presence of cytopathic effect on the cell culture monolayer. RESULTS: Mosquitoes were permissive to orally acquire infections, to virus dissemination, and transmission of SLEV in the saliva. Culex saltanensis and Cx. interfor are potential vectors of SLEV. CONCLUSIONS: Our results demonstrate that in Argentina both Culex saltanensis and Culex interfor are susceptible to SLEV and competent for its transmission. Moreover they are abundant during SLEV epidemic period in urban area, positive for this virus in nature, and found to feed on natural hosts.

Morphological variation in diagnostic features for two Culex (Culex) species of the Neotropical Region (Diptera: Culicidae).

The accurate identification of the 198 species included in the subgenus Culex is dependent on a few morphological characters, principally of the male genitalia (Harbach et al. 2012). Most species within Culex ( Culex ) have been described based on male genitalia due to their low intraspecific variability compared to characters of females and larvae. Male genitalia provide synapomorphic characters useful in establishing phylogenetic relationships in Culicidae (Reinert et al. 2008, Reidenbach et al. 2009, Harbach et al. 2012, Laurito & Almiron 2013), as well as autapomorphic traits allowing specific identifications.

Phylogenetic relationships of Culex ( Culex ) species (Diptera, Culicidae) from Argentina based on morphological characters
Zootaxa, 2013Co-Authors: Magdalena Laurito, Walter Ricardo AlmironAbstract:The purpose of this study was for the first time to establish phylogenetic hypotheses among Culex ( Culex ) species from Argentina, and to evaluate the relationships of this subgenus with subgenera Phenacomyia and Phytotelmatomyia . We em-ployed maximum parsimony to analyse morphological characters derived from the morphology of the adult, pupal and larval stages. More than 30 figures, including photographs, illustrating 87 characters are provided. The cladistic analysis of 61 terminal taxa and 95 characters was undertaken, under implied weighting method, with a range of K values from 6 to 9. Three unstable taxa were detected therefore a reduced strict consensus tree was created. The subgenus Culex appears polyphyletic relative to subgenus Oculeomyia . The internal classification of subgenus Culex is shown to be chaotic; the Pipiens Group is polyphyletic relative to the Sitiens and Coronator Groups, the latter being polyphyletic with respect to the Pipiens Group. Phylogenetic analysis combining characters provided by morphology and molecular sources are need-ed to interpret more fully the relationships in the group.

Of the bird species tested, Western scrub jays, Aphelocoma coerulescens, produced the highest viremia response, followed by house finches, Carpodacus mexicanus, and house sparrows, Passer domesticus. Most likely, few mourning, Zenaidura macroura, or common ground, Columbina passerine, doves and no California quail, Callipepla californica, or chickens would infect blood-feeding Culex mosquitoes. All Western scrub jays and most house finches succumbed to infection. All avian hosts produced a lower viremia response and survived after infection with an endemic strain of St. Louis encephalitis virus. Culex species varied in their susceptibility to infection with both viruses, with Culex stigmatosoma Dyar generally most susceptible, followed by Culex tarsalis Coquillett, and then Culex p. quinquefasciatus Say....
