Immunogenicity of precocious strain developed from Shanxi strain of Eimeria tenella
Chinese journal of veterinary science, 2008Co-Authors: Cao Hong-qingAbstract:One precocious line was derived from the Shanxi strain of Eimeria tenella,and its immunogenicity was studied.The results suggested that the precocious strain had the same immunogenicity as the parental strain,but the pathogenicity of the precocious strain was found to be attenuated markedly;Its perfect immune dose for first time should be 1 500 oocytes per chicken and 2 000 for the second time.Result indicated that after secondary immunization it can provide over 3 months long protection.Dot-ELISA results indicated that the second immunity can induce the dynamic changes of sera specific IgG in chickens inoculated with Eimeria tenella,which is consistent with anticoccidial ability.

Infection of chicken is initiated by ingestion of sporulated oocysts. The oocyst wall has two typical layers: a smooth outer layer formed by wall-forming body I (WF I) and an inner layer originating from WF II. We observed that the inner layer is circumscribed by a hitherto undocumented linear suture and propose its role in the process of infection.

A simple method for the purification of Eimeria tenella sporozoites.

Mattig, Drössigk U, Rolf EntzerothAbstract:A rapid, simple and cheap method for the purification of Eimeria tenella sporozoites has been developed using commercially available filter paper (595 filter paper circles, Schleicher & Schuell, 3354 Dassel, Germany, order 311610). Yield and purity are of the same value as by purification with a leukopac column (Bontemps & Yvore 1974). The described method can be used for the purification of Eimeria tenella sporozoites under sterile conditions for subsequent in vitro cultivation.

We screened the Eimeria tenella genome database for proteases, classified these into families and determined their stage specific expression.

Study on Pathogenicity of Eimeria tenella YL Strain after Several Passages
Progress in Veterinary Medicine, 2011Co-Authors: Yu San-keAbstract:To study the pathogenicity of multiple passaged Eimeria tenella,Seven-day-old chickens were infected with sporulated oocysts of 36 passage Eimeria tenella YL strain at different dosages.The pathogenicity was assessed by clinical symptom,mortality,degree of bloody diarrhea,latent period,cecal lesions,average weight gain(AWG),and oocysts per gram faeces(OPG).The results revealed that the bloody stools were discharged from infected chickens at 3-4 days post-inoculation and the latent period showed to be 110 h.The relative weight gain(RWG) of high dosage group(1×106/per bird),middle dosage group(1×105/per bird) and low dosage group(1×104/per bird) were 72%,74%,77%,respectively,compared with control group.The cecal lesion scores of high dosage,middle dosage,and low dosage were 2,1.68,and 0.86,respectively.The oocyst yields of different dosages roughly showed normal distribution.However,middle dosage group yielded the most oocysts among different treatments.Result showed that Eimeria tenella YL strain had high pathogenicity in chickens although it was passaged for many times and the infective dosage showed positive correlation with the pathogenicity.The oocyst output from infected chickens showed the "crowd effect".

Detection of Antibodies Against UV-attenuated Eimeria tenella YL Strain Vaccine by ELISA
Progress in Veterinary Medicine, 2010Co-Authors: Yu San-keAbstract:No coccidial chicks were randomly divided into two groups,and immunized with UV-attenuated Eimeria tenella YL strain vaccine twice on the 9th day and 16th day respectively.ELISA was used to detect the changes of seral antibodies against Eimeria tenella YL strain.The test result showed that the dynamic change of antibody level in chicks presented an overall upward trend and reached the peak value(0.398) on the 11th day after the second immunization,and the antibody level stayed at a high level afterwards.The antibody level declined smoothly in control group.The dynamic changes of antibody level showed that UV-attenuated Eimeria tenella YL strain vaccine induced a strong immune protection.
