    Abstract We examined the impact of Pleistocene glacial cycles on geographical distribution and genetic structure of the mitten crab Eriocheir sensu stricto in East Asia using sequence variation of mitochondrial cytochrome c oxidase I and cytochrome b gene segments. Phylogenies revealed four distinct but shallow structured lineages in Eriocheir s. s. Three lineages dominated the East China Sea-Yellow Sea, the Sea of Japan and the South China Sea on the margins of the region, and one lineage occurred on Okinawa Island. This geographical distribution represents a general phylogeographic pattern in East Asia, which is closely associated with the fluctuations of marginal seas and islands during the Pleistocene. The four lineages are estimated to have diverged during the mid-Pleistocene. Demographic expansions were observed in each lineage, starting within the second-to-latest interglacial period in the marginal sea lineages (∼70–130 ka) and within the last glacial period in the Okinawa lineage (∼25–80 ka). Expansions have probably taken place northward along the coast of the East China Sea-Yellow Sea, following the rise of sea levels. Centered on the southern Korean Peninsula, expansions have likely occurred northward along the west coast and eastward along the south coast of the Sea of Japan. Each marginal sea has served as a single refugium during glacial periods. Two secondary contact regions were identified, one of the East China Sea-Yellow Sea and South China Sea lineages, and another of the East China Sea-Yellow Sea and Sea of Japan lineages. Phylogeography of Eriocheir s. s. provides insights into the evolutionary history and mechanism for generating biodiversity in East Asia.

    Abstract:

    The classical role of Vitellogenin (Vg) is providing energy reserves for developing embryos, but its roles appear to extend beyond this nutritional function, and its importance in host immune defense is garnering increasing research attention. However, Vg-regulated immunological functions are dependent on three different domains within different species and remain poorly understood. In the present study, we confirmed three conserved VG domains - LPD_N, DUF1943 and VWD - in the Chinese mitten crab (Eriocheir sinensis), highlighting functional similarities of Vg in vertebrates and invertebrates. Of these three domains, DUF1943 and VWD showed definitive bacterial binding activity via interaction with the signature components on microbial surfaces, but this activity was s not exhibited by the LPD_N domain. Antibacterial assays indicated that only the VWD domain inhibits bacterial proliferation, and this function may be conserved between different species due to the conserved amino acid residues. To further explore the relationship between Vg and polymeric immunoglobulin receptor (pIgR), we expressed EspIgR and the three Eriocheir sinensis-Vg (EsVg) domains in HEK293T cells, and co-immunoprecipitation assay demonstrated that only the DUF1943 domain interacts with EspIgR. Subsequent experiments demonstrated that EsVg regulates hemocyte phagocytosis by binding with EspIgR through the DUF1943 domain, thus promoting bacterial clearance and protecting the host from bacterial infection. To the best of our knowledge, our work is the first to report distinct domains in Vg inducing different immunological outcomes in invertebrates, providing new evidence that pIgR acts as a phagocytic receptor for Vg.

    Abstract:

    B52 is a member of the classical serine/arginine (SR)-rich proteins, which are phylogenetically conserved and play significant roles in mRNA maturation, including alternative splicing. In the present study, the docking site, selector sequences and locus control region of the Chinese mitten crab (Eriocheir sinensis) Down syndrome cell adhesion molecule (EsDscam) were identified. Alternative splicing of Dscam is essential to generate different isoforms. We also isolated and characterised the B52 gene from E. sinensis (EsB52). The 876 bp open reading frame of EsB52 encodes a 291 amino acid residue polypeptide, and EsB52 has two RNA recognition motifs (RRMs) at the N-terminus and an arginine/serine-rich domain at the C-terminus. Each RRM contains two degenerate short submotifs, RNP-1 and RNP2. Analysis of tissue distribution revealed that EsB52 mRNA expression was widespread in all tested tissues, and especially high in brain and hemocytes. In hemocytes, EsB52 was upregulated significantly after stimulation with pathogen-associated molecular patterns and bacteria. Furthermore, EsB52 RNAi decreased the number of Ig7 inclusion in mRNA rather than Ig2 or Ig3. Taken together, these findings suggest that EsB52 acts as an alternative splicing activator of EsDscam.

    Abstract Accessory sex gland (ASG) in Eriocheir sinensis is the important component of the male reproductive system; its function is still unknown. Here we report that ASG protein can digest the spermatophore wall effectively. By optimizing the incubation temperatures, durations, and secreted protein concentrations using the L30 (52) orthogonal method, we find the protein concentration was the important factor affecting spermatophore wall digestion. In order to find out the digestion effect and spermatozoa quality, we observed the acrosin activity and survival rate of free spermatozoa obtained by different methods. We found that the survival rate of ASG protein digestion is the highest (98.06%), and its acrosin activity of free spermatozoa was significantly higher (4.290 ± 0.095 µIU/106) than those obtained by traditional trypsinase digestion (2.446 ± 0.251 µIU/106) or mechanical homogenate (1.423 ± 0.109 µIU/106) methods, but it was still significantly lower than those obtained from the spermatheca of ma...

    Abstract:

    In order to elucidate the functional and molecular mechanisms employed during accessory sex gland seasonal development, we performed suppression subtractive hybridization (SSH) experiments in the crab, Eriocheir sinensis, by constructing forward and reverse cDNA libraries enriched with up- or down-regulated transcript genes. A total of 175 ESTs were obtained from 180 randomly picked clones in the two libraries, with an average insert size of 450 bp, and a cloning efficiency . 90%. Comparative sequence analysis of ESTs with sequences reported in public genomic databases identified 88 unigenes, 35 of which were homologous to reported proteins, with the remainder unidentified. Unigene Annotation identified some unigenes via the Gene Ontology classification. Some of these unigenes had greater activity in the early stage of gland development, with an inferred function in cellular processes (involved in cell proliferation and differentiation), and the other genes with inferred functions in protein synthesis, energy metabolism, and signal transduction were more prevalent during the peak stage of gland development. Hence, genes elevated during the early stage may play roles in the process of rapid cell growth, while those elevated during the peak stage may play important roles in protein synthesis and secretion, as well as sperm capacitation and fertilization. SSH results were validated by the real-time PCR analysis of five candidate genes, whose expression correlated well with the SSH results, and exhibited relative expression levels that were five fold greater than those observed in the SSH libraries, confirming the SSH success.

    Abstract Spiroplasma Eriocheiris, has been identified as a novel lethal pathogen of Eriocheir sinensis tremor disease (TD, typically paroxysmal tremors of the pereiopod). The thoracic ganglion of E. sinensis infected by S. Eriocheiris was the direct reason of the TD. This study was designed to understand of TD processes in crab and the immune response of thoracic ganglion at the transcriptomic levels. The transcriptomic profiles of thoracic ganglion in the experimental group (S. Eriocheiris inject, termed SI) and the control group (R2 medium inject, termed RI) were obtained using Illumina HiSeq 2500. In total, there were 60,159,089 and 47,613,445 high-quality clean reads assembled from SI and RI, respectively. In addition, 39,636 unigenes were obtained with an N50 length of 575 bp. Moreover, by RPKM analysis, a total of 8049 differentially expressed genes (SI vs RI) were detected, including 927 up-regulated and 7122 down-regulated genes. The transcription levels of gene in prophenoloxidase-activating system (proPO), Wnt signaling pathway, phagocytosis, Toll-like system, Ca2+ regulation related process, etc. of SI were significantly changed compare with RI. Finally, 12 selected differentially expressed genes (Serpin, TLR, FZ4, SPI2, Tax1, TLSP, AI, CTLR, Rab11A, LAMP-1, Trx1 and PI3K) were verified using qRT-PCR showed similar tendency with the RNA-seq. The results shown that the proPO, Wnt signaling pathway, phagocytosis, Toll-like system, Ca2+ regulation related process, glutamate signal related process, etc. were involved in the process of the S. Eriocheiris infection.

    This study mainly investigated the composition of adult male Chinese mitten crab (Eriocheir sinensis) from four grades/sizes (Grade I: 200–249 g; Grade II: 175–199 g; Grade III: 150–174 g; Grade IV: ≤ 150 g). The results showed that the grade III crabs had the largest gonadsomatic index (GSI), which was significantly higher than the grade I and grade II crabs, no significant difference was found with the grade IV crab. Significant differences in moisture and total lipid contents were observed among various edible parts from different grades of male Eriocheir sinensis. In particular, grade II crabs had the highest total lipid and dry matter content for hepatopancreas. A balanced amino acids composition and a high essential amino acids score (EAAS) were found in the muscle and gonads of grade III crabs. The levels of poly-unsaturated fatty acids (PUFA), n-3 PUFA, n-6 PUFA and docosahexaenoic acid (DHA) in the hepatopancreas, as well as the contents of PUFA, highly-unsaturated fatty acids (HUFA), n-3 PUFA, arachidonic acid (ARA), and eicosapentaenoic acid (EPA) in the gonads were significantly increased in the grade II crabs. Taken together, it can generally be concluded that adult male Eriocheir sinensis of 150–200 g (Grade II–III) weight have the highest nutritional quality even though they are not the largest crabs.

    In pond cultures of juvenile Eriocheir sinensis, limb autotomy stress seriously affects and restricts the quality and economic benefits of aquaculture. This study was designed to evaluate the effects of dietary supplementation of L-tryptophan on E. sinensis under the cheliped autotomy stress. In the present study, 252 crabs were divided into four groups: dietary L-trp supplementation with 0.28, 0.40, 0.53, and 0.70%, and their hematological immunity, antioxidant capacity, anti-stress, and antibacterial ability were evaluated after 14 days of using biochemical analysis, flow cytometry, and molecular biology techniques. First, we counted the mortality after 14 days of feeding and found that compared with other treatments, dietary supplementation of 0.53 and 0.70% L-trp significantly lowered the mortality of E. sinensis. Moreover, the total hemocyte count (THC), hemocyanin, and glutathione (GSH) content, and glutathione peroxidase (GSH-Px) activity significantly increased at 7 and 14 d with dietary supplementation of 0.53 and 0.70% L-trp, in contrast with the significant decrease in malondialdehyde (MDA) content at 14 d in the same dietary groups (P
