The chloroplast genome sequence from Eugenia uniflora, a Myrtaceae from Neotropics

The complete chloroplast (cp) genome sequence of Eugenia uniflora , a member of the Neotropical Myrtaceae family, is reported here. The genome is 158,445 bp in length and exhibits a typical quadripartite structure of the large (LSC, 87,459 bp) and small (SSC, 18,318 bp) single-copy regions, separated by a pair of inverted repeats (IRs, 26,334 bp). It contains 111 unique genes, including 77 protein-coding genes, 30 tRNAs and 4 rRNAs. The genome structure, gene order, GC content and codon usage are similar to the typical angiosperm cp genomes. Comparison of the entire cp genomes of E. uniflora L. and three other Myrtaceae revealed an expansion of 43 bp in the intergenic spacer located between the IRA/large single-copy (LSC) border and the first gene of LSC region. Simple sequence repeat (SSR) analysis revealed that most SSRs are AT rich, which contribute to the overall AT richness of the cp genome. Additionally, fewer SSRs are distributed in the protein-coding sequences compared to the noncoding regions. Phylogenetic analysis among 58 species based on 57 cp genes demonstrated a closer relationship between E. uniflora L. and Syzygium cumini (L). Skeels compared to the Eucalyptus clade in the Myrtaceae family. The complete cp genome sequence of E. uniflora reported here has importance for population genetics, as well as phylogenetic and evolutionary studies in this species and other Myrtaceae species from Neotropical regions.

An analysis of phylogeographic population structure and demography was conducted on E. uniflora from 46 localities in natural environments across the distribution range of the species based on three plastid markers. ENM was also performed to predict suitable environments and areas of dramatic decrease in future suitability for the species under distinct representative concentration pathways (RCPs). Eugenia uniflora exhibited higher haplotype and nucleotide diversity in the southern part of its distribution than in the northern part. Two divergent lineages were revealed in the phylogenetic analysis of haplotypes, with an estimated divergence at c. 4.9 Mya. The populations in the northern and central regions of the range probably experienced population growth, whereas populations in the southern region are marked by historical demographic stability. ENM results indicate that the distribution of E. uniflora was fragmented in cool periods and was broader and more connected during warm periods during Pleistocene. The results suggest distinct evolutionary histories in southern to northern populations, indicating region-specific responses to changes.

Eugenia uniflora is a plant native to tropical America with pharmacological and ecological importance, and there have been no previous studies concerning its gene expression and regulation. To date, no miRNAs have been reported in Myrtaceae species. Results Small RNA and RNA-seq libraries were constructed to identify miRNAs and pre-miRNAs in Eugenia uniflora. Solexa technology was used to perform high throughput sequencing of the library, and the data obtained were analyzed using bioinformatics tools. From 14,489,131 small RNA clean reads, we obtained 1,852,722 mature miRNA sequences representing 45 conserved families that have been identified in other plant species. Further analysis using contigs assembled from RNA-seq allowed the prediction of secondary structures of 25 known and 17 novel pre-miRNAs. The expression of twenty-seven identified miRNAs was also validated using RT-PCR assays. Potential targets were predicted for the most abundant mature miRNAs in the identified pre-miRNAs based on sequence homology. Conclusions This study is the first large scale identification of miRNAs and their potential targets from a species of the Myrtaceae family without genomic sequence resources. Our study provides more information about the evolutionary conservation of the regulatory network of miRNAs in plants and highlights species-specific miRNAs.

Chloroplast DNA variation and phylogeography of Eugenia uniflora L. (Myrtaceae) in the Brazilian Atlantic forest

This biome covers an area of more than one million square kilometres along the Brazilian coast and extending to eastern Paraguay and northeastern Argentina [2]. AF is considered one of the most threatened ecosystems on Earth due to intense disturbance, having been reduced to only 7.5% of its original area [1]. Despite an increase in research efforts in the past few years, studies of AF species diversification and knowledge about its evolutionary history is still scarce. Vegetation changes in the Atlantic Forest related to climatic changes during the Pleistocene have been registered in paleopalinological studies, with the replacement of large areas of forests by subtropical grasslands and savannas during cooler and drier conditions [3,4]. Also, studies of paleoclimatic models, predicted the presence of historically stable areas (refugia) in the Atlantic Forest during the Late Quaternary [5]. Eugenia uniflora L. (Myrtaceae), a shrubby tree with edible cherry-like fruits which is locally known as pitanga or Brazilian cherry. This species is one of the key species in the Atlantic rain forest geomorphological domain, which includes the Atlantic forest and the adjacent Restinga ecosystem [6]. E. unifloraoccurs in areas of medium and large levels of rainfall and can also be found in different vegetation types and ecosystems. This species present economic and folk medicinal applications and is an important pioneer species in the Restinga ecosystem and has been used to recover and manage disturbed and fragmented areas. Our aim in this study was to investigate the phylogeography and genetic diversity of Eugenia uniflora to help elucidate the evolutionary history of this species as a model for gain insights into past vegetation patterns in the Brazilian Atlantic Forest.

(pitanga) is widely distributed in tropical areas. It is present in coastal vegetation from Ceara, in northeastern Brazil, to Rio Grande do Sul, at the southern tip of the country. Eugenia uniflora is of ecological importance, both as colonizing species on disturbed land and as food supplier for a wide variety of insects, birds and mammals. Pitanga plays a role in the maintenance of shrubby coastal ecosystems, especially at disturbed sites, and in 'restinga' ecosystems, at the interface between low forest and strand vegetation. To investigate the genetic diversity residing within the species, three neighboring populations at a distance of less than 24 km from each other, with varying degrees of human impact, were studied. The level of genetic diversity within and between populations was assessed with amplified fragment length polymorphism (AFLP) methodology. A total of 532 AFLP markers were analyzed in 66 individual trees. The polymorphism level varied from 61.2 to 96% depending on the primer combination used. Intra- and inter-population genetic diversity analysis showed that more than 88% of the variation resided within the populations, with a Gst of 0.123. Nevertheless, using neighbor joining (NJ) and principal component analysis (PCA), on the genetic distance (GD) data, permitted the three analyzed populations to be differentiated.

The role of extracts from Eugenia uniflora L. against metal stress in eukaryotic and prokaryotic models

Ethanolic extract, as well as tannic and flavonoid fractions, were prepared from samples obtained from the species which were analyzed by LC-MS. The antioxidant activity through the FRAP method showed that the tannic fraction had the lowest EC50. The suggested compounds identified in the extract and fractions were quercetin and the derivatives, myricetin, gallic acid, vanillic acid, quinic acid and catechin. Regarding the flavonoid fraction, a reduction in the fungicidal effect was observed. When the ethanolic extract was assayed in the presence of mercury chloride, all fractions showed a cytoprotection in relation to this metal. The sub-allelopathic concentration used was 64 μg/mL. The flavonoid fraction showed cytoprotective activity against aluminum for caulicles and radicles, whereas for the mercury against the radicles at all concentrations. Meanwhile the tannic fraction against aluminum showed a better development in the size of the radicles, the mercury there were differences in the sizes of the radicles and caulicles. The results demonstrated in this work indicate that natural products of Eugenia uniflora are promising sources of natural products with antioxidant and chelating activity.

Eugenia uniflora leaves essential oil induces toxicity in drosophila melanogaster involvement of oxidative stress mechanisms

(Myrtaceae family), also known as “pitanga”, is a tree species widely used in popular medicine. Despite the well documented beneficial effects of the extracts and essential oils from this plant, little is known about its toxicity. We performed a phytochemical fingerprinting and evaluated the toxicity induced by the Eugenia uniflora leaves essential oil in a Drosophila melanogaster model. In order to understand the biochemical mechanisms involved in E. uniflora essential oil toxicity, changes in the Nrf2 signaling as well as the hallmarks of oxidative stress were measured. The exposure of adult flies to the essential oil via a fumigant method resulted in increased mortality and locomotor deficits. In parallel, an oxidative stress response signaling, evidenced by changes in ROS production, lipid peroxidation, alterations in the activity of antioxidant enzymes and expression of Nrf2 protein targets occurred. In the light of our findings, attention is drawn to the indiscriminate use of this plant for medicinal purposes. In addition, a potential bio-insecticidal activity of Eugenia uniflora volatile compounds is suggested, a fact that needs to be further explored.

Materiales y metodos: En este contexto, se evaluaran in vitro, los potenciales antioxidantes, de los extractos de Eugenia uniflora y Psidium sobraleanum, ademas de cuantificar fenoles y flavonoides presentes en los extractos. Resultados: De acuerdo con los resultados obtenidos, se observo una mejor actividad antioxidante con el extracto de Eugenia uniflora, en el test de TBARS con fofolipidos de huevo, los extractos redujeron los niveles basales en el proceso de peroxidacion lipidica; y cuando fue inducida con Fe+2, el extracto de Psidium sobraleanum se mostro mas eficiente. Conclusiones: Por lo tanto, a traves de estos ensayos se pudo comprobar que los extractos de las hojas de las especies Eugenia uniflora y Psidium sobraleanum presentan actividad antioxidante, directamente relacionada con sustancias fenolicas producidas a partir de sus metabolitos secundarios.

Ethanol extract from Eugenia uniflora was assayed, for its antifungal activity, either alone or combined with four selected chemotherapeutic antimicrobial agents, including anphotericin B, mebendazole, nistatin, and metronidazole against these strains. The obtained results indicated that the association of the extract of E. uniflora to metronidazole showed a potential antifungal activity against C. tropicalis. However, no synergistic activity against the other strains was observed, as observed when the extract was associated with the other, not enhancing their antifungal activity.

Accordingly, secondary metabolites from plants with antioxidant activity represent an interesting alternative. The aim of this work was to evaluate the cytoprotective effect of the ethanol extract of Eugenia uniflora against mercury chloride. The main antioxidant phytochemicals were quantified by HPLC. The chelating effect was evaluated using the O-phenanthroline method. The cytoprotective effect was assayed using bacterial and plant models. The cytotoxic and the negative geotaxis effect was determined using Drosophila melanogaster. The ethanol extract of E. uniflora demonstrated chelating effect against iron, and these results can be related to the total phenols (1079 mg/g) and flavonoids (946.9 mg/g), detected by HPLC and quantified. The same extract demonstrated a cytoprotective effect in both models and a non-toxic effect against D. melanogaster , with low mortality and low inhibition of geotaxis, demonstrating that the extract can reduce the toxicity of this heavy metal against prokaryotic and Eukaryotic organisms. Due the results, we can conclude that the phytocompounds in the ethanol extract of E. uniflora , possibly phenols and flavonoids, could be interesting agents to protect different organisms against the heavy metal damages by a chelating or an antioxidative mechanism. Other studies are still necessary; however, these findings can open a new perspective to the recuperation of environments contaminated by heavy metals.

When the ethanolic extract was assayed in the presence of mercury chloride, all fractions showed a cytoprotection in relation to this metal. The sub-allelopathic concentration used was 64 μg/mL. The flavonoid fraction showed cytoprotective activity against aluminum for caulicles and radicles, whereas for the mercury against the radicles at all concentrations. Meanwhile the tannic fraction against aluminum showed a better development in the size of the radicles, the mercury there were differences in the sizes of the radicles and caulicles. The results demonstrated in this work indicate that natural products of Eugenia uniflora are promising sources of natural products with antioxidant and chelating activity.

Accordingly, secondary metabolites from plants with antioxidant activity represent an interesting alternative. The aim of this work was to evaluate the cytoprotective effect of the ethanol extract of Eugenia uniflora against mercury chloride. The main antioxidant phytochemicals were quantified by HPLC. The chelating effect was evaluated using the O-phenanthroline method. The cytoprotective effect was assayed using bacterial and plant models. The cytotoxic and the negative geotaxis effect was determined using Drosophila melanogaster. The ethanol extract of E. uniflora demonstrated chelating effect against iron, and these results can be related to the total phenols (1079 mg/g) and flavonoids (946.9 mg/g), detected by HPLC and quantified. The same extract demonstrated a cytoprotective effect in both models and a non-toxic effect against D. melanogaster , with low mortality and low inhibition of geotaxis, demonstrating that the extract can reduce the toxicity of this heavy metal against prokaryotic and Eukaryotic organisms. Due the results, we can conclude that the phytocompounds in the ethanol extract of E. uniflora , possibly phenols and flavonoids, could be interesting agents to protect different organisms against the heavy metal damages by a chelating or an antioxidative mechanism. Other studies are still necessary; however, these findings can open a new perspective to the recuperation of environments contaminated by heavy metals.

Eugenia uniflora leaves essential oil induces toxicity in drosophila melanogaster involvement of oxidative stress mechanisms

(Myrtaceae family), also known as “pitanga”, is a tree species widely used in popular medicine. Despite the well documented beneficial effects of the extracts and essential oils from this plant, little is known about its toxicity. We performed a phytochemical fingerprinting and evaluated the toxicity induced by the Eugenia uniflora leaves essential oil in a Drosophila melanogaster model. In order to understand the biochemical mechanisms involved in E. uniflora essential oil toxicity, changes in the Nrf2 signaling as well as the hallmarks of oxidative stress were measured. The exposure of adult flies to the essential oil via a fumigant method resulted in increased mortality and locomotor deficits. In parallel, an oxidative stress response signaling, evidenced by changes in ROS production, lipid peroxidation, alterations in the activity of antioxidant enzymes and expression of Nrf2 protein targets occurred. In the light of our findings, attention is drawn to the indiscriminate use of this plant for medicinal purposes. In addition, a potential bio-insecticidal activity of Eugenia uniflora volatile compounds is suggested, a fact that needs to be further explored.

Materiales y metodos: En este contexto, se evaluaran in vitro, los potenciales antioxidantes, de los extractos de Eugenia uniflora y Psidium sobraleanum, ademas de cuantificar fenoles y flavonoides presentes en los extractos. Resultados: De acuerdo con los resultados obtenidos, se observo una mejor actividad antioxidante con el extracto de Eugenia uniflora, en el test de TBARS con fofolipidos de huevo, los extractos redujeron los niveles basales en el proceso de peroxidacion lipidica; y cuando fue inducida con Fe+2, el extracto de Psidium sobraleanum se mostro mas eficiente. Conclusiones: Por lo tanto, a traves de estos ensayos se pudo comprobar que los extractos de las hojas de las especies Eugenia uniflora y Psidium sobraleanum presentan actividad antioxidante, directamente relacionada con sustancias fenolicas producidas a partir de sus metabolitos secundarios.
