Anticonvulsant properties of Euterpe Oleracea in mice.

Monteiro, Hervé RogezAbstract:Abstract Acai ( Euterpe Oleracea Mart.), a highly consumed fruit in Amazon, is from a common palm with remarkable antioxidant properties. Because oxidative stress and seizures are intimately linked, this study investigated the potential neuroprotective and anticonvulsant effects of commercial clarified acai juice (EO). EO did not alter spontaneous locomotor activity. Four doses of EO were sufficient to increase latencies to both first myoclonic jerk and first generalized tonic–clonic seizure and significantly decrease the total duration of tonic-clonic seizures caused by pentylenetetrazol administration. Also, electrocortical alterations provoked by pentylenetetrazol were prevented, significantly decreasing amplitude of discharges and frequencies above 50 Hz. EO was also able to completely prevent lipid peroxidation in the cerebral cortex, showing a potent direct scavenging property. These results demonstrate for the first time that E. Oleracea significantly protects against seizures and seizure-related oxidative stress, indicating an additional protection for humans who consume this fruit.

Spontaneous postharvest fermentation of açai (Euterpe Oleracea) fruit

These fruit contain a high bacterial load and are transported on boards stowed inside or outside the holds of small boats. In this context, postharvest parameters were assessed under conditions that simulated these two methods of EOF transport: stowage in closed polystyrene boxes, simulating the inside of cargo holds, i.e., transport in a closed system; and open baskets, simulating transport in an open environment, i.e., transport in the prow or bow of the boat. EOF suffered spontaneous fermentation of alcoholic, acetic, and lactic types in the closed system, which is the most common type of transportation of this fruit. In the closed system, there was a predominance of lactic acid bacteria over acetic acid bacteria, with 82% and 95% of the initial content of d -glucose and d -fructose being consumed, respectively, after 27 h of experiment. The weight loss reached 1.7% and there was a logarithmic decrease of the major phenolic compounds of the fruit in the closed system, with losses of 78% of cyanidin-3-rutinoside, 88% of cyanidin-3-glucoside, 78% of homorientin, and 72% of orientin after 27 h, which was higher than in the open system (58%, 66%, 73% and 62%, respectively). Trueness (<6.76% relative bias), repeatability (<4.6% RSD), intermediate precision (<5.3% RSD), selectivity, response function and linearity for major anthocyanins, cyanidin-3-glucoside and cyanidin-3-rutinoside, were evaluated. The concentration range validated was 1-48 μg/mL for both compounds. In addition two cyanidin-di-O-glycosides were detected for the fist time in this fruit. We also showed that a first extraction of the fruits with ethyl acetate removes the lipophilic compounds and allows an easier extraction by methanol and quantification of anthocyanins in this extract.

(acai) seed extract (ASE) prevents maternal cardiovascular changes and intrauterine growth restriction (IUGR) in experimental preeclampsia (PE).Methods: ASE administration (200 mg/kg/day) during mid to late pregnancy in a rat model of L-NAME-induced PE.Results: ASE impaired the maternal hypertension and microalbuminuria as well as the lower fetal and placental weight in experimental PE. ASE also prevented the maternal vascular dysfunction and lipoperoxidation in this model.Conclusion: ASE protected against maternal cardiovascular changes and IUGR in the L-NAME-induced PE. The protective effect of ASE may be partly explained by its antioxidant property.

We assessed the effects of Euterpe Oleracea Mart. (acai) seed extract (ASE) rich in proanthocyanidins and catechin on vascular dysfunction and oxidative stress associated with hypertension in spontaneously hypertensive rats (SHR). SHR and control rats were treated with ASE (200 mg/kg/day) or vehicle for 10 weeks. In the rat mesenteric arterial bed (MAB), acetylcholine response, endothelial nitric oxide synthase (eNOS) and superoxide dismutase 1 (SOD 1) expressions were studied. The antioxidant enzyme activity, oxidative damage and nitrite quantification were assessed in MAB and heart homogenates. eNOS immunohistochemistry and histological analysis was carried out on aortic sections. ASE was able to attenuate the hypertension and prevent the endothelial dysfunction in MAB of SHR. The increased levels of protein carbonylation and associated low levels of nitrite in MAB and heart of SHR were attenuated by ASE. The up-regulation of eNOS and SOD1 expression and the increased activity of SOD in MAB from SHR were normalized by ASE. In aorta from SHR, ASE prevented the increase in media thickness, media:lumen ratio and the decrease in the percentage of elastic fibers. Our results suggest that ASE produces antihypertensive effect and prevents the vascular dysfunction in SHR, through mechanisms involving antioxidant effects and NO production. Industrial relevance. Euterpe Oleracea Mart. (Acai) has been considered one of the most important medicinal plants of the Amazon by its beneficial effects in the treatment of fever, pain, inflammation and anemia. More recently, Euterpe Oleracea Mart. has been reported to reduce blood pressure with an important antioxidant property. The present research reports the protective effect of the extract of the seeds from Euterpe oleraca Mart. against high blood pressure and the associated vascular structural and functional changes. The test for antihypertensive and vascular anti-hypertrophic effect of the hydro-alcoholic extract of the seed of Euterpe Oleracea Mart. provides scientific data to confirm the potentials of Euterpe Oleracea Mart as an antihypertensive medicinal plant. Considering the large supply of Acai fruit in Brazil to obtain the extract, it provides a new therapeutic possibility of low cost for hypertensive patients. Keywords. Acai is an abundant and much consumed fruit by the Amazon local population, and studies have demonstrated that it is rich in phytochemicals with antioxidant, anti-inflammatory, and anticancer activities. Therefore, the aim of this study was to test this plant for anticancer activity in different human malignant cell lines. Cell lines derived from breast and colorectal adenocarcinomas were treated with 10, 20, and 40 μg/mL of bark, seed, and total acai fruit hydroalcoholic extracts for 24 and 48 h. After treatment, cell viability was measured using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays, and cell morphological features were observed by light and transmission electron microscopy. The type of cell death was also evaluated. The data were analyzed statistically by one-way analysis of variance (ANOVA), followed by Dunnett’s or Tukey’s post hoc tests, as appropriate.
