    Abstract:

    Many trematode parasites cause infection in humans and are thought to be a major public health problem. Their ecological diversity in different regions provides challenging questions on evolution of these organisms. In this report, we perform transcriptome analysis of the giant intestinal fluke, Fasciolopsis Buski, using next generation sequencing technology. Short read sequences derived from polyA containing RNA of this organism were assembled into 30677 unigenes that led to the annotation of 12380 genes. Annotation of the assembled transcripts enabled insight into processes and pathways in the intestinal fluke, such as RNAi pathway and energy metabolism. The expressed kinome of the organism was characterized by identifying all protein kinases. We have also carried out whole genome sequencing and used the sequences to confirm absence of some of the genes, not observed in transcriptome data, such as genes involved in fatty acid biosynthetic pathway. Transcriptome data also helped us to identify some of the expressed transposable elements. Though many Long Interspersed elements (LINEs) were identified, only two Short Interspersed Elements (SINEs) were visible. Overall transcriptome and draft genome analysis of F. Buski helped us to characterize some its important biological characteristics and provided enormous resources for development of a suitable diagnostic system and anti-parasitic therapeutic molecules.

    Helminths include both parasitic nematodes (roundworms) and platyhelminths (trematode and cestode flatworms) that are abundant, and are of clinical importance. The genetic characterization of parasitic flatworms using advanced molecular tools is central to the diagnosis and control of infections. Although the nuclear genome houses suitable genetic markers (e.g., in ribosomal (r) DNA) for species identification and molecular characterization, the mitochondrial (mt) genome consistently provides a rich source of novel markers for informative systematics and epidemiological studies. In the last decade, there have been some important advances in mtDNA genomics of helminths, especially lung flukes, liver flukes and intestinal flukes. Fasciolopsis Buski, often called the giant intestinal fluke, is one of the largest digenean trematodes infecting humans and found primarily in Asia, in particular the Indian subcontinent. Next-generation sequencing (NGS) technologies now provide opportunities for high throughput sequencing, assembly and annotation within a short span of time. Herein, we describe a high-throughput sequencing and bioinformatics pipeline for mt genomics for F. Buski that emphasizes the utility of short read NGS platforms such as Ion Torrent and Illumina in successfully sequencing and assembling the mt genome using innovative approaches for PCR primer design as well as assembly. We took advantage of our NGS whole genome sequence data (unpublished so far) for F. Buski and its comparison with available data for the Fasciola hepatica mtDNA as the reference genome for design of precise and specific primers for amplification of mt genome sequences from F. Buski. A long-range PCR was carried out to create an NGS library enriched in mt DNA sequences. Two different NGS platforms were employed for complete sequencing, assembly and annotation of the F. Buski mt genome. The complete mt genome sequences of the intestinal fluke comprise 14,118 bp and is thus the shortest trematode mitochondrial genome sequenced to date. The noncoding control regions are separated into two parts by the tRNA-Gly gene and don’t contain either tandem repeats or secondary structures, which are typical for trematode control regions. The gene content and arrangement are identical to that of F. hepatica. The F. Buski mtDNA genome has a close resemblance with F. hepatica and has a similar gene order tallying with that of other trematodes. The mtDNA for the intestinal fluke is reported herein for the first time by our group that would help investigate Fasciolidae taxonomy and systematics with the aid of mtDNA NGS data. More so, it would serve as a resource for comparative mitochondrial genomics and systematic studies of trematode parasites

    Food-borne fluke infections/trematodiases are emerging as a major public health problem worldwide with over 40 million people affected and over 10% of world population at risk of infection. The major concentration of these infections is in Southeast Asian and Western Pacific Regions, where the epidemiological factors (including the prevalent socio-cultural food habits) are conducive for transmission of these infections. The preponderance of these infections is usually in food deficit poor communities that lack access to proper sanitary infrastructure. While targeting health for all, especially the poor rural tribal communities, it is imperative to take these infections into account. Bayesian analysis phylogeny of food-borne trematode parasites under study showed that they are closely related phylogenetic groups. To focus the control strategies at the target populations, the aim of the present study was to establish molecular methods for accurate discrimination between common food-borne trematodes parasites Paragonimus (lung fluke), Fasciolopsis (giant intestinal fluke) and Fasciola (liver fluke), the infections of which commonly prevail in NE India. In the first step, we amplified and sequenced the second internal transcribed spacer (ITS2) region of ribosomal DNA, utilizing nucleotide differences between the multiple sequence alignments of the parasites under study. Based upon the differences in nucleotide sequences of conserved regions, we designed species-specific primers that can unequivocally discriminate one species from another. ITS2 sequence motifs allowed an accurate in-silico distinction of the trematodes. The data indicate that ITS2 motifs (≤ 50 bp in size) can be considered promising tool for trematode species identification. Using molecular morphometrics that is based on ITS2 secondary structure homologies, phylogenetic relationships with various isolates of several trematode species have been discussed. The present results suggest that the ITS2 specific primers can be used for epidemiological investigations of the prevalence of trematodiasis.

    Objective: Alpinia nigra (Zingiberaceae) is a traditionally used medicinal plant of North-east India against helminthiases. In view of its medicinal property the present study was designed to see the anthelmintic property of its major bioactive compound astragalin, compared to crude extract and praziquantel against Fasciolopsis Buski . Methods: Parasites were exposed to 2, 5, 10 and 20 mg/ml concentrations of plant extract and 0.025, 0.05 and 0.10 mg/ml concentration of astragalin in phosphate buffer saline. Paralyzed parasites were processed for histochemical, biochemical and morphological studies. Two important tegumental enzymes acid-and alkaline phosphatase were studies for its alterations in kinetic parameters on exposure to extracts and astragalin. Results: Treated parasites revealed complete paralysis followed by death in a dose-dependent manner. Histochemical and biochemical studies on enzymes showed alterations in the activities. Morphologically, destruction and deformation were seen in the surface architecture of F. Buski on exposure to different treatment. Acid-and alkaline phosphatase enzyme showed optimum activity at pH 6 and 9.5 respectively. The Vmax changed from 11.02 and 10.24 µM/min/mg protein in control to as low as 6.98 and 6.54 µM/min/mg tissue proteins in treated parasite for acid and alkaline phosphatase, respectively. Alterations in the Km values have also been noticed between the control and treated parasites. Conclusion: The altered enzyme activities, tegumental architecture and kinetic parameters as observed under the influence of the plant extract and astragalin clearly indicate that the botanicals of the tested plant and its bioactive glycoside, astragalin acts as an anthelmintic agent against the tested fluke F. Buski .

    Abstract:

    Alpinia nigra belonging to the family Zingiberaceae is an ethnomedicine with established anthelmintic property. In view of its medicinal property, the present study was designed to investigate the anthelmintic property of astragalin, a bioactive compound of A. nigra against fluke parasite, Fasciolopsis Buski. In vitro exposure of flukes at varying concentrations 0.025 – 0.1 mg/ml of PBS a dose-dependent effect on motility and mortality was observed. Stereoscan observations of the astragalin exposed fluke revealed deformity of the surface architecture including shrinkage and eroded scales leading to scar formation as well as loss of the scales. Ultrastructural observations revealed distortion of glycocalyx, musculature region, basal layer and disintegration of the nucleus, nucleolus, nuclear membrane, Golgi complexes and mitochondria. A large number of vacuole formations could also be seen in flukes exposed to astragalin compared to control. The result therefore indicates the anthelmintic property of astragalin, the bioactive compound of A. nigra against the fluke parasite F. Buski.

    Alpinia nigra (Geartn) Burtt. is a traditionally used medicinal plant, aqueous extract of which is consumed by the tribes of Tripura, India, against helminthiasis. In order to explore the effects of A. nigra plant extract on the neuromuscular activity of parasites, inhibition of acetylcholinesterase (AchE) in Raillietina echinobothrida and Fasciolopsis Buski was investigated. The interaction of the enzyme with the substrate and the plant extract was characterized by the kinetic parameters Km and Vmax which were determined from Lineweaver-Burk plots. The ethanolic shoot-extract of A. nigra inhibited the AchE activity in dose-dependent manner with IC50 values of 1.4 mg/ml and 0.91 mg/ml in R. echinobothrida and F. Buski, respectively. Km and Vmax of R. echinobothrida and F. Buski were found to be 1.23 mM, 1.45 µmole/h/mg proteins and 1.09 mM, 2.27 µmole/h/mg proteins, respectively. The interaction of plant extract reduced the Vmax values by 33.10% and 31.28% while the Km values were increased by 81.30% and 79.82% in R. echinobothrida and F. Buski, respectively. Determination of the inhibitory parameters of A. nigra on AchE activity in R. echinobothrida and F. Buski revealed that the plant extract acted as a mix and non-competitive type of enzyme inhibitor.

    Fasciolopsis Buski is one of the biggest trematode parasite which can be infected the human being because the worm live in lumen intestine. Human can be infected because eat raw or unwell cooked water plants. In Indonesia, this worm diseases only found in some village in Hulu Sungai Utara district, with prevalence between 1,2-7,8%. The research design is a cross sectional with collecting epidemiology data, such as parasite examination on the reservoir (it suppose to buffalo swamp), the intermediate host (some species snails), water plants of parasite encystation (water lily, water chesnut, water cress). Result of the examination of stool, 7,8% (121/1.555 person) have positive egg of F. Buski. The collecting result of epidemiology data for looking the water plants of metacercariae eocystations, the intermediate host of cercariae growth and host reservoir has been collected 206 specimens but can not find the water plants of encystations or the intermediate host. Keywords: Fasciolopsis Buski, parasite, cercariae

    Abstract:

    Fasciolopsiosis is a worm disease which is caused by Fasciolopsis Buski. The disease is endemic at villages in Hulu Sungai Utara Regency with prevalence between 1.2-7.8%. The sufferer are children whose age around 1-10 years old. In the year 2002, the data base on epidemiological and socio-cultural aspect, were collected by employing cross sectional research design. In 2003 intervention was done to the community in 3 villages (Padang Bangkal, Kalumpang Dalam and Talaga Mas) by doing the Islamic religion approach campaign in the mosque or public places using poster. To find out the result of intervention, the researcher evaluated parasitological and socio cultural especially knowledge aspects. Total respondents were 335 family chief in 6 villages (3 intervened villages and 3 non­ intervened villages). The result of parasitological data collecting in 2003 showed a prevalence rate of 4.6%. This prevalence rate indicated the decrease of 3.2% compared to those of 2002 prevalence rate which was 7.8%. Community knowledge about Buski disease was increased at intervened villages on average 40.0% from respondents after intervention, while at the non-intervention villages the increase was only 7.5%. There was knowledge improvement about the Buski disease at intervened villages on average 46.1% from respondents after intervention, the non-intervened villages the rise was only 0.6%. Knowledge improvement about transmission of Buski disease at intervened villages on average was 49.9% and while the rise of non-intervened villages only 1.9% (p<0,05). The researcher suggested that improving community knowledge can be done under religious approach especially Islam by referring to Al Qur'an and Hadist.     Fasciolopsiosis is worm disease which is caused Fasciolopsis Buski. The disease is an endemic at some villages in Hulu Sungai Utara Regency with prevalence 1.2-7.8%.The patients are children with age around 1-15 years old. In the year 2002-2003 have been carried out a research "Model of Control Fasciolopsis Buski at South Kalimantan with Socio-Cultural Approach". In the first year (2002) of research, the data base of epidemiological and socio-cultural aspect, with cross sectional research design, have been collected. And in the second year (2003) has been done the intervention to the basic school students in three villages (Kalumpang Dalam, Putat Atas, and Talaga Mas), by doing a health campaign using comic books with Banjar language (local language). To find out the intervention has success, the researcher evaluated parasitological and socio cultural data collecting, especially knowledge, attitude and practice (KAP) aspect. Data collecting about KAP aspect was executed to the basic school students (grade 3-5). Total samples are 160 basic school students in those three villages. The result of parasitological data collecting in 3 villages after inten>ention is found a prevalence rate 5.2% positive fasciolopsiosis. There is decreased about 6.5% from 11.7% on the first year (2002) into 5.2% at second year (2003). The result of socio-cultural data collecting for KAP aspect as quantitative is found increase. There is knowledge increasing about the worm disease, where before intervention only 86.8% students who know the Buski disease, and after intervention become 96.8%. While on practice aspect also increase, before intervention 73.1% basic school students use drugs worm (albendazol) and 84.3% after intervention, before intervention 48.7% students never eat raw water plants, after intervention become 19.9%, before intervention 57.5% students wash their hands with soap but after intervention raised up become 85.0% (p<0,05). The Conclusion is there is correlation between the intervention by doing health campaign using comic "Buski" book (by local "Banjar" language) to basic school students and KAP aspect increasing. The suggestion of the research is there is a necessary needs to socialize aspects of good health living and "Buski" disease (fasciolopsiosis) at basic school students through information which is appropriate with target and condition of the local area.

    Fasciolopsiasis adalah penyakit kecacingan yang disebabkan oleh cacing Fasciolopsis Buski. Cacing ini merupakan salah satu parasit trematoda terbesar yang dapat menginfeksi manusia dan bermanifestasi di dalam lumen usus.  Manusia terinfeksi cacing ini dikarenakan memakan tumbuhan air yang mentah atau yang tidak dimasak dengan baik. Secara endemik, penyakit kecacingan ini di Indonesia hanya ditemukan  di beberapa desa di kabupaten Hulu Sungai Utara dengan prevalens antara 1,2-7,8%. Sampai saat ini angka prevalens penyakit tidak menunjukkan kecenderungan turun, akan tetapi menunjukkan adanya penyebaran penyakit ke wilayah lainnya. Rancang penelitian adalah cross sectional dengan mengumpulkan data parasitologis untuk mengetahui besarnya prevalens  fasciolopsiasis yang dilakukan dengan cara pemeriksaan tinja penduduk pada 7 desa di 3 kecamatan pada kabupaten Hulu Sungai Utara. Hasil survei parasitologis yang dilaksanakan pada 7 desa penelitian (Sei Papuyu, Kalumpang Dalam, Sarang Burung, Talaga Mas, Putat Atas, Padang Bangkal dan Sapala-Bararawa) yang mencakup 1.555 penduduk diperoleh angka prevalens sebanyak 7,8% positif fasciolopsiasis. Anak-anak usia di bawah 10 tahun merupakan penderita fasciolopsiasis yang paling banyak yaitu 67,8% atau 5,2% dari total penduduk yang diperiksa. Oleh karena itu perlu dikembangkan suatu upaya surveilans untuk memantau prevalens fasciolopsiasis dengan pemeriksaan tinja dan pengobatan penderita yang difokuskan kepada anak­ anak sekolah dasar. Pelaksanaannya 2 kali dalam setahun yaitu pada pertengahan musim hujan (Februari-Maret) dan pertengahan musim kemarau (Agustus-September).

    Fasciolopsis Buski adalah salah satu parasit trematoda yang dapat menginfeksi manusia di karenakan memakan tumbuhan air yang mentah atau tidak di masak dengan baik. Penyakit kecacingan ini ditemukan di beberapa desa di kabupaten Hulu Sungai Utara dengan prevalensi antara 1,2 - 7,8 %. sampai saat ini angka prevalens penyakit tidak menunjukkan kecenderungan turun, bahkan terindikasi dengan semakin meluasnya penyebaran penyakit ke wilayah lainnya. Bagi para pelaksana program kebutuhan data penanggulangan dan pemberantasan fasciolopsiosis (penyakit kecacingan Buski). Pada tahun kedua pelaksanaan penelitian dikembangkan model intervensi penyuluhan yang berbasiskan aspek religius masyarakat setempat. Pelaksanaan penelitian "Model penangulangan Fasciolopsis Buski di Kalimantan Selatan dengan Pendekatan Sosial Budaya" pada tahun kedua dengan menggunakan model rancangan ulang non random (non-randomized pretest posttest control group design). Intervensi dilakukan pada 3 desa (Kalumpang dalam, Pusat Atas dan Talaga Mas) melalui upaya penyuluhan dengan menggunakan media komik bergambar berbahasa daerah (Banjar) untuk anak SD, poster dalam bahasa daerah dan ceramah dengan materi yang meliputi upaya pencegahan dan pemberantasan fasciolopsiasis yang dikaitkan dengan Al Qur'an dan hadist. Sebagai desa pembanding (Sei Papuyu, Padang, Bangkal dan sarang Burung) pelaksanaan penyuluhan dilakukan dalam kegiatan rutin Puskesmas. Untuk mengetahui keberhasilan pelaksanaan intervensi dilakukan pengumpulan data parasitologi dan sosial budaya khususnya aspek pengetahuan dan perilaku. Pengumpulan data tentang aspek pengetahuan dan perilaku positif dilaksanakan pada anak SD (kelas 3-6) dan kepala keluarga. Jumlah sampel untuk anak SD sebanyak 160 anak dan untuk kepala keluarga banyak 350 KK dari 6 desa (3 desa intervensi dan 3 desa pembanding). Dari 350 KK yang direncanakan, sampel yang memenuhi syarat dan dapat dianalisis sebesar 335 KK. Hasil pengumpulan data parasitologis yang dilaksanakan pada 6 desa penelitian (Sei Papuyu, Kalumpang Dalam, Sarang Burung, Talaga MAs, Putat Atas dan Padang Bangkal) setelah pelaksanaan intervensi diperoleh angka sebanyak 4,6 % positif fasciolopsiasis. Terdapat penurunan sebesar 3,2 % dari semula pada tahun pertama 7,8% menjadi 4,6% pada tahun kedua. Hasil pengumpulan data sosial budaya untuk mengetahui aspek pengetahuan dan perilaku positif pada anak SD dan aspek pengetahuan pada kepala keluarga (KK) secara kuantitatif diperoleh peningkatan yang bermakna. Aspek pengetahuan dan perilaku positif pada anak Sd di desa intervensi meningkat yaitu 5,8% untuk aspek pengetahuan dan 27,0% untuk aspek perilaku setelah pelaksanaan intevensi (P<0.005). Sedangkan untuk para KK yang berada di desa intervensi terdapat peningkatan aspek pengetahuan sebesar 41,9% dibanding dengan para KK di desa pembanding yaitu 3,4 % (p<0,005). Kesimpulan yang diperoleh dari penelitian ini adalah adanya intervensi yang berupa penyuluhan dengan menggunakan bahasa daerah terdapat anak SD dan penyuluhan kepada kepala keluarga (KK) yang dikaitkan dengan aspek keimanan dalam lingkup keislaman dapat meningkatkan pengetahuan dan perilaku positif anak SD dan meningkatkan pengetahuan kepala keluarga (KK) sehingga prevalens fasciolopsiasis secara bermakna juga dapat diturunkan. Saran yang dapat diajukan adalah perlunya dilakukan sosialisasi tentang aspek-aspek yang berhubungan dengan hidup sehat dan penyakit kecacingan Buski (fasciolopsiasis) kepada seluruh tingkatan usia dalam koridor religius keislaman.

    Alpinia nigra (Geartn) Burtt. is a traditionally used medicinal plant, aqueous extract of which is consumed by the tribes of Tripura, India, against helminthiasis. In order to explore the effects of A. nigra plant extract on the neuromuscular activity of parasites, inhibition of acetylcholinesterase (AchE) in Raillietina echinobothrida and Fasciolopsis Buski was investigated. The interaction of the enzyme with the substrate and the plant extract was characterized by the kinetic parameters Km and Vmax which were determined from Lineweaver-Burk plots. The ethanolic shoot-extract of A. nigra inhibited the AchE activity in dose-dependent manner with IC50 values of 1.4 mg/ml and 0.91 mg/ml in R. echinobothrida and F. Buski, respectively. Km and Vmax of R. echinobothrida and F. Buski were found to be 1.23 mM, 1.45 µmole/h/mg proteins and 1.09 mM, 2.27 µmole/h/mg proteins, respectively. The interaction of plant extract reduced the Vmax values by 33.10% and 31.28% while the Km values were increased by 81.30% and 79.82% in R. echinobothrida and F. Buski, respectively. Determination of the inhibitory parameters of A. nigra on AchE activity in R. echinobothrida and F. Buski revealed that the plant extract acted as a mix and non-competitive type of enzyme inhibitor.

    Abstract The alcoholic extract of Lysimachia ramosa Wall (Primulaceae) was tested in vitro against helminth parasites, Fasciolopsis Buski and Ascaris suum, from porcine hosts and Raillietina echinobothrida from domestic fowl. The live adult parasites, collected from a freshly autopsied host, were exposed to different concentrations (5–50 mg) of the test plant extract in physiological phosphate-buffered saline (PBS) having 0.1% dimethyl sulphoxide (DMSO) at 37 ± 1 °C. The treated parasites revealed complete inactivation and flaccid paralysis that was followed by death at varying periods of time. A dose-dependent loss of motility and mortality was observed in all the treated parasites. Scanning electron microscopic observations revealed conspicuous deformity of the surface architecture in all the parasites exposed to the test plant extract. The general tegument in F. Buski showed shrinkage and loss of scale-like spines; proglottides all along the strobilar length in R. echinobothrida appeared shrunken and deformed and the cuticular surface of A. suum appeared disorganised, having lost transverse striations. The botanicals of the test plant seem to be effective against all the three types of helminth parasites.

    Abstract:

    Ethanolic shoot extract of Alpinia nigra , a traditionally used anthelmintic medicinal plant among the Tripuri tribes of north-east India, was tested in vitro to determine its anthelmintic efficacy in gastrointestinal trematode Fasciolopsis Buski , using alterations in the activity of vital tegumental enzymes viz. acid phosphatase (AcPase), alkaline phosphatase (AlkPase) and adenosine triphosphatase (ATPase). Live adult F. Buski treated in vitro with different concentrations of the plant extract showed significant decline in the visible stain histochemically and enzyme activities. Quantitatively, the total enzyme activity of AcPase, AlkPase and ATPase was found to be reduced by 45, 41 and 43%, respectively compared to the control. The reference drug, praziquantel also showed more or less similar effect like that of the plant extract. The results suggest that the tegumental enzymes of the parasite may be an important target of action for active component(s) of A. nigra , which appears to act transtegumentally.

    The ultrastructural alterations in the tegument of Fasciolopsis Buski in response to incubation in the alcoholic extract of Alpinia nigra were evaluated using transmission electron microscopy. The body tegument of the trematode is composed of an external syncytial layer, musculature, and an inner layer containing tegumental cells. The syncytium comprises various organelles like mitochondria, lysosomes, and tegumentary bodies of the type 2 kind with rare sighting of the type 1. Severe distortion and disorganization of the tegument was revealed in the parasite exposed to the A. nigra extract in the current study. The extent of vacuolization was such that vacuoles proceeded down to the basal lamina causing the syncytium to separate from the tegument at different places. There was depletion of parenchyma material and loss of connecting tubules running down from the syncytium to the tegumental cells causing the cells to be deprived of any proper boundaries. Microsc.     Abstract. Fasciolopsiasis is a trematode worm disease of Fasciolopsis Buski’s species, which is only endemic in some areas in Hulu Sungai Utara, South Kalimantan. Buski life cycle requires an intermediate host such as snails and plants that live in the aquatic environment. Fasciolopsiasis endemic areas in HSU is a marshy area, however not all regions that have the same characteristics or directly adjacent to an endemic area fasciolopsiasis. This research is a descriptive, aims to describe the aquatic environment through the parameters of temperature, acidity (pH), salinity, the value of Dissolved Oxygen (DO), Biological Oxygen Demand (BOD), and Chemical Oxygen Demand (COD), which supports Buski’s habitation in endemic areas. Cross-sectional study design, and implemented in the village of Kalumpang Dalam, Sungai Papuyu, and Telaga Mas, in the month of June to September 2012. The results showed swamp water environment in the research site with a surface water temperature of 25oC, pH 6, salinity 0‰, DO value between 3,49-88,7 mg O2/L, BOD value between 0,05-3,64 mg O2/L, and COD values between 3,016- 39,51 mg O2/L, which indicate the presence of marshy water pollution and still found life freshwater snails as an intermediate host of F.Buski : Lymnaea and Indoplanorbis Keywords : Fasciolopsis Buski, fasciolopsiasis, swamp water, intermediate host, HSU Abstrak.     Abstract. Fasciolopsiasis is intestinal disease caused by consuming contaminated water plant by metacercaria of Fasciolopsis Buski. Many fasciolopsiasis patients are children because their knowledge and their awareness of health remains low. This study aims to know characteristics and behaviour in family sufferer's family. This study was conducted from March to December 2012 with cross sectional study design. Examination of stool samples to determine infection F. Buski using centrifugation method (Formalin Ether Technique). The results showed fasciolopsiasis patients mostly found in Sungai Papuyu Village as many as 9 people, sex most patients are women and the most age range between 6-12 year old. Based on the results of interview known family knowledge is still low, defecate habit directly into the swamp and have not been taking anthelmintic. Kebanyakan penderita fasciolopsiasis adalah anak-anak karena pengetahuan dan kesadaran mereka akan kesehatan masih rendah. Studi ini bertujuan untuk melihat karakteristik dan perilaku keluarga penderita fasciolopsiasis. Penelitian ini dilaksanakan pada bulan Maret hingga Desember 2012 dengan rancangan penelitian cross sectional. Pemeriksaan sampel tinja untuk mengetahui infeksi cacing F. Buski dengan menggunakan centrifugation method (Formalin Ether Technique). Hasil penelitian ini menunjukkan penderita fasciolopsiasis terbanyak ditemukan di Desa Sungai Papuyu yaitu sebanyak 9 orang, perempuan dan dengan kisaran umur antara usia 6-12 tahun. Berdasarkan hasil wawancara diketahui pengetahuan keluarga masih rendah, kebiasaan BAB langsung ke air rawa dan belum pernah mengkonsumsi obat cacing.     Fasciolopsiasis di Indonesia endemik terjadi di Kabupaten Hulu Sungai Utara, Provinsi Kalimantan Selatan. Dasar utama pengendalian fasciolopsiasis adalah memutuskan siklus hidup cacing, untuk itu semua bentuk stadium cacing harus dikenali serta faktor penting yang mempengaruhi penularannya yaitu jenis keong yang berperan sebagai hospes perantara pertama. Untuk menentukan jenis keong hospes perantara F. Buski perlu dikonfirmasi serkaria yang ditemukan pada keong tersebut merupakan serkaria F. Buski. Identifikasi serkaria secara mikroskopis belum bisa memastikan serkaria pada keong di daerah endemis merupakan serkaria F. Buski sehingga perlu digunakan metode yang lebih akurat yaitu Polymerase Chain Reaction (PCR). Penelitian ini ditujukan untuk mengidentifikasi serkaria F. Buski serta mengkonfirmasi jenis keong hospes perantara pertama dari F. Buski. Penelitian berupa studi observasional di lapangan dan laboratorium, dilaksanakan pada Maret-Desember 2014. Pengambilan sampel dilakukan di Desa Sungai Papuyu dan Desa Kalumpang Dalam, uji PCR sampel serkaria dilakukan di Laboratorium Biologi Molekuler FMIPA Universitas Lambung Mangkurat Banjarbaru. Pengambilan sampel mendapatkan 6 genus keong yaitu Pomacea, Bellamya, Indoplanorbis, Lymnaea, Gyraulus dan Melanoides, didapatkan 3 jenis serkaria yaitu : Echinostome cercariae pada keong Lymnaea dan Indoplanorbis, Brevifurcate-pharyngeate cercariae pada keong Lymnaea dan Sulcatomicrocercous cercariae pada keong Bellamya. Hasil PCR menujukkan sampel positif F. Buski merupakan Echinostome cercariae yang terdapat pada keong Lymnaea dan Indoplanorbis sehingga dapat dikonfirmasi bahwa keong tersebut merupakan hospes perantara pertama dari F. Buski.
