    Abstract:

    The present study aimed to describe the structural and ultrastructural morphological characteristics of the lingual epithelium and the connective tissue cores (CTCs) of wild boar (Sus scrofa). The tongues were processed for light microscopy, scanning electron microscopy, and transmission electron microscopy. In this study, we revealed the filiform, fungiform, Foliate, and vallate Papillae. The filiform Papilla is elongated with a conical shape and its CTC has a conical shape; the fungiform Papilla is rounded with a dome-shape and its CTC is flower bud; the Foliate Papilla is formed by four pairs of epithelial folds and irregular grooves, and its CTC is thin with adjacent conjunctive projections, and taste buds and serous glands in the epithelial layer have been evidenced; and the vallate Papilla is oval surrounded by a groove with increases of epithelium surface, and the CTC is formed by numerous connective projections lined. Also noted were serous gland and taste buds on the medial wall of the vallate Papilla. The epithelium has the keratinized, granular, spinous, basal, and lamina propria layers. In conclusion, we found new descriptions and shapes of the CTCs of the lingual Papillae. In addition, we demonstrated the epithelium structural characteristics, the nuclear distribution between the epithelial layers, and the ultrastructural aspects of the dorsal epithelium of the tongue.

    The tongue is an important organ in species due to its feeding functions, and its structure is influenced by the habitat and diet. The Collared Peccary (Pecari tajacu, Linnaeus, 1758) is a terrestrial mammal that is distributed on the American continents and has an omnivorous diet. This study aimed to describe the morphological characteristics of the tongue, lingual Papillae and the connective tissue cores (CTCs) of the Collared Peccary. Eight tongues were collected from the Wild Animals Multiplication Center. The samples (n = 6) were processed for three-dimensional analysis of their dorsal epithelium, and their CTCs by scanning electron microscopy and the other samples (n = 2) were used to observe ultrastructural characteristics by transmission electron microscopy. Filiform Papillae were observed in the lingual apex and body with their conical CTC demonstrating ripples in their extent. Two types of fungiform Papillae were observed, the first in the apex and body with a dome-shaped CTC and the second forming a dorsolateral line with a radial pattern CTC. The vallate Papillae were bilateral in the caudal region with the CTCs, characterised by numerous projections, and Foliate Papilla were not observed. We found a new type of Papilla anteriorly to the vallate Papilla with an irregular groove and a CTC formed by triangular-shaped projections with a duct opening at the top. We concluded that the lingual Papillae of the Collared Peccary resemble those Papillae of other mammals, however, were revealed another form of their CTCs and a new lingual Papillae morphological description.

    The structural elements, tridimensional aspects of the lingual Papillae, yours connective tissue cores (CTCs) and the ultrastructure of the dorsal epithelium surface of the tongue of guinea pig are examined. Filiform Papillae and their CTCs lie throughout the tongue surface, featuring subtypes and different shapes. Fungiform Papillae are dome-shaped, and their CTCs are few, with heterogenic disposition among the filiform Papillae in the lingual apex and body and in grooves stretching to the side margin among the Foliate Papillae towards the lingual apex. Foliate Papillae are well developed, and their CTCs are composed of five pairs of parallel seams limited by deep grooves placed bilaterally in the caudal region. Two vallate Papillae are observed in the caudal region and reveal specialized structures which are developed and differentiated when they present compound elements by a double epithelial projection, in a column, similar to the shape of the Foliate Papilla. Further, the epithelium is organized by layers with ultrastructural components. Therefore, we conclude that the structural descriptions, dimensional aspects of tongue Papillae, their CTCs and the ultrastructure of the tongue epithelium of guinea pigs are similar to other rodent mammals, however, with new descriptions as double epithelial projection constituting each vallate Papilla.

    The characteristics of the interface epithelium-connective tissue of the lingual mucosa of Calomys callosus was studied by employing scanning electron microscopy after removal of the epithelial cells. In the anterior part, the connective tissue Papillae of filiform Papillae are distributed at random in their original configuration, and the round-shaped fungiform Papillae are scattered among the filiform Papillae. In the middle part, the filiform Papillae present several rod-shaped projections. In the posterior part, the vallate Papilla are oval in shape, and the connective tissue reveals a large ridge separated by deep lateral grooves. The posterior adjacent area is a flat surface and shows several openings of salivary gland ducts. The Foliate Papillae are formed by several small ridges separated by grooves. In the areas adjacent to the Foliate Papilla, small pointed and laminar Papillae were observed.

  • Glutamate-induced Cobalt Uptake Elicited by Kainate Receptors in Rat Taste

    Glutamate-induced cobalt uptake reveals non-N-methyl-D-aspartate (non-NMDA) glutamate receptors (GluRs) in rat taste bud cells. However, it is not known which type of non-NMDA glutamate receptors is involved. We used a cobalt staining technique combined with pharmacological tests for kainate or a-amino-3-hydroxy-5-methyl-isoxazole-propionic acid (AMPA) receptors and/or immunohistochemistry against subunits of GluRs to examine the presence of non-NMDA receptors in rat Foliate tastebud cells. Cobalt uptake into taste cells was elicited by treating taste buds with glutamate, kainate or SYM 2081, a kainate receptor agonist. Treating taste buds with AMPA or fluorowillardiine did not stimulate significant cobalt uptake. Moreover, 6-cyano-7-nitro-quinoxaline-2, 3-dione significantly reduced cobalt staining elicited by glutamate or kainate receptor agonists, but SYM 2206, an AMPA receptor antagonist, did not. Immunohistochemistry against subunits of GluRs reveals GluR6 and KA1-like immunoreactivity. Moreover, most glutamate-induced cobalt-stained cells showed GluR6 and KA1-like immunoreactivity. These results suggest that glutamate-induced cobalt uptake in taste cells occurs mainly via kainate type GluRs.     Glutamate-induced cobalt uptake reveals that non-NMDA glutamate receptors (GluRs) are present in rat taste bud cells. Previous studies involving glutamate induced cobalt staining suggest this uptake mainly occurs via kainate type GluRs. It is not known which of the 4 types of taste bud cells express subunits of kainate GluR. Circumvallate and Foliate Papillae of Sprague-Dawley rats (45~60 days old) were used to search for the mRNAs of subunits of non-NMDA GluRs using RT-PCR with specific primers for GluR1-7, KA1 and KA2. We also performed RT-PCR for GluR5, KA1, PLCβ2, and NCAM/SNAP 25 in isolated single cells from taste buds. Taste epithelium, including circumvallate or Foliate Papilla, express mRNAs of GluR5 and KA1. However, non-taste tongue epithelium expresses no subunits of non-NMDA GluRs. Isolated single cell RT-PCR reveals that the mRNAs of GluR5 and KA1 are preferentially expressed in Type II and Type III cells over Type I cells.
