    Abstract:

    Intergeneric somatic hybrids between embryogenic callus-derived protoplasts of round kumquat ( Fortunella japonica Swingle) and ‘Morita navel’ orange ( Citrus sinensis Osbeck) were produced by electrofusion. Among the eight different fusion strains obtained, six showed normal morphology, whereas the remaining two showed malformation. All the regenerated plants were intermediate in leaf morphology and had thick and round leaves, which are typical characteristics of polyploids. Ploidy analyses by flow cytometry and chromosome counting in root-tip cells revealed that these plants are amphidiploid (2 n =4×=36). Hybridity of the fusion products was confirmed by random amplified polymorphic DNA and cleaved amplified polymorphic sequence (CAPS) analyses. Furthermore, analyses of chloroplast (cp) and mitochondrial (mt) DNA by CAPS showed that these somatic hybrids contained cp- and mt-DNA of round kumquat without recombination in the regions analyzed.

    The fruits of Fortunella Swingle are widely consumed as fresh fruits and traditional medicine in China. China is the origin center and has the largest cultivated area of the genus Fortunella. In this study, the chemical compositions of ethanol extracts of the major Fortunella cultivated types including Fortunella japonica Swingle, Fortunella margarita Swingle, Fortunella crassifolia Swingle 1 (Lanshang) and Fortunella crassifolia Swingle 2 (Liuyang) were determined using ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC Q-TOF/MS) method, and their antioxidant activities were evaluated. 12 compounds were identified and 5 compounds were tentatively characterized. The results showed that the chemical compositions of the ethanol extracts of 4 Fortunella cultivated types were largely the same. 3', 5'-di-C-glucopyranosylphloretin was the predominant flavonoid in Fortunella fruits, and Fortunella margarita Swingle had higher contents of flavonoids than other species. In addition, the data demonstrated high antioxidant activities of Fortunella fruits. The developed method could be available to rapidly analyze the chemical compounds in Fortunella fruits and its products. This study will provide information for further quality assessment and utilization of Fortunella resources.

  • Comparative analysis on the distribution of heterochromatin in Citrus, Poncirus and Fortunella chromosomes

    Double fluorochrome staining with chromomycin A_3 (CMA) and 4′-6-diamidino-2-phenylindole (DAPI) was used to characterize and compare the distribution of constitutive heterochromatin along chromosomes of Citrus, Poncirus and Fortunella species. Only CMA-positive bands were distinguishable in metaphase chromosomes. Preferential distribution of heterochromatin in terminal regions, mainly of the long arm, and centromeric regions of a few long chromosomes was a common feature of these genera. Heteromorphism between possible homologous chromosomes was present in the majority of species. Citrus and Poncirus revealed some remarkably uniform chromosomes without any intensively fluorescing region, whereas Fortunella cultivars were differentiated by the presence of CMA bands in all chromosomes. Through measurements assisted by a computer, amounts of CMA-positive regions were shown to be highest in Fortunella . Similarities between Citrus and Poncirus suggest little heterochromatin diversification among karyotypes of these genera, whereas Fortunella , with higher amounts and more homogenous distribution of heterochromatin, is more divergent.

    Abstract:

    Somatic hybrids were obtained by the symmetric fusion of embryogenic callus cells from tetraploid 'Mame' kumquat [Fortunella hindsii (Champ.) Swing.] and mesophyll cells from diploid trifoliate orange [Poncirus trifoliata (L.) Raf.]. Southern blot analysis of three regenerants revealed that they carried specific rDNA fragments from both fusion partners, thereby confirming their hybridity. In contrast, mitochondrial DNA (mtDNA) and chloroblast DNA (cpDNA) were unidirectionally transmitted from the callus parent without any evidence of recombination. No differences in the restriction fragment patterns of rDNA, mtDNA or cpDNA could be detected among the regenerants. Flow cytometry showed that two regenerants were hexaploids, as expected, but that one was pentaploid, probably due to elimination of chromosomes prior to the regeneration of this plant.

    Branch internodes of mature plants and stem internodes of seedlings of Fortunella hindsii flowered in vitro on half-strength MT (Murashige and Tucker 1969) basal medium supplemented with benzyladenine, adenine, 6-γ-γ-dimethylallylaminopurine and kinetin. The highest percentage of flowering was achieved with explants originating from branch internodes of flowering plants close to the apex on half-strength MT basal medium containing 5% sucrose and 0.01 mg 1^−1 BA in light. Exposure to darkness for more than 3 weeks followed by re-exposure to light reduced flowering. Flowering required a 4-day exposure to BA, but shoot formation could be initiated even without exposure to BA. First branch internode segments on MT basal medium containing 5% sucrose were prolific in flower (85%) production. The sucrose treatment affected the flower bud size distribution. There were about 13 flower buds per culture in the largest size category (>5 mm).

    Abstract Nine isozyme systems were investigated by polyacrylamide gel electrophoresis as a means of identifying the six species of Fortunella . By examining the characteristic isoenzymatic patterns, it was found that there was considerable variability between the different species. The relationships among the six species of Fortunella were inferred from the similarity matrix cata. The results indicate that no pairs of species are identical and the degree of relationship between the species was low, suggesting that the six species of Fortunella are independent.

    Abstract Intergeneric hybrids between Microcitrus (Microcitrus inodora) × Eremocitrus (Eremocitrus glauca), Microcitrus (Microcitrus warburgiana) × Fortunella (Fortunella margarita), Fortunella (F. margarita) × Microcitrus (M. warburgiana) , and Citrus (Citrus grandis cultivar ‘Tosa-buntan’) × Poncirus (Poncirus trifoliata) were developed by controlled pollination. Hybridization was confirmed by isozyme analysis using polyacrylamide gel electrophoresis. Genetic analysis of glutamate-oxaloacetate transaminase (GOT) clearly indicated that a dimeric nature of GOT isozymes in ‘true citrus fruit trees’ of the subtribe ‘Citrinae’ and their genetic control by two loci with six alleles at Got-1 and four alleles at Got-2 . Four new species-specific alleles were discovered, one for E. glauca (Got-1 ‘E’) and three for M. warburgiana (Got-1 ‘W’, Got-2 ‘W’ and ‘G’). Common alleles were observed among the genera of Microcitrus, Fortunella and Eremocitrus and homologous loci were observed among the studied genera.
