    Abstract:

    O arsênio é um metalóide tóxico que se tornou um problema de saúde pública em todo o mundo. Como forma de reduzir a contaminação ambiental por este metalóide, a qual é proveniente de atividades antropogênicas e naturais, a utilização de microorganismos em processos de biorremediação se tem mostrado uma estratégia promissora. A cianobactéria filamentosa homocitada, Geitlerinema unigranulatum UFV-E01, pertencente à ordem Pseudanabaenales, foi isolada de um ambiente contaminado por arsênio, sugerindo uma habilidade em lidar com o efeito tóxico deste metalóide. Com vista nisso, o presente trabalho objetivou caracterizar a resistência ao arsenato de sódio e quantificar o arsênio total extracelular da cianobactéria G. unigranulatum UFV-E01. As análises de resistência ao arsenato de sódio revelaram que a cianobactéria foi capaz de crescer em até 50 mM por 20 dias. Além disso, a cianobactéria G. unigranulatum UFV-E01 acumulou arsenato de sódio por 10 dias, reduzindo em até 67% o arsênio extracelular. Pelos dados obtidos neste estudo, a cianobactéria G. unigranulatum UFV-E01 foi capaz de resistir a altas concentrações de arsenato de sódio, no entanto outras análises, como a caracterização das vias metabólicas envolvidas no processo de resistência, devem ser realizadas para considerar sua aplicação em ambientes impactados por arsênio.ABSTRACT: Arsenic is a toxic metalloid that has become a public health problem worldwide.     Geitlerinema amphibium (C. Agardh ex Gomont) Anagn. and G. unigranulatum (Rama N. Singh) Komarek et M. T. P. Azevedo are morphologically close species with characteristics frequently overlapping. Ten strains of Geitlerinema (six of G. amphibium and four of G. unigranulatum) were analyzed by DNA sequencing and transmission electronic and optical microscopy. Among the investigated strains, the two species were not separated with respect to cellular dimensions, and cellular width was the most varying characteristic. The number and localization of granules, as well as other ultrastructural characteristics, did not provide a means to discriminate between the two species. The two species were not separated either by geography or environment. These results were further corroborated by the analysis of the cpcB-cpcA intergenic spacer (PC-IGS) sequences. Given the fact that morphology is very uniform, plus the coexistence of these populations in the same habitat, it would be nearly impossible to distinguish between them in nature. On the other hand, two of the analyzed strains were distinct from all others based on the PC-IGS sequences, in spite of their morphological similarity. PC-IGS sequences indicate that these two strains could be a different species of Geitlerinema. Using morphology, cell ultrastructure, and PC-IGS sequences, it is not possible to distinguish G. amphibium and G. unigranulatum. Therefore, they should be treated as one species, G. unigranulatum as a synonym of G. amphibium.

    SUMMARY The taxonomic study of 14 strains of Geitlerinema amphibium (Ag. ex Gom.) Anagnostidis and Geitlerinema unigranulatum (R.N. Singh) Komarek and Azevedo, coming from several localities was undertaken. Use was made of morphological data and molecular data were obtained by means of the DNA fingerprinting technique using highly iterated palindrome (HIP1) sequences. The employed morphological characteristics were those used for species taxonomic identification belonging to the Geitlerinema genus, namely, cell dimensions, shape of the apical cell, motility, number and localization of cyanophycin granules in the cell. The two species revealed as polymorphic were discriminated only by means of the average cellular diameters. In spite of this, minima and maxima values of the cellular diameters overlapped. It was found from molecular analysis that a high genetic diversity and the formation of two clusters consisted of G. amphibium and G. unigranulatum, plus a sole strain keeping itself isolated from the remaining. Also, these clusters were not related to the geographic location; they encompassed strains from water bodies distant from each other by as much as 3500 km, or Brazilian and Spanish strains. Molecular and morphological data support the possibility that G. unigranulatum could be considered a synonym for G. amphibium. HIP1 fingerprinting is a powerful tool for the study of genetic of cyanobacteria closely related taxa. This study points to the necessity of using other than morphological data in the taxonomic revision of cyanobacteria, as well as in the proposition of new taxons.

    Abstract Most of the knowledge about cyanobacteria toxin production is traditionally associated with planktonic cyanobacterial blooms. However, some studies have been showing that benthic cyanobacteria can produce cyanotoxins. According to this, we aimed to evaluate the production of microcystins and saxitoxins in benthic cyanobacteria isolated from aquatic ecosystems in the Northeast of Brazil and to use a polyphasic approach for their identification. Forty-five clonal strains were isolated from rivers and water supply reservoirs, and identified using morphological and molecular phylogenetic characteristics. In order to evaluate the toxins production, the strains were screened for genes involved in the biosynthesis of microcystins and saxitoxins, positive results were confirmed and cyanotoxins quantified using HPLC. Eight species were identified belonging to the Phormidiaceae, Pseudanabaenaceae and Nostocaceae families. This is the first study in Brazil that shows that strains from the Geitlerinema genus correspond to at least three phylogenetic lineages, which possibly correspond to three distinct species to be subsequently reclassified. The strains that showed one of the genes involved in the cyanotoxins production were analyzed by HPLC and Geitlerinema amphibium, Geitlerinema lemmermannii, Cylindrospermum stagnale and Phormidium uncinatum were identified as producing one or more saxitoxins variants. Thus, this is the first report of saxitoxins production for those first three species and the first report in Brazil for P. uncinatum.

    This study analysed vertical and temporal variations of cyanobacteria in a potable water supply in northeastern Brazil. Samples were collected from four reservoir depths in the four months; September and December 2007; and March and June 2008. The water samples for the determination of nutrients and cyanobacteria were collected using a horizontal van Dorn bottle. The samples were preserved in 4% formaldehyde for taxonomic analysis using an optical microscope, and water aliquots were preserved in acetic Lugol solution for determination of density using an inverted microscope. High water temperatures, alkaline pH, low transparency, high phosphorous content and limited nitrogen content were found throughout the study. Dissolved oxygen stratification occurred throughout the study period whereas temperature stratification occurred in all sampling months, with the exception of June. No significant vertical differences were recorded for turbidity or total and dissolved forms of nutrients. There were high levels of biomass arising from Planktothrix agardhii, Cylindrospermopsis raciborskii, Geitlerinema amphibium and Pseudanabaena catenata. The study demonstrates that, in a tropical eutrophic environment with high temperatures throughout the water column, perennial multi-species cyanobacterial blooms, formed by species capable of regulating their position in the water column (those that have gas vesicles for buoyancy), are dominant in the photic and aphotic strata.O presente estudo analisou as variações vertical e temporal de cianobactérias em um reservatório de abastecimento de água no nordeste do Brasil. As amostras foram coletadas em quatro profundidades no reservatório, durante quatro meses (setembro e dezembro de 2007, março e junho de 2008). As amostras de água para a determinação de nutrientes e de cianobactérias foram coletadas por meio de garrafa horizontal de Van Dorn. As amostras foram preservadas em formol 4% para posterior análise taxonômica utilizando um microscópio óptico, e alíquotas de água foram preservadas em solução de Lugol acético para determinação da densidade através de um microscópio invertido. Altas temperaturas da água, pH alcalino, baixa transparência, alto teor de fósforo e limitação de nitrogênio foram encontrados ao longo do estudo. Durante todo o estudo, foi verificada estratificação do oxigênio dissolvido enquanto que estratificação térmica só não foi observada em junho. Diferenças verticais não foram registradas para a turbidez e formas totais e dissolvidas de nutrientes. Ocorreram altas biomassas de Planktothrix agardhii, Cylindrospermopsis raciborskii, Geitlerinema amphibium e Pseudanabaena catenata.   • Cyanobacterial blooms in freshwaters bodies in a semiarid region, northeastern Brazil: A review

    Harmful cyanobacterial blooms have caused several problems in freshwater environments due to their prolific growth and the harmful cyanotoxins produced by some species. The occurrence of these organisms has increased in recent decades due to climate change and eutrophication, although most studies are from temperate regions in the Northern hemisphere. This review presents data about cyanobacteria occurrence, dominance, and toxicity events in freshwater bodies in a semiarid region of Northeast Brazil, in the tropical Southern hemisphere. We performed a literature survey of cyanobacteria publications from 1930 to 2016. We made a list of all the dominant species registered in each state, noted their distribution and occurrence of dominance events involving one or more species, and the registered records of toxic blooms, including information about the toxins involved and the range of values. We selected 102 publications that described cyanobacteria occurrence from states in Northeast Brazil; these publications included relevant contributions regarding cyanobacteria distribution, richness, density, and biomass. Forty-nine dominant species were recorded, with the most representation found in the state of Pernambuco (30 spp.). The genera with the highest occurrences were Microcystis, Cylindrospermopsis, Planktothrix, Dolichospermum (=Anabaena), and Geitlerinema, especially the species Cylindrospermopsis raciborskii, Microcystis aeruginosa, and Planktothrix agardhii. Episodes of toxic blooms were observed in four states. Microcystins, cylindrospermopsin, saxitoxins, and anatoxin-a(S) were found to be associated with these blooms. In Northeast Brazil, harmful cyanobacterial blooms are common in urban and public reservoirs. However, in recent years, cyanobacterial blooms in this region have been more intense and perennial, with high biomass occurring throughout the year

PC-IGS sequences indicate that these two strains could be a different species of Geitlerinema. Using morphology, cell ultrastructure, and PC-IGS sequences, it is not possible to distinguish G. amphibium and G. unigranulatum. Therefore, they should be treated as one species, G. unigranulatum as a synonym of G. amphibium.

    Abstract:

    SUMMARY The taxonomic study of 14 strains of Geitlerinema amphibium (Ag. ex Gom.) Anagnostidis and Geitlerinema unigranulatum (R.N. Singh) Komarek and Azevedo, coming from several localities was undertaken. Use was made of morphological data and molecular data were obtained by means of the DNA fingerprinting technique using highly iterated palindrome (HIP1) sequences. The employed morphological characteristics were those used for species taxonomic identification belonging to the Geitlerinema genus, namely, cell dimensions, shape of the apical cell, motility, number and localization of cyanophycin granules in the cell. The two species revealed as polymorphic were discriminated only by means of the average cellular diameters. In spite of this, minima and maxima values of the cellular diameters overlapped. It was found from molecular analysis that a high genetic diversity and the formation of two clusters consisted of G. amphibium and G. unigranulatum, plus a sole strain keeping itself isolated from the remaining. Also, these clusters were not related to the geographic location; they encompassed strains from water bodies distant from each other by as much as 3500 km, or Brazilian and Spanish strains. Molecular and morphological data support the possibility that G. unigranulatum could be considered a synonym for G. amphibium. HIP1 fingerprinting is a powerful tool for the study of genetic of cyanobacteria closely related taxa. This study points to the necessity of using other than morphological data in the taxonomic revision of cyanobacteria, as well as in the proposition of new taxons.
