    An attenuated Line of LeisHmania infantum (tHe H-Line), developed tHrougH exposure to gentamicin, Has been sHown to protect dogs against canine visceral leisHmaniasis. A specific diagnostic test to differentiate dogs vaccinated witH tHe attenuated Line from dogs infected witH L. infantum wild-type (L. infantum WT) could be a valuable tool in evaluating tHe effectiveness of canine vaccination. In tHis study, 28 HealtHy dogs were allocated into four groups. In Group I and Group II (eigHt dogs per group), dogs were immunized subcutaneously (s.c.) witH L. infantum H-Line, and tHe dogs of Group II cHallenged s.c. witH L. infantum WT, at 2 montHs post-immunization. In Group III, eigHt animals were cHallenged s.c. witH L. infantum WT, and four dogs of Group IV were injected s.c. witH PBS. We found tHat sera from vaccinated dogs recognize a 21 kDa antigen of promastigotes of L. infantum H-Line but not of L. infantum WT, wHereas sera from unvaccinated dogs cHallenged witH L. infantum WT, recognized a 21 kDa antigen of promastigotes of L. infantum WT but not of L. infantum H-Line. Sera from dogs cHallenged witH L. infantum WT witH prior vaccination witH L. infantum H-Line, recognized a 21 kDa antigen of botH L. infantum WT and L. infantum H-Line. THese results suggest tHat tHe Western blot analysis of antibodies against 21 kDa antigens of L. infantum H-Line and WT may be a useful tecHnique for distinguisHing between dogs vaccinated witH L. infantum H-Line and dogs naturally infected witH L. infantum WT.

    Abstract:

    We Have previously described an attenuated Line of LeisHmania infantum (H-Line), selected by culturing promastigotes in vitro in tHe presence of gentamicin. To elucidate tHe molecular basis for tHis attenuation, we undertook a comparative proteomic analysis using multiplex 2-dimensional (2D) difference gel electropHoresis. EigHteen proteins tHat sHowed significant and reproducible cHanges in expression were identified. Many of tHese were components of tHe tHiol-redox control system in LeisHmania and tHis observation, validated by Western blot, prompted us to investigate tHe sensitivity of tHe attenuated Line to oxidative stress. THe attenuated Line was found to be significantly more susceptible to Hydrogen peroxide, a cHange wHicH may explain tHe loss of virulence. In a direct assay of trypanotHione-dependent peroxidase activity, Hydrogen peroxide metabolism in tHe H-Line was significantly lower tHan in wild type. FurtHermore, trypanotHione reductase activity was significantly lower in tHe H-Line, suggesting tHat gentamicin selection may result in pleiotropic affects on tHiol metabolism in LeisHmania. A putative RNA-binding protein was very strongly up-regulated in tHe attenuated Line, suggesting a possible target for gentamicin in LeisHmania.

    An attenuated Line of LeisHmania infantum (L. infantum H-Line) Has been establisHed by culturing promastigotes in vitro under gentamicin pressure. Here, we sHow tHat L. infantum H-Line induced significantly HigHer levels of IFN-γ and lower levels of IL-10 compared witH tHose in dogs infected witH L. infantum wild type (WT). Anti-LeisHmania-specific total IgG, IgG1, and IgG2 antibodies were present in tHe serum of all infected dogs, witH levels of IgG2 subclass HigHest in tHe sera of dogs inoculated witH L. infantum H-Line. Relatively HigH levels of IgG1 were found in tHe sera of dogs infected witH L. infantum WT. Six of seven dogs immunized intradermally (i.d.) witH tHe attenuated Line later sHowed a positive skin test to leisHmanin, wHereas tHe dogs infected witH L. infantum WT did not. No clinical abnormalities were observed, and no parasites found in tHe visceral organs of tHe dogs inoculated intravenously (i.v.) witH L. infantum H-Line over 24 montHs post-inoculation. Dogs wHicH Had been immunized witH L. infantum H-Line i.d. 12 montHs previously were protected against cHallenge witH L. infantum WT.     An attenuated Line of LeisHmania major (L. major H-Line) Has been establisHed by culturing promastigotes in vitro under gentamicin pressure. A modification of tHe previously described metHod for tHe generation of attenuated L. major is described, giving rise to attenuated parasites after 8 ratHer tHan 12 subpassages. No lesions developed in BALB/c mice infected witH L. major H-Line, wHereas L. major wild-type (WT) induced a TH2 like response witH progressive lesions. Analysis of splenocyte IFN-γ and IL-4 production following stimulation witH promastigotes sHows tHat tHe L. major H-Line preferentially induces TH1-like responses and possibly down-regulates TH2 responses in BALB/c mice. L. major H-Line parasites remained localized in tHe skin and draining lympH node, wHereas L. major WT parasites disseminated into tHe visceral organs of BALB/c mice. Mice infected witH L. major H-Line acquired some resistance against L. major WT.     THe clinicopatHological cHanges following infection witH an attenuated Line of LeisHmania infantum (L. infantum H-Line) were evaluated in mixed breed dogs. Two groups of dogs were infected intravenously (i.v.) or intradermally (i.d.) witH L. infantum H-Line and two control groups were infected i.v. or i.d. witH L. infantum wild-type (L. infantum WT). None of tHe dogs, wHicH were infected i.v. or i.d. witH L. infantum H-Line, sHowed any abnormalities during tHe observation period. In contrast, two out of tHree dogs, wHicH were infected i.v. witH L. infantum WT, developed clinical signs of disease. In addition, no HistopatHological cHanges were seen in tHe liver and spleen of tHe dogs infected witH tHe attenuated Line of parasite, wHereas tHe HistopatHological cHanges in tHe two dogs infected i.v. witH L. infantum WT were severe in form and manifested by infiltration of HigH numbers of inflammatory cells. No promastigotes were found in cultures set up from spleens and livers of dogs infected witH L. infantum H-Line at 12 montHs post-infection, wHereas promastigotes were seen in tHe spleen and liver cultures from 2 dogs infected i.v. witH L. infantum WT.     Objective To observe tHe morpHology cHaracteristics of lips and cHin in skeletal Class Ⅲ malocclusion patients and tHe soft tissue cHanges after different ortHognatHic surgery patterns. MetHods THe 94 adult surgery patients witH skeletal Class Ⅲ malocclusion were selected, wHicH comprised of 31 mandible-only cases, 10 maxilla-only cases and 53 two-jaw cases. THe gender difference on soft tissue morpHology and cHanges of lips following different ortHognatHic surgeries were evaluated by cepHalometric analysis. Results THe gender differences were found only on tHe lengtH of upper lip, tHe deptH and degree of mentolabial fold. In tHe mandible-only group, tHe Li-LL、B'-B、LLL、B'-(LL-Pog')、B'-LL-FH、LL-B' -Pog'、UL-SnPog'、LL-SnPog'、B'- SnPog'、Sn-H Line、Li-H Line、Sm-H Line、Ns-H Line、Ls-NsPog' in botH gender Have cHanged significantly. In maxilla-only group, tHe deptH of tHe upper lip and ULL、A'-(Sn-UL)、Cotg-Sn-Ls、A'-UL-FH、B' -LL-FH and tHe distance from some points to different reference Lines Have cHanged significantly. And in tHe two-jaw group, besides tHe differences observed in mandible-only group, tHe Cotg-Sn-Ls、A'-UL-FH also Have a significant cHange. Conclusions THe gender difference is not significant in skeletal Class Ⅲ patients in tHe tHickness of lip soft tissue, but is significant in tHe lengtH of lips and tHe morpHology of lips and cHin. Two-jaw surgery is good at improving lip and cHin morpHology and is better for patients witH unsatisfied nasolabial and mentolabial morpHology botH.   • Observational SearcHes for CHromospHeric g-Mode Oscillations from CaII H-Line Observations

    We Have used a HigH spatial and temporal resolution of long time sequence of spectra in CaII H-Line obtained at tHe Vacuum Tower Telescope (VTT) of tHe Sacramento Peak Observatory on a quiet region at tHe center of tHe solar disk over a large number of brigHt points and network elements to searcH for atmospHeric (cHromospHeric) g-mode oscillations. An important parameter of tHe H-Line profile, intensity at H2v(IH2V), Has been derived from a large number of Line profiles. We derived tHe ligHt curves of all tHe brigHt points and network elements. THe ligHt curves represent tHe main pulse witH large intensity amplitude and followed by several follower pulses witH lower intensity amplitudes. THe ligHt curves of tHese brigHt points would give an impression tHat one can as well draw curves towards and away from tHe HigHest peak (main pulse) sHowing an exponential growtH and decay of tHe amplitudes. An exponential decaying function Has been fitted for all tHe ligHt curves of tHe brigHt points to determine tHe damping time of tHe modes tHat are more or less tHe same, and one value of tHe coefficient of exponent can represent reasonably well tHe decay for all tHe cases. THe FFT analysis of temporal variation of botH tHe brigHt points and tHe network elements indicates around 10-min periodicity. We speculate tHat tHis longer period of oscillation may be related to cHromospHeric g-mode oscillations.

    We Have analysed a 35-min-long time sequence of spectra in tHe Ca ii H Line, Na i D1 and D2 Lines, and in a large number of strong and weak Fe i Lines taken over a quiet region at tHe center of tHe solar disk. THe time series of tHese spectra Have been observed simultaneously in tHese Lines under HigH spatial, spectral, and temporal resolution at tHe Vacuum Tower Telescope (VTT) of tHe Sacramento Peak Observatory. We Have derived tHe Line profiles and tHeir central intensity values at tHe sites of tHe cHromospHeric brigHt points, wHicH are visible in tHe H Line for easy identification. We Have done a power spectrum analysis for all tHe Lines, using tHeir central intensity values to determine tHe period of oscillations. It is sHown tHat tHe 3 Fe i Lines, present ∼ 23 Å away from tHe core of tHe H Line representing tHe pure pHotospHeric Lines, Na i D1 and D2 Lines, 6 Fe i Lines at tHe wings of H Line, and Can H Line exHibit 5-min, 4.05-min, 3.96-min, and 3.2-min periodicity in tHeir intensity oscillations, respectively. Since all tHese Lines form at different HeigHts in tHe solar atmospHere from low pHotospHere to middle cHromospHere and sHow different periodicities in tHeir intensity oscillations, tHese studies may give an idea about tHe spatial and temporal relation between tHe pHotospHeric and cHromospHeric intensities. THerefore tHese studies will Help to better understand tHe pHysical mecHanisms of solar oscillations. It is clearly seen tHat tHe period of intensity oscillations decreases outward from tHe low pHotospHere to tHe middle cHromospHere. Since we Have studied a single feature at a time on tHe Sun (i.e., brigHt points seen in tHe H Line) in all tHese spectral Lines simultaneously, tHis may explain about tHe footpoints of tHe brigHt points, tHe origin of 3-min oscillations, and tHe relation to otHer oscillations pertaining to tHese locations on tHe Sun. We Have concluded tHat 80% of tHe brigHt points are associated witH dark elements in tHe true continuum, and tHey may seem to Have a relationsHip witH tHe dark intergranular lanes of tHe pHotospHere, after carefully examining tHe brigHtness (brigHt tHreads) extending from tHe core to tHe far wings of tHe H Line at tHe locations of a large number of brigHt points, using tHeir time sequence of spectra.

    From a 35-min time series of pHotograpHic spectra in tHe Ca ii H-Line obtained at tHe Vacuum Tower Telescope (VTT) of tHe Sacramento Peak Observatory under HigH spatial, spectral, and temporal resolution, we Have derived a large number of H-Line profiles at tHe sites of tHe brigHt points in tHe interior of tHe supergranulation cells, and at tHe network elements, on a quiet region at tHe centre of tHe solar disc. It is sHown tHat tHe brigHt points are associated witH 3-min periodicity in tHeir intensity oscillations wHereas tHe network elements exHibit ∼7-min periodicity. It is surmised tHat tHe large difference in periods of tHe intensity oscillations, tHe strengtH of tHe magnetic fields, and tHe intensity enHancements at tHe sites of tHe brigHt points and tHe network elements tHemselves may probably be taken as evidence to argue tHat tHe mecHanisms of Heating in tHe two cases are dissimilar, irrespective of tHe sizes of tHese structures.

    We study Hinode/SOT-FG observations of intensity fluctuations in Ca II H-Line and G-band image sequences and tHeir relation to simultaneous and co-spatial magnetic field measurements. We explore tHe G-band and H-Line intensity oscillation spectra botH separately and comparatively via tHeir relative pHase differences, time delays and cross-coHerences. In tHe non-magnetic situations, botH sets of fluctuations sHow strong oscillatory power in tHe 3 - 7 mHz band centered at 4.5 mHz, but tHis is suppressed as magnetic field increases. A relative pHase analysis gives a time delay of H-Line after G-band of 20\pm1 s in non-magnetic situations implying a mean effective HeigHt difference of 140 km. THe maximum coHerence is at 4 - 7 mHz. Under strong magnetic influence tHe measured delay time sHrinks to 11 s witH tHe peak coHerence near 4 mHz. A second coHerence maximum appears between 7.5 - 10 mHz. Investigation of tHe locations of tHis doubled-frequency coHerence locates it in diffuse rings outside pHotospHeric magnetic structures. Some possible interpretations of tHese results are offered.

  • Space-Time Distribution of G-Band and Ca II H-Line Intensity Oscillations in Hinode/SOT-FG Observations

    Abstract:

    We study tHe space-time distributions of intensity fluctuations in 2 - 3 Hour sequences of multi-spectral, HigH-resolution, HigH-cadence broad-band filtergram images (BFI) made by tHe SOT-FG system aboard tHe Hinode spacecraft. In tHe frequency range 5.5 10 mHz. By looking at G-band frequencies above 10 mHz we find tHat tHe oscillatory power, botH at tHese frequencies and at lower frequencies too, lies in a mesH pattern witH cell scale 2 - 3 Mm, clearly larger tHan normal granulation, and witH correlation times on tHe order of Hours. THe mesH pattern lies in tHe dark lanes between stable cells found in time-integrated G-band intensity images. It also underlies part of tHe brigHt pattern in time-integrated H-Line emission. THis discovery may reflect dynamical constraints on tHe sizes of rising granular convection cells togetHer witH tHe turbulence created in strong intercellular downflows.
