Out-of-season artificial reproduction of common dace (leuciscus leuciscus L.) under controlled conditions

The results indicate there is the possibility of initiating the reproducion in individuals of this before the natural spawning season. There could be induction of spermiation by altering the environmental conditions. For females, hormonal injections were necessary for induction of final oocyte maturation and ovulation. Generally, there were high percentages of spermiation (100%) and ovulation (87%–100%) as well as low mortality (0%–11% and 7%–13% for males and females, respectively) among the induced spawners. The greatest embryo survival occurred when Ovaprim (84.4%) or hCG (89.6%) was administered, although the latency time using hCG was at least twice as long compared to when other spawning agents were used (84–92 hrs and 30–44 hrs). The results from present study could be applicable for optimization of breeding stock numbers in aquaculture enterprises and in providing insights for conservation of L. leuciscus endangered populations.

Early development and allometric growth patterns of rheophilic cyprinid common dace leuciscus leuciscus (Cyprinidae: Leuciscinae)

Based on the external morphology, three different periods and eight characteristic steps (ES9–JS1; ES—embryonic steps, JS—juvenile steps) were identified between hatching and 30 days post-hatch at 25°C. This study shows that all allometric changes occurred after notochord flexion across a wide range of body lengths (15.35–21.78 mm TL) reflecting the larvae–juvenile transition. Observations on the early development of common dace confirmed the relatively uniform development of cyprinid species and support the hypothesis of differential growth patterns for primary functions during early ontogeny. Differences in relative growth between common dace and other species might be attributed to their different size at hatching, behaviour, and environmental cues.

INFLUENCE OF FEEDING NATURAL AND FORMULATED DIETS ON CHOSEN RHEOPHILIC CYPRINID LARVAE

The aim of the work was to determine the impact of commercial feed used as an alternative diet for larval fish of the genera leuciscus on growth and survival during rearing under controlled conditions. Larvae of chub, leuciscus cephalus (L.), ide, leuciscus idus (L.), and orfe (its yellow pigmented form), and dace, leuciscus leuciscus (L.), were obtained from breeding under controlled conditions and reared for 21 days in a closed recirculating system. The control group was comprised of larvae fed Artemia nauplii. The experimental group was fed Artemia for the first seven days of rearing and then this was partially replaced with formulated feed. The experimental group was fed from day 14 exclusively with formulated feed. The results obtained indicated that the feeding treatment was highly effective. While the body sizes of the larval dace from the control group were similar to those of the experimental group, the larvae of the other species that received the mixed diet attained greater weights (chub, ide) or greater weights and lengths (orfe) by the end of the experiment

The aim of the study was to obtain information regarding the spontaneous formation of gynogens in ide, leuciscus idus (L.). Oocytes obtained from orfe and activated with the milt of dace, leuciscus leuciscus (L.), were used for the study. Several control groups were established to determine the quality of gametes and the quality of the diluted semen. Ploidy in the offspring was determined by analyzing the number of nucleoli in the cells. Spontaneous diploid individuals appeared in groups in which the biological quality of the oocytes was poor. The percentage of these individuals was not large, and the average was a maximum of 0.4%. The effectiveness of hormonal preparations (Ovopel, Ovaprim, LHRHa, hCG and CPE) in stimulating spermiation in dace leuciscus leuciscus (L.)

The control group (Control) included fish which milt was taken after 0.9% NaCl injection. The parameters of sperm movement were analysed with the CASA system and included: sperm motility (MOT, %), progressive motility (PRG, %), curvilinear velocity (VCL, μm s−1) and straightlinear velocity (VSL, μm s−1) of sperm, linearity of movements (LIN = 100 × VSL/VCL,%), wobbling (WOB, %), amplitude of lateral head displacement (ALH, μm) and beat cross frequency (BCF, Hz). Milt was taken from all fish (n = 12 per group) 48 h after hormonal stimulation and selected milt and sperm parameters were determined. The best results in volume of milt was found after Ovaprim treatment and TVM and VOM values were statistically higher compared to the values recorded in the Control (P 0.05). The highest VCL and VSL values were recorded after LHRHa treatment but there was no statistical differences between Control, Ovaprim and CPE groups. Isolation and characterization of polymorphic microsatellite loci for the dace complex: leuciscus leuciscus (Teleostei: Cyprinidae)

Four multiplex polymerase chain reaction sets were optimized in order to genotype 26 polymorphic loci in all, including 16 previously published cyprinid-specific loci. The level of genetic diversity was assessed in 142 dace individuals. We also successfully applied 26 of the microsatellites to 10 related species. These primers thus will be useful to assess population structure of the dace and other cyprinid species, with application for conservation issues and phylogeographical approaches.

Heterogeneous colonization pattern of European Cyprinids, as highlighted by the dace complex (Teleostei: Cyprinidae).

Guillard, André GillesAbstract:The dace (leuciscus leuciscus), with a very large geographic distribution all over Europe, represents an interesting species model for studies of the global mechanisms underlying aquatic system biodiversity. To assess the congruence with the past colonization process hypothesis of the freshwater fauna in Western Europe, we investigated the evolutionary history of this species, by integrating morphological variation (eight meristic characters), mitochondrial (cytochrome b, 16S rDNA and control region, over a total of 2169 bp) and nuclear (12 allozymes loci) phylogenetic relationships, and investigating population dynamics via expansion, migration, bottleneck, and divergence time analyses. We carried out nested clade phylogeographic analysis for a total of 663 specimens from 31 populations taken from all over the distribution area. Unlike previous studies, we found that L. leuciscus is currently constituted by five lineages belonging to two clades (yielding 6.3% of pairwise divergence). The relationships between these lineages were accounted for by complex biogeographical patterns due to Pliocene and Pleistocene paleoclimatic events, validating the identification of new glacial refuges for freshwater fish in Western Europe. Finally, we demonstrated hybridization between L. leuciscus and leuciscus idus.

Différenciation génétique et hybridation chez le cyprin Leudscus leudscus

Intraspecific sequence variation within one portion of two mitochondrial genes, cytochrome b and the control region, in L. leuciscus was examined in 21 individuals from France. Analysis of the sequence data using maximum parsimony and neighbor joining algorithms separated the taxa into two lineages corresponding to a part of the Allier basin and to a set of Cyprinidae collected from the Aquitain basin. In our analysis populations in the Aquitain basin and populations in the Allier basin constitute two taxonomic entities. These results suggest that morphological and genetic evolution may be decoupled in L. leuciscus , resulting in morphological similarity between the two basins despite significant genetic divergence. Moreover, we identified two haplotypes belonging to Rutilus rutilus and Alburnus alburnus lineages. Molecular data suggests that genetic differentiation is sufficient to consider and manage them as two taxonomic entities and as separate populations. A more precise phylogenetic description should affect future conservation management decisions about L. leuciscus .

Individual genetic tagging for teleosts: an empirical validation and a guideline for ecologists

Vacquie-garcia, Géraldine LootAbstract:The efficiency of individual genetic tagging was determined by using passive integrated transponders (PIT) as a comparative conventional tagging method. Fifty-five common dace leuciscus leuciscus were captured in the wild, PIT tagged and fin clipped (for DNA analysis). Thirty fish were recaptured on three occasions and tissue samples were collected. Using 18 microsatellite loci, 79-94% of the recaptures were correctly assigned. Experience with scoring L. leuciscus microsatellites led to more individuals correctly assigned. Allowing matches that differed by one or two alleles resulted in 100% of all recaptures successfully assigned irrespective of the observer. Reducing the set of loci to five to six loci appropriately selected did not affect the assignment rate, demonstrating that costs can be subsequently reduced. Despite their potential benefits, the application of genetic tags for teleosts has been limited. Here, it was demonstrated that genetic tagging could be applied, and a clear guideline (flowchart) is provided on how this method can be developed for teleosts and other organisms, with subsequent practical applications to ecology, evolutionary biology and conservation management.

Evidence for host variation in parasite tolerance in a wild fish population

Disentangling between tolerance and resistance is important for predicting the evolutionary outcomes of host-parasite interaction. Dace (leuciscus leuciscus) are often parasitized by the ectoparasite Tracheliastes polycolpus which feeds on (and destroys) fins, reducing thus the host’s condition. We tested the hypothesis that genetically-based variation in ectoparasite tolerance exists in a wild dace population. We found that moderately heterozygous dace, which are less resistant than highly heterozygous or homozygous dace, tolerated better the effect imposed by T. polycolpus for a given parasite burden. However, tolerance also varied upon environmental conditions, suggesting that genetic and environmentally-based variation exists for both resistance and tolerance in this natural host-parasite system. Moreover, a negative genetic correlation may exist between tolerance and resistance, and hence several evolutionary outcomes are possible in this interacting system.

Species-specific responses to landscape fragmentation: implications for management strategies

Here, we compared the genetic structure of four freshwater fish species differing in their body size (leuciscus cephalus; leuciscus leuciscus; Gobio gobio and Phoxinus phoxinus) between a fragmented and a continuous landscape. We tested if, overall, fragmentation affected the genetic structure of these fish species, and if these species differed in their sensitivity to fragmentation. Fragmentation negatively affected the genetic structure of these species. Indeed, irrespective of the species identity, allelic richness and heterozygosity were lower, and population divergence was higher in the fragmented than in the continuous landscape. This response to fragmentation was highly species-specific, with the smallest fish species (P. phoxinus) being slightly affected by fragmentation. On the contrary, fish species of intermediate body size (L. leuciscus and G. gobio) were highly affected, whereas the largest fish species (L. cephalus) was intermediately affected by fragmentation. We discuss the relative role of dispersal ability and effective population size on the responses to fragmentation we report here. The weirs studied here are of considerable historical importance. We therefore conclude that restoration programmes will need to consider both this societal context and the biological characteristics of the species sharing this ecosystem

The effects of the ectoparasite Tracheliastes polycolpus (Copepoda: Lernaeopodidae) on the fins of rostrum dace (leuciscus leuciscus burdigalensis)

Samples from five study sites along the river revealed different patterns of parasite infestation. Heavily infested fish were found at the upper study sites whereas much lower infestation levels were observed at the lower study sites. The copepods showed an aggregated dispersion pattern on host fins. The results showed significantly preferred microhabitats, with adult females being more abundant on the anal, pelvic and along the external part of the pectoral fins. The anal and pelvic fins were damaged by the parasite with a loss of their surface area. These fin alterations may reduce the fish’s swimming ability and therefore affect the rostrum dace population. Our findings highlight the need to study the effects of parasites on stream fish populations.

Effects of temperature on survival, deformations rate and selected parameters of newly hatched larvae of three rheophilic cyprinids (genus leuciscus).
Polish Journal of Natural Science, 2010Co-Authors: Krzysztof Kupren, Andrzej Mamcarz, Dariusz KucharczykAbstract:Experiments were conducted to determine full range of tolerated and optimal water condition for eggs incubation of three species from genus leuciscus i.e: dace leuciscus leuciscus (L.), ide leuciscus idus (L.), and chub leuciscus cephalus (L.). Spawners were caught from rivers of the northern (Pasleka River drainage) and central (Pilica River drainage) parts of Poland. Fertilized eggs were incubated under controlled conditions at ten different constant temperatures ranging from 4.5 to 29.0 o C. The optimal temperature ranges for the incubation of dace, ide and chub eggs were 7.5 to 12.3oC; 15.7oC and 19.0 to 23.0oC, respectively (considering hatching percentage, incidence of abnormalities and size of hatched larvae), which are close to the water temperature during spawning season. This study also reveals that embryos of studied species can adapt to increasing water temperature due to global warming up to 23.0 o C (dace and ide) and 27.5 o C (chub). Besides, some inter-population differences in the response to temperature were observed.

Application of Ovopel and Ovaprim and their combinations in controlled reproduction of two reophilic cyprinid fish species.

The experiment was conducted under controlled conditions. During the experiment the percentage of ovulating females, ovulation time and survival of embryos to the eyed stage were recorded. Stimulation with Ovopel resulted in shorter ovulation appearance time in females of both species (48–50 h for dace and 36–38 h for ide) while stimulation with Ovaprim in a higher percentage of live embryos in the eyed stage (almost 80 and 60% for dace and ide respectively). The highest hormonal stimulation effectiveness was recorded in the group where a combination of those two hormonal substances was applied as shortening of the time for achievement of spawning readiness (48 and 36 hours for dace and ide respectively), a significant ovulation synchronization and the highest embryos survival rates (almost 80 and 70% for dace and ide respectively) were recorded.

Polish Journal of Natural Science, 2008Co-Authors: Krzysztof Kupren, Andrzej Mamcarz, Dariusz Kucharczyk, Maja Prusińska, Slławomir KrejszeffAbstract:Wild living spawners of dace leuciscus leuciscus (L.), ide L. idus (L.) and chub L. cephalus (L.), were obtained from rivers of central (the Pisa River basin) and northern (the Pasleka River basin) Poland and next transported to a hatchery for artificial reproduction. The obtained eggs were dry fertilized and next incubated in water as different constant temperatures ranging from 4.5 to 29.0 o C. The hatched embryos were kept at the same temperatures until the moment of complete yolk sac resorption and later fed ad libitum on live nauplial Artemia sp. stages. The power function was applied for determination of the relation between the temperature and time until and achievement of each of the thirteen characteristic stages of embryonic development. It was shown that the pace of embryonic development increased with the temperature increase. In the majority of cases, at identical thermal conditions, both the incubation time and the pace of embryonic development were much less diversified among individual of the same species originating from different population than between the individual species. The obtained data finds application in optimization of early raring process and as a consequence will be an important tool in protection of endangered species.

CHANGES IN MORPHOMETRIC PARAMETERS IN SELECTED EARLY ONTOGENIC STAGES OF THREE FISH SPECIES FROM THE GENUS leuciscus (TELEOSTEI, CYPRYNIDAE)
Archives of Polish Fisheries, 2008Co-Authors: Krzysztof Kupren, Andrzej Mamcarz, Dariusz Kucharczyk, Maja PrusińskaAbstract:Observations were conducted of the changes in the measurements of selected morphometric characters in embryos and larvae of three closely-related fish species (dace, leuciscus leuciscus (L.), ide, leuciscus idus (L.), European chub, leuciscus cephalus (L.)). It was confirmed that individual development immediately following hatching is fast with rapid changes in body proportions that stem from the development of particular systems. It was also observed that in dace and ide the values of the biometric characters observed were very similar, as were the trends of these changes. Although chub differed insignificantly from the other two species, statistically significant relative values and the character of growth were noted especially with regard to the lowest height of the tail fold, head depth, body depth, and yolk sac length. Differences in body proportions can be explained by the different maintenance strategies of these species during the earliest stages of ontogenesis.
