    Abstract:

    Ulat pemakan daun kelapa sawit adalah hama utama atau hama kunci di perkebunan kelapa sawit karena dapat menyerang sawit hampir setiap waktu. Penelitian ini bertujuan untuk mempelajari dan mengetahui keragaman jenis ulat pemakan daun pada perkebunan kelapa sawit di Hartono Plantation Indonesia (HPI)-Agro PT. Peniti Sungai Purun (PSP). Penelitian ini bersifat deskriptif dengan cara melakukan pengamatan langsung di lapangan. Hasil pengamatan menunjukan bahwa terdapat 19 jenis ulat pemakan daun kelapa sawit yaitu: Setora nitens, Darna trima, Darna (Ploneta) diducta, Thosea vetusta, Birthosea bisura, Hidari irava, Mahasena corbetti, Clania sp., Amathusia phidippus, Orgyia antiqua, Dasychira inclusa, Spilosoma libericipeda, Calliteara horsfieldii, Elymnias panthera, Artornis sp., Euproctis virgunncula, Euproctis flexuosa serta terdapat 2 jenis yang belum diketahui secara pasti spesiesnya dari famili Lymantriidae. Spesies yang ditemukan termasuk dalam 6 famili, yaitu 5 spesies dari famili Limacocididae, 2 spesies dari famili Psychidae, 8 spesies dari famili Lymantriidae, 2 spesies dari famili Nymphalidae, 1 spesies dari famili Hesperiidae dan 1 spesies dari famili Arctiidae. Spesies yang paling dominan adalah Clania sp. sekitar 49% kemudian disusul oleh Setora nitens 30.97% dan spesies lainnya dibawah 10%. Kata Kunci : Kelapa Sawit, Keragaman, Ulat Pemakan Dau

    Abstract:

    Background Outbreaks of the casuarina moth, Lymantria xylina Swinehoe (Lepidoptera: Lymantriidae), which is a very important forest pest in Taiwan, have occurred every five to 10 years. This moth has expanded its range of host plants to include more than 65 species of broadleaf trees. LyxyMNPV (L. xylina multiple nucleopolyhedrovirus) is highly virulent to the casuarina moth and has been investigated as a possible biopesticide for controlling this moth. LdMNPV-like virus has also been isolated from Lymantria xylin a larvae but LyxyMNPV was more virulent than LdMNPV-like virus both in NTU-LY and IPLB-LD-652Y cell lines. To better understand LyxyMNPV, the nucleotide sequence of the LyxyMNPV DNA genome was determined and analysed.

    Abstract:

    Abstract Four new cell lines, designated as NTU-LY-1 to -4, respectively, were established from the pupal tissues of Lymantria xylina Swinhoe (Lepidoptera: Lymantriidae). These cell lines have been cultured approximately 80 passages during 2 years in TNM–FH medium supplemented with 8% fetal bovine serum, at a constant temperature of 28 °C. Each line consists of three major morphological types: round cells, spindle-shaped cells, and giant cells. The characterization of these cell lines showed that they are different from previously established lines derived from related Lepidopteran species. All new lines were susceptible to the L. xylina multiple nucleopolyhedrovirus (LyxyMNPV) and appeared to have a good potential for studying this virus.

    Abstract:

    A new cell line, designated NTU-Pn-HF, was established from the pupal ovary of Perina nuda Fabricius (Lepidoptera: Lymantriidae) and characterized by distinct morphological properties and molecular markers. Pn-HF cells are highly susceptible to P. nuda nucleopolyhedrovirus (PenuNPV). Surprisingly, some of the sub-cultural Pn-HF cells suffered an obvious cytopathic effect (CPE) and proceeded to die, while most of the cells remained healthy with good nutrition. Under electron microscopy, most of the Pn-HF cells were found to be carriers of a picorna-like virus. A specific anti-P. nuda picorna-like virus (PnPV) antiserum and RT-PCR with a primer set designed from the sequence of a putative PnPV’s helicase gene confirmed that the picorna-like virus in Pn-HF cells is PnPV (Wang et al., 1999, J. Invertebr. Pathol. 74: 62‐68), and Pn-HF cells are persistently infected with PnPV. This finding implies that the Pn-HF cell line can be a PnPV supporter for further work, and also a convenient tool in studying the pathogenesis of insect picornavirus and the mechanism of the picornavirus-persistent infection.

    Abstract A continuous cell line, designated NTU-PN-HH, was established from the pupal ovary ofPerina nudaFabricius (Lepidoptera: Lymantriidae). The cells have been through more than 300 passages during 4 years in TNM-FH medium supplemented with 10% FBS, at a constant temperature of 28°C. The cell line consists of four major morphologic types: polymorphic cells, spindle-shaped cells, round cells, and squamous cells. The characterization of this cell line showed that NTU-PN-HH is a newly established cell line. It is the first cell line that is permissive to PnNPV (P. nudamultiple nuclear polyhedrosis virus).
