    Abstract:

    Total hemocyte number,quantity and morphological change of plasmatocyte and granulocyte of Musca Domestica third larvae infected by colon bacillus after 4 h,8 h,16 h and 24 h were observed by inverted microscope and Giemsa stained method to provide a scientific basis for further studying the function of hemocyte and cell immunological mechanism.The results showed that the plasmatocyte and granulocyte are main immune cells in immune response of Musca Domestica larvae,the total hemocytes,plasmatocytes and granulocytes of Musca Domestica third larvae after infection for 4 h all increased and were up to the peak after infection for 8 h,and then began to decrease.The plasmatocytes and granulocytes of different treatment groups after infection formed piles,and the cytomorphosis,vacuole,and congregation of grains in the nucleus also occurred.There was cell division among the plasmatocytes of the Musca Domestica larvae in control group.The granulocyte division and plasmatocyte division occurred in different treatment groups after infection,and the self-division of hemocytes may be the primary source.

    [Objective] The research aimed to observe the effects of Escherichia coli infection on the morphology of hemocytes of the 3rd stage larva of Musca Domestica in vitro and understand the hemocytes types that take part in the cell immunity of Musca Domestica larval.[Method] The hemcytes of the 3rd stage larva of Musca Domestica were cultured in vitro and the hemocyte morphology was observed about 2,4,6,8 h after culture in vitro.After Escherichia coli were injected into the hemocytes of the 3rd stage larva of Musca Domestica in vitro,the morphology changes of hemocytes were observed at different time after infection.[Result] The hemocytes of of the 3rd stage larva of Musca Domestica was divided into five types about 2 h after hemoculture.The hemocytes partly adherence was seen about 6 h after hemoculture.The vacuolation and morpholysis was found in plasmatocytes after being infected by E.coli and a great quantity bacterium were gathered around granulocyte,but the morphology changes of hemocytes were not found in the prohemocyte,shprulocyte and oenocytoid.[Conclusion] The plasmatocyte and granulocyte were primary participants of the cell immunity of Musca Domestica larval,but the prohemocyte,sphrulocyte and oenocytoid do not participate in the cell immune reactions.

    Objective To examine the antibacterial activity of antibacterial peptides produced from the cultural embryonic cell of the Musca Domestica induced by lipopolysaccharide(LPS).Methods Embryonic cell lines of the Musca Domestica(MDEC-07114) were induced with LPS.Culture supernatant was collected at 4,8,12,16,20 and 24 h after induction.The antibacterial peptides were recovered by trichloroacetic acid precipitation method,and the antibacterial activity was determined using the zone inhibition assay.Results Embryonic cell at the logarithmic growth phase was induced by LPS.There was no significant change in the cell number and the cell morphology.Antibacterial peptides was produced at 8 h and reached a peak level at 16 h after stimulation.The antibacterial activity could still be detected at 24 h after the treatment.Conclusion Antibacterial peptides can be produced by the LPS-treated embryonic cell lines of the Musca Domestica(MDEC-07114).

    Objective To observe the rule of antibacterial activity and the antibacterial characterization of the antibacterial proteins in the Musca Domestica larvae by different inducements.To provide the experiment proof for the antibacterial proteins producing and application of the Musca Domestica larvae.Methods The Musca Domestica larvae were induced by four factors,antibacterial proteins were separated from the tissue homogenate at the different time,detected their antibacterial activity to observe the peak of the antibacterial proteins.Several different bacteria were used as detecting index to detect the antibacterial characterization of antibacterial proteins which induced by four inducements.Results The antibacterial proteins of antibacterial activity of four induced groups were better than control group,and the peak of antibacterial activity appeared at 36~48?h after induced by four inducements.These antibacterial proteins could inhabit many bacteria(including some Grame-positive,Grame-negative bacteria and rifampin resistant E.coli),but antibacterial activity were different.Conclusion The peak of antibacterial proteins in the Musca Domestica larvae by different inducements appeared at different time,and could inhibit diversiform bacteria.

  • Cloning and Sequence Analysis of the Defensins Gene from Musca Domestica Larva

    Abstract:

    Objective To clone and sequence analysis the defensins genes from Musca Domestica larva. Methods Total RNA was extracted from Musca Domestica larva. The PCR was carried out, and the product of PCR were puified and cloned into plasmid T vector. The recombinant plasmid was sequenced and analyzed. Results The amplified products from Musca Domestica larva were about 310bp. The sequence analysis showed that the amplified fragment from Musca domestic larva had a high homology compared with the published defensins sequence from Musca Domestica.There has a single nucleotide variation at signal peptide region. Conclusions The defensins cDNA of Musca Domestica larva have been cloned successfully.This will lay the foundation for the research of expression of the recombinant defensins and its antibiotic activity.

    Objective To observe the changes of carbohydrate in the hemlymph of Musca Domestica larva before and after in the diapause.To explore the biochemistry mechanism in the diapause of Musca Domestica larva.Methods The diapause of the Musca Domestica larva was inducted by low temperature.The contents of total carbohydrate,trehalose and glycogen in the hemlymph of Musca Domestica larva in the diapause 10d,15d,20d groups and the nondiapause group were determimined by anthrone chromatometry,respectively.Results Compared with the diapause groups and the nondiapause group,the content of the hemlymph total carbohydrate was increased significantly in the diapause 10d group,but were decreased in the 15d and 20d groups;the contents of the hemlymph trehalose were increased significantly in the diapause 10d and 20d groups,and there were not differences in the diapause 15d group;the contents of the hemlymph glycogen were decreased in the diapause 10d and 15d groups,but was increased significantly in the 20d group.Conclusion The accumulation model of carbohydrate alcohol in the diapause larva of the Musca Domestica was belonged to the model of trehalose.

  • Construction of SSH library of Musca Domestica larvae upon infection with Pasteurella mutocida of bovine

    Abstract:

    Three-day-old Musca Domestica larvae were pricked with a needle dipped in Pasteurella multocida of Bovine.The total RNA was extracted from Musca Domestica larvae,and we got the high quality mRNA after isolation and purification.Suppression subtractive hybridization(SSH) was performed to generate a subtracted cDNA library,and the differentially expressed genes were screened by reverse Northern hybridization technology.The results showed that the sizes of positive clonal fragments that picked randomly were between 200bp and 750bp and 183 genes differentially expressed were screened by reverse Northern hybridization technology.A large number of genes that associated with the immune defensive of Musca Domestica and a portion of the non-homologous genes were identified by BLAST.
