    Abstract:

    The pine processionary moth (PPM), Thaumetopoea wilkinsoni Tams. (Lepidoptera: Thaumatopoeidae), is an important forest pest in the Mediterranean area, and urticating hairs of the caterpillars of this species cause allergic reactions on skin of humans and animals. In the present study, the larvicidal activities of Turkish oregano (Origanum Onites L.) essential oil and its four major components, carvacrol, gamma-terpinene, terpinen-4-ol and thymol, were evaluated against fourth/fifth-instar larvae of PPM under laboratory conditions. The essential oil was larvicidal to PPM with an LD50 value of 3800 µL L−1 when 0.1 mL was applied per larva. Carvacrol was the most toxic component found in the essential oil (LD50 = 3100 µL L−1), followed by thymol (LD50 = 5500 µL L−1). The other two components, gamma-terpinene and terpinen-4-ol, were less effective. The results showed that Turkish oregano essential oil and its two components, carvacrol and thymol, could be potential alternatives to synthetic insecticides for the control of PPM. Bu yuzden yiyeceklerde bir baharat olarak da tercih edilir. Terpen sentaz-6 ( TPS-6 ), aromatik bitkilerde sesqui-terpenlerin sentezinden sorumlu bir enzimdir. O. Bu calismada, O. Onites ’de TPS-6 geninin kismi olarak klonlanmasi ve dizilenmesi amaclandi. O. Onites ’den toplam RNA izolasyonu TRI Reagent solusyonu ile gerceklestirildi. O.     Abstract:

    Origanum Onites L. (oregano) is an aromatic plant used in medicine, cosmetics and different industrial fields. It includes essential oil and is extensively cultivated as aromatic plant in the world. Therefore, it is also preferred as a spice in foods. Terpene synthase-6 (TPS-6) is an enzyme responsible for synthesis of sesqui-terpenes in aromatic plants. The investigation of gene sequence information and expression characteristics of this enzyme in O. Onites may contribute to increase of synthesis of terpenes as raw materials in medicine and cosmetic etc. industry. In this study, partial cloning and sequencing of the TPS-6 gene in the O. Onites was aimed. Total RNA isolation from O. Onites was performed with TRI Reagent solution. The mRNAs were transformed to cDNAs by Revers Transcription Method. The middle region of TPS-6 gene was amplified by PCR method using degenerative primers designed for TPS-6. Amplified DNA fragments were obtained at 481bp. size and ligated to PGEM T Easy plasmid vector with T4 DNA ligase. The recombinant plasmid vector was transferred to the XL1-Blue host cells (E. coli) and the cloned DNA was sequenced. The partial TPS-6 DNA sequence was confirmed in NCBI GenBank database and registered with an accession number (MF983853). The obtained DNA sequence was analyzed with InterProScan and Clustal Omega software in EBI database. The analysis results showed that this DNA sequence has a specific motif (DXXD) for TPS-6 catalyzing cyclization reactions. The phylogenetic dendrogram was drawn for comparison with other plant TPS-6 genes. O. Onites TPS-6 sequence was determined to be very close to O. vulgare TPS-6 on the dendrogram than other plant TPS-6s. The obtained gene sequence information will guide biotechnological researches in the productivity increase of this gene using expression analysis

    Abstract:

    Plants are constantly exposed to a variety of environmental (biotic and abiotic) stresses and they develop sophisticated signaling pathways to adapt the changing environment. Mitogen activated protein kinase (MAPK) cascade in plants is a common mechanism used to translate the external stimuli into cellular responses and composed of triple kinases modules namely MAPKKK-MAPKK-MAPK. In this study, a MAPKK (OoMAPKK1) has been cloned from Origanum Onites L. (Lamiaceae) which is a commercially cultivated aromatic plant in the world. The OoMAPKK1 gene is 1,059 bp long, encodes a 353 amino acid protein with an estimated molecular mass of 39.416 kDa. The secondary structure of the OoMAPKK1 protein was analyzed by SOPMA and the 3D crystal structure of OoMAPKK1 protein was created by using Cn3D 4.1 software. The expression levels of OoMAPKK1 under different abiotic stresses (cold, wounding and abscisic acid) were investigated by semi-quantitative RT-PCR. Changes in the expression of OoMAPKK1 under cold stress, wounding and abscisic acid (ABA) application may suggest that OoMAPKK1 is involved in the cross-talk of multi-environmental stress responses.

    Abstract:

    We successfully used the guanidine isothiocyanate method for isolation of total RNA from leaf, stem, and root tissues of the aromatic plant Origanum Onites. The RNA was extracted with TRI Reagent at room temperature and was recovered by isopropanol precipitation. The isolated RNA was capable of reverse transcription. The extraction method described here does not require ultracentrifugation, and it is fast, simple, and effective. The procedure can be completed within 3 hours and may be applicable to other aromatic medicinal plants containing high amounts of phenolic compounds.

    Abstract:

    Origanum Onites is an economically important medicinal plant with high essential oil content. Lack of an appropriate DNA isolation procedure is a limiting factor for any molecular study of this plant. We have used a protocol for genomic DNA isolation based on a hexadecyltrimethylammonium bromide (CTAB) method described for other plant species. The method involves mortar grinding of leaf tissue, modified CTAB extraction using high salt concentrations and polyvinyl pyrrolidone, and successive isoamyl alcohol/chloroform extractions. The yield was approx. 20 microg DNA per 200 mg of initial fresh plant material. The genomic DNA obtained by this method was suitable to be used in restriction digests, inter simple sequence repeat (ISSR) and randomly amplified polymorphic DNA (RAPD) reactions. This extraction method should facilitate the molecular analysis of Origanum chemotypes.
