    Abstract:

    A new asteltoxin analog, named asteltoxin H (1), was isolated by the solid-state fermentation of the fungus Pochonia suchlasporia var. suchlasporia TAMA 87. The chemical structure of 1 was deduced by spectroscopic methods, including 1D and 2D NMR, HRESIMS, and UV-Vis analyses. Compound 1 showed insecticidal activity against prepupae of the blowfly, Lucilia sericata, with an LD50 value of 0.94 µg/mg prepupal body weight.

    A new polyhydroxylated pyrrolizidine alkaloid designated as pochonicine (1) was isolated from a solid fermentation culture of the fungal strain Pochonia suchlasporia var. suchlasporia TAMA 87. The structure of 1 was determined using NMR and MS techniques as (1R*, 3S*, 5S*, 6S*, 7R*, 7a S*)-5-acetamidomethyl-3-hydroxymethyl-1,6,7-trihydroxypyrrolizidine. Pochonicine (1) showed potent inhibition against beta-N-acetylglucosaminidases (GlcNAcases) of various organisms including insects, fungi, mammals, and a plant but no inhibition against beta-glucosidase of almond, alpha-glucosidase of yeast, or chitinase of Bacillus sp. The GlcNAcase inhibitory activity of pochonicine (1) was comparable to nagstatin, a potent GlcNAcase inhibitor of natural origin.

    Abstract Nematodiosis are responsible for financial losses in cattle production systems due to treatment costs, falling production rates and animal deaths. The incorporation of sodium alginate pellets containing nematophagous fungi in the bovine diet is a method for the control of nematodiosis. The aims of this study were to evaluate the viability of Arthrobotrys cladodes and Pochonia chlamydosporia contained in sodium alginate pellets after passage through the bovine gastrointestinal tract, as well as to evaluate the effects of single and combined use of these fungi against bovine parasitic nematodes. The reduction in parasitic nematode infective larvae of bovines by the combined use of Arthrobotrys cladodes and Pochonia chlamydosporia was more than the reduction in infective larvae by Arthrobotrys cladodes or Pochonia chlamydosporia alone in the collections 12, 24 and 36 h after giving the pellets to the animals. Arthrobotrys cladodes and Pochonia chlamydosporia combined promoted maximum reduction of infective larvae of 86.3%. Therefore, the combination of Arthrobotrys cladodes and Pochonia chlamydosporia may be an effective method to control gastrointestinal nematodiosis affecting grazing cattle.

    The objective of this study was to evaluate the infectivity of Toxocara canis eggs after interacting with isolated nematophagous fungi of the species Duddingtonia flagrans (AC001) and Pochonia chlamydosporia (VC4), and test the predatory activity of the isolated AC001 on T. canis second stage larvae after 7 days of interaction. In assay A, 5000 embryonated T. canis eggs previously in contact with the AC001 and VC4 isolated for 10 days were inoculated into domestic chickens ( Gallus gallus domesticus ), and then these animals were necropsied to collect material (digested liver, intestine, muscles and lungs) at 3-, 7-, 14-, and 21-day intervals after inoculation.     Abstract The aim of this work was to analyse the enzymatic activity and in vitro ovicidal effect of Pochonia chlamydosporia (VC1 and VC4) and Paecilomyces lilacinus (PL1) on Oxyuris equi eggs of horses. The growth of isolates and their enzymatic production were evaluated on agar media supplemented with gelatin (GA), casein (CA), olive oil (OOA) or starch (SSA). The ovicidal effect was evaluated according to morphological alterations. Following, the P. chlamydosporia crude extract production and proteolytic content was evaluated (VC1 and VC4) in liquid medium at the interval of 15, 30, 45 and 60 min during incubation of P. chlamydosporia and P. lilacinus which grew and showed enzymatic activity on agar media (GA, CA, OOA and SSA). At the 15th day, VC1, VC4 and PL1 showed results on GA, CA, OOA and SSA media, for Type 3 effect of (78, 54, 52 and 68%), (72, 50, 58 and 78%) and (62, 57, 65 and 63%).     Abstract In vitro effects of nematophagous fungi Duddingtonia flagrans (AC001), Monacrosporium thaumasium (NF34) and Pochonia chlamydosporia (VC1) were evaluated against eggs and third-stage infective larvae (L3) of horse cyathostomin (Nematoda: Strongylidae). The following percentage reductions compared with the control group were observed after a 20-day exposure period: AC001, 61.6%; NF34, 66.1%; VC1, 73.2%; group AC001 + VC1, 86.8%; NF34 + VC1, 77.3%; AC001 + NF34, 92.4%. The results showed that the fungal isolates (VC1, AC001 and NF34), acting alone or in conjunction, were efficient in controlling horse cyathostomin under in vitro conditions.

    Ascaris suum is a gastrointestinal nematode parasite of swines. The aim of this study was to observe Pochonia chlamydosporia fungus on biological control of A. suum eggs after fungus passage through swines gastrointestinal tract. Eighteen pigs, previously dewormed, were randomly divided into three groups: group 1, treated with the fungus isolate VC4; group 2, treated with the fungus isolate VC1 and group 3 did not receive fungus (control). In the treated groups, each animal received a 9 g single dose of mycelium mass containing P. chlamydosporia (VC1 or VC4). Thereafter, animal fecal samples were collected at the following intervals: 8, 12, 24, 36, 48, 72 and 96 h after treatment beginning and these were poured in Petri dishes containing 2% water-agar culture medium. Then, 1,000 A. suum eggs were poured into each dish and kept in an incubator at 26°C and in the dark for 30 days. After this period, approximately 100 eggs were removed from each Petri dish and morphologically analyzed under light microscopy following the ovicidal activity parameters. The higher percentage observed for isolated VC4 eggs destruction was 57.5% (36 h) after fungus administration and for isolate VC1 this percentage was 45.8% (24 h and 72 h) (p > 0.01). P. chlamydosporia remained viable after passing through the gastrointestinal tract of swines, maintaining its ability of destroying A. suum eggs.

    INTRODUCTION: Ancylostoma sp is a potentially zoonotic geohelminth. METHODS: This study aimed to evaluate in vitro the action of crude enzyme extract of Pochonia chlamydosporia (VC4) on eggs of Ancylostoma sp in 2% water-agar and in fecal cultures. RESULTS: The percentage reduction in Ancylostoma sp egg eclosion was 76.8% in Petri dishes of the treated group compared to the control group. CONCLUSIONS: The crude enzyme extract of Pochonia chlamydosporia was effective at reducing Ancylostoma sp egg eclosion and can be used as biological control of this nematode.
