    Abstract:

    Abstract The aim of this study was to evaluate simultaneously a battery of biomarkers in Prochilodus lineatus fish in order to obtain a holistic overview of silver nanoparticles (AgNP) toxicity. Juveniles were exposed to 2.5 and 25.0 µgAgNP L−1 for 5 and 15 days. We analyzed silver accumulation in tissues (liver, intestine and brain), morphometric and hematological parameters, oxidative stress markers (antioxidant enzymes, lipid peroxidation, and antioxidant capacity against peroxyl radicals-ACAP-), and metabolic responses in plasma (glucose, proteins, triglycerides and cholesterol), liver and muscle (glycogen, proteins, and lipids). Additionally, after 15-day exposure, the number of colony forming units (CFU) of bacteria living at the skin mucus secretion was analyzed to corroborate the AgNP biocide properties. Tissue Ag accumulation was concentration- and time-dependent: liver > intestine > brain. The hepatosomatic index increased at both periods of exposure at 25.0 µgAgNP L−1. Hematological and plasma biochemical parameters increased after 5 days of exposure to the highest concentration. When fish were exposed for 15 days to 25.0 µgAgNP L−1, many changes were observed: glycogen content increased in liver and muscle, muscle protein concentration decreased, all antioxidant enzymes activities were enhanced and ACAP decreased in liver. The CFU decreased in the mucus of fish exposed to both AgNP treatments when compared to the control group. The multivariate analysis revealed a clear difference in physiological profiles of individuals exposed to the different treatments. Our results showed that AgNP in short and subchronic exposures threaten the health of fish.

    This study evaluated the histopathological changes in gills and liver of Prochilodus lineatus inhabiting the Salado River basin. Fish were collected in four different sampling stations. The histological lesions in the tissues were examined under light microscopy and evaluated with quantitative analyses. The morphometric analysis of the gills showed a significant shortening of secondary lamellae and a lower percentage of area for gas exchange in fish from station 1 (an urban area, located near the mouth of the Salado River) in comparison with fish gills from the reference site (station 4, a relatively pristine area). Moreover, a significantly higher area occupied with necrotic foci and the occurrence of an important inflammatory response were observed in fish liver of station 1 than the samples caught from other stations. Thus, histopathological evidences showed differences among sites, which could be related to different environmental conditions.

    This field study assessed water quality of Salado River basin by using a set of biomarkers in the fish Prochilodus lineatus. Multiple biomarkers were measured, including morphological indexes (condition factor, liver somatic index), hematological (red and white blood cells) and biochemical (glucose, total protein and cholinesterase activity) parameters. Besides, detoxication and oxidative stress markers (antioxidant enzymes, lipid peroxidation) were measured in liver, gills and kidney. Despite water quality assessment did not show marked differences among sites, biomarkers responses indicate that fish are living under stressful environmental conditions. According to multivariate analysis glucose, glutathione S-transferase activity, lipid peroxidation levels and the count of white blood cells are key biomarkers to contribute to discrimination of sites. So, we suggest use those biomarkers in future monitoring of freshwater aquatic systems.

    Abstract:

    Evidence that migration prevents population structure among Neotropical characiform fishes has been reported recently but the effects upon species diversification remain unclear. Migratory species of Prochilodus have complex species boundaries and intrincate taxonomy representing a good model to address such questions. Here, we analyzed 147 specimens through barcode sequences covering all species of Prochilodus across a broad geographic area of South America. Species delimitation and population genetic methods revealed very little genetic divergence among mitochondrial lineages suggesting that extensive gene flow resulted likely from the highly migratory behavior, natural hybridization or recent radiation prevent accumulation of genetic disparity among lineages. Our results clearly delimit eight genetic lineages in which four of them contain a single species and four contain more than one morphologically problematic taxon including a trans-Andean species pair and species of the P. nigricans group. Information about biogeographic distribution of haplotypes presented here might contribute to further research on the population genetics and taxonomy of Prochilodus.

    : Within the genus Prochilodus Agassiz, 1829, five species are known to carry B chromosomes, i.e. chromosomes beyond the usual diploid number that have been traditionally considered as accessory for the genome. Chromosome microdissection and mapping of repetitive DNA sequences are effective tools to assess the DNA content and allow a better understanding about the origin and composition of these elements in an array of species. In this study, a novel characterization of B chromosomes in Prochilodus costatus Valenciennes, 1850 (2n=54) was reported for the first time and their sequence complementarity with the supernumerary chromosomes observed in Prochilodus lineatus (Valenciennes, 1836) and Prochilodus argenteus Agassiz, 1829 was investigated. The hybridization patterns obtained with chromosome painting using the micro B probe of P. costatus and the satDNA SATH1 mapping made it possible to assume homology of sequences between the B chromosomes of these congeneric species. Our results suggest that the origin of B chromosomes in the genus Prochilodus is a phylogenetically old event.

    The genus Prochilodus includes individuals ranging in size from medium to large, being highly relevant for commercial and subsistence fishing. Prochilodus species have a diploid number of 2 n  = 54 chromosomes and up to seven supernumerary (B) microchromosomes. Previous research has shown that B frequency increased drastically in the Mogi-Guaçu river population of Prochilodus lineatus in the early 1980s, whereas it remained about constant in the 1990s. Here we analyses B frequency in this population during the 2003–2007 period and have found that frequency has not changed significantly since 1987, and that these B chromosomes do not show the intra-individual variation in number that characterized them in the 1980s. This indicates that these B chromosomes have been neutralized, after their invasion, through their mitotic stabilization.

    Within the genus Prochilodus, the species Prochilodus lineatus is certainly the most studied one from a cytogenetic point of view. In this study, a cytogenetic characterization of specimens of P. lineatus from Mogi-Guacu River was performed in the period from 2003 to 2007, through utilization of cytogenetic markers, such as Giemsa, Ag-NOR, C-banding and cytogenetical-molecular markers (FISH) to detect both 5S and 18S ribosomal genes. All analyzed individuals presented 2n=54 meta/submetacentric chromosomes, besides bearing up to 7 supernumerary microchromosomes. Polymorphic NORs were detected on a single chromosomal pair. The constitutive heterochromatin was distributed at centromeric region of all chromosomes in the A complement, while the microchromosomes were totally heterochromatic. A syntenic distribution of 5S and 18S ribosomal genes was detected, comprising the NOR-bearing chromosomal pair. No additional ribosomal clusters in other chromosomes were observed. Although the karyotype features are commonly conserved within the genus Prochilodus, the few differences on the distribution of both ribosomal genes and heterochromatin when compared to specimens of P. lineatus from other localities can be associated to the evolutionary changes that these repetitive sequences have undergone through the years.

    Abstract:

    Summary Meiotic analysis performed on a sample of 10 specimens of Prochilodus lineatus revealed continuous filaments of different sizes stained by AgNO3, corresponding to the bivalent of the normal complement. Small supernumerary chromosomes were observed as isolated and well stained bodies, scattered among the other elements. Synaptonemal complex studies have shown that the beginning of chromosome pairing process in P. lineatus usually occurs from the telomeres to the pericentromeric region. At the end of the pachytene 27 bivalents are perfectly paired and the small supernumerary chromosomes of this species are seen as bivalents, trivalents, or tetravalents. The central region of these small chromosomes show a trick staining when they formed bivalents or tetravalents. This portion seems to correspond to the pericentromeric region of the regular chromosomes with the heterochromatic characteristic of B chromosomes.

    The population of Prochilodus lineatus found in the Mogi-Guacu River is karyotypically polymorphic, carrying acrocentric, metacentric, and submetacentric B chromosomes. The analysis of each B chromosome frequency in this species revealed a variation in the distribution pattern, with the metacentric type having the highest frequency (73.30%), followed by submetacentric (25.22%) and acrocentric B chromosomes (1.48%). The transmission pattern of the supernumerary chromosomes was identified by controlled crosses, and it was shown that the acro- and submetacentric B chromosomes have a transmission pattern below the Mendelian rate (kB = 0.333 and kB = 0.385, respectively), but the metacentric variant has a cumulative transmission pattern (kB = 0.587). These results indicate that the acro- and submetacentric B chromosomes are undergoing an extinction process, while the metacentric B chromosomes appear to be accumulating in frequency with each generation.

    Abstract:

    B chromosomes have attracted the attention of Neotropical fish cytogeneticists in recent years, both for their remarkable occurrence in this group and also because of the interest in studies of the genetic structure and role played in the genome of these organisms. The aim of this study was to report the first occurrence of supernumerary chromosomes in Prochilodus argenteus (Agassiz, 1829), this being the fifth carrier species among thirteen within the genus Prochilodus (Agassiz, 1829). The extra elements identified in this species are small sized heterochromatic chromosomes characterized by a low mitotic instability index, being very similar to other supernumerary chromosomes described in the species of the genus Prochilodus. Morphology, structure and dispersion of the supernumerary genomic elements which occur in species of this genus are discussed aiming to better understand aspects involved the origin of supernumerary chromosomes and the differentiation process and relationships among species of this family.
