  • the complete sequence and gene organization of the mitochondrial genome of the gadilid Scaphopod siphonondentalium lobatum mollusca
    Molecular Phylogenetics and Evolution, 2004
    Co-Authors: Hermann Dreyer, Gerhard Steiner
    Abstract:

Comparisons of mitochondrial gene sequences and gene arrangements can be informative for reconstructing high-level phylogenetic relationships. We determined the complete sequence of the mitochondrial genome of Siphonodentalium lobatum, (Mollusca, Scaphopoda). With only 13,932 bases, it is the shortest molluscan mitochondrial genome reported so far. The genome contains the usual 13 protein-coding genes, two rRNA and 22 tRNA genes. The ATPase subunit 8 gene is exceptionally short. Several transfer RNAs show truncated TψC arms or DHU arms. The gene arrangement of S. lobatum is markedly different from all other known molluscan mitochondrial genomes and shows low similarity even to an unpublished gene order of a dentaliid Scaphopod. Phylogenetic analyses of all available complete molluscan mitochondrial genomes based on amino acid sequences of 11 protein-coding genes yield trees with low support for the basal branches. None of the traditionally accepted molluscan taxa and phylogenies are recovered in all analyses, except for the euthyneuran Gastropoda. S. lobatum appears as the sister taxon to two of the three bivalve species. We conclude that the deep molluscan phylogeny is probably beyond the resolution of mitochondrial protein sequences. Moreover, assessing the phylogenetic signal in gene order data requires a much larger taxon sample than is currently available, given the exceptional diversity of this character set in the Mollusca.

  • molecular phylogeny of Scaphopoda mollusca inferred from 18s rdna sequences support for a Scaphopoda cephalopoda clade
    Zoologica Scripta, 2003
    Co-Authors: Gerhard Steiner, Hermann Dreyer
    Abstract:

The phylogenetic relationships of the Scaphopoda, one of the ‘lesser’ molluscan classes, with the other conchiferan taxa are far from clear. They appear either as the sister-group to the Bivalvia (Diasoma concept) or to a Gastropoda–Cephalopoda clade or to the Cephalopoda alone (helcionellid concept). We compiled a 18S rDNA sequence dataset of 48 molluscan species containing 17 Scaphopods to test these hypotheses and to address questions regarding high-level relationships with the Scaphopoda. Both parsimony and maximum likelihood trees show low branch support at the base of the Conchifera, except for the robust clade uniting Scaphopoda and Cephalopoda. This result is corroborated by spectral analysis and likelihood mapping. We also tested alternative topologies which scored significantly worse both in tree length and in likelihood. The 18S rDNA data thus reject the Diasoma in favour of a Scaphopoda–Cephalopoda clade as proposed in the helcionellid concept. When plotted on the molecular tree, the pivotal morphological characters associated with the burrowing life style of the Bivalvia and Scaphopoda, i.e. mantle/shell enclosure of the body and the burrowing foot with true pedal ganglia, appear convergent in these groups. In contrast, the prominent and tilted dorsoventral body axes, multiple cephalic tentacles and a ring-shaped muscle attachment on the shell are potential synapomorphies of Scaphopoda and Cephalopoda. Most of the higher taxa within the Scaphopoda are supported by the molecular data. However, there is no support for the families Dentaliidae and Gadilidae. The basal position of the Fustiariidae within the Dentaliida is confirmed.

  • Hypervariability of Ascidian Mitochondrial Gene Order: Exposing the Myth of Deuterostome Organelle Genome Stability
    Molecular biology and evolution, 2009
Co-Authors: Carmela Gissi, Graziano Pesole, Fabio Iannelli, Francesco Mastrototaro, Vanessa Guida, Francesca Griggio
    Abstract:

The few sequenced mitochondrial (mt) genomes of the class Ascidiacea (Chordata, Tunicata), mostly belonging to congeneric species of the Phlebobranchia order, show extraordinary gene order rearrangements. In order to assess if this hypervariability in gene order is a general feature of Ascidiacea, we report here the gene arrangement of five ascidians belonging to the Aplousobranchia and Stolidobranchia orders. Our data show that Ascidiacea are characterized by: 1) extensive gene order rearrangements both within and between the three major lineages; 2) lack of significant similarities to the gene order of other deuterostomes; and 3) an extent of rearrangements comparable with that of Mollusca (especially the Gastropoda, Bivalvia, and Scaphopoda classes), a phylum with highly rearranged mtDNAs. The only conserved feature is the location of all genes on the same strand, which suggests that selective constraints are related to the mt transcription. Finally, a higher mobility of the tRNA genes is undetectable because of saturation effect, and only the partially conserved cox2-cob gene block seems to retain some phylogenetic signals.
