Kajian Fitokimia Fraksi Etil Asetat dari Lichen Stereocaulon massartianum Hue. dan Uji Aktivitas Antibakteri dengan Metode KLT-Bioautografi

Reports on phytochemical studies and pharmacological activities of this species are still limited, based on literature studies that have been carried out. This research was conducted to determine their secondary metabolites and potential antibacterial activity. The air-dried thallus of lichen S. massartianum was macerated successively using n-hexane, ethyl acetate and methanol solvents. The compounds were separated by chromatography and recrystallization methods, then analyzed by spectroscopy (UV-Vis, FTIR, 1H and 13C-NMR). Furthermore, the antibacterial activity assay was performed by agar diffusion method on ethyl acetate extract, and TLC-Bioautography for the isolated compound. Three compounds have been isolated from ethyl acetate extract, i.e. atranorin (1), stictic acid (2) and norstictic acid (3). The results of antibacterial assay from the extract showed antagonistic activity against pathogenic bacteria S. aureus, E. faecalis, E. coli, and P. aeruginosa at concentrations of 10 and 20%, while TLC-bioautography of compound 3 exhibited growth inhibition area in all test bacteria.Penelitian ini merupakan kegiatan lanjutan dalam inventori lichen/lumut kerak genus Stereocaulon dengan fokus pada Stereocaulon massartianum Hue yang dikoleksi di daerah bebatuan Danau Diatas, Sumatera Barat, Indonesia. Berdasarkan penelusuran literatur, kajian fitokimia, dan aktivitas farmakologis dari spesies ini masih sedikit. Penelitian ini dilakukan untuk mengetahui kandungan metabolit sekunder dan potensi aktivitas antibakterinya. Thallus kering lichen S. massartianum dimaserasi bertingkat dengan pelarut n-heksan, etil asetat dan metanol. Kemudian pemisahan senyawa dengan metode kromatografi dan rekristalisasi. Senyawa-senyawa hasil isolasi dianalisis secara spektroskopi (UV-Vis, FTIR, 1H dan 13C-NMR). Uji aktivitas antibakteri dilakukan dengan metode difusi agar terhadap ekstrak etil asetat, dan KLT-Bioautografi untuk senyawa hasil isolasi. Tiga senyawa berhasil diisolasi dari ekstrak etil asetat, yaitu atranorin (1), asam stiktat (2) dan asam norstiktat (3). Hasil uji antibakteri dari ekstrak tersebut menunjukkan aktivitas antagonis terhadap bakteri patogen S. aureus, E. faecalis, E. coli, dan P. aeruginosa pada konsentrasi 10 dan 20 %, dan KLT-bioautografi dari senyawa 3 memperlihatkan daerah hambat pertumbuhan pada semua bakteri uji

These isolated compounds have been tested for their cytotoxic activities against human breast adenocarcinoma cancer cell (MCF-7) and human oral squamous carcinoma cancer cell (HSC-3) using MTT assay with a slight modification. Some methanolic extracts of the leaves of Elephantopus scaber, Piper betle, Andrographis paniculata, rhizomes of Boesenbergia pandurata, Alpinia galanga, Kaempferia galanga and isolated compounds; ethyl haematommate (2), olivatolic acid (3), deoxyelephantopin (5), andrographolide (6), hydroxychavicol (7), eugenol (8), curcumin (9), pinostrobin (10), pinocembrin (11), alpinetin (12), α-mangostin (13), angiopteroside (14), and stevioside (15) from Sumatran traditional medicinal plants have also been tested

However, the scientific study of this tricky genus is still limited, making it a challenge to study the group further. Detailed morphological studies are essential to discriminate closely shaped species which is illustrated through personal data focused on phyllocladia, apothecia and spores of nine species. Secondary metabolites isolated from Stereocaulon species are mostly some depsides, depsidones, diphenylethers and dibenzofurans which can have a taxonomic value. The use of Stereocaulon lichens as a traditional medicine in several regions of the world and pharmacological studies of extracts and isolated compounds have been compiled. The complete 1H and 13C NMR spectral assignments of stictic acid derivatives are reported with some revisions. Five depsidones belonging to the stictic acid chemosyndrome were superoxide anion scavengers as potent as ascorbic acid and with no toxicity on two human cell lines

The structure of stereocalpin A (1) was mainly determined by analysis of the NMR spectroscopic data and by chemical methods. Stereocalpin A (1) is a unique cyclic peptide incorporating an unprecedented 5-hydroxy-2,4-dimethyl-3-oxo-octanoic acid in the structure. Stereocalpin A (1) shows moderate cytotoxicity against three human solid tumor cell lines.

The structures of 1–3 were determined by analysis of their spectroscopic data (NMR, UV, MS) and by chemical methods. The known compound usnic acid (4) was also obtained. Compounds 1–4 showed moderate inhibitory activity against therapeutically targeted protein tyrosine phosphatase 1B (PTP1B).
