    Abstract:

    The Antidiarrhoeal activity of the seed extract of Albizzia lebbeck (Benth.) was investigated employing conventional rodent models of diarrhoea, i.e. castor oil-induced diarrhoea, upper gastrointestinal transit (u.g.t.) and fluid secretion. It was found that the aqueous methanol extract of Albizzia lebbeck seeds (2.5-5 mg/kg i.p.) possessed Antidiarrhoeal activity which strengthens the earlier use of the seeds in the treatment of diarrhoea and dysentery. The Antidiarrhoeal dose of the extract was at least 10-30 times less than the LD(50) dose. The extract (2.5-5 mg/kg i.p.) potentiated the Antidiarrhoeal activity of loperamide (1 mg/kg i.p.). Nalaxone (0.5 mg/kg i.p.) significantly inhibited the Antidiarrhoeal activity of the extract as well as loperamide, thus indicating a role of the opioid system in the Antidiarrhoeal activity of the extract.

    Background The root-bark of the medicinally important plant Zanthoxylum budrunga (ZBRB) brooks a variety of uses in ethnobotanical and ethnomedicinal practice in Bangladesh and thus demands biological investigation to reveal its therapeutic potentiality. So, the present study was perpetrated to explore antioxidant, analgesic, Antidiarrhoeal, and a cytotoxic activity of ethanolic root-bark extract of Zanthoxylum budrunga and was also to quantify the major bioactive polyphenolic constituents by HPLC analysis. Methods Total phenolic content was measured spectrophotometrically by using Folin Chiocalteu’s reagent while in vitro antioxidant activity was determined by means of 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay and reducing power assay. HPLC analysis was performed to identify and quantify the major bioactive polyphenolic constituents present in the extract. Acetic acid induced writhing test and hot-plate test were conducted to evaluate the analgesic activity of the crude extract. On the other hand, in vivo antidiarrheal potentiality was investigated by using experimentally castor oil induced diarrhoea in mice and brine shrimp lethality bio-assay was implemented to check the cytotoxic potentiality of the crude extract. Results Zanthoxylum budrunga showed DPPH scavenging activity (IC_50 54.27 μg/mL), while the total phenolic content was 647.91 mg GAE/100 g of extract.     The different fractions of crude ethanol extract of leaf and stem of S. caseolaris (Linn.) (Sonneratiaceae) were screened for its analgesic and Antidiarrhoeal activities. The different fraction of crude extract was obtained by using four different solvent systems. The different fractions of crude extract produced significant writhing inhibition in acetic acid induced writhing in mice at dose of 250 and 500mg/kg BW comparable to the standard drug diclofenac sodium at the dose of 25mg/kg BW. When tested for its Antidiarrhoeal effects on castor oil induced diarrhea in mice, it increased mean latent period and decreased the frequency of defecation significantly at the dose of 250 and 500mg/kg BW comparable to the standard drug loperamide at the dose of 50mg/kg BW. The overall results tend to suggest the analgesic and Antidiarrhoeal activities of the different fractions of crude extract. Both ethyl acetate fraction of stem and chloroform fraction of leaf have significant analgesic activity. Again between the two fractions of crude ethanol extract ethyl acetate fraction of S.caseolaris stem have most significant Antidiarrhoeal activity.     The methanol extract of Xylocarpus granatum bark was studied for its Antidiarrhoeal properties in experimental diarrhoea, induced by castor oil and magnesium sulphate in mice. At the doses of 250 and 500 mg/kg per oral, the methanol extract showed significant and dose-dependent Antidiarrhoeal activity in both models. The extracts also significantly reduced the intestinal transit in charcoal meal test when compared to atropine sulphate (5 mg/kg; i.m.). The results showed that the extracts of Xylocarpus granatum bark have a significant Antidiarrhoeal activity and supports its traditional uses in herbal medicine.

    The methanol extract of Cyperus rotundus rhizome, given orally at the doses of 250 and 500 mg/kg b.w., showed significant Antidiarrhoeal activity in castor oil induced diarrhoea in mice. Among the fractions, tested at 250 mg/kg, the petroleum ether fraction (PEF) and residual methanol fraction (RMF) were found to retain the activity, the latter being more active as compared to the control. The ethyl acetate fraction (EAF) did not show any Antidiarrhoeal activity.

    Abstract The methanol (MeOH) extract of the barks of Xylocarpus moluccensis , and different fractions of this extract were studied for Antidiarrhoeal activity using castor oil- and magnesium sulphate-induced diarrhoea models in mice. At the doses of 250 and 500 mg/kg, the MeOH extract showed significant Antidiarrhoeal activity in both models. The EtOAc fraction (EAF) and the residual MeOH fraction (RMF) exhibited similar activity using a dose of 250 mg/kg in both models. No Antidiarrhoeal activity was observed with the chloroform fraction (CHF) at the test doses. When tested for antibacterial effect, the MeOH extract displayed moderate inhibitory activity against Escherichia coli , Vibrio cholera , Staphylococcus aureus , Staphylococcus epidermis , Shigella dysentery , Staphylococcus pyogenes , Salmonella typhi , Pseudomonas aeruginosa and Enterobacter aerogenes . While the CHF inhibited the growth of Escherichia coli , Vibrio cholerae , Shigella dysenteriae , Shigella sonnei , Staphylococcus epidermis , Staphylococcus pyogenes and Pseudomonas aeruginosa , the EAF was active against all test organisms except Vibrio cholera and Staphylococcus epidermis . The RMF inhibited the growth of all the test organisms with moderate zone of inhibition. On the basis of these findings, it can be assumed that Xylocarpus moluccensis could be a potential source for novel ‘lead’ discovery for Antidiarrhoeal drug development.

  • Efficacy of Aegle marmelos L. Corr. (Rutaceae) in Ayurvedic Antidiarrhoeal formulation

    Aegle marmelos L. Corr. (Rutaceae) is an active ingredient of most of the ayurvedic Antidiarrhoeal formulations. A study was undertaken to evaluate the effectiveness of Aegle marmelos in polyherbal Antidiarrhoeal formulation by comparing its Antidiarrhoeal and antispasmodic effect with Mebarid; an Antidiarrhoeal Ayurvedic formulation. An aqueous extract of Aegle marmelos (AAM) was tested for its Antidiarrhoeal and antispasmodic activities. Antidiarrhoeal effect of AAM was evaluated in castor oil induced diarrhea and intestinal secretion in mice and antispasmodic effect was evaluated by charcoal meal test in mice at a dose of 25, 50, 100 mg/kg. Antidiarrhoeal and antispasmodic effect of Mebarid was evaluated at the dose of 10 ml/kg. The % inhibition of diarrhoea was 31.81 %, 43.93 %, 54.54 % and 92.45 % for AAM 25, 50, 100 mg/kg (po) and Mebarid 10 ml/kg dose (po) respectively. The % inhibition of intestinal secretion was 29.30 %, 34.05 %, 46.73 %% and 81.58 % for AAM 25, 50, 100 mg/kg (po) and Mebarid 10 ml/kg dose (po) respectively. AAM 25, 50, 100 mg/kg (po) and Mebarid 10 ml/kg dose (po) produced 10.95 %, 21.65 %, 29.19 % and 47.15 % inhibition of intestinal transit respectively. These results suggest that AAM is an active ingredient of Mebarid in treating diarrhea by its antisecretory and antimotility effect.

    Antidiarrhoeal effect of Kutajarishta, an ayurvedic formulation was evaluated with different agonist and antagonist in castor oil induced diarrhoea in mice. Antidiarrhoeal effect of Kutajarishta was decreased by Isosorbide dinitrite (nitric oxide donor) and Glibenclamide (potassium channel blocker). Yohimbine (α2 adrenergic receptor blocker) did not produce any significant effect on the Antidiarrhoeal activity of Kutajarishta. The results obtained suggest that Antidiarrhoeal effect of Kutajarishta was through potassium channels and nitric oxide pathway and not through α2 adrenergic receptors.

    In Maharashtra (India) Myristica fragrans (Myristic aceae) is used as Antidiarrhoeal and antispasmodic in new born infants and children traditionally. Aqueous Myristi ca fragrans extract (MF) was tested for its antidia rrhoeal effect in castor oil induced diarrhoea in mice and antispa smodic effect on contractions induced by acetyl cho line, nicotine and histamine in isolated guinea pig ileum. MF prod uced a significant and dose dependent antidiarrhoea l effect and also inhibited the acetyl choline, nicotine and his tamine induced contractions of isolated guinea pig ileum. It can be concluded that MF possesses Antidiarrhoeal and anti spasmodic activity may be due to its anticholinergi c and antihistaminic effect. Thus the study provided a sc ientific basis for the use of Myristica fragrans as an ingredient of traditional polyherbal formulations in the treatmen t of diarrhoea.

    Piperine was extracted and isolated from Piper nigrum. Antidiarrhoeal effect of piperine and aqueous extract of Piper nigrum was examined in castor oil and magnesium sulphate induced diarrhoea in mice. Piperine has shown significant inhibition of diarrhoea when compared with Antidiarrhoeal effect of aqueous extract of Piper nigrum. The results indicate that piperine is the active Antidiarrhoeal constituent of Piper nigrum.

  • Antispasmodic effect of Cyperus Rotundus L. (Cyperaceae) in diarrhoea

    Cyperus rotundus L. (Cyperaceae) is used as an active ingredient of most of the ayurvedic Antidiarrhoeal formulations. A study was undertaken to evaluate the effectiveness of Cyperus rotundus as a antispasmodic in diarrhoea. An aqueous extract of tubers of Cyperus rotundus (ACR) was tested for its Antidiarrhoeal and antispasmodic activity. Antidiarrhoeal effect of ACR was evaluated in castor oil induced diarrhea in mice and antispasmodic effect was evaluated by charcoal meal test in mice at a dose of 125, 250, 500 mg/kg. The % inhibition of diarrhoea was 30.36 %, 37.90 %, 45.45 % and 92.45 % for ACR 125, 250, 500 mg/kg (po) and Loperamide 2 mg/kg dose (po) respectively. ACR 125, 250, 500 mg/kg (po) and Atropine Sulphate 2 mg/kg dose (po) produced 24.35 %, 31.48 %, 36.75 % and 55.94 % inhibition of intestinal transit respectively. These results indicate that ACR produces its Antidiarrhoeal effect through decreasing intestinal secretions and antispasmodic effect by inhibiting the intestinal motility.
