Mother tinctures (85% ethanol extracts) of T. aphrodisiaca have also been used for the treatment of central nervous system disorders. In the present investigation, T. aphrodisiaca mother tinctures formulated by three reputed manufacturers of homoeopathic medicines (NLK, DWSG and SBL) were evaluated for their Anxiolytic Activity. Dried mother tinctures of T. aphrodisiaca were subjected to Anxiolytic Activity evaluation at various doses, i.e. 50, 75, 100, 125 or 150 mg/kg p.o. in mice using elevated plus maze apparatus. Dried mother tinctures exhibited significant Anxiolytic Activity at 50 mg/kg (NLK), 75 mg/kg (DWSG) and 125 mg/kg (SBL), respectively, with reference to control as well as standard (diazepam, 2 mg/kg p.o.). Mother tinctures of T. aphrodisiaca available in the market, have significant Anxiolytic Activity. Amongst the three mother tinctures of T. aphrodisiaca analyzed, the dry residue of NLK possesses the highest amount of Anxiolytic constituent(s). To ensure uniformity and consistency of biological effects in herbal formulations, these should be standardized on the basis of bioactive markers. The authors are actively involved in isolating the bioactive constituent(s) from T. aphrodisiaca so that the plant can be standardized on the basis of biologically active constituent(s).

In the present investigation, petroleum ether (60–80°C), chloroform, methanol, and water extracts of T. aphrodisiacaaerial parts were evaluated for anti-anxiety Activity in mice using elevated plus-maze apparatus. Among all the extracts, only methanol exhibited significant anti-anxiety Activity at a dose of 25 mg/kg with respect to control as well as standard (diazepam, 2 mg/kg). The bioactive methanol extract was shaken with petroleum ether, chloroform, and n-butyl alcohol, and all the shakings as well as the remaining methanol extract (RME) were further evaluated for Anxiolytic Activity. Butanol fraction and RME were found to exhibit Anxiolytic Activity in mice at the dose of 10 mg/kg and 75 mg/kg, respectively.

Materials and Methods: The extracts of P. incarnata were further subdivided in four doses, i.e., 100, 200, 300, and 400 mg/kg, suspended in a vehicle, and administered orally to groups of mice. Methanol extract of P. incarnata (125 mg/kg, orally) was taken as a standard. Anxiolytic Activity was measured using the elevated plus-maze model. All treatments were given orally. Forty-five (45) minutes after the treatments, mice were placed on the center of the elevated plus-maze and the number of entries in open arms were measured for 5 minutes. Anxiolytic Activity of aerial and underground parts of Passiflora incarnata.

The methanol extracts of leaves, stems, flowers, and whole plant exhibited Anxiolytic effects at 100, 125, 200 and 300 mg/kg, respectively. The roots were practically devoid of Anxiolytic effects. These results show that roots and flowers of P. incarnata act as natural adulterants by causing a significant increase in the Anxiolytic dose. Therefore, separation of these parts is recommended prior to any pharmacological, phytochemical and standardization studies on P. incarnata.

Plant-Based Medicines for Anxiety Disorders, Part 2: A Review of Clinical Studies with Supporting Preclinical Evidence

CamfieldAbstract:Research in the area of herbal psychopharmacology has revealed a variety of promising medicines that may provide benefit in the treatment of general anxiety and specific anxiety disorders. However, a comprehensive review of plant-based Anxiolytics has been absent to date. Thus, our aim was to provide a comprehensive narrative review of plant-based medicines that have clinical and/or preclinical evidence of Anxiolytic Activity. We present the article in two parts. In part one, we reviewed herbal medicines for which only preclinical investigations for Anxiolytic Activity have been performed. In this current article (part two), we review herbal medicines for which there have been both preclinical and clinical investigations of Anxiolytic Activity. A search of MEDLINE (PubMed), CINAHL, Scopus and the Cochrane Library databases was conducted (up to 28 October 2012) for English language papers using the search terms ‘anxiety’ OR ‘anxiety disorder’ OR ‘generalized anxiety disorder’ OR ‘social phobia’ OR ‘post-traumatic stress disorder’ OR ‘panic disorder’ OR ‘agoraphobia’ OR ‘obsessive compulsive disorder’ in combination with the search terms ‘Herb*’ OR ‘Medicinal Plants’ OR ‘Botanical Medicine’ OR ‘Chinese herb*’, in addition to individual herbal medicines. This search of the literature revealed 1,525 papers, of which 53 plants were included in the review (having at least one study using the whole plant extract). Of these plants, 21 had human clinical trial evidence (reviewed here in part two), with the other 32 having solely preclinical evidence (reviewed in part one). Support for efficacy was found for chronic use (i.e. greater than one day) of the following herbs in treating a range of anxiety disorders in human clinical trials: Piper methysticum, Matricaria recutita, Ginkgo biloba, Scutellaria lateriflora , Silybum marianum, Passiflora incarnata, Withania somniferum, Galphimia glauca, Centella asiatica, Rhodiola rosea, Echinacea spp., Melissa officinalis and Echium amoenum . For several of the plants studied, conclusions need to be tempered due to methodological issues such as small sample sizes, brief intervention durations and non-replication. Current evidence does not support Hypericum perforatum or Valeriana spp. for any anxiety disorder. Acute Anxiolytic Activity was found for Centella asiatica, Salvia spp., Melissa officinalis, Passiflora incarnata and Citrus aurantium. Bacopa monnieri has shown Anxiolytic effects in people with cognitive decline. The therapeutic application of psychotropic plant-based treatments for anxiety disorders is also discussed, specifically Psychotria viridis and Banisteriopsis caarti (ayahuasca), Psilocybe spp. and cannabidiol-enriched (low tetrahydrocannabinol (Δ^9-THC)) Cannabis spp.

Plant-based medicines for anxiety disorders, part 1: A review of preclinical studies

An open-ended, language-restricted (English) search of MEDLINE (PubMed), CINAHL, Scopus and the Cochrane Library databases was conducted (up to 28 October 2012) using specific search criteria to identify herbal medicines that have been investigated for Anxiolytic Activity. This search of the literature revealed 1,525 papers, from which 53 herbal medicines were included in the full review (having at least one study using the whole plant extract). Of these plants, 21 had human clinical trial evidence (reviewed in part 2), with another 32 having solely preclinical studies (reviewed here in part 1). Preclinical evidence of Anxiolytic Activity (without human clinical trials) was found for Albizia julibrissin, Sonchus oleraceus, Uncaria rhynchophylla, Stachys lavandulifolia, Cecropia glazioui, Magnolia spp., Eschscholzia californica, Erythrina spp., Annona spp., Rubus brasiliensis, Apocynum venetum, Nauclea latifolia, Equisetum arvense, Tilia spp., Securidaca longepedunculata, Achillea millefolium, Leea indica, Juncus effusus, Coriandrum sativum, Eurycoma longifolia, Turnera diffusa, Euphorbia hirta, Justicia spp., Crocus sativus, Aloysia polystachya, Albies pindrow, Casimiroa edulis, Davilla rugosa, Gastrodia elata, Sphaerathus indicus, Zizyphus jujuba and Panax ginseng. Common mechanisms of action for the majority of botanicals reviewed primarily involve GABA, either via direct receptor binding or ionic channel or cell membrane modulation; GABA transaminase or glutamic acid decarboxylase inhibition; a range of monoaminergic effects; and potential cannabinoid receptor modulation. Future research should focus on conducting human clinical trials on the plants reviewed with promising Anxiolytic Activity.

We now describe a specific binding site on GABA(A) receptors with nM affinity for valerenic acid and valerenol, common constituents of valerian. Both agents enhanced the response to GABA at multiple types of recombinant GABA(A) receptors. A point mutation in the beta2 or beta3 subunit (N265M) of recombinant receptors strongly reduced the drug response. In vivo, valerenic acid and valerenol exerted Anxiolytic Activity with high potencies in the elevated plus maze and the light/dark choice test in wild type mice. In beta3 (N265M) point-mutated mice the Anxiolytic Activity of valerenic acid was absent. Thus, neurons expressing beta3 containing GABA(A) receptors are a major cellular substrate for the Anxiolytic action of valerian extracts.
