    Abstract:

    Covariation of individual responses to different fear-eliciting situations is expected to manifest in general “fearfulness syndrome.” We tested for the existence of such a syndrome in wild-caught captive greenfinches. We assessed the propensity to give distress calls at handling, latency to feed at the presence of a predator image, and changes in locomotor activity in response to distress calls of conspecifics. Additionally, we measured the frequency of flapping flight movements against cage bars and tendency to damage tail feathers in captivity as indicators of the ability of birds to cope with captive conditions. As a proxy of neuroendocrine activity (an expected covariate of fearfulness), we measured the amount of stress hormone corticosterone deposited into feathers grown during captivity. All the behavioral traits were individually repeatable in time, but there were no correlations between them. Lack of the behavioral syndrome for fearfulness was also revealed by structural equation modelling. The findings of this experiment challenge the concept of a single internal variable responsible for fearfulness and support the proposed multidimensional nature of fear responses.

    Abstract:

    Assessment of costs accompanying activation of immune system and related neuroendocrine pathways is essential for understanding the selective forces operating on these systems. Here we attempted to detect such costs in terms of disruption to redox balance and interference between different immune system components in captive wild-caught greenfinches (Carduelis chloris). Study birds were subjected to an endotoxin-induced inflammatory challenge and temporary exposure to a psychological stressor (an image of a predator) in a 2*2 factorial experiment. Injection of bacterial endotoxin resulted in up-regulation of two markers of antioxidant protection – erythrocyte glutathione, and plasma oxygen radical absorbance (OXY). These findings suggest that inflammatory responses alter redox homeostasis. However, no effect on markers of oxidative damage to proteins or DNA in erythrocytes could be detected. We found no evidence that the endotoxin injection interfered with antibody production against Brucella abortus antigen or the intensity of chronic coccidiosis. The hypothesis of within-immune system trade-offs as a cost of immunity was thus not supported in our model system. We showed for the first time that administration of endotoxin can reduce the level of corticosterone deposited into feathers. This finding suggests a down-regulation of the corticosterone secretion cascade due to an endotoxin-induced immune response, a phenomenon that has not been reported previously. Exposure to the predator image did not affect any of the measured physiological parameters.

    Carotenoid and melanin pigments in the plumage of birds are hypothesized to be sensitive to oxidative stress. We manipulated oxidative status of captive greenfinches (Carduelis chloris L.) by the administration of buthionine sulfoximine (BSO), a selective inhibitor of the synthesis of glutathione (GSH), an intracellular antioxidant. Half of the birds in the treated group, as well as in the control group, also received dietary carotenoid (lutein) supplementation. BSO treatment reduced erythrocyte GSH levels and caused oxidative damage as indicated by the increased concentration of plasma malondialdehyde (MDA), an end product of lipid peroxidation. BSO treatment also reduced the brightness (i.e. increased blackness) of the tips of tail feathers grown during the experiment. These results show that a low systemic GSH level is required for development of eumelanin plumage coloration and that such a low GSH level is also potentially dangerous for the organism. Carotenoid supplementation increased plasma carotenoid levels and chroma of the yellow parts of the feathers grown during the experiment. However, carotenoid supplementation did not reduce plasma MDA levels. Manipulation of GSH did not affect plasma carotenoids or carotenoid-based plumage coloration.     Measurement of phytohaemagglutinin (PHA)-induced skin swelling is the most popular assay of immune function in avian studies. The mechanisms causing swelling have been relatively well studied; however, very little is known about the potential long term physiological effects of PHA. Here we show that injection of PHA into patagium of captive greenfinches Carduelis chloris increases the concentration of heterophils (phagocytic cells of the innate immune response) in the peripheral blood for at least 30 days. Such long-term consequences should be taken into account when using PHA skin test in studies monitoring changes in individual physiological condition and/or immune status.

    It is expected that the costs of exploiting an organism's immune system form the basis of many life-history trade-offs. However, the processes responsible for generating these costs have rarely been quantified in terms of energy expenditures and physiological rearrangements in the organism during the immune response. To determine the physiological changes caused by the immune response, we recorded various condition indices of male captive greenfinches (Carduelis chloris) 1, 4, and 8 days after immune challenge with a nonpathological antigen (sheep red blood cells; SRBC). Measured variables included estimates of total and differential leukocyte counts, serum protein concentrations and profile, serum triglyceride concentration, basal metabolic rate, body mass, spontaneous locomotory activity, and serum total antioxidant capacity.     The amount and distribution of genetic variation in seven introduced New Zealand populations of Carduelis chloris were assessed at 40 loci using starch gel electrophoresis and compared with those in native European populations. Fewer alleles (1.45) and fewer polymorphic loci (33 per cent) were detected across the introduced populations than across native populations (1.75, 55 per cent), reflecting the narrow geographical origin of the introduced populations. There was no evidence for severe inbreeding or genetic drift as the levels of average heterozygosity (H = 0.025), percentage polymorphic loci (P = 16.9) and average number of alleles per locus (A = 1.22) were indistinguishable from levels observed within European populations (77 = 0.025, P =14.1, A = 1.19). Furthermore, introduced populations were genetically less weakly differentiated (F_ST = 0.022) than native populations (F_ST = 0.041), indicating that little genetic drift has been involved in the colonization of the new range. Similar levels of genetic variability in native and introduced greenfinch populations are consistent with theoretical expectations as the founder population size was relatively large (>60 individuals) and a rapid increase in population size directly after colonization was documented. A review of earlier studies on introduced birds also revealed that reductions in levels of genetic variability seem to be inversely proportional to the size of the founder population, and that less variation has been lost if the rate of population growth directly after introduction was fast.

    Levels of fluctuating asymmetry (FA) in 12 bilateral skeletal traits were estimated from 12 populations of greenfinches (Carduelis chloris) collected along a north-south gradient across Europe. Average FA of measured traits was positively correlated with latitude indicating that the younger and genetically less diverse northern European populations are developmentally less stable than the older and genetically more diverse southern populations. Levels of FA differed significantly between different traits being lowest for functionally important traits (limb and wing bones) and highest for functionally less important traits such as foramina (apertures through bones)- a pattern that was highly concordant across different populations. Males tended to exhibit higher levels of FA than females, a finding consistent with the suggestions that males are more prone to developmental perturbations than females. Age differences in levels of FA were relatively clear, but inconsistent across traits with different degree of functionality. Individual heterozygosity - as enumerated from variation in allozyme loci - was unrelated to individual FA. No evidence for existence of individual asymmetry parameter (IAP) was found although traits related to locomotion indicated some degree of integration, which was expressed by correlations in the signed asymmetry. Nevertheless, an individual's overall asymmetry was poorly predicted by asymmetry of individual characters. Evidence for existence of population asymmetry parameter (PAP) was clear since all traits exhibited a similar degree of association with latitude. That the latitudinal cline of increasing FA towards north coincided with decreasing levels of genetic variability across the cline could be indicative of break down of developmental stability in the recently established and genetically impoverished populations. To what extent a reduced heterozygosity, the break up of co-adapted gene complexes and/or environmental differences contributed to this process cannot be distinguished from our data.

  • Testis size variation in the greenfinch Carduelis chloris: relevance for some recent models of sexual selection

    Interspecific evidence that testis size responds to selection caused by sperm competition has been obtained from many taxa. However, little is known about the sources of intraspecific variation in testis size, although such variation may have functional significance. Variation in testis size and asymmetry was studied within and between eight geographically separated (and genetically differentiated) populations of greenfinches Carduelis chloris. The relationships between testis size and plumage brightness (degree of yellowness) and the prevalence of haematozoan infections were also investigated in three of these populations, as they related to the predictions of the immunocompetence handicap hypothesis, and Moller's hypothesis relating directional testis asymmetry to phenotypic quality. There were large differences between populations in testis size, with males from northern populations having larger testes than those from southern populations. Within populations, large testes were associated with larger body size and greater age. When the influence of these factors was removed statistically, males with large testes were more likely to be infected with haematozoan parasites, and had brighter yellow plumage. No evidence was found that directional asymmetry in testis size was related to either of these measures of phenotypic quality. These results are consistent with the hypothesis that males with large testes, while signalling higher phenotypic quality as revealed by increased plumage brightness, also pay a cost in terms of reduced immunocompetence, revealed by the increased probability of infection in these males. That these patterns were similar in three different populations adds further strength to these conclusions. Our results suggest that studying the sources of variation in testis size among individuals can reveal interesting processes in sexual selection.

    Quantitative trait divergence and variability among 12 greenfinch populations across continental Europe was examined and compared to divergence in neutral genetic markers (allozymes). The added among locality variance component for 16 skeletal traits was large (mean 28%, range 4–48%) equalling a divergence of up to three SD units. The divergence in quantitative traits (Qst = 0.04-0.48) greatly exceeded that in alloymes (FST= 0.01-0.07), indicating the differentiation in quantitative traits to be larger than expected by mutation and drift alone. This conclusion was consistent also with results from the multivariate approach of Rogers & Harpending. However, genetic and morphometric distances between populations were positively correlated, even when controlling for the geographic distance separating pairs of populations. In concordance with Bergmann's rule, most traits were strongly and positively correlated with latitude, indicating latitudinally ordered genetic or/and environmental effects. However, the correlation between lower mandible width and latitude was strongly negative, demonstrating an inverse relationship between beak size and body size across the populations. These results are interpreted to reflect the re-colonization of history of northern Europe (genetic and geographic distances correlated) which has been paralleled by selection acting on quantitative traits (QST>FST)- In particular, the counter-gradient variation in beak width, a functionally important trait, is suggestive of an adaptive basis for quantitative trait divergence.
