MATERIALS AND METHODS Wounds were made in the tongue using a punch tool in 64 male Sprague-Dawley rats, randomized into four groups (n = 16 per group): group 1 (control), group 2 (CAE), group 3 (AUBM mixed with orabase), and group 4 (orabase). No product was applied in group 1 and groups 2-4 received three daily topical applications. The animals were weighed on day 0 and at the time of euthanasia. Four rats in each group were euthanized at days 2, 7, 14, and 21 and the tongues were processed for: macroscopic morphometric analysis, myeloperoxidase (MPO) and malondialdehyde (MDA) levels, histological wound repair (degree of reepithelialization and inflammation), and CD31 positivity. RESULTS The animals' weight gain, histological wound repair, and CD31 positivity from greatest to least were: AUBM > CAE > orabase > control. Percentage of tongue occupied by wound, MPO, and MPA levels from least to greatest were: AUBM < CAE < orabase < control, whereby the AUBM group showed significant differences (p ≤ 0.05) in comparison with the other groups on days 2, 7, 14, and 21 for percentage of tongue occupied by wound and MDA and on days 7, 14, and 21 for MPO. CONCLUSIONS CAE is effective for oral tissue regeneration, while AUBM is an even more potent means of oral mucosa regeneration. CLINICAL RELEVANCE AUBM may be beneficial to patients with oral wounds; this finding requires further clinical and laboratory investigation.

This study demonstrated that CA Extract facilitates the wound-repair process by promoting fibroblast proliferation and collagen synthesis and exhibits antibacterial activity. Gelatin nanofibres containing C. Asiatica Extract were fabricated via electrospinning and were shown to exhibit dermal wound-healing activity in a rat model. The wound areas of rat skin treated with electrospun gelatin membranes containing C. Asiatica (EGC) presented the highest recovery rate compared with those treated with gauze, neat gelatin membranes and commercial wound dressings. The results of the histopathological examination support the outcome of the wound models. Contact-angle and water-retention measurements confirmed that the addition of C. Asiatica Extract did not significantly affect the hydrophilicity of the EGC membranes. The measured weight loss revealed that the EGC membranes are biodegradable. The findings suggest that EGC membranes are a promising material for the treatment of skin wounds. Copyright © 2015 John Wiley & Sons, Ltd.

Among the 'pegaga’, Centella Asiatica has been used as a remedy for diabetes mellitus. Thus, we decided to validate this claim by evaluating the in vivo antidiabetic property of C. Asiatica (CA) on T2DM rat model using the holistic 1 H NMR-based metabolomics approach. Method In this study, an obese diabetic (mimic of T2DM condition) animal model was developed using Sprague–Dawley rats fed with a high-fat diet and induced into diabetic condition by the treatment of a low dose of streptozotocin (STZ). The effect of C. Asiatica Extract on the experimental animals was followed based on the changes observed in the urinary and serum metabolites, measured by 1 H NMR of urine and blood samples collected over the test period. Results A long-term treatment of obese diabetic rats with CA Extract could reverse the glucose and lipid levels, as well as the tricarboxylic acid cycle and amino acid metabolic disorders, back towards normal states. Biochemical analysis also showed an increase of insulin production in diabetic rats upon treatment of CA Extract. Conclusion This study has provided evidence that clearly supported the traditional use of CA as a remedy for diabetes. NMR-based metabolomics was successfully applied to show that CA produced both anti-hyperglycemic and anti-hyperlipidemic effects on a rat model. In addition to increasing the insulin secretion, the CA Extract also ameliorates the metabolic pathways affected in the induced diabectic rats. This study further revealed the potential usage of CA Extract in managing diabetes mellitus and the results of this work may contribute towards the further understanding of the underlying molecular mechanism of this herbal remedy.

Increased of Type 1-Collagen Expression in Vaginal Wall of the Ovariectomized Rats (Rattus novergicus) Strain Wistar After Ovariectomy on Centella Asiatica Extract

This condition will result in an inhibition of the epithelial cell maturation and reduced collagen synthesis so that vaginal elasticity will also decrease. This condition can be improved by the administration of estrogen or phytoestrogen . Centella Asiatica is one of the plants that contain phytoestrogens with several essential components such as asiaticoside, Asiatic acid and madecassic acid could increase synthesis collagen. The objective of this study was to investigate the thickening and increasing collagen of the vaginal wall of ovariectomized Wistar rats treated with Centella Asiatica Extract . Ovariectomized rats were divided into 4 treatment groups, where rats in P1 group was not administered the Extract of the Centella Asiatica ; Meanwhile rats in P2, P3 and P4 wsa administered the Centella Asiatica Extract at dose of 30, 60 and 120 mg/kg BW respectively per day for 40 days. On day 61, the rats were sacrificed and examined immunohistochemically for the assessment of their type-1 collagen expression. In conclusion, the expression of the type-1 collagen producing cells increased significantly by the administration of Centella Asiatica Extracts at doses of 30 , 60 and 120 mg/ kg BW daily for 40 day.. However, the 120 mg one was the most optimal dose that was able to enhance type-1 collagen synthesis. This proved that Centella Asiatica Extract contained phytoestrogen , which could increase the thickening and elevate the collagen of vaginal walls of the ovariectomized rats.

This can lead to domestic disharmony because of divorce, affairs and domestic violence. To prevent these complaints needs estrogen therapy or phytoestrogens. Centella Asiatica is a phytoestrogen that increase proliferation and collagen thickening in vaginal wall. However, the mechanism of thickening and increasing collagen of vaginal wall has not been clear. This study aims to explain the mechanism of thickening due to proliferation and maturation of epithelial and collagen synthesis increased vaginal wall of Rattus norvegicus that experienced a given ovariectomy.The research method is experimental design with posttest only control group design, by 35 female rats of Rattus norvegicus age of 4 months, weight 290-300 grams, conducted randomized into 4 groups, each group was seven rats. All groups performed ovariectomy. Group-1 weren’t given Extract of Centella Asiatica. Group-2, 3, 4 were given Extracts of Centella Asiatica on day 22 post-ovariectomy with a dose of 30mg, 60mg, 120mg/Kbw/day for 40 days orally. Immunohistochemical examination to see the expression of estrogen receptor beta-producing cells (ER-s) and collagen-producing cells. Histological examination to see thickening of the vaginal wall. Results were analyzed using univariate analysis showed an increase in epithelial proliferation, maturation of the vaginal wall was significant (p
