Almost 90% of the nematocidal activity of Chenopodium ambrosioides infusions was due to a hydrophilic component different from ascaridole. Synthetic ascaridole and the ascaridole from infusions, extracted into hexane, caused a reduction of carbachol-induced contractions in rat gastrointestinal smooth muscle at concentrations required to kill Caenorhabditis elegans (L.). The herbal infusion and the ascaridole-free hexane-extracted aqueous residue of the above infusion, at nematocidal concentrations, had no detectable effect on smooth muscle contraction in the above system. It would appear that the traditional form of usage of Chenopodium ambrosioides infusions as a vermifuge is safer than the use of the herb's essential oil.

For this purpose, the molecular affinity between the total proteins from erythrocytes infected with Plasmodium falciparum and HCE or chloroquine was evaluated by surface plasmon resonance (SPR). Subsequently, the plasmodicidal potential of HCE was assessed in a P. falciparum culture. Using BALB/c mice infected with Plasmodium berghei intraperitoneally (ip.), we evaluated the effects of ip. treatment, for three consecutive days (day 7, 8, and 9 after infection), with chloroquine (45 mg/kg) or HCE (5 mg/kg), considering the survival index and the parasitaemia. The groups were compared to an untreated control group that receives only PBS at the same periods. The results indicated that HCE could bind to the total proteins of infected erythrocytes and could inhibit the parasite growth in vitro (IC50 = 25.4 g/mL). The in vivo therapeutic treatment with HCE increased the survival and decreased the parasitaemia in the infected animals. Therefore, the HCE treatment exhibited a significant antiplasmodial effect and may be considered as a potential candidate for the development of new antimalarial drugs.

Chenopodium ambrosioides L., popularmente conhecido como mastruz, tem sido utilizado no Maranhao para o tratamento topico de ulceras leishmanioticas. O trabalho avaliou a eficacia das fracoes do extrato hidroalcoolico das folhas de C. ambrosioides contra formas promastigotas da especie Leishmania amazonensis. O fracionamento foi realizado pela particao sequencial do extrato hidroalcoolico de folhas de C. ambrosioides com solventes de polaridade crescente (hexano, cloroformio e acetato de etila). As formas promastigotas foram cultivadas em meio RPMI 1640 com e sem as fracoes. Apos 24h de incubacao a 26oC, o numero de promastigotas viaveis foi contado pelo metodo direto em câmara de Neubauer. As concentracoes que inibem o crescimento de 50% (CI50 ) das promastigotas de L. amazonensis foram calculadas a partir da avaliacao da mortalidade das promastigotas in vitro. De acordo com escores padroes nas fracoes de acetato de etila e hidroalcoolica remanescente nao foi observada atividade leishmanicida significativa, mas as fracoes hexânica e cloroformica apresentaram-se ativas. Os resultados obtidos demonstram uma acao leishmanicida promissora das fracoes. Estudos futuros sao necessarios para investigar a eficacia destas fracoes no tratamento da leishmaniose em modelos experimentais in vivo. Abstract: In vitro leishmanicidal activity of fractions obtained from hydroalcoholic extracts of Chenopodium ambrosioides’ leaves. Leishmaniasis is an infectious disease caused by protozoa of the genus Leishmania and represents serious public health problem in Africa, Asia and Latin America. Chenopodium ambrosioides, popularly known as “mastruz”, has been used in Maranhao for the topical treatment of leishmanial ulcers. The study evaluated the effectiveness of the fractions from hydroalcoholic extract of C. ambrosioides’ leaves against the promastigotes of Leishmania amazonensis. Fractionation as accomplished by partition sequential extract of leaves of C. ambrosioides with solvents of increasing polarity (hexane, chloroform and ethyl acetate). The promastigotes forms were cultured in supplemented RPMI 1640 with or without fractions. After 24h incubation at 26°C, the number of viable promastigotes was counted by the direct method in a Neubauer chamber. The concentrations that inhibit growth of 50% (IC50 ) of L. amazonensis’ promastigotes were calculated from the assessment of the mortality of promastigotes in vitro. According to standard scores, in the ethyl acetate and hydroalcoholic remaining fractions there was no leishmanicidal activity, while the hexane and chloroform fractions were actives. The results showed a promissor leishmanicidal activity of fraction. Future studies are necessary to investigate the effectiveness of these fractions in the treatment of these fractions in experimental models in vivo. (Chenopodiaceae), conhecida como erva-de-Santa-Maria ou mastruz, representa especie exotica, de grande ocorrencia no estado do Maranhao, muito utilizada na pratica popular para diversos fins terapeuticos. Estudos farmacologicos tem comprovado o potencial da especie para desenvolvimento de bioprodutos como alternativa terapeutica. Assim, o objetivo deste trabalho foi desenvolver metodologia analitica para padronizacao dos extratos de C. ambrosioides empregando ensaios quimicos e biologicos de atividade anti-Giardia, visando o desenvolvimento de tecnologia farmaceutica na obtencao de novos farmacos no tratamento da giardiase. Foram obtidos extratos das folhas de C. ambrosioides por planejamento fatorial dos parâmetros: natureza do solvente (etanol 70% e agua), operacao de extracao (maceracao, percolacao e extracao por Soxhlet) e hidromodulo; submetidos a analise quimica e avaliacao da atividade giardicida in vitro. Os resultados permitem concluir que a obtencao dos extratos hidroalcoolicos das folhas de C. ambrosioides pelo procedimento extrativo de percolacao e maceracao representam os extratos com melhor perfil quimico e/ou atividade giardicida; evidenciando que a natureza do solvente, o procedimento extrativo e o hidromodulo sao variaveis que infuenciam na obtencao dos extratos da especie em estudo. Abstract: Chenopodium ambrosioides L., Chenopodiaceae, (herb-of-Santa-Maria or mastruz), represents high occurrence alien species, in the State of Maranhao, widely used in popular practice for many therapeutic purposes. Pharmacological studies have demonstrated the potential of species to develop therapeutic bioproducts such as new alternatives. Thus, the goal of this work was to developanalytical methodology for standardized extracts of Chenopodium. ambrosioides employing chemical and biological tests of giardicidal activity, aiming at the development of pharmaceutical technology in getting new drugs for Giardia treatment. Were obtained extracts of leaves of C. ambrosioides by factorial planning factors: nature of solvent (ethanol 70 and water), operation of extraction (maceration, percolation and Soxhlet extraction) and hydromodule (plant:solvent); subjected to chemical analysis and evaluation of giardicidal activity in vitro. The results suggest that the obtaining of hydroalcoholic extracts from the leaves of C. ambrosioides extractive of percolation and maceration represents the extract with better chemical profle and/or giardicidal activity; evidencing that the nature of the solvent, extractive procedure and hydromodule are variables that infuence in obtaining extracts of the species under study. The chemotherapy to this disease is not always effective and can cause several side effects. Chenopodium ambrosioides L. (Chenopodiaceae) is used by the native people in the treatment of cutaneous ulcers caused by different species of Leishmania. The aim of this study was to investigate the effect of the treatment with a hydroalcoholic crude extract (HCE) from the leaves of Chenopodium ambrosioides on the murine infection with Leishmania amazonensis. MATERIAL AND METHODS: The mice were treated for 4-6 weeks post-infection (p.i.) with HCE (5 mg/kg) or meglumine antimoniate (Sb(v)) (28 mg/kg) either by the oral route, once a day, for 15 days or by five intralesional (IL) injections at intervals of 4 days. The thickness of the infected paws was determined weekly and the parasite load evaluated in the draining lymph nodes (LN), the spleen and in the footpad after 7 weeks of infection. The nitric oxide (NO) production was evaluated in cultures with cells from peritoneum or LN. RESULTS: The IL treatment increased the NO production in the LN and peritoneum cultures and reduced the parasite load from the footpad, spleen and LN. On the other hand, the oral treatment decreased did alter neither the NO production nor the parasite load. CONCLUSIONS: IL HCE treatment was more efficient than the oral HCE treatment since the former was able to control the dissemination of infection. This effect can be due to either a direct leishmanicidal effect of HCE or the improvement in the NO production by HCE-stimulated macrophages. The results could justify the topical use of the Chenopodium ambrosioides' leaves in the treatment of the ulcers caused by Leishmania.

(Chenopodiaceae), a plant known in Brazil as ‘mastruz’, have been used by native people to treat parasitic diseases. Experimentally it was shown that Chenopodium ambrosioides inhibits the Ehrlich tumor growth, what could be due to an immunomodulatory effect of this product. The aim of this study was to investigate the effect of hydroalcoholic crude extract (HCE) from leaves of Chenopodium ambrosioides on macrophage activity and on lymphoid organs cellularity. C3H/HePas mice received the HCE (5 mg/kg) by intraperitoneal via and were sacrificed 2 days later. HCE treatment did not alter the cell number in bone marrow, but it increased the cell number in peritoneal cavity, spleen and lymph node. The spreading and phagocytosis activity, the PMA-induced hydrogen peroxide (H 2 O 2 ) release and the nitric oxide (NO) production were also increased when compared to control group. Similar results were obtained with concanavalin A (Con A), used as a positive control, with exception of the NO production that was only detected in HCE-derived macrophages. The in vitro treatment with HCE induced a dose-dependent NO production by resident macrophages, but did not enhance the NO production by HCE-derived macrophage, which however, was enhanced by Con A, suggesting that HCE and Con A induce NO production by different routes. In conclusion, HCE-treatment was able to increase the macrophages activity and also the cellular recruitment to secondary lymphoid organs, what could explain the previously related anti-tumor activity of Chenopodium ambrosioides .

Pest Management Science, 2011Co-Authors: Jin Feng HuAbstract:BACKGROUND: In a screening programme for new agrochemicals from Chinese medicinal herbs, Chenopodium ambrosioides L. was found to possess strong fumigant activity against the maize weevil Sitophilus zeamais (Motsch.). Essential oil of C. ambrosioides was obtained by hydrodistillation, and the constituents were determined by GC-MS analysis. The active compounds were isolated and identified by bioassay-directed fractionation. RESULTS: Five active compounds [(Z)-ascaridole, 2-carene, ρ-cymene, isoascaridole and α-terpinene] were isolated and identified from the essential oil from Chinese C. ambrosioides. The LC₅₀ values (fumigation) of the crude essential oils and the active compound (Z)-ascaridole against S. zeamais adults were 3.08 and 0.84 mg L⁻¹ air respectively. The LD₅₀ values (contact toxicity) of the crude essential oil and (Z)-ascaridole against S. zeamais adults were 2.12 and 0.86 µg g⁻¹ body weight respectively. CONCLUSION: The findings suggested that the essential oil of Chenopodium ambrosioides and its main active constituent, (Z)-ascaridole, may be explored as a natural potential fumigant.

Southwest China Journal of Agricultural Sciences, 2020Co-Authors: Zhang Hong, Liao Ying, Wang Yu, Ma DanweiAbstract:Allelopathy of decomposition liquids from the residue of Chenopodium ambrosioides after removing essential oil on Triticum aestivum,Brassica rapa and Vigna radiate were studied.The result showed that there was no significant effect of decomposition liquids from the residue of this plant after removing essential oil on seedling growth of receptors,but significantly inhibited seed germination of receptors and delayed germination.The allelopathic effects were enhanced when concentration increased.The sensitivity of three receptors on decomposition liquids from the residue of Chenopodium ambrosioides after removing essential oil ordinal was Triticum aestivum,Vigna radiate and Brassica rapa.

Antifungal activity of volatile oil from invasive weed, Chenopodium ambrosioides L., on dermatophytes.
Southwest China Journal of Agricultural Sciences, 2020Co-Authors: Liao Ying, Li Yuan, Ma DanweiAbstract:Using 2-fold serial dilution method and fumigating method,the activity of volatile oil from an invasive weed,Chenopodium ambrosioides L.on Microsporum canis,Epidermophyton floccosum and Trichophyton rubrum were investigated.The results indicated that volatile oil from C.ambrosioides could effectively inhibite M.canis,E.floccosum and T.rubrum directly and indirectly.The minimum inhibitory concentration(MIC) and the minimal bactericidal concentration(MBC) was 1-2 and 4-8 mL·L-1,respectively.This study provided scientific basis for the further development of C.ambrosioides.

Response of root border cells of soybean to cultured Chenopodium ambrosioides root exudates
Soil and Environmental Sciences, 2020Co-Authors: Ma DanweiAbstract:By using the techniques of tissue culture and aeroponic culture,response of root border cells of soybean(Giycine max L.) to cultured Chenopodium ambrosioides root exudates was studied.The results showed the number and activity of root border cells decreased consecutively when treated with cultured C.ambrosioides root exudates.Some granules gathered around free root border cells would result from chelation between root border cells exudates with that of the cultured root.The cultured root exudates could stimulate the activity of pectin methylesterase of the root cap of soybean,but the effect gradually lowered down along with the increasing of treatment concentration and treatment time.The results suggested that the cultured root exudates had stress on root border cells of soybean and root border cells of soybean alleviated the allelopathic effects of the cultured root exudates on root tips though programmed cell death and chelating.

Antifungal activity of essential oil from Chenopodium ambrosioides L. growing in sichuan against plant-pathogenic fungi
Soil and Environmental Sciences, 2020Co-Authors: Ma DanweiAbstract:The antifungal activity and antifungal mechanism of essential oil from Chenopodium ambrosioides growing in Sichuan was evaluated against five species of plant-pathogeni fungi by the poison assay and fumigating assay.Compared to the corresponding control,the IC50 of Rhizoctonia solani Kuha,Curvularia lunata(walk)Boed,Pyricularia oryzae Cav.,Gibberelle zeae(Schw.)Petch, and Pestalotia funerea were 0.734μL·mL-1 ,0.342μL·mL-1 ,0.435μL·mL-1 ,0.370μL·mL-1 and 0.332μL·mL-1 ,respectively.When a dose of 1μL essential oil was used,growth of R.solani,C.lunata,P.oryzae,and Pestalotia funerea were promoted,and growth of those five fungi were inhibited when the essential oil was beyond 1μL dose.Fungi treated by essential oil occurred some changes including damage of cellularity,swarm of protoplast,decrease or increase of kinds of proteins,but there was no any change observed on DNA level.

Methods: Cytotoxicity was characterized by 50 % inhibition (IC50) of human breast cancer cell lines (MCF-7) using 3-(4,5-dimethylthaizol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Apoptosis was analysed by Hoechst33258 staining and DNA ladder. MCF-7 cellular superoxide dismutase (SOD), catalase (CAT) vitality and malondialdehyde (MDA) content were evaluated. Results: The essential oil was cytotoxic to MCF-7 cell line. A dose- and time-dependent inhibition was observed with IC50 values of 18.75, 9.45 and 10.50 μg/ml at 6, 24 and 48 h, respectively. Analyses by Hoechst33258 staining and DNA ladder indicate that the essential oil induced apoptosis. SOD vitality significantly decreased (p< 0.05) by 51 % when the concentration of the essential oil increased from 1.25 to 12.5 μg/ml while CAT vitality significantly increased (p < 0.05) by 71 % when essential oil concentration was similarly increased. The MDA content of each treatment group, when compare to control, did not show any significant difference (p < 0.05). Conclusion: The essential oil of C. ambrosioides was cytotoxic to MCF-7 cell line and induced apoptosis.
