In dieser Zeit hatte eine schwangere Frau (Graviditatswoche 19/20) Kontakt mit den abortierenden Ziegen. In der Folge erkrankte sie an einer schweren Allgemeininfektion und erlitt einen Abort. In der Plazenta der Patientin konnte Chlamydophila abortus immunhistologisch und mittels PCR nachgewiesen werden. Ziel des Fallberichtes ist es TierarztInnen auf dieses potenzielle Zoonoserisiko aufmerksam zu machen.

In one of three investigated caprine abortions Chlamydophila abortus could be identified as aetiology. During this time a pregnant woman (pregnancy week 19/20) had contact with aborting goats. She developed a severe generalized infection and aborted. The placenta contained Chlamydophila abortus shown by immunohistochemistry and PCR. The aim of the present case report is to alert medical doctors about the potential zoonotic risk of ovine/caprine abortions.

Chlamydophila spp infection in horses with recurrent airway obstruction similarities to human chronic obstructive disease
Respiratory Research, 2008Co-Authors: Dirk Theegarten, Konrad Sachse, Kerstin Fey, Britta Mentrup, Helmut Hotzel, O AnhennAbstract:Recurrent airway obstruction (RAO) in horses is a naturally occurring dust-induced disease mainly characterized by bronchiolitis which shows histological and pathophysiological similarities to human chronic obstructive pulmonary disease (COPD). In human COPD previous investigations indicated an association with Chlamydophila psittaci infection. The present study was designed (1) to clarify a possible role of this infectious agent in RAO and (2) to investigate the suitability of this equine disorder as a model for human COPD. Clinico-pathological parameters of a total of 45 horses (25 horses with clinical signs of RAO and 20 clinically healthy controls) were compared to histological findings in lung tissue samples and infection by Chlamydiaceae using light microscopy, immunohistochemistry, and PCR. Horses with clinical signs of RAO vs. controls revealed more inflammatory changes in histology (p = 0.01), and a higher detection rate of Chlamydia psittaci antigens in all cells (p < 0.001) and bronchiolar epithelial cells alone (p < 0.001) by immunohistochemistry. The abundance of chlamydial inclusions increased with the severity of disease. PCR was positive in 60% of horses with RAO vs. 45% of the controls (p = 0.316). OmpA sequencing identified Chlamydophila psittaci (n = 9) and Chlamydophila abortus (n = 13) in both groups with no significant differences. Within the group of clinically healthy horses subgroups with no changes (n = 15) and slight inflammation of the small airways (n = 5) were identified. Also in the group of animals with RAO subgroups with slight (n = 16) and severe (n = 9) bronchiolitis could be formed. These four subgroups can be separated in parts by the number of cells positive for Chlamydia psittaci antigens. Chlamydophila psittaci or abortus were present in the lung of both clinically healthy horses and those with RAO. Immunohistochemistry revealed acute chlamydial infections with inflammation in RAO horses, whereas in clinically healthy animals mostly persistent chlamydial infection and no inflammatory reactions were seen. Stable dust as the known fundamental abiotic factor in RAO is comparable to smoking in human disease. These results show that RAO can be used as a model for human COPD.

Two of the dead animals were subjected to post mortem examination. From different organs of the rats, chlamydiae were isolated and confirmed by PCR and DNA sequencing as Chlamydophila psittaci. The results show that infection with Chlamydophila psittaci may occur in laboratory rats. Such outbreaks may have influence on the results of experimental studies. Chlamydial infections in laboratory animals also pose risk to humans (zoonosis).

As samples served of each moment of collection and sex of the animals 10 cloacal swabs which were taken at the age of 1, 4, 8 and 12 (females) or 16 weeks (males) and at the time of slaughter at the age of 16 or 20 weeks. Spleen samples were taken at the time of slaughter, additionally. These were pooled making 1 pool out of 5 individual samples. The cloacal and spleen pools were examined by nested PCR (nPCR), Capture-ELISA and Capture Blocking-ELISA directly as well as after isolation attempts in cell cultures. The most sensitive method to detect chlamydia, with 6 isolates proved to be the isolation by cell culture followed by detection using nPCR. Not corresponding to the results of the nPCR were 4 positive reactions found by the Capture-ELISA which could in no case be affirmed by Capture-Blocking-ELISA. The direct examination of cloacal swab pools by nPCR proved positive in only 2 cases. In contrast to this the examination of these samples by Capture-ELISA showed a high percentage of 71,9% positive results, of which only 2 cases were confirmed by nPCR and none by Capture- Blocking- ELISA. Of the 8 Chlamydia positive results in the nPCR 7 could be classified by DNA sequencing to Cp. abortus and only one to Cp. psittaci

Macroscopic inspection of fetus and placenta was completed by histopathology and bacteriological isolation of agents. In addition, immunohistologically the following antigens were labeled in formalin-fixed and paraffin-embedded tissue sections: Toxoplasma gondii, Neospora caninum, Chlamydophila abortus (formerly Chlamydia psittaci serovar 1) and Border Disease Virus. From farms with abortions caused by Chlamydophila abortus specific data were recorded. In 75% of abortion cases in sheep and in 59% of cases in goats an etiologic diagnosis could be substantiated. Chlamydophila abortus is the most commonly involved agent in the etiology of caprine and ovine abortion (sheep 39%, goats 23%), followed by Toxoplasma gondii (sheep 19%, goats 15%) and Coxiella burnetti (sheep 1%, goats 10%). All other agents are of minor importance. An infectious cause of abortion based on histopathologic findings without isolation of agents was observed in sheep (10%) and goats (21%). Malformation occurred in sheep (2%) and goats (3%) and lesions suggestive for Vitamin E/Selenium deficiency were seen in goats only (2%).

They are caused respectively by strictly intracellular and Gram negative bacterium Chlamydophila abortus (Cp. abortus) and Coxiella burnetii (C. burnetii). Chlamydophila pecorum (Cp. pecorum) is commonly isolated from the digestive tract of clinically inconspicuous ruminants but the abortive and zoonotic impact of this bacterium is still unknown because Cp. pecorum is rarely suspected in abortion cases of small ruminants. We have developed a multiplex PCR (m-PCR) for rapid simultaneous differential detection of Cp. abortus, Cp. pecorum and C. burnetii in clinical samples taken from infected animals.

Bernard, Annie RodolakisAbstract:The efficacy of Chlamydophila abortus vaccine strain 1B in protecting against two selected Chlamydophila pecorum strains, isolated from an aborted goat (M14) in Morocco and a ewe (AB10) in France, was investigated in a mouse model, by comparing the reduction in number of bacteria in the placentas of vaccinated mice challenged intraperitoneally at 11 days of pregnancy with the reference C. abortus (AB7) and C. pecorum (M14, or AB10) strains, to those of unvaccinated mice. Vaccine 1B was shown to provide effective protection against the field strains of C. pecorum, since it significantly reduced the placental Chlamydophila colonisation. The two C. pecorum strains were not sufficiently abortifacient in mice to use reduction in abortion as a criterion of protection.

Revue d'élevage et de médecine vétérinaire des pays tropicaux, 2004Co-Authors: A H Remmal, Annie Rodolakis, A Souriau, A El H IdrissiAbstract:In order to improve the direct diagnosis of abortive chlamydiosis in small ruminants, 225 vaginal swabs collected from aborting sheep and goats were tested for Chlamydophila DNA by two types of polymerase chain reactions (PCR): chlamydial-infection specific PCR (CTU/CTL), and speciesspecific PCR (CPS/CPC) for Chlamydophila abortus and Cp. pecorum . The omp 1 gene coding for the major outer membrane protein of Chlamydiales was found in 65 (29%) of the analyzed samples, of which 80% had been collected within four weeks postabortion. The majority of PCR (CTU/CTL) positive samples (69%) came from herds found positive by the complement fixation test. The species-specific PCR applied to PCR (CTU/CTL) positive samples did not allow detecting the expected fragments of Cp. abortus (1800 bp) and Cp. pecorum (580 bp), except in four samples out of 65 tested. The remaining samples were either negative (63%) or non conclusive (31%) with non-specific bands of intermediate molecular weights. The enzyme restriction analysis of the omp1 gene after DNA amplification revealed much genetic diversity among the 65 samples tested. Typical genetic patterns of Cp. abortus were found only in 15 samples, but the majority (77%) of the samples showed patterns attributed to Cp. pecorum or closely related to it. Culture on McCoy cells of 26 vaginal swabs and abortion products from animals found with chlamydial infection (serology and PCR) revealed the presence of Chlamydophila in eight samples: seven were characterized as Cp. abortus and one as Cp. pecorum . The remaining samples gave no cytopathogenic effect after five serial passages, or they were contaminated. A Cp. pecorum strain, designated M14, isolated for the first time in Morocco, was further investigated to test its virulence in mice. These data suggest there is genetic variation in abortive strains and a possible link between Chlamydophila pecorum and abortions in sheep and goats in Morocco. This genetic diversity needs to be further investigated in order to gear vaccination strategies for control of abortive chlamydiosis in Morocco.

Veterinary Research, 2003Co-Authors: Celine Hechard, Olivier Grépinet, Annie RodolakisAbstract:In order to enhance the quantity and the protective properties of the antibodies induced by DNA vaccination with the heat shock protein dnaK gene of Chlamydophila abortus AB7 as well as to elicit an efficient cellular immune response, we vaccinated mice with a DNA prime followed by a boost with the recombinant DnaK protein. In non-pregnant mice, this strategy induced the same predominance of the IgG2a isotype as DNA immunization alone with a substantial increased antibody level. The induced antibodies had no in vitro neutralizing properties on C. abortus infectivity. Moreover, the proteic boost probably failed to elicit an efficient cellular immune response since the pregnant or non-pregnant mice were not protected against the bacterial challenge.
