Genetically engineering Crambe abyssinica- A potentially high-value oil crop for salt land improvement

Krens, Eibertus N. Van LooAbstract:Crambe abyssinica (Crambe) is a new industrial oil crop that can grow on saline soil and tolerates salty water irrigation. Genetically engineered Crambe in which the seed‐oil composition is manipulated for more erucic acid and less polyunsaturated fatty acid (PUFA) would be highly beneficial to industry. In this research, lysophosphatidic acid acyltransferase 2 RNA interference (CaLPAT2‐RNAi) was introduced into the Crambe genome to manipulate its oil composition. The result showed in comparison with wild type, CaLPAT2‐RNAi could significantly reduce linoleic and linolenic acid content, simultaneously increasing erucic acid content. Systematic metabolism engineering was then carried out to further study CaLPAT2‐RNAi, combined with the overexpression of Brassica napus fatty acid elongase (BnFAE), Limnanthes douglasii LPAT (LdLPAT), and RNAi of endogenous fatty acid desaturase 2 (CaFAD2‐RNAi). Oil composition analysis on the tranformants' seeds showed that (a) with CaFAD2‐RNAi, PUFA content could be dramatically decreased, in comparison with BnFAE + LdLPAT + CaFAD2‐RNAi, and BnFAE + LdLPAT + CaFAD2‐RNAi + CaLPAT2‐RNAi seeds showed lower linolenic acid content; (b) BnFAE + LdLPAT + CaFAD2‐RNAi + CaLPAT2‐RNAi could increase the erucic acid content in Crambe seed oil from less than 66.6% to 71.6%, whereas the highest erucic acid content of BnFAE + LdLPAT + CaFAD2‐RNAi was 79.2%; (c) although the four‐gene combination could not increase the erucic acid content of seed oil to a higher level than the others, it led to increased carbon resource deposited into C22:1 and C18:1 moieties and lower PUFA. Summarily, the present research indicates that suppression of LPAT2 is a new, promising strategy for seed‐oil biosynthesis pathway engineering, which would increase the value of Crambe oil.

However, as an understudied crop, improvement of Crambe has been hampered by the lack of genetic and genomic information to enhance its yield, oil quality and resistance against biotic and abiotic stress. Development of molecular markers is therefore of great significance to facilitate genetic improvement of Crambe. In this study, high-throughput sequencing was performed to generate sequences for the transcriptome and genome of a widely planted Crambe cultivar, Galactica. A total of 186,778 expressed sequence tag (EST) contigs as 8,130,350 genomic contigs were assembled as well. Altogether, 82,523 pairs of primers were designed in the flanking sequences of the simple sequence repeat (SSR) within these contigs. Virtual PCR analysis showed that a fraction of these primers could be mapped onto the genomes of related species of Brassica, including Brassica rapa, B. oleraceae and B. napus. Genetic diversity analysis using a subset of 166 markers on 30 independent C. abyssinica accessions exhibited that 1) 95 % of the designed SSRs were polymorphic among these accessions; 2) the polymorphism information content (PIC) value of the markers ranged from 0.13 to 0.89; 3) the genetic distances (coefficient NEI72) between accessions varied from 0.06 to 0.36. Cluster analysis subsequent on the accessions demonstrated consistency with Crambe breeding history. F-statistics analysis revealed a moderate level of genetic differentiation in C. abyssinica (Gst = 0.3934) and a accordingly low estimated gene flow (Nm = 0.7709). Application of high-throughput sequencing technology has facilitated SSR marker development, which was successfully employed in evaluating genetic diversity of C. abyssinica as demonstrated in our study. Results showed these molecular markers were robust and provided powerful tools for assessing genetic diversity and estimating Crambe breeding history. Moreover, the SSR primers and sequence information developed in the study are freely available to the research community.

Fries) is an ideal crop for industrial oil production because of its high erucic acid content (C22:1, approx. 60 %) in its seed oil. The value of Crambe oil can be improved by increasing C22:1 content or reducing polyunsaturated fatty acids (PUFA). The FAD2 gene plays a critical role in PUFA biosynthesis. To identify targets for breeding, we characterized FAD2 in Crambe for copy number and expression profile. Seven copies of FAD2 were detected in allohexaploid Crambe. Three genes (CaFAD2-C1, -C2 and -C3) were expressed in multiple tissues, including root, seedling, leaf, flower, bud and developing seeds. In developing seeds, the expression of these genes was upregulated with CaFAD2-C3, being expressed predominantly with a peak at 20 days after pollination. This gene is therefore a promising candidate gene for determining PUFA levels in seed oil. Four other FAD2 genes were considered to be “pseudo-genes” as they harbour internal stop codons and were not expressed. Among the six Crambe varieties with consistent variation in oil composition, no nucleotide polymorphisms were found in the CaFAD2-C1, -C2 and -C3 genes. In seeds at 30 days after pollination, statistically significant expression level polymorphisms for only one gene, CaFAD2-C2, was found among the varieties. However, although significantly different, the difference in expression was small and did not explain the variation in oil composition. Given the absence of genetic variation in CaFAD2 genes in Crambe breeding lines, breeding for high erucic acid content calls for a molecular breeding approach whereby mutations are chemically induced to increase the genetic variation.

One alternative to solve the problem is energy generation using vegetal-oil-based fuels, made of local raw materials that are renewable and have a lower impact on the environment. The objective of the present work was to assess the performance and emissions of a diesel engine for energy generation that operated with soybean, linseed, and Crambe vegetal-oil-based fuels. The fuels were tested in a 5-kVA generator engine at a load from 750 W to 3000 W. Biofuels were obtained from a mixture of conventional diesel oil with soybean, linseed, and Crambe oil at incremental levels of 10%, 30%, 50%, and 70% of vegetal oil in the fuel mixture composition. The increase in soybean oil proportion at low load (750 W) resulted in lower specific consumption. At a high load (3000 W), there was a reduction in specific consumption when compared to conventional diesel fuels. Crambe biodiesel resulted in a reduction in emissions of CO, CO2, and NOx as function of the increase in Crambe oil in the proportion. The results of these studies indicate that conventional diesel blends with low (10%) and medium (30%) concentrations of soybean, linseed, and Crambe vegetal oil are viable technical solutions for the partial substitution of conventional diesel.

The objective of this work was to compare the efficiency of two solvents in different moisture contents in the extraction of peanut oil (Arachis hypogaea L.), sunflower (Helianthus annuus L.), soybean (Glycine max), Crambe (Crambe abyssinica) and canola (Brassica napus L.). The experiment was conducted in a 5 × 2×2 factorial scheme, and five grain moisture contents (9.44%, 7.76%, 6.68%, 6.82% and 7.77%) were tested for canola, Crambe, peanut, sunflower and soybean, respectively, two solvents (petroleum ether and hexane) and four repetitions, totaling 80 experimental units. The solvents presented have no influence on the content of sunflower oil and soybean, while extractions of dry grains result in an increase in the content of sunflower oil, regardless of the solvent used. The interaction between the solvent of hexane (without moisture in the grains) optimizes the extraction of canola oil. It was also verified the highest percentage of peanut oil content (43.49%) extracted with petroleum ether with moisture in the grains (6.69%). For the grains of Crambe, the humidity of (7.76%) presented the best percentage with 11.95% of oil.

Oilseed crop Crambe as a source of renewable energy in Brazil

Ex R. E. Fries) is an oilseed with the potential for cultivation in Brazil as a source of renewable energy in areas with tropical conditions. Crambe oil contains erucic acid, giving it economic importance for use in the electricity sector. Crambe oil can be used in insulating fluids and in the manufacture of chemical products and biodiesel without competing with human food production. As it is a new crop in Brazil, the scientific literature on Crambe is still scarce. This review discusses general aspects of agricultural production and the potential applications of Crambe associated mainly with the manufacture of biodiesel. The topics discussed in this review include: (i) genetic improvement of Crambe; (ii) agronomic practices of production, pests, diseases, harvesting, storage, and economic viability; (iii) industrial use of Crambe oil as an electrical insulator and in biodiesel; (iv) detoxification of the co-product generated during oil extraction for use in animal feed and use of the co-product in the natural bioremediation of contaminants; and (v) the challenges of increasing Crambe production. Crambe has a remarkable future as a source of renewable energy in Brazil. Strategies to boost its cultivation, such as the identification of improved varieties and optimization of production system logistics, marketing, and resource allocation, should be adopted.

Oil crops, such as soybeans, canola, jatropha and Crambe stand out in the west of Parana state, Brazil. The Crambe ('Crambe abyssinica' H.) is a promising plant in the production of oil in Brazil and is an alternative to the crop rotation system. Several studies have been conducted on Crambe as there is currently little information regarding this crop in the west of Parana. Also, studies on plant density are also required, as this factor is important to avoid competition for soil nutrients and reduced grain yield. This study aimed to assess Crambe growth and production under different sowing densities. The experiment was conducted on an experimental farm in the city of Cascavel - PR. The spatial arrangement proposed in this study varied according with sowing densities of 8, 12, 17.5 and 24 kg ha-1 Crambe seeds with spacing of 0.45 m between rows and 5 replications in an experimental design with randomized blocks. Data obtained were subjected to analysis of variance (ANOVA) and to regression test in ASSISTAT software. Results showed that plant height, fresh mass, dry mass, plants per linear meter and grain yield were influenced by variations in plant sowing density. Plant height (110.08 cm) and yield (2118.57 kg ha-1) showed the best results with sowing density of 8 kg ha-1. Fresh mass (67.95 t ha-1), dry mass (22.52 t ha-1) and plants per linear meter (55.60 units) reached the best results with a sowing density of 24 kg ha-1.

Crambe oil yield and soil physical properties responses to no tillage cover crops and chiseling

However, some Brassicas, such as Crambe (Crambe abyssinica Hochst), may show tolerance to moderately-compacted soils. The objective of this study was evaluate the effect of cover crops and chiseling on the soil physical properties, grain yield and oil content of Crambe. Five summer cover crops, pear millet (Pennisetum americanum), sunn hemp (Crotalaria juncea), pigeonpea (Cajanus cajan), sunn hemp (Crotalaria spectabilis), and velvet bean (Mucuna pruriens) and two soil management systems (chiseling and no-tillage) were evaluated for three years in Brazil. Chiseling soil management decreased soil bulk density and increased total porosity, but chiseling had short-term effects on soil physical properties (12 months). Cover crops showed no difference regarding the effects on soil physical properties. The traditional no-tillage system presented higher soil bulk density and lower total porosity, which did not affect Crambe grain and oil yield. Crambe presented expected grain and oil yield regardless of the adopted management and cover crops.

Two experiments were conducted in a heated greenhouse. Experiments were arranged in a completely randomized design, in a factorial, with two irrigation treatments (well-irrigated or drought stress conditions, imposed at the flowering and grain filling stages) and six soil bulk density levels (0.90, 0.99, 1.08, 1.17, 1.26, and 1.35 Mg m –3 ), with four replicates. The restriction of the water supply during the stages of flowering and grain filling affects the growth and productivity of Crambe. The roots of the Crambe plant are not significantly affected by drought conditions or the interaction between drought conditions and soil compaction. Crambe shoot growth was not significantly affected by soil compaction, at soil densities up to 1.35 Mg m –3 . Crambe is more sensitive to drought conditions

De Souza, Flavio GurgaczAbstract:The search for renewable sources of energy gains new options of raw materials every day, such as crops from agriculture. Oil crops, such as soybeans, canola, jatropha and Crambe stand out in the west of Parana state, Brazil. The Crambe ('Crambe abyssinica' H.) is a promising plant in the production of oil in Brazil and is an alternative to the crop rotation system. Several studies have been conducted on Crambe as there is currently little information regarding this crop in the west of Parana. Also, studies on plant density are also required, as this factor is important to avoid competition for soil nutrients and reduced grain yield. This study aimed to assess Crambe growth and production under different sowing densities. The experiment was conducted on an experimental farm in the city of Cascavel - PR. The spatial arrangement proposed in this study varied according with sowing densities of 8, 12, 17.5 and 24 kg ha-1 Crambe seeds with spacing of 0.45 m between rows and 5 replications in an experimental design with randomized blocks. Data obtained were subjected to analysis of variance (ANOVA) and to regression test in ASSISTAT software. Results showed that plant height, fresh mass, dry mass, plants per linear meter and grain yield were influenced by variations in plant sowing density. Plant height (110.08 cm) and yield (2118.57 kg ha-1) showed the best results with sowing density of 8 kg ha-1. Fresh mass (67.95 t ha-1), dry mass (22.52 t ha-1) and plants per linear meter (55.60 units) reached the best results with a sowing density of 24 kg ha-1.

Os graos de Crambe foram colhidos com 10,2 % de teor de agua e armazenados em sacos de papel, acondicionados em condicoes laboratoriais. As analises foram realizadas assim que os graos foram colhidos (tempo 0) e aos 4, 8 e 12 meses de armazenamento. Apos obtencao dos dados, os resultados foram submetidos a analise de variância e as medias comparadas pelo teste de Tukey (p≤0,05). Conclui-se que a quantidade de lipideos nao variou significativamente, ao longo dos 12 meses armazenamento. No entanto, levando em consideracao o aumento a pigmentacao e a degradacao dos acucares soluveis totais, sugere-se o armazenamento dos graos de Crambe ate 6 meses apos a colheita.

Entretanto, as prensas continuas com pequena capacidade, tem um grande potencial de utilizacao nas pequenas comunidades rurais, apresentam baixa eficiencia de extracao, deixando uma quantidade significativa de oleo na torta. A operacao de retirada das cascas dos frutos de Crambe, antes da prensagem, constitui-se em um fator importante que pode otimizar a extracao mecânica do oleo de Crambe, reduzindo o teor de oleo que se perde pelo processamento do grao com casca. O objetivo deste trabalho foi avaliar o rendimento de oleo de Crambe extraido mecanicamente, contendo diferentes percentagens de retirada de casca. Foram realizados os testes de rendimento de extracao do oleo pelo metodo mecânico de prensagem a frio, a partir do Crambe submetido aos seguintes tratamentos de descasque: graos sem casca, graos com 10% de casca, graos com 20% de casca e graos com casca (controle). Para avaliar o efeito do descasque foi utilizado um delineamento experimental inteiramente casualizado, com quatro tratamentos e quatro repeticoes. Os resultados foram submetidos a analise de variância e as medias comparadas pelo teste de Tukey a 5% de probabilidade. Os resultados obtidos neste trabalho permitiram observar o efeito positivo e significativo do descasque dos graos sobre a extracao do oleo de Crambe, atingindo o maximo rendimento com a ausencia de casca.PALAVRAS-CHAVE: Crambe, descasque, rendimento de extracao, prensagem mecânica, eficiencia de extracao. DEHULLING PROCESS OF Crambe [Crambe Hyspanica subesp. Abyssinica (Hochst. ex R. E. Fr.) PRINA] AIMING TO OIL EXTRACTIONABSTRACT: The mechanical screw pressing (expeller) is the method most commonly used for extracting oil from various types of oilseeds, constituting of a simple operation that requires no skilled labor and is easily adaptable to various types of grain. However, continuous presses with small capacity have great potential of use in small rural communities, and have low extraction efficiency, leaving a significant amount of oil in the Crambe cake. To remove the hull from Crambe fruits before pressing is an important factor that can optimize the mechanical extraction of Crambe oil, reducing oil waste. The purpose of this work was to evaluate the yield of mechanically extracted oil from Crambe fruits containing different percentages of hull. The oil extraction yield tests were performed by the cold pressing method, submitting the Crambe to the following hulling treatments: totally dehulled, partially dehulled with 10% hull, partially dehulled with 20% hull, and whole grains (control). To evaluate the dehulling effect, a completely randomized experimental design was used, with four treatments and four replications. The results were submitted to analysis of variance and the averages were compared by the Tukey’s test at 5% probability. The results obtained in this work allowed observing the positive and significant effect of hulling to Crambe oil extraction. Crambe seeds of FMS Brilhante cultivar were produced in the Lageado experimental farm (UNESP, Botucatu, Brazil) in 2012. The drying methods were: artificial drying with hot air, artificial drying with fresh air, natural yard drying, natural shade drying and natural field drying. The water content of seeds was monitored before, during and after drying. Germination, first germination count, abnormal seedlings, GSI, germination synchronization, frequency and germination mean time, electrical conductivity, fatty acid index and oil content were evaluated in order to know the physiological quality. The experimental design was randomized and the averages compared by the t-test at 5% significance. Results showed that artificial hot air drying favored with higher germination and vigor of Crambe seeds when compared to other drying methods and demanded shorter time to reduce the water content. The natural shade drying method is feasible for Crambe seeds despite taking longer.

Os metodos de secagem podem influenciar, dependendo do manejo adotado, a qualidade do oleo bruto extraido dos graos. O trabalho objetivou avaliar a influencia de diferentes metodos de secagem sobre a qualidade do oleo bruto extraido dos graos de Crambe. O delineamento experimental empregado foi inteiramente casualizado, com cinco tratamentos (secagem com ar aquecido, ar natural, terreiro, sombra e campo), com quatro repeticoes. Para verificar a qualidade dos graos foi realizada a analise da acidez graxa, e no oleo bruto, foram realizadas as analises de indice de iodo, indice de acidez, teor de agua, massa especifica a 20oC e viscosidade cinematica a 40oC. Apos obtencao dos dados, os resultados foram submetidos a analise de variância e quando significativo as medias foram comparadas pelo teste “t” (p≤0,05). A secagem com ar aquecido proporcionou menor indice de acidos graxos nos graos. Os metodos de secagem avaliados nao comprometeram a qualidade do oleo extraido dos graos da cultura do Crambe. Crambe (Crambe abyssinica HOCHST) GRAINS CRUDE OIL QUALITY UNDER DIFFERENT DRYING METHODS ABSTRACT: The Culture of Crambe became promising since its grains presents high oil content. The methods of drying may influence the quality of crude oil extracted from the beans, depending on the way it is managed. This study goal was to evaluate the influence of different drying methods on the quality of the crude oil extracted from Crambe grain. The experimental design was completely randomized with five treatments (drying with heated air, natural air, yard, shade, and at the field), and with four tests for each. In order to check the quality of the grains, we performed an analysis of fat acidity, and to evaluate the crude oil quality, we conducted analysis of iodine value, acid value, water content, density at 20oC, and a kinematic viscosity at 40°. After obtaining the data, the results were submitted to analysis of variance. When significant means were detected, they were compared by t-test (p ≤ 0.05). The method of drying with heated air provided a lower rate of fatty acid in grains. The drying methods evaluated did not compromise the quality of the oil extracted from the Crambe grain. Nucleus, 2013Co-Authors: Samir Paulo Jasper, Marco Antonio Martin Biaggioni, Paulo Roberto Arbex SilvaAbstract:Atualmente, a producao de biodiesel no mundo vem crescendo de forma acelerada, este interesse e demanda por biodiesel promovem um aumento da demanda por materia-prima, ou seja, lipidios. O biodiesel e um substituto do diesel de petroleo obtido pela transesterificacao, acida ou basica, dos lipidios presentes nos oleos e gorduras. O Crambe abyssinica Hochst e uma especie vegetal que tem despertado interesse dos produtores brasileiros, devido ao teor de oleo, rusticidade, cultivo mecanizado e, principalmente, por ser uma cultura de inverno tornando, assim, mais uma opcao ao agricultor neste periodo. Objetivou-se, neste trabalho, a caracterizacao fisico-quimica do oleo e do biodiesel proveniente do Crambe abyssinica Hochst, de acordo com a Resolucao n° 7 da ANP. A analise de acidos graxos do oleo do Crambe indicou elevada concentracao de acidos graxos insaturados, o que pode nao ser adequado para o uso do biodiesel em regioes muito frias, caso utilizado puro ou em misturas elevadas com oleo diesel. O biodiesel produzido a partir do Crambe abyssinica Hochst revelou estar dentro das normas estabelecidas pela Agencia Nacional do Petroleo, Gas Natural e Biocombustiveis.
