This study evaluated the levels of metallic micronutrients in Crassostrea rhizophorae (oyster) and Mytella guyanensis (mangrove mussel) collected in southern Bahia, Brazil, in a stretch of 280 km long. The metal contents (Cu, Fe, Mn and Zn) of the edible part of the species were evaluated with inductively coupled plasma optical emission spectrometry (ICP OES). Mean concentrations in mg kg-1 found for C. rhizophorae and M. guyanensis, respectively, were: Cu (2.09 ± 0.90 and 2.31 ± 0.63); Fe (32.03 ± 4.18 and 213.36 ± 51.67); Mn (1.75 ± 0.44 and 13.61 ± 2.83) and Zn (129.54 ± 25.88 and 12.13 ± 1.13). Both bivalves were relevant in the concentration of micronutrients investigated, with emphasis on the high values of Zn in C. rhizophorae and Fe in M. guyanensis.

The objective of this study was to assess the bacterial composition of the edible oyster Crassostrea rhizophorae in urban and preserved estuaries. Particularly, we assessed the presence of pathogenic bacteria, investigated antibiotic susceptibility in bacterial isolates, and quantified β-lactam antibiotic resistance genes (blaTEM, blaSHV, and blaKPC) via quantitative PCR of oyster DNA. Our results detected total coliforms, Escherichia coli, and enterobacteria in the oysters from urban estuaries, which is indicative of poor water quality. In addition, our detection of the eaeA and stxA2 virulence genes in 16.7% of E. coli isolates from oysters from this region suggests the presence of multiantibiotic-resistant enteropathogenic and enterohemorrhagic E. coli strains. Foram analisadas as ostras Crassostrea brasiliana de um cultivo em espinhel e C. rhizophorae de um manguezal adjacente, na localidade de Porto do Campo, Baia de Camamu, Bahia. As coletas (n = 100) foram efetuadas em outubro de 2012. Em laboratorio, as ostras foram medidas e abertas para a retirada da carne, que foi macerada em gelo seco. Para a extracao das proteinas, foi adotada a adaptacao de um protocolo utilizado para mexilhoes, apos o que foi realizada a separacao na primeira dimensao, por focalizacao isoeletrica (IEF). Os peptideos foram transferidos para um Espectrometro de Massas. Os espectros obtidos foram analisados no software ProteinLynx Global Server 4.2 e tambem pela ferramenta MASCOT (Matrix Science) e comparados com os bancos de dados do SWISSPROT e do NCBI, respectivamente. A identificacao foi evidenciada por meio da beta-tubulina, homologia Perkinsus marinus ATCC 50983 e codigo da proteina no banco de dados NCBI = gi|294889481. Este e o primeiro registro de P. marinus na Bahia e o quarto no Brasil.

Fragments of gills and rectum of the oysters (n = 150) were incubated in Ray’s fluid thioglycollate medium (RFTM). Genus Perkinsus-specific PerkITS85/750 PCR assays were performed and their amplicons were sequenced by the Sanger method. The RFTM assays confirmed Perkinsus spp. The sequencing of the amplified fragments from the rDNA internal transcribed spacers (ITS) of Perkinsus spp. confirmed Perkinsus chesapeaki. Neighbor-Joining analyzes place P. chesapeaki identified in this study in a well-supported clade with other isolates of the same species. This is the first record of P. chesapeaki infecting C. rhizophorae in South America.

Brazilian Journal of Veterinary Research and Animal Science, 2016Co-Authors: Mariane Dos Santos Aguiar Luz, Guisla BoehsAbstract:This study investigated the infection of Perkinsus beihaiensis (Perkinsozoa) in the oyster Crassostrea rhizophorae, both from a long-line cultivation system and from a nearby intertidal zone of mangrove, both in the state of Bahia, northeastern Brazil. The collections were performed in October and November 2012, and in January 2013. The oysters (n = 300) were measured, examined macroscopically for signs of infection and then submitted to the following laboratory techniques: histology, Ray's fluid thioglycollate medium assay (RFTM), polymerase chain reaction (PCR) and sequencing, which confirmed the identification of the pathogen. Histological and RFTM analyses showed, respectively, a mean prevalence of 93.3% and of 69%. The infection was usually mild or very mild. There was no significant difference (p > 0.05) between the environments in terms of infection prevalence or severity. This is the first record of P. beihaiensis in the state of Bahia and the second in oysters from Brazil and South America.

Even though, many studies have been carried out to establish its culture, there has not been established which and how environmental factors influence its growth under culture condition. Spats with a mean size of 30 mm were suspended in “Spanish baskets” at a depth of 4 m during 6 months. The experiment was carried on at La Restinga lagoon, Margarita Island, Venezuela. Each month oysters were measured (ventral-dorsal and antero-posterior), and weighted (dry weight from conch, tissue and fouling). Survivorship was also evaluated. Water temperature was recorded continuously and weekly measurements of Secchi depth, salinity, oxygen, total, organic and inorganic seston, and chlorophyll a were determined. Results show 89% increase in ventral-dorsal size reaching sizes of 54.5 mm. Dry weight of tissue at the end of the study period was 0.48 g after a decrease during the warmer months of august and september, indicating temperature as an important factor controlling spawning. The high and constant phytoplanktonic biomass present in La Restinga lagoon showed that food availability was not a limiting factor, which together with the small variability in the studied environmental variables, made impossible to identify factors controlling growth and survival of Crassostrea rhizophorae in long-line suspended culture. The exception was the occasional predation by Cymatium snails.

Effects of mass and position of artificial fouling added to the upper valve of the mangrove oyster Crassostrea rhizophorae on its growth and survival

HimmelmanAbstract:Abstract We ran an experiment on mangrove oysters Crassostrea rhizophorae to evaluate the effects of adding different masses of artificial fouling to the upper valve, either to the umbo region or the ventral edge of the shell. Growth and survival were quantified after a 30 d period in suspended culture in the La Restinga Lagoon, Venezuela. The artificial fouling was cement weighing 0.5, 1, 2 or 3 fold the mass of the upper valve. No fouling was added to a control group. Fouling mass, but not the position of the artificial fouling, affected growth in shell length. However, only the heaviest fouling (3 times the mass of the upper valve) had a significant effect. In contrast, there was no affect of either fouling mass or position on tissue growth. Finally, our data indicated that mortality could be affected by the position where we added artificial fouling (greater mortality when fouling was added to the ventral edge of the shell), but not by fouling mass. Our study indicates it is unlikely that the levels of natural fouling that develop on oysters in suspended culture would be sufficient to affect either growth or survival.

2006Co-Authors: César Lodeiros, Esperanza Buitrago, Alejandro Guerra, Isla MargaritaAbstract:We evaluated the growth and survival of the mangrove oyster Crassostrea rhizophorae (30 mm maximum length) reared in two suspended-culture systems (raft and long-line) using three types of enclosures (oyster enclosures, lantern nets and enclosures based on crates used to transport fish), at a density of 750 ind m –2 , in La Restinga Lagoon, Margarita Island, Venezuela, over the course of six months. No significant differences were found in shell growth (length and mass) due to the system and enclosures used; however, raft-cultured oysters showed better tissue growth. At the end of the study, the oysters in lantern nets suspended from rafts obtained a tissue mass that was around 40% higher than that of the rest of the oysters cultured in the other enclosures. Significant differences in oyster survival were not observed for the raft and long-line systems used, but the cumulative survival at the end of the study was significantly higher for the oyster enclosures (52–56%) than for the lantern nets (36–39%) and fish crates (24–25%). We therefore recommend employing rafts and discarding the use of crate-type enclosures in the culture of C. rhizophorae in lagoon systems.

Health assessment of the oyster Crassostrea rhizophorae on the southern coast of Bahia, northeastern Brazil

A total of 180 oysters (30/sampling point/season) were examined macroscopically for the presence of pathogens and anatomical changes. The specimens were subsequently fixed in Davidson solution, processed for paraffin embedding, sectioned and stained with Harris' hematoxylin and eosin. Histological analysis revealed the presence of Rickettsia-like organisms (RLOs), Ancistrocoma, Trichodina, Sphenophrya, Nematopsis, Urastoma, Bucephalus in the sporocyst phase, a nonspecific metacercaria, and a metacestode of genus Tylocephalum. The prevalence of infection was low except for parasitism by Nematopsis sp. which also caused histopathological changes. The presence of Bucephalus sp. caused parasitic castration. First report of the protozoan parasite Perkinsus marinus in South America, infecting mangrove oysters Crassostrea rhizophorae from the Paraíba River (NE, Brazil)

Perkinsosis was detected by incubation of oyster gill pieces in Ray's fluid thioglycollate medium. The monthly prevalence values were all above 70%, thus infection was not likely to be a transient event. Perkinsus sp. parasites isolated from eight oysters were propagated in vitro. PCR-RFLP analysis of in vitro cultured cells as well as the sequences of the rDNA ITS region allowed the identification of the in vitro propagated parasites as Perkinsus marinus. Phylogenetic analyses using rDNA ITS region sequences strongly supported the Perkinsus sp. from Paraiba in a monophyletic group with P. marinus. Thus, the results confirmed the species affiliation of Paraiba Perkinsus sp. as P. marinus. This is the first report of P. marinus in Brazil and South America and the first report of P. marinus naturally infecting C. rhizophorae.

The oysters (n = 900) were collected in February-March and July-August 2010. The Ray's fluid thioglycollate medium (RFTM) analysis of gills and rectum revealed hypnospores of Perkinsus sp. with a high mean prevalence (63%). The infection intensity varied from very light to advanced. The polymerase chain reaction confirmed Perkinsus in 87.2% of the RFTM-positive oysters. Histological analysis showed trophozoites and schizonts phagocytized by hemocytes, mainly in the intestine and the stomach epithelium.

Despite the socioeconomic importance of this natural resource, there are few studies on the health of the oysters in this region. This study aimed to survey pathological changes in the mangrove oyster C. rhizophorae in the estuary of the Pacoti River, CE. Adult oysters were collected in August 2008 (N = 450) and December 2009 (N = 450) at three sites of the Pacoti estuary and in 2010 (N = 600) samplings were done quarterly at one site which has showed the higher prevalence de Perkinsus. Macroscopical and histological analyses were used to evaluate pathological changes, Ray’s Fluid Thioglycollate Medium (RFTM) to detect Perkinsus spp. and polymerase chain reactions (PCR) and DNA sequencing to identify Perkinsus species. In 2009, RFTM assay detected Perkinsus sp. infecting the tissues of C. rhizophorae with low prevalences of 1.3%, 6.7% e 7.3% in sites 1, 2 and 3, respectively, and in 2010, in site 3, prevalence was 2% (12 of 600 oysters). PCR did not confirm any positive case in 2009 and only 5 in 2010. The phylogenetic analyses strongly indicate that the Perkinsus species infecting oysters C. rhizophorae of this study belongs to Perkinsus beihaiensis. The histology confirmed 11 cases of Perkinsus sp. infecting the C. rhizophorae in 2009, and only two cases in 2010. Nematopsis sp. was the protozoan observed with greater prevalence (up 96.7%). Other found protozoa were: Trichodina, Sphenophrya, Ancistrocoma – like and an unknown ovarian parasite. The metazoa found were the polychaete Polydora with high prevalences, a turbellarian, possibly of the genus Urastoma, an unidentified digenean metacercariae and larvae of cestode Tylocephalum. A continuous monitoring of diseases in bivalves from this natural population is recommended, since the phylogenetic analyses indicate the occurrence of P. beihaiensis infecting oysters C. rhizophorae whose pathogenic potential is unknown.

In the present study we describe for the first time a parasite of the genus Perkinsus infecting the mangrove oyster Crassostrea rhizophorae from the Brazilian coast. Prevalence of this parasite was low in the Pacoti River estuary (Ceara, northeast Brazil) and absent in oysters from southern Brazil. Oyster gill and rectum tissues incubated in Ray's fluid thioglycollate medium (RFTM) revealed the presence of spherical hypnospores (5 to 55 microm diam.). Histological analysis showed the occurrence of typical signet-ring trophozoites and schizonts (3 to 6 microm diam.) infecting connective tissues of several organs and digestive epithelia. PCR assays specific to the genus Perkinsus, followed by cloning and sequencing of the internal transcribed spacer (ITS) region of the ribosomal ribonucleic acid (rRNA) gene complex, confirmed a close phylogenetic relationship between Brazilian Perkinsus sp. and P. beihaiensis infecting Chinese oysters.

The model of growth in length was represented by the equation Lt = 103.41 [1 - exp (-1.59 (t-0, 31))], showing a fast growth which allows the oyster to reach commercial size (60 mm ) at 6 months, with a maximum estimated longevity of 2 years

Para ello se realizaron muestreos mensuales y aleatorios de las ostras. La composicion anual de talla estuvo integrada por organismos de 15 a 95 mm de longitud total, con un mayor numero de individuos en el intervalo de 45 y 60 mm. La relacion talla-peso presento un coeficiente de correlacion r = 0,66 y una pendiente b = 1,80, senalando que la especie tiene un crecimiento alometrico minorante. El modelo de crecimiento en longitud estuvo representado por la ecuacion Lt = 103,41 [1 - exp (-1,59 (t-0,31))], mostrando un crecimiento acelerado alcanzando a los 6 meses la talla comercial (60 mm), con una longevidad maxima estimada de 2 anos. Each month oysters were measured (ventral-dorsal and antero-posterior), and weighted (dry weight from conch, tissue and fouling). Survivorship was also evaluated. Water temperature was recorded continuously and weekly measurements of Secchi depth, salinity, oxygen, total, organic and inorganic seston, and chlorophyll a were determined. Results show 89% increase in ventral-dorsal size reaching sizes of 54.5 mm. Dry weight of tissue at the end of the study period was 0.48 g after a decrease during the warmer months of august and september, indicating temperature as an important factor controlling spawning. The high and constant phytoplanktonic biomass present in La Restinga lagoon showed that food availability was not a limiting factor, which together with the small variability in the studied environmental variables, made impossible to identify factors controlling growth and survival of Crassostrea rhizophorae in long-line suspended culture. The exception was the occasional predation by Cymatium snails.

Effects of mass and position of artificial fouling added to the upper valve of the mangrove oyster Crassostrea rhizophorae on its growth and survival

HimmelmanAbstract:Abstract We ran an experiment on mangrove oysters Crassostrea rhizophorae to evaluate the effects of adding different masses of artificial fouling to the upper valve, either to the umbo region or the ventral edge of the shell. Growth and survival were quantified after a 30 d period in suspended culture in the La Restinga Lagoon, Venezuela. The artificial fouling was cement weighing 0.5, 1, 2 or 3 fold the mass of the upper valve. No fouling was added to a control group. Fouling mass, but not the position of the artificial fouling, affected growth in shell length. However, only the heaviest fouling (3 times the mass of the upper valve) had a significant effect. In contrast, there was no affect of either fouling mass or position on tissue growth. Finally, our data indicated that mortality could be affected by the position where we added artificial fouling (greater mortality when fouling was added to the ventral edge of the shell), but not by fouling mass. Our study indicates it is unlikely that the levels of natural fouling that develop on oysters in suspended culture would be sufficient to affect either growth or survival.

2006Co-Authors: César Lodeiros, Esperanza Buitrago, Alejandro Guerra, Isla MargaritaAbstract:We evaluated the growth and survival of the mangrove oyster Crassostrea rhizophorae (30 mm maximum length) reared in two suspended-culture systems (raft and long-line) using three types of enclosures (oyster enclosures, lantern nets and enclosures based on crates used to transport fish), at a density of 750 ind m –2 , in La Restinga Lagoon, Margarita Island, Venezuela, over the course of six months. No significant differences were found in shell growth (length and mass) due to the system and enclosures used; however, raft-cultured oysters showed better tissue growth. At the end of the study, the oysters in lantern nets suspended from rafts obtained a tissue mass that was around 40% higher than that of the rest of the oysters cultured in the other enclosures. Significant differences in oyster survival were not observed for the raft and long-line systems used, but the cumulative survival at the end of the study was significantly higher for the oyster enclosures (52–56%) than for the lantern nets (36–39%) and fish crates (24–25%). We therefore recommend employing rafts and discarding the use of crate-type enclosures in the culture of C. rhizophorae in lagoon systems.
