    Abstract:

    Climate change can benefit individual species, but when pest species are enhanced by warmer temperatures agricultural productivity may be placed at greater risk. We analyzed the effects of temperature anomaly on arrival date and infestation severity of potato leafhopper, Empoasca Fabae Harris, a classic new world long distance migrant, and a significant pest in several agricultural crops. We compiled E. Fabae arrival dates and infestation severity data at different states in USA from existing literature reviews and agricultural extension records from 1951–2012, and examined the influence of temperature anomalies at each target state or overwintering range on the date of arrival and severity of infestation. Average E. Fabae arrival date at different states reveal a clear trend along the south-north axis, with earliest arrival closest to the overwintering range. E. Fabae arrival has advanced by 10 days over the last 62 years. E. Fabae arrived earlier in warmer years in relation to each target state level temperature anomaly (3.0 days / °C increase in temperature anomaly). Increased temperature had a significant and positive effect on the severity of infestation, and arrival date had a marginal negative effect on severity. These relationships suggest that continued warming could advance the time of E. Fabae colonization and increase their impact on affected crops.

    Abstract:

    The potato leafhopper, Empoasca Fabae, is a pest of economic crops in the United States and Canada, where it causes damage known as hopperburn. Saliva, along with mechanical injury, leads to decreases in gas exchange rates, stunting and chlorosis. Although E. Fabae saliva is known to induce plant responses, little knowledge exists of saliva composition at the molecular level. We subjected the salivary glands of E. Fabae to Roche 454-pyrosequencing which resulted significant number (30,893) of expressed sequence tags including 2805 contigs and 28,088 singletons. A high number of sequences (78%) showed similarity to other insect species in GenBank, including Triboliumcastaneum, Drosophilamelanogaster and Acrythosiphonpisum. KEGG analysis predicted the presence of pathways for purine and thiamine metabolic, biosynthesis of secondary metabolites, drug metabolism, and lysine degradation. Pfam analysis showed a high number of cellulase and carboxylesterase protein domains. Expression analysis of candidate genes (alpha amylase, lipase, pectin lyase, etc.) among different tissues revealed tissue-specific expression of digestive enzymes in E. Fabae. This is the first study to characterize the sialotranscriptome of E. Fabae and the first for any species in the family of Cicadellidae. Due to the status of these insects as economic pests, knowledge of which genes are active in the salivary glands is important for understanding their impact on host plants.

    Behavior and reproduction of potato leafhopper, Empoasca Fabae (Harris) (Homoptera: Cicadellidae), were examined in the laboratory in response to varying densities of leafhoppers, a host legume (alfalfa, Medicago sativa L.), and a non-host grass (large crabgrass, Digitaria sanguinalis L.). A series of caged treatments with increasing crabgrass density per cage resulted in a corresponding series of decreasing oviposition and decreasing number of primary oocytes per female. Oviposition and number of primary oocytes were reduced by approximately 30% at the highest grass density. However, adult activity as measured by percentage of leafhoppers observed on alfalfa, in flights per minute, and on sides of cage, remained unchanged with increasing crabgrass density. When responses given at crabgrass densities were compared with pure alfalfa of equivalent vegetation density (plant surface area), activity in the presence of crabgrass was two to four times larger than alfalfa alone of equivalent vegetation density, w...
