    Abstract:

    Objetivo:Comparar el efecto in vitro del aceite esencial de Eucalyptus Globulus con el aceite esencial de Origanum vulgare sobre el crecimiento de Candida albicans aislada de paciente con candidiasis vulvovaginal. Material y métodos:Candida albicans aislada de paciente con candidiasis vulvovaginal para las pruebas in vitro.La actividad antifúngica se determinó por el método Kirby y Bauer o de difusión de discos; la concentración inhibitoria mínima (CIM) y la concentración mínima fungicida (CMF) mediante el conteo de las unidades formadoras de colonias (UFC). Principales medidas de resultados: Halos de inhibición, CIM, CMF. Resultados: se determinó que la diferencia de las medias entre los diámetros de los halos de inhibición de las concentraciones al 25%, 50%, 75% y 100% del aceite esencial de Eucalyptus Globulus y del aceite esencial de Origanum vulgare es significativa, ya que p valor .0000, siendo menor que el nivel de significancia (p < 0.05), con lo que se determinó que el primero presentó menor efecto inhibitorio que el segundo. Se determinó que la CMI del aceite esencial de Eucalyptus Globulus fue de 50% y del aceite esencial de Origanum vulgare fue de 25%, en los cuales, se observó ausencia de turbidez. Se observó que solamente el aceite esencial de Origanum vulgare presentó a partir de la concentración 75% (UFC = 0), el cual fue considerado como la concentración mínima fungicida (CMF).     Este estudio se realizó con el objetivo de evaluar el efecto antimicótico del aceite esencial de Eucalyptus Globulus sobre Trichophyton rubrum, Microsporum canis y Epidermophyton floccosum, comparado con Clotrimazol, in vitro. Se planteó un diseño experimental que tuvo como muestra 10 cultivos de cada uno de los dermatofitos. El aceite esencial se obtuvo por arrastre con vapor de agua y se utilizó al 100%. Se evaluó el efecto del aceite esencial de Eucalyptus Globulus in vitro, mediante el método de difusión con pozos en agar Sabouraud.     Abstract Eucalyptus Globulus Labill. is managed in commercial plantations for the pulp and paper industry as a coppice system. For stand reforestation the stumps are uprooted and at present used as biofuel. In this study, the biomass of stumps collected in six eucalypt stands in Portugal was evaluated in terms of physical, chemical and fuel properties. Stumps had basic density between 0.55 g cm−3 and 0.71 g cm−3, and the chemical composition showed 15.1% total extractives (mainly ethanol soluble extractives, 10.5%), 24.8% lignin and 67.0% holocellulose. In terms of thermal properties the stumps presented 18.9 MJ kg−1 for higher heating value (HHV) and 0.6%, 81.1% and 18.3% respectively for ash, volatile matter and fixed carbon. The elemental composition (C = 51.0%; H = 5.8%; O = 45.3%; Cl = 0.04%) has adequate values for use as woodfuel.

    The variation in extractives content in sapwood and heartwood was investigated among 12 trees in each of four commercial plantations of Eucalyptus Globulus in central Portugal. The study was carried out at the 15% height level and extractions used successively dichloromethane, ethanol and water. At all sites, heartwood had significantly more extractives than sapwood, on average 3.8 and 2.4%, respectively. Most extractives consisted of ethanol soluble material (on average 52% of total extractives). Among the sites, there was a statistically significant difference in the content of extractives but the most important source of variation was the within-tree variation between sapwood and heartwood. Differences in the content of extractives were also observed among trees. A strong relation between extractives content and heartwood proportion was found. The potential loss of pulp yield and problems associated with accumulation of extractives are directly related to the heartwood proportion in the eucalypt stems. Forest management should take into account heartwood development and selection for minimising heartwood extractives.

    Wood properties relevant for pulping were measured in Eucalyptus Globulus trees from plantations with different spacings (3x2, 3x3, 3x4, 4x4 and 4x5 m), at 18 years of age, on discs taken at breast height. The wood density varied from 565kg/m 3 to 594 kg/m 3 , respectively, in the closer (3x2) and the wider (4x5) spacings. There was no effect of spacing on mean fibre length (on average 1.02 mm), fibre width (21.3m) and wall thickness (5.7m). The wood chemical composition was similar for all spacings except for extractives, which increased from 5.4% to 8.7% with increasing spacing. Pulp yield (% on o.d. wood) in the closer spacings was higher and with lower residual lignin than in wider spacings (e.g. 57.0% and 54.1% at Kappa 15 for 3x2 and 4x5, respectively). The chemical quality of wood therefore decreased in the wider spacings and management for pulping should take this into account.

    The effects of hydrothermal treatments of Eucalyptus Globulus wood have been evaluated in terms of the solid yield and the composition of the treated wood. The contents of cellulose, xylan, and Klason lignin in treated solids were determined and interpreted by means of kinetic models. Particular attention was paid to the degree of polymerization (DP) lowering of cellulose caused by hydrothermal treatments, which was assessed by measurements of intrinsic viscosity.

    Abstract:

    Xylose solutions have been produced from Eucalyptus Globulus wood by autohydrolysis (with hot, compressed water) and post-hydrolysis (in presence of sulfuric acid). This two-stage process led to solids enriched in cellulose and lignin (suitable as a substrate for pulping) and liquors containing xylose as the major component. The liquid phase from post-hydrolysis also contained other sugars (glucose, arabinose) and acetic acid. Neutralized liquors (as obtained, or after membrane concentration), were employed (directly or after detoxifi cation by ion exchange) as fermentation media for the production of hemicelluosic bioethanol with the yeast Pichia stipitis CECT 1922T. Under the best conditions assayed (fermentation of neutralized, concentrated and detoxifi ed two-stage hydrolysis liquors), bioconversion took place at nearly stoichiometric yield, with a volumetric productivity of 0.37 g l -1 h -1 .

The purified XOS product was assayed for composition and characterised by HPLC-RI, HPAEC-PAD, HPSEC, MALDI-TOF-MS and NMR techniques. The results suggested the presence of neutral and acidic XOS with a degree of acetylation of about 0.6. The fermentability of the refined XOS product by faecal inocula was assessed by measuring both substrate consumption and formation of short-chain fatty acids.

    Abstract This work provides an assessment on the fractionation of Eucalyptus Globulus wood by sequential stages of autohydrolysis (to cause the solubilization of hemicelluloses) and organosolv pulping (to dissolve lignin, leaving solids enriched in cellulose). With this approach, valuable products (hemicellulose-derived saccharides, sulphur-free lignin fragments and cellulosic substrates with low contents of residual hemicelluloses) are obtained in separate streams, according to the biomass refinery approach. Autohydrolysis was carried out under optimized operational conditions, and organosolv pulping was performed using uncatalyzed ethanol–water solutions. The effects of the most influential operational variables (autohydrolysis severity, delignification temperature and ethanol concentration in the organosolv stage) on solid yield, solid composition, cellulose susceptibility and recovery of the various fractions was assessed using statistical methods, which enabled the identification of the most favourable operational conditions.
