On the basis of morphological characters and phylogenetic evidence of DNA sequences of multiple loci including the internal transcribed spacer (ITS) regions, the large subunit nuclear ribosomal RNA gene (nLSU), the small subunit nuclear ribosomal RNA gene (nSSU), the small subunit mitochondrial rRNA gene sequences (mtSSU), the translation elongation factor 1-α gene (tef1) and the second subunit of RNA polymerase II (rpb2), six new genera, Fragifomes, Niveoporofomes, Piptoporellus, RhodoFomitopsis, Rubellofomes and Ungulidaedalea are established. Four new species, Buglossoporus eucalypticola, Daedalea allantoidea, Piptoporellus hainanensis and P. triqueter are descibed from China. Illustrated descriptions of the novel species are provided. Identification keys to Fomitopsis and its related genera, as well as keys to the species of each genus are provided.

This fungus is characterized by perennial, effused-reflexed to pileate basidiomes, concentrically sulcate pileal surface, pinkish brown to vinaceous brown pore surface, fusoid cystidioles, small, oblong-ellipsoid basidiospores (4–5 × 1.9–2.5 μm), and occurrence on gymnosperms. Morphologically, Fomitopsis subfeei is similar to F. feei and F. incarnata, but distinct from the latter two species phylogenetically. Phylogeny based on the internal transcribed spacer regions and nuclear large subunit ribosomal RNA gene regions supported Fomitopsis subfeei as a distinctive new species in Fomitopsis.

Fomitopsis cystidiata sp. nov. is unique due to its annual and conchoidal to flabelliform basidiomata, orange brown to fuscous and concentrically zonate pileus and purple pink pore surface when fresh, large angular pores (0.5–1 per mm), thick-walled cystidia and cylindrical to oblong-ellipsoid basidiospores at 5–6.2 × 2–2.8 μm. Fomitopsis fragilis sp. nov. is characterized by its annual, ungulate and fragile basidiomata with a dark brown crust, large angular pores (1–2 per mm), densely septated skeletal hyphae, and oblong-ellipsoid basidiospores at 4–5.2 × 2.2–2.8 μm. Phylogenetic analysis based on ITS+nLSU rRNA gene regions suggested two new species belonging to Fomitopsis sensu lato of the Antrodia clade.

Both species have annual, effused-reflexed basidiomata with several small imbricate pilei protruding from a large resupinate part. F. cana is characterized by its mouse-grey to dark grey basidiomata, pores 5–8 per mm, and small cylindrical to oblong-ellipsoid basidiospores (5–6.2 × 2.1–3 μm). F. subtropica is characterized by its white, cream, straw-yellow to more or less flesh-pink basidiomata which was easily separable from the substrate, smaller pores (6–9 per mm) and smaller basidiospores (3.2–4 × 1.8–2.1 μm), and presence of yellowish oil-like substances in trama. Phylogenetic analysis based on combined ITS and nLSU sequences suggest that the two new species belong in the Fomitopsis sensu stricto group within the Antrodia clade.

The structures of the compounds were elucidated based on their spectral properties. All triterpenes demonstrated antibacterial activity against S. aureus but possessed no antiradical activity against DPPH radicals.

Carbohydrate Polymers, 2014Co-Authors: Deepa Deswal, Preeti Nandal, Rishi Gupta, Ramesh Chander KuhadAbstract:Abstract The sugarcane bagasse was biologically pretreated with three white-rot fungi; Pleurotus florida, Coriolopsis caperata RCK 2011 and Ganoderma sp. rckk-02, individually under solid-state fermentation. P. florida, C. caperata RCK 2011 and Ganoderma sp. rckk-02 degraded lignin up to 7.91, 5.48 and 5.58%, respectively. The lignocellulolytic enzymes produced by these fungi were also monitored during solid state fermentation of sugarcane bagasse. The fungal fermented sugarcane bagasse when hydrolyzed with crude cellulases from brown-rot fungus, Fomitopsis sp. RCK2010, released comparatively 1.5–2.4 fold higher sugars than in case of untreated sugarcane bagasse. The study demonstrated that white-rot fungal pretreatment improved the amenability of plant material for enzymatic hydrolysis.

Enhanced exoglucanase production by brown rot fungus Fomitopsis sp. RCK2010 and its application for cellulose saccharification.

RCK2010 was optimized under solid-state fermentation using Plackett–Burman design (PBD) and response surface methodology (RSM). Four fermentation variables (moisture, inoculum level, casein, and Triton X-100) were identified to effect cellulase production significantly by PBD, which were further optimized using RSM of central composite design. An overall 130 % increase in enzyme production was achieved by the optimization of variables using statistical approaches. Moreover, crude cellulase from Fomitopsis sp. RCK2010 was applied to saccharify pretreated Prosopis juliflora (cellulosic fraction), which resulted in the release of 327.35 mg/g of reducing sugars that could further be utilized for bioethanol production.

RCK2010 were optimized under solid state fermentation. An initial pH of 5.5 and moisture ratio of 1:3.5 (solid:liquid) were found to be optimal for maximum enzyme production. Of the different carbon sources tested wheat bran gave the maximum production of CMCase (71.526 IU/g), FPase (3.268 IU/g), and β-glucosidase (50.696 IU/g). Among the nitrogen sources, urea caused maximum production of CMCase (81.832 IU/g), where as casein and soyabean meal gave the highest FPase (4.682 IU/g) and β-glucosidase (69.083 IU/g) production, respectively. Among amino acids tested glutamic acid gave the highest production for CMCase (84.127 IU/g); however 4-hydroxy- l -proline stimulated maximum FPase production (6.762 IU/g). Saccharification of pretreated rice straw and wheat straw by crude enzyme extract from Fomitopsis sp. RCK2010 resulted in release of 157.160 and 214.044 mg/g of reducing sugar, respectively.

Here, we present the genome sequence of Fomitopsis pinicola GR9-4 which represent different F. pinicola clade than the previously sequenced North American isolate FP-58527 SS1. The genome was sequenced by using a paired-end sequence library of Illumina and a 2.5k and 5k mate-pair library (ABI SOLiD). The final assembly adds up to a size of 45 Mb (including gaps between contigs), with a GC-content of 56%. The gene prediction resulted in 13,888 gene models. The genome sequence will be used as a basis for understanding population genomics, genome-wide association studies and wood decay mechanisms of this brown-rot fungus.

Population structure of the wood decay fungus Fomitopsis pinicola
Heredity, 1999Co-Authors: Nils Hogberg, Ottmar Holdenrieder, Jan StenlidAbstract:Three populations of the wood decay fungus Fomitopsis pinicola, one from each of three countries (Sweden, Russia and Lithuania), were studied by means of arbitrary primed PCR. The genetic structure of the populations was assessed by inferring the genotype of the genets by studying the haplotypes of several single-spore isolates from one sporocarp for each individual. Heterozygotes could therefore be detected with a dominant genetic marker. The amplified band and the null allele of all loci segregated in a way that was in agreement with a 50:50 ratio. Genetic analysis showed that the total population as well as the subpopulations had heterozygote frequencies in agreement with Hardy–Weinberg expectations. No population differentiation was detected in spite of large geographical distances among the populations studied. We also compared the methods of somatic incompatibility and AP-PCR in terms of their value in detecting fungal genets. This was tested for a sample of dikaryotic mycelia from Switzerland. For the tested material the two methods gave congruent results.
