preliminary phytochemical and physicochemical characterization of Gynura segetum lour merr compositae leaf

Methods: Preliminary phytochemical and physicochemical studies using chromatographic techniques,were carried out, Parameters evaluated include ash value, loss on drying and extractive value, amongst others. . Results: Phytochemical results revealed that the leaves of Gynura segetum contain alkaloid, terpene, flavonoid, tannin and saponin. Mean ash value (%) was 13.5 (total), 2.49 (acid-insoluble ash) and 6.14 (water-soluble ash). Loss on drying was 10.70 % while extractive value was 20.60 % in water and 14.13 % in ethanol. Thin layer chromatography (TLC) analysis indicates the presence of rutin in the methanol extract of G. segetum and this was supported by high performance liquid chromatography (HPLC) studies. Conclusion: Useful quantitative and descriptive data essential for identifying and characterizing the plant for the purpose of quality control are presented.

(Compositae) is a well recognized medicinal plant in Indonesia and Malaysia. It is believed to have an anticoagulant effect and is used in treating snake-bites, inflammations and other skin afflictions. This study was carried out to evaluate the antimicrobial effects of Gynura segetum leaves extracts and its fractions. The chemical compositions of the active extracts were also determined. The antimicrobial activities of different solvent extracts of leaves of Gynura segetum were evaluated using the agar well-diffusion method. The Minimum Inhibitory Concentration (MIC) of the active subfractions was determined by the tube dilution method. Gas Chromatography-mass spectrometry (GC-MS) analysis was carried out to identify the chemical compositions of the active extracts. The ethyl acetate fraction and its subfraction E4 performed potent antimicrobial activities and fifteen known chemical constituents were identified by GCMS analysis as 4-vinylphenol, 1-tetradecene, phenol, 2,4-bis(1,1-dimethylethyl), 1-hexadecene, E-15-heptadecenal, hexadecanoic acid, 1,2-benzenedicarboxylic acid, dibutyl ester, 1-docosene, octadecanoic acid, 1-eicosene, cyclotetracosane, 1,2-benzenedicarboxylic acid, bis(2-ethylhexyl)ester, butanedioic acid, monomethyl ester, niacin and 4-hydroxy-benzoic acid. The results of this study suggested a connection between the antimicrobial activities and the chemical structures. The plant may be used as a potential source for antimicrobial agents.

Aim of the study This study was carried out to evaluate the anti-angiogenic effect of Gynura segetum leaves extracts and its fractions. The chemical compositions of the active extracts were also determined. Materials and methods The anti-angiogenic activity of Gynura segetum leaves extracts and its fractions was evaluated in vivo using the chick embryo chorioallantoic membrane (CAM) assay. Gas chromatography–mass spectrometry (GC–MS) analysis was carried out to identify the chemical compositions of the active extracts. Results The CAM treated with Gynura segetum leaves extracts and its fractions (100 μg/disc) showed a significantly greater anti-angiogenic effect compared to the positive control suramin (50 μg/disc). Chemical analysis of the active extracts from the leaves of Gynura segetum yielded nine known compounds: undecane (1), neophytadine (2), hexadecanoic acid, methyl ester (3), 9,12-octadecadienoic acid, methyl ester (4), 9,12,15-octadecatrienoic acid, methyl ester (5), phytol (6), tetradecanal (7), octadecanoic acid, methyl ester (8) and γ-sitosterol (9). Conclusions These results suggested that Gynura segetum has anti-angiogenic activity. The plant may be used as a potential source for protection against cancer.

However, there is little toxicological information available regarding safety following repeated exposure. The present investigation describes the toxicity of a methanol extract of Gynura procumbens leaves in rats. Materials and methods For acute toxicity studies, a methanol extract of Gynura procumbens was orally administered to Sprague–Dawley rats (female and male) at a dose range of 1000–5000 mg/kg. For sub-chronic toxicity studies, the rats were orally administered the methanol extract of Gynura procumbens at the doses of 125, 250, and 500 mg/(kg day) for a period of 13 weeks. The animals were sacrificed, followed by examination of their organs and blood serum. Results and conclusions Administration of the methanol extract from Gynura procumbens leaves at 1000–5000 mg/kg did not produce mortality or significant changes in the general behaviour, body weight, or organ gross appearance of rats. There were no significant differences in the general condition, growth, organ weights, haematological parameters, clinical chemistry values, or gross and microscopic appearance of the organs from the treatment groups as compared to the control group. Therefore, the NOAEL for the Gynura procumbens methanol extract is 500 mg/(kg day) administered orally for 13 weeks.

The aim of this retrospective study was to identify independent prognostic markers for survival in patients with Gynura segetum-induced HVOD and to evaluate the effect of anticoagulants and transjugular intrahepatic portosystemic shunt (TIPS) on survival rate. Clinical data including symptoms, signs, imaging characteristics, laboratory test results, results of liver tissue biopsies, type of treatment during follow-up and clinical outcomes were collected. Univariate, multivariate and time-dependent Cox regression analyses were performed. Survival rates were 91% (95% confidence interval [CI], 82–95%), 64% (95% CI, 53–69%) and 57% (95% CI, 51–65%) at 1, 3 and 60 months, respectively. Total bilirubin, albumin and hepatic encephalopathy were independent prognostic markers of survival. Anticoagulants were administered to 76% of the patients. Among 75 patients treated with anticoagulants, 49 patients (65.3%) were cured, whereas 26 patients (34.7%) died; the cure rate in anticoagulant-treated patients was higher than that of those not treated with anticoagulants (χ2 = 9.129, P = 0.004). Cure rate of the anticoagulation + TIPS treatment group was 64.3%, which was also higher than that of the non-anticoagulation group; however, this was not significantly different (χ2 = 3.938, P = 0.096). The presence of hepatic encephalopathy, serum bilirubin and albumin levels were major prognostic factors for Gynura segetum-induced HVOD. Anticoagulation therapy significantly increased the cure rate; however, TIPS treatment did not have a beneficial effect on the cure rate.

Only a few articles on HVOD induced by Gynura root have been reported in the literature. It is suspected that pyrrolizidine alkaloids in Gynura root might be responsible for HVOD. In this paper, we report a case of HVOD and review the literature.
