GhoshAbstract:A study to evaluate cypermethrin resistance in Rhipicephalus (Boophilus) microplus and Hyalomma anatolicum collected from Muktsar and Mansa districts of Punjab state, India, was conducted by using adult immersion test (AIT). The regression graphs of probit mortality of ticks plotted against log values of concentrations of cypermethrin was utilized for the determination of slope of mortality, lethal concentration for 50% (LC50), and the resistance factor (RF). On the basis of the data generated on variables (mortality, egg mass weight, reproductive index, and percentage inhibition of oviposition), the resistance levels were categorized. Resistance to cypermethrin was categorized as level II and I in R. (B.) microplus collected from Muktsar and Mansa districts, respectively, whereas, H. anatolicum from both locations showed a susceptible status. The RF values of Muktsar and Mansa field samples of engorged R. (B.) microplus (5.48 and 2.18, respectively) were much higher as those of engorged H. anatolicum (1.12 and 0.82, respectively) indicating a lower level and slower rate of development of cypermethrin resistance in multi-host ticks. The data generated in the current study might be of immense help in formulating suitable control measures against ticks and tick-borne diseases of animals.

Results obtained by the “larval packet test” (LPT) showed a low grade resistance (level-I, RF <5) in the tick species to both deltamethrin and cypermethrin in 10 areas and higher grade resistance (level-II, RF <25) to deltamethrin in one area, where intensive use of synthetic pyrethroids are practiced for tick control. Low grade resistance to diazinon (level I) was recorded in six areas where organophosphates compounds are extensively used for agricultural practices allowing increased exposure of the moulting instars of the ticks to these chemicals. Biochemical analysis of the samples suggested involvement of esterase and alterations of acetylcholinesterase in the resistance mechanisms.

GhoshAbstract:Summary In an attempt to develop vaccine against Hyalomma anatolicum anatolicum, the protective efficacy of rHaa86 was evaluated against experimental challenge infestations of homologous tick species and lethal dose of Theileria annulata. Following challenge, a significant difference of 20.9% (P < 0.01) in the dropping per cent of ticks fed on immunized and control animals was recorded. A statistically significant reduction of 49.6 mg (P < 0.01) in the weight of ticks fed on immunized animals in comparison with control was noted. The ticks dropped from immunized animals laid fewer eggs and a reduction of 68.1 mg (P < 0.05) in comparison with the ticks fed on control animals was noted. The DT%, DO%, DR% and E% were calculated as 73.8, 31.3, 15.8 and 82.3% respectively. In all the calves fever (rectal temperature ≤39.5°C) was detected after a mean period of 7.2 days in immunized calves and on 5.8 days in control calves following lethal challenge with T. annulata. The mean Maximum Macroschizonts Index was 7.8% and 10.6% in the immunized and control calves respectively. Two calves (identification no. 351 and 354) died in the immunized group while all five calves died in the control group. The data demonstrated that rHaa86 antigen-based vaccine could serve as one of the effective components of the integrated control of H. a. anatolicum and T. annulata.

As a component of integrated control measures against the tick vector, attempts have been made to identify candidate protein molecules for development of an anti-tick vaccine in the different stages of this tick species. By strategic methods of isolation of the targeted molecules using affinity purification of proteins showing reactivity with immunoglobulins of animals previously immunized with different sources of tick antigen, six proteins were isolated in a significantly pure form. The recovery percentage of the candidate proteins was very low in the range of 1.8-8.0%. The protective potentiality of the antigens was tested in immunization and challenge trials and maximum potential was observed in the proteins isolated from total larval extracts, nymphal extracts and in larval glycoprotein. One of the antigens with a molecular weight of 37kDa isolated from larvae of H. a. anatolicum was found to have some adverse effect on development of Theileria annulata in the vector tick. The progress in the development of immunoprophylactic measures against H. a. anatolicum is discussed.

By comparing the efficacy of rHaa86 and rBm86, it was observed that vaccine based on rHaa86 will be more effective in controlling homologous challenge infestations (68AE7% against larvae and 45AE8% against adults). The results of this trial demonstrated that species-specific antigens are the better choice for vaccine development and could serve as an effective tool for the integrated control of H. a. anatolicum.

Thirty conserved genus specific nucleotide change were observed in Hyalommid and Boophilid ticks. These conserved sequences were sufficient to identify embryonic stages of the ticks. These conserved sequences at the genus level could act as biomarker for identification of ticks during epidemiological studies of tick borne diseases, transmitted by Hyalommid and Rhipicephalid ticks.

Immunization of cross-bred cattle against Hyalomma anatolicum anatolicum by purified antigens

SharmaAbstract:Extracts prepared from unfed larvae of Hyalomma anatolicum anatolicum were purified by immunoaffinity chromatography using anti-gut IgG as ligand. Affinity purified antigen (Aff-GHLAg) was used to immunize cross-bred (Bos taurus × B. indicus) calves of 6–7 months of age. Immunized calves rejected 70.6% larvae, 54.5% nymphs and 61.9% adults. No significant changes in the engorged weight of females was observed; however, significant decrease in the engorgement weight of larvae and nymphs was recorded. There was a significant decrease in the emerging nymphs (p < 0.05) and adults (p < 0.01) of the tick stages fed on immunized animals. SDS-PAGE analysis revealed three antigenic proteins of 100, 59.4 and 37 kDa responsible for induction of resistance in the host.

Acaricide resistance status of Rhipicephalus (Boophilus) microplus and Hyalomma anatolicum collected from Haryana

Tick control strategy involves mainly the use of synthetic acaricides which results into development of acaricide resistance. In the present study, the tick samples were collected from organized and unorganized farms of Hisar (HSR) and Fatehabad (FTH) districts of Haryana state to determine the status of development of acaricide resistance. The discriminating doses (DD) of deltamethrin, cypermethrin and diazinon were established against reference susceptible IVRI-I (national registration no. NBAII/IVRI/BM/1/1998) and IVRI-II (national registration no. NBAII/ IVRI/HA/1/1998) lines of Rhipicephalus (Boophilus) microplus and Hyalomma anatolicum , respectively, using adult immersion and larval packet tests. The results indicated that FTH isolates of R (B) microplus and H anatolicum developed resistance against all the 3 acaricides while the HSR isolates of both the tick species are slowly developing resistance. The data on field status of acaricide resistance in two sampled districts of Haryana will be helpful to adopt suitable strategy to overcome the process of development of resistance in ticks.
