    Abstract:

    Abstract Computer assisted movement tracking was used to characterize the motility of two marine microalgae, Isochrysis galbana and Tetraselmis chui , and to investigate the toxicity of Cu, Pb, and Cd on motile percentage, curvilinear velocity, average path velocity, straight line velocity, linearity, straightness, and wobble. Except for motile percentage, all other motility parameters differed significantly between I. galbana and T. chui . Based on relative motile percentage data, the median effective concentration (EC 50 ) of Cu on the motility of I. galbana and T. chui was 31.4 and 1.3 μmol/L, respectively, while for Pb it was 37.8 and 10.9 μmol/L and for Cd it was 121.6 and 37.8 μmol/L, respectively. Compared to I. galbana , T. chui was more sensitive to all tested metals. The toxic effect of the heavy metals on motility exhibited the following decreasing order for both species: Cu > Pb > Cd. Our results indicate that I. galbana and T. chui motility is sensitive to heavy metals and can be used as an indicator for toxicology bioassays.

    Abstract:

    Salah satu penyebab rendahnya pertumbuhan dan kelulushidupan pada  perkembangan awal larva sampai spat tiram mutiara ( Pinctada maxima ) adalah pemberian pakan alami yang kurang tepat. Tujuan penelitian ini adalah untuk menganalisis pemberian pakan alami Isochrysis galbana dan Chaetoceros amami dan kombinasinya terhadap pertumbuhan dan kelulushidupan veliger-spat Pinctada maxima .  Penelitian ini dilaksanakan di PT. Autore Pearl Culture, Sumbawa, NTB. Rancangan Acak Lengkap dengan 5 perlakuan dan 3 kali ulangan diterapkan pada penelitian ini, yaitu pemberian pakan alami 100% I. galbana ); kombinasi 75% I. galbana dan 25% C. amami , kombinasi 50% I. galbana dan 50% C. amami , kombinasi 25% I. galbana dan 75% C. amami , dan 100% C. amami , dengan kepadatan 7 x 106 sel/mL. Untuk mengetahui pengaruh perlakuan dilakukan analisis ragam (ANOVA) yang dilanjutkan dengan uji wilayah ganda Duncan.. Hasil penelitian menunjukkan bahwa selama pemeliharaan veliger-spat dengan pakan  I. galbana , C. amami dan kombinasinya berpengaruh sangat nyata (P<0,01) terhadap pertumbuhan panjang mutlak cangkang dan berpengaruh nyata (P<0,05) terhadap kelulushidupan. Pertumbuhan panjang mutlak dorsal-ventral dan anterior-posterior cangkang tertinggi dicapai pada perlakuan kombinasi 25% I. galbana dan 75% C. amami, yaitu 1.623,7µm dan 2.217,11 µm. Kelulushidupan tertinggi dicapai oleh pemberian 100% I. galbana sebesar 6,85%. The highest dorsal-ventral and anterior-posterior shell growth (1,623.7 µm and 2,217.11 µm) reached by spat fed on combination of 25% I. galbana and 75% C. amami .     Abstract:

    Oithona sp. adalah salah satu dari jenis copepoda yang sering diberikan sebagai pakan alami di dalam pembenihan ikan dan udang. Kajian tentang pemanfaatan pakan alami untuk budidaya Oithona sp. baik secara volume terbatas maupun massal belum banyak dilakukan. Penelitian ini bertujuan untuk mengetahui pengaruh pakan alami (Chaetoceros calcitrans dan Isochrysis galbana) dengan dosis yang berbeda terhadap nilai ingestion rate dan peforma pertumbuhan Oithona sp. Pengaruh pakan alami (Chaetoceros calcitrans dan Isochrysis galbana) dengan dosis yang berbeda terhadap nilai ingestion rate dan peforma pertumbuhan telah dilakukan pengkajian pada penelitian ini. Metode eksperimen telah digunakan pada penelitian ini dimana menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 3 ulangan. Perlakuan yang diujikan berbasis pada jumlah berat yang sama antara C. calcitrans dan I. galbana dengan perbandingan berat masing–masing 50%:50%. Perlakuan tersebut adalah A (304.737:137.741 sel/mL), B (373.607:168.871 sel/mL), C (442.478:200.000 sel/mL), D (511.349:231.129 sel/mL) dan E (580.219:262.259 sel/mL). Copepoda Oithona sp. dipelihara dalam botol gelas ukuran 50 mL yang diisi air laut 10 mL dengan kepadatan 1 ind. Oithona sp. per mL. Oithona sp. dipelihara selama 21 hari dengan kondisi suhu 26,67˚C; salinitas 25,67‰; dan pH 9. Hasil penelitian menunjukkan dosis fitoplankton yang berbeda berpengaruh nyata (P<0,05) terhadap ingestion rate (Ir) dan peforma pertumbuhan Oithona sp. Berdasarkan hasil penelitian disimpulkan bahwa dosis fitoplankton, C. calcitrans dan I. galbana (50%:50%) sebanyak 511.349:231.129 sel/ml dapat meningkatkan peforma pertumbuhan Oithona sp. yaitu kepadatan total (11,83 ± 0,419 ind./mL), laju pertumbuhan populasi (0,124 ± 0,00215/hari), produksi telur (32,67 ± 2,08 telur/betina) dan memiliki nilai ingestion rate (256.597 ± 14.595,729 sel/mL) yang tinggi dibanding perlakuan lain.     Abstract:

    Abstract This study investigated the growth inhibition effects of three phenicol antibiotics on microalgae used in aquaculture. Different dose levels of chloramphenicol (CAP), florfenicol (FF), and thiamphenicol (TAP) were added to cultures of one freshwater green alga, Chlorella pyrenoidosa , and two marine algae, Isochrysis galbana and Tetraselmis chui . For the two marine algae, FF showed higher toxicity levels (EC 50 , 1.3–8 mg l −1 ) than CAP (4–41 mg l −1 ) and TAP (38–158 mg l −1 ). CAP was more toxic to the freshwater algae (EC 50 , 14 mg l −1 ) than FF (215 mg l −1 ) and TAP (1283 mg l −1 ). TAP was the least toxic to the three algae, but maintained the highest stability during the test period. Among the tested algae, T. chui was the species most sensitive to the three antibiotics. This study demonstrates that all three phenicol antibiotics can inhibit growth of the three microalgae and should be carefully used in aquaculture.
