comparative proteomic profiling during ovarian development of the shrimp Metapenaeus ensis
Molecular Biology Reports, 2014Co-Authors: Ju Cui, Ka Hou ChuAbstract:Two-dimensional electrophoresis and mass spectrometry were used to identify proteins that are differentially expressed during ovarian maturation in Metapenaeus ensis. 87 spots with consistently significant quantitative differences (≥1.5-fold for vol%) among stage I, III and V ovaries were chosen for MS/MS analysis. 45 spots were significantly matched to known proteins in the database (Mascot score >40). Half of them were down-regulated, in contrast to 9 out of 45 proteins that were up-regulated as ovarian maturation proceeded. Functionally, these identified proteins could be classified into five major groups, including cytoskeleton (11 %), metabolism (18 %), signal transduction (32 %), gene expression (14 %) and immune response (7 %). Among the differentially expressed reproduction-related proteins, the mRNA expression level of cellular retinoic acid/retinol binding protein in M. ensis (MeCRABP) during ovarian maturation was further characterized by quantitative real-time PCR. It was down-regulated during ovarian maturation. In situ hybridization further revealed that MeCRABP transcript was localized in ooplasm of previtellogenic oocytes but not in vitellogenic oocytes. These results demonstrate the application of proteomic analysis for identification of proteins involved in shrimp ovarian maturation and they provide new insights into ovarian development.

Phylogenetic relationships among the genera of the Penaeidae (Crustacea: Decapoda) revealed by mitochondrial 16S rRNA gene sequences

The analysis generally supports the three-tribe scheme proposed by Burkenroad (1983) but it is not consistent with the five-group classification of Kubo (1949). Three clades are resolved: (Penaeus sensu stricto + Fenneropenaeus + Litopenaeus + Farfantepenaeus + Marsupenaeus + Melicertus + Funchalia + Heteropenaeus), (Metapenaeus + Parapenaeopsis + Xiphopenaeus + Rimapenaeus + Megokris + Trachysalambria) and (Metapenaeopsis + Penaeopsis + Parapenaeus), corresponding to the Penaeini, Trachypenaeini and Parapenaeini respectively, while the affinities of Atypopenaeus and Trachypenaeopsis are obscure. The molecular data support that Miyadiella represents the juvenile stage of Atypopenaeus. Within the Trachypenaeini, Trachypenaeus sensu lato is clearly paraphyletic, while the monophyly of Penaeus sensu lato in the Penaeini is questionable.

In Metapenaeus ensis, there are six naupliar, three protozoeal and three mysid larval instars, followed by postlarval development. Irregular heartbeat begins late in naupliar instar 6. Co-ordinated beating at 400–600 beats min−1 commences in the first protozoeal instar and continues throughout larval life. Initially, the contractile region is located more posteriorly in the cephalothorax and has a single pair of ostia, and the arterial distribution is limited to a single anterior vessel. In later mysid instars, a second cardiac pumping site develops posterior to, but connected with, the original site. This extension is more muscular, contains additional ostia and develops additional distribution vessels supplying the cephalothorax and abdominal areas. The original site is gradually merged into the new extension and only small refinements in the circulation occur in postlarval and juvenile life. Changes in physiological responses of the heart also occur throughout development. Responses to intra-pericardial microinjection of 5-hydroxytryptamine change drastically during development, as do cardiac responses to ambient hypoxia. Similarly, heartbeat of later juvenile instars is inhibited by injection of tetrodotoxin, while heartbeat of larval and early juvenile instars is not, suggesting that neurogenic regulation via the cardiac ganglion arises later in development. Our present studies attempt to integrate the anatomical and physiological changes in the development of the crustacean heart.

Morphometric analysis indicated that infested male and female shrimp have slightly, but significantly, lower body weight than uninfested shrimp of the same carapace length. While the parasites sterilize their hosts, the low infestation rate (0·53%) appears not to pose a threat to the shrimp fisheries in the Zhujiang estuary.

High variability in survival rates were observed among animals from different spawners. Larvae exposed to 0.06 and 0.10 mg Cu l−1 over a 10-day period had lower survival rates and exhibited slower development than control animals. No significant treatment effects were found at 0.04 and 0.08 mg Cu l−1. Effects of copper on survival and growth of postlarvae were studied over an 8-day period. Copper concentrations lower than 0.10 mg l−1 did not affect survival rates. Slower growth rates were observed in postlarvae exposed to 0.08 and 0.10 mg Cu l−1.

Antimicrobial activities of the chitosans dissolved in acetic, lactic, formic and hydrochloric acid at different concentrations (1.00, 0.50, 0.25, 0.10 and 0.05%) were tested in vitro by using the disk diffusion method with standard microorganisms (Pseudomonas putida, Pseudomonas fluorescens, Vibrio parahaemolyticus, Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli 1, Escherichia coli 2, Listeria monocytogenes, Enterecoccus faecalis, Aeromonas caviae). On the other hand, gentamicin was used as positive control. Antimicrobial test results indicated that chitosan demonstrated different effects depending on the chitosan concentration, solvent material and bacteria type. In the present study, both the extracted and commercial chitosans were observed to have antimicrobial effects on nearly all types of the bacteria.

In addition, the physicochemical characteristics of the chitosan extracted from M. stebbingi shells were compared to commercial chitosan. The degree of deacetylation was calculated by the titration method and elemental analysis. The molecular weight was determined by viscosimetric methods. The results of the study indicate that shrimp shells are a rich source of chitosan as 17.48% of the shell’s dry weight is consisted of this material. Extracted chitosan exhibited a lower molecular weight, higher degree of deacetylation, higher viscosity and higher water and fat binding capacities compared to the commercial chitosan.

For both species, the highest protein values were recorded during the summer (p < 0.05). The most abundant amino acids were, in decreasing order, glutamic acid, aspartic acid, arginine, lysine, leucine, glycine and alanine. The ratio of essential to nonessential amino acids (mean value) for green tiger shrimp and speckled shrimp were 0.60 and 0.59, respectively. Mineral contents of the two species varied with season (p < 0.05), except for the Ca content in green tiger shrimp.

Aquaculture, 2004Co-Authors: Subramaniyan Manivannan, Bob Kennedy, Indrani KarunasagarAbstract:Viral diseases remain a major threat to the shrimp aquaculture industry because some can cause massive disease outbreaks. Samples of shrimp were collected from the offshore waters of southwest coast of India during November 1999 to August 2000. The samples consisted of 30 individuals (n=30) belonging to different species such as Metapenaeus monoceros, Metapenaeus elegans, Metapenaeus dobsoni, Solenocera choprai and Parapenaeopsis stylifera. The samples were screened for the presence of monodon baculovirus by histopathology and polymerase chain reaction (PCR). Of the 30 individuals examined, 4 specimens of M. monoceros and 1 specimen of M. elegans showed multiple occlusion bodies in the hepatopancreatic cells, suggestive of infection by MBV-like viruses. Samples positive for MBV by histopathology were found to be positive by nested PCR, confirming that the occlusions were indeed due to MBV. The results suggest that Metapenaeus spp. could serve as a reservoir of the virus in the natural environment. This study documents the presence of MBV in wild caught M. monoceros and M. elegans for the first time.

The incidence of the virus in non-cultured crustaceans from shrimp farms was also studied. The results indicate that wild-caught asymptomatic marine shrimp such as Metapenaeus dobsoni, Parapenaeopsis stylifera, Solenocera indica and Squilla mantis carry WSSV. This virus could be detected in apparently healthy marine crabs Charybdis annulata, C. cruciata, Macrophthalmus sulcatus, Gelasimus marionis nitidus and Metopograpsus messor. The virus could also be detected in asymptomatic Macrobrachium rosenbergii cultured inland far away from coast. Detection of carrier animals required two-step nested PCR.

Hasil perhitungan menunjukkan bahwa ukuran udang jari pertama tertangkap apong (lc) pada panjang karapas 14.5 mm. Pertumbuhan allometrik negatif (b < 3). Panjang karapas takhingga (L∞) adalah 42.6 mm, dengan indeks kurva pertumbuhan (K) sebesar 1.3/tahun dan to=-0,017 tahun. Waktu terjadinya pertumbuhan maksimum (tmb) pada umur 0.59 tahun, pada panjang karapas 23 mm. Laju kematian total (Z) sebesar 8.19/tahun, laju kematian alami (M) sebesar 1.43/tahun dan laju kematian penangkapan (F) adalah 6.76/tahun. Puncak penambahan baru terjadi pada bulan Juni, yaitu sebesar 17.86%. Laju pengusahaan (E) sebesar 0.83/tahun,menunjukan tingkat pengusahaan berlebih atau terjadi growth-overfishing, sehingga perlu pengendalian eksploitasi. Penelitian bertujuan untuk mengetahui beberapa aspek reproduksi dan pengkajian musim dan daerah pemijahan udang M. elegans di perairan Segara Anakan. Metode peneltian yang digunakan survei post facto. Daerah penelitian dibedakan menjadi 3 wilayah perairan. Penentuan zona berdasarkan dua aspek, yaitu aspek ekologis dan aspek daerah penangkapan (fishing ground) alat tangkap apong. Pada setiap zona ditentukan tiga lokasi sampling. Sampel udang pada setiap stasiun diambil 30 ekor udang betina terbesar, sehingga terkumpul 270 ekor udang sampel. Hasil penelitian menunjukkan bahwa M. elegans memijah sepanjang tahun, dengan dua puncak musim pemijahan yaitu bulan April/Mei dan November/Desember. Daerah pemijahan udang jari ( M. elegans ) adalah perairan laguna bagian tengah. Meskipun pada lokasi pemijahan terjadi pendangkalan akibat laju sedimentasi yang tinggi dan menjadikan perairan keruh serta kondisi salinitas yang berfluktuasi tinggi, namun pemijahan tetap terjadi di perairan tersebut. Kata kunci : aspek reproduksi, daerah pemijahan, udang jari, laguna Segara Anakan. Study on the reproduction and spawning ground of Metapenaeus elegans in Segara Anakan Lagoon Cilacap was conducted from March to December 2004. This study was aimed at investigating some reproductive aspects and to discover the spawning ground M. elegans. The research was used survey post facto method. The researh area was divided into three waters zone. The zone was determinated based on two aspect, i.e. ecological aspect and fishing ground aspect. Three sampling station determined of each zone. The shrimp sample was taken from three unit apong (set net) cathces. 30 biggest male shrimp were took off from in each, so 270 male shrimp collected. The result showed that M. elegans spawned along the year with two peaks of spawning season, i.e. April/May and November/December. Spawning ground of fine shrimp ( M. elegans ) was found in lagoon waters, despite high turbidity, high sedimentation rate and high salinityfluctuation in the water.
