    Abstract The small lungworm Muellerius capillaris is very prevalent in goats and causes production losses. Its control is particularly difficult. The nematophagous fungus Duddingtonia flagrans has been shown to be effective in trapping a large range of gastro-intestinal nematode larvae but its trapping activity against small lungworm remains to be assessed. The purpose of this work was firstly, to evaluate the ability of first-stage larvae of M. capillaris (L1) to induce trap formation in in vitro conditions and secondly, to determine the effect of D. flagrans on the L1 infectivity to snails. In experiments on agar, the presence of L1 failed to induce any D. flagrans traps whereas in the same conditions, gastro-intestinal third-stage larvae induced 44–135 traps/cm 2 depending on the species. Moreover, when the traps were pre-induced by Haemonchus contortus larvae, the L1 of M. capillaris were not trapped. For the in vivo trial, two goats naturally infected with M. capillaris received D. flagrans chlamydospores at the daily dose rate of 5 × 10 5 spores/kg BW for 8 days. Faeces were collected individually before, during and 11 days after spore administration. On each day of harvest, the initial larval output was determined. The remaining faeces were subjected to coproculture at 21 °C for 7 days. At the end of this period, L1 were collected and used to infect snails (30 snails per goat isolate each snail given 40 L1 by direct deposit of the larvae on the foot of the snail). These snails were artificially challenged in contrast to others that were exposed to natural infection by exposure to faeces carrying first-stage M. capillaris larvae. The natural infection used the same number of snails, i.e. 30 snails deposited on the faeces of each goat. After 3 weeks at room temperature, the infective larvae present in the snail foot were counted. There was no difference in the survival of the L1 in faeces after coproculture whether the faeces contained D. flagrans or not. The infectivity of the extracted larvae from the two goats before and after fungal administration was the same. The number of infective larvae per snail obtained after “natural” infection showed variations that were not related to the presence of D. flagrans mycelium in faeces. These trials clearly indicate that D. flagrans was unable to trap or to alter the infectivity of M. capillaris first-stage larvae and thus cannot be considered as a non-chemotherapeutic alternative approach to the control of the small lungworm in goats.

    The survival capacities of Muellerius capillaris (Nematoda: Protostrongylidae) free-living larval stages (L1) of an Israeli desert isolate (Nubian isolate) (92 mm rain per annum) and a French isolate from a temperate habitat (650 mm rain per annum) were compared under humid and extreme dry conditions. Under the dry conditions (7% relative humidity [r.h.] and 33% r.h. at 23 °C), both isolates exhibited the same remarkable survival capacity for periods of 17 and 28 days, respectively (>92% survival, P >0·1). However, during and after recovery from the anhydrobiotic state, different behaviour patterns of L1 were observed in each isolate. Under humid conditions (97% r.h.) for 10 days, both isolates exhibited similarly low survival percentages (13·4±4·6%, S.E. , survival for the Nubian isolate, 3±3% for the French isolate, P >0·05). In water, the French isolate was less active, survived better (52±8·4% vs 28·7±3·7, by day 28, P 0·05).     Les larves du premier stade (L1) du nematode Muellerius capillaris sont resistantes a la deshydratation au sein des feces. Des L1 dans des feces ont ete soumises a des conditions de deshydratation croissantes (non-deshydrate; 70% de matiere seche (MS), 0 h; 70 % MS, 240 h; 90 % MS, 240 h; 95 % MS, 48 h) avant l'infestation. L'infestation des mollusques hotes intermediaires est moins importante et les larves 13 sont moins grandes dans les lots infestes par des L1 soumises au prealable a la deshydratation. La taille des 13 pourrait etre un indicateur des stress subis par les larves L1 avant l'infestation des hotes intermediaires.

    One hundred and eighty-seven dairy goats originating from 7 farms in western France were treated individually with febantel (probenzimidazole drug) at a dose rate of 5 mg/kg in spring 1991. Individual faecal samples were collected on d 0, 7 and 21 post-treatment and processed for counting first stage larvae of Muellerius capillaris (LPG: larvae per gram of faeces). Eggs of digestive tract strongyles were also counted on d 0 and 7 (EPG: eggs per gram of faeces). Blood samples were taken on d 0 to assess pepsinogen concentration related to abomasal damage caused by strongyle infection. The individual characteristics of the goats were recorded. The procedure was repeated on the same animals in autumn 1991. The faecal larval count reduction (FLCR) was mainly related to farm and season: farm 1 in the spring and farm 6 in the autumn showed a higher proportion of high responder (HR) goats (FLCR > 80%), whereas farm 3 exhibited a lower proportion of HR goats in the autumn. On the other hand, the characteristics of the goats (breed, physiological status, age, weight, presence or absence of wattles and horns) as well as the initial level of parasitism (LPG, EPG and pepsinogen concentration) all had a role which was limited or zero on the variability of response to treatment. Goats could not be individually categorized according to their response level because the FLCR values obtained in the spring were not repeated in the autumn, suggesting that other environmental parameters could be involved.

    The high prevalence of benzimidazole-resistant nematodes in French grazing dairy goat flocks led to a search for nematode-control schemes based on integrated approaches with non-chemical options, like vaccination, grazing management, or biological control using nematophagous fungi. The effect of the daily feeding of goats with spores of the nematophagous fungus Duddingtonia flagrans on third-stage larvae (L3) of Teladorsagia circumcincta was examined in faecal cultures. In addition, the effect of D. flagrans on the survival of first-stage larvae (L1) of Muellerius capillaris was tested. Twenty-two culled dairy goats previously raised in a zero-grazing system were twice infected at monthly intervals with 5,000 and then 7,500 T. circumcincta L3. Eight animals were infected with a benzimidazole-susceptible (BZs) strain while the remainder received a benzimidazole-resistant one (BZr). Six culled goats naturally infected with M. capillaris were purchased from private farms. All the goats were divided in two groups, one group receiving daily 5×105 chlamydospores of D. flagrans/kg body weight per goat for seven consecutive days in the food, the other group acting as control. For T. circumcincta-infected goats, individual egg counts and coprocultures (13 days, 25 °C) followed by L3 extraction with the Baermann method were performed. For M. capillaris-infected goats, extraction of L1 with the Baermann apparatus was individually performed on day 0 and after coprocultures on days 7, 10 and 14. Reductions in percentage development of T. circumcincta L3 in fungus groups compared with control groups ranged from 84% (BZs strain) to 90% (BZr strain). A decrease in M. capillaris L1 recovery was noted on days 7 and 10 (a reduction of 70% compared with day 0) and on day 14 (85%), but this pattern was similar in both groups, whether receiving the fungus or not. At the dosage of 5×105 spores/kg body weight, D. flagrans was highly effective in reducing the larval development of T. circumcincta in goats faeces. In contrast, the fungus did not reduce M. capillaris L1 survival in faeces in our conditions.

    Seven dairy-goat farms, located in central, western France, were studied in order to assess the variability in susceptibility of the lungworm Muellerius capillaris first-stage larvae (L1) to 3 different anthelmintics in relation to farm origin, by means of motility tests. The motility tests were performed by mixing larval suspensions with pyrantel (PYR), thiabendazole (TBZ) or ivermectin (IVE) solutions (3 concentrations x 3 incubation durations). The same anthelmintic tests were repeated 7 and 21 d after febantel (probenzimidazole) treatment of the goats. Before the treatment of goats, the average ratio (x 100) of L1 motility compared with control were very different for the 3 anthelminthics: 44, 30 and 59, respectively for TBZ, PYR and IVE. The ratios of L1 motility were the same at days 7 and 21 after the treatment of goats. The susceptibility of L1 to anthelmintics varied a great deal from 1 farm to another when goats had not been previously treated, whereas L1s from the same farms were the same after treatment of goats. The occurrence of different motilities of L1 with the anthelmintic tests from 1 farm to another should be related to the existence of different populations of M capillaris. The L1 motilities were reduced after anthelmintic treatment of goats suggesting a temporary effect on populations of M capillaris females shedding L1.

    The survival capacities of Muellerius capillaris (Nematoda: Protostrongylidae) free-living larval stages (L1) of an Israeli desert isolate (Nubian isolate) (92 mm rain per annum) and a French isolate from a temperate habitat (650 mm rain per annum) were compared under humid and extreme dry conditions. Under the dry conditions (7% relative humidity [r.h.] and 33% r.h. at 23 degrees C), both isolates exhibited the same remarkable survival capacity for periods of 17 and 28 days, respectively (> 92% survival, P > 0.1). However, during and after recovery from the anhydrobiotic state, different behaviour patterns of L1 were observed in each isolate. Under humid conditions (97% r.h.) for 10 days, both isolates exhibited similarly low survival percentages (13.4 +/- 4.6%, S.E., survival for the Nubian isolate, 3 +/- 3% for the French isolate, P > 0.05). In water, the French isolate was less active, survived better (52 +/- 8.4% vs 28.7 +/- 3.7, by day 28, P < 0.05) and retained a better morphological appearance (shrunk less) than the Nubian isolate. Larval development of the French isolate in the land snail Theba pisana was significantly faster than that of the Nubian isolate, reaching a higher proportion of infective stages (L3) on day 36 post-infection (78.6% vs 56% P > 0.05). However, similar infection intensities were recovered in snails infected with either of the isolates (59.8 +/- 10.3, S.E., n = 14, for the French isolate, 53 +/- 6, n = 13, for the Nubian isolate, P < 0.05).
