    Abstract:

    We have identified a transcript derived fragment (TDF) corresponding to SGT1 in a study of differential gene expression on the resistant wild peanut, Arachis diogoi, upon challenge from the late leaf spot pathogen, Phaeoisariopsis personata, and cloned its full-length cDNA followed by subsequent validation through q-PCR. Sodium nitroprusside, salicylic acid, ethephon and methyl jasmonate induced the expression of AdSGT1, while the treatment with abscisic acid did not elicit its up-regulation. AdSGT1 is localized to both nucleus and cytoplasm. Its overexpression induced hypersensitive-like cell death in tobacco under transient conditional expression using the estradiol system, and this conditional expression of AdSGT1 was also associated with the up-regulation of NtHSR203J, HMGR and HIN1, which have been shown to be associated with hypersensitive response in tobacco in earlier studies. Expression of the cDNA in a susceptible cultivated peanut variety enhanced its resistance against the late leaf spot pathogen, Phaeoisariopsis personata, while the heterologous expression in tobacco enhanced its resistance against Phytophthora parasitica var. nicotianae, Alternaria alternata var. nicotianae and Rhizoctonia solani. Constitutive expression in peanut was associated with the co-expression of resistance-related genes, CC-NB-LRR and some protein kinases.

    The wild relatives of peanut are resistant to various economically important diseases including late leaf spot (LLS) caused by Phaeoisariopsis personata, compared with the susceptible cultivated peanut (Arachis hypogaea L.). The interaction of the late leaf spot pathogen, Phaeoisariopsis personata and the highly resistant, diploid peanut wild species, Arachis diogoi was analyzed at the molecular level by differential gene expression studies. Genes up-regulated with in 48 h of pathogen challenge were isolated as partial cDNAs. Some of the isolated genes, which are shown to be involved in the first line of defense in plants, were further characterized with respect to their transcriptional regulation in response to pathogen. Among the isolated clones, two were found to encode cyclophilin like proteins. One of the two isolated partial cDNAs encoding cyclophilin like proteins was extended using 5' RACE. The full length cDNA, designated as AdCyp, was 886 bp in length and encodes a polypeptide of 172 amino acids. Southern hybridization suggests that AdCyp is possibly coded by a single gene and at least one more identical gene is present in Arachis diogoi genome. AdCyp exhibits evolutionary conservation across the kingdoms. Phylogenetic analysis showed that AdCyp belongs to the subgroup I of Group I in cyclophilins. A translational fusion of GFP-AdCyp was found to localize to both cytosol and nucleus. AdCyp transcripts were found to accumulate in response to the treatments with pathogen as well as phytohormones. Constitutive heterologous expression of AdCyp resulted in enhanced resistance to Ralstonia solanacearum and reduced susceptibility towards Phytophthora parasitica var. nicotianae in transgenic tobacco and the resistance was associated with higher transcript levels of various defense related genes.

    Angular leaf spot of bean caused by Phaeoisariopsis griseola is a serious foliar disease of French bean in northern India.  The relative efficacies of different fungicides including six systemic and non-systemic fungicides and four combi-products were evaluated against P griseola under in vitro and in vivo conditions. The in vitro evaluation studies indicated that fenamidone (10%) + mancozeb (50%) (Sectin WG), pyraclostrobin (5%) + metiram (55%) (Cabrio Top WG), azoxystrobin (20%) + difenoconazole (12.5%) (Amistar Top SC), trifloxystrobin (25%) + tebuconazole (50%) (Nativo WG) and azoxystrobin (25%) (Amistar SC) alone were potent growth inhibitors of P griseola. Field evaluation for two consecutive years (2011 and 2012) revealed that four periodic sprays of Sectin at 15 days interval were most effective in controlling the disease and enhancing the green pod yield.

    Abstract:

    Angular leaf spot caused by Phaeoisariopsis griseola (Sacc) Ferraris is one of the most important foliar diseases of French bean in mid-hills of Himachal Pradesh. Survey and surveillance studies conducted revealed that the disease occurred in moderate to severe form in different French bean growing localities of Solan, Shimla and Sirmour districts of the state. The in vitro evaluation of botanicals against the mycelial growth P griseola revealed that the extract of garlic bulb at 10 per cent concentration caused significant growth inhibition (84.62%) of the test pathogen followed by neem seed kernel extract. Amongst the different fungal antagonists evaluated in vitro Trichoderma harzianum proved most inhibitory to P griseola while testing its antagonistic efficacy through dual culture or through its toxic substances.

    Abstract:

    Angular leaf spot of Phaseolus vulgaris is a serious disease caused by Phaeoisariopsis griseola, in which two major gene pools occur, namely Andean and Middle-American. Sequence analysis of the SSU region of nrDNA revealed the genus Phaeoisariopsis to be indistinguishable from other hyphomycete anamorph genera associated with Mycosphaerella, namely Pseudocercospora and Stigmina. A new combination is therefore proposed in the genus Pseudocercospora, a name to be conserved over Phaeoisariopsis and Stigmina. Further comparisons by means of morphology, cultural characteristics, and DNA sequence analysis of the ITS, calmodulin, and actin gene regions delineated two groups within P. griseola, which are recognised as two formae, namely f. griseola and f. mesoamericana.

    Abstract:

    Angular leaf spot caused by Phaeoisariopsis griseola is an economically important disease of beans (Phaseolus vulgaris) in southern Africa. The success of local programmes breeding for resistance to this disease depends to a large extent on the genetic variation within the pathogen population. To assess variability within the pathogen, 28 isolates of P. griseola from various localities were compared using isozyme analysis by means of starch gel electrophoresis. Thirteen loci were identified in 10 enzyme systems. Using UPGMA, three electrophoretic types were detected. The most common type included the South African isolates, namely seven from the Mpumalanga and KwaZulu-Natal provinces, respectively, 10 from Malawi, and one from Portugal. Two isolates from Bembeke in Malawi, and one from the Netherlands, differed from the rest. An isolate of Phaeoramularia angolensis, used as an outgroup, differed from the P. griseola isolates in all enzyme systems tested. The high homology of banding patterns among isolates of P. griseola from southern Africa suggests the local population to be uniform.
