Influence of leaf temperature and blue light on the accumulation of rosmarinic acid and other phenolic compounds in Plectranthus scutellarioides (L.)

Zimmermann, Stine Kögler, Heiko MibusAbstract:Abstract In this study, the influence of different lamp types on physiology and secondary metabolites of Plectranthus scutellarioides (Solenostemon scutellarioides, Coleus blumei) was examined. The following four lamp systems were tested: a new microwave plasma lamp (MPL) emitting artificial sunlight, a commercial high-pressure sodium lamp (HPS), a ceramic metal halide lamp (CDM), and light-emitting diodes (LEDs). The lamps differed not only in the spectral properties of the emitted light but also in the emission of infrared radiation. The aim of this study was to investigate both the influence of the spectral quality and the influence of the leaf temperature from four different lamp systems on morphology and secondary metabolites in P. scutellarioides. Total phenolic compounds were quantified with colorimetric methods. For detailed description of the phenolic compounds, qualitative and quantitative analysis of selected phenolic compounds were performed by HPLC. Stem elongation was increased in plants grown with MPL and LED light, which was attributed to the higher amount of far-red light in the emission spectra. On the other hand, higher infrared radiation from MPL and HPS lamps led to increased leaf temperatures compared to plants grown with LED or CDM light, resulting in faster plant development indicated by greater leaf pair formation. In addition to rosmarinic acid (RA), which is typical for members of the family Lamiaceae, luteolin and apigenin glycosides were detected by mass spectrometry. The results demonstrated that P. scutellarioides leaves contained three acylated cyanidin diglycosides, one of them containing just a coumaroyl residue, the other two containing one and two additional malonyl residues, whereby malonylated compounds appeared in higher quantities. Leaves grown with LED and CDM light contained the highest amount of RA and flavone glycosides per dry weight, which was attributed to a lower leaf temperature compared to leaves developed under MPL or HPS light. Further, an increased blue light content in the emission spectra led to thicker leaves and consequently to a higher accumulation of secondary metabolites per leaf area.

Di Indonesia, tanaman ini umumnya dikenal dengan nama daerah jawer kotok. Secara etnofarmakologi telah digunakan dalam berbagai pengobatan seperti postpartum, dermatitis, sakit perut, batuk, nyeri pada otot, asma, gangguan pencernaan dan lain-lain. Berdasarkan studi farmakognosi dan kegunaannya secara tradisional, P. scutellarioides berpotensi secara terapetik dalam pengembangan obat herbal. Review ini menjelaskan aktivitas farmakologi. P scutellarioides pada studi in vitro mapun in vivo. Aktivitas farmakologi tersebut yaitu antiinflamasi, imunomodulator, antioksidan, antihistamin dan antidiabetes.

AKTIVITAS ANTIDIABETES EKSTRAK ETANOL DAUN ILER (Plectranthus scutellarioides (L.) R.Br.) PADA TIKUS PUTIH GALUR WISTAR DENGAN METODE INDUKSI ALOKSAN

In addition to synthetic drugs, traditional medicine widely used by diabetic patients. Research of several species from Plectranthus genus showed that Plectranthus amboinicus and P. esculentus have antidiabetic activity in rat. Therefore, it is necessary to do antidiabetic activity research and its effective dose of plants iler (Plectranthus scutellarioides (L.) R.Br) which derived from the same genus and has been used empirically as oral antidiabetic. Antidiabetic activity test conducted using white male Wistar rats which given diabetogen alloxan. Rat who had developed diabetes later given different dose of iler leaves ethanol extract and glibenclamide 0,5 mg/kgBB as a positive control. Blood glucose level was measured using amperometric method utilizing enzymatic reaction of glucose dehydrogenase which measured by glucometer. As a result, a dose of 200 mg / kg dose had the highest antidiabetic activity, followed by a dose of 300 mg / kg, the percentage decrease in relative blood glucose are 21,52% and 3,64% respectively, but a dose of 100 mg / kg didn’t have antidiabetic activity. Antidiabetic activity of 200 mg/kg dose of extract didn’t have any significant difference with glibenclamide, which has percentage decrease in relative blood glucose 21,35%.

This research was aimed to identify the antioxidant potency of ethanolic extract of Plectranthus scutellaroides leaves in vitro by using spectrophotometric methods with DPPH (1,1-diphenyl-2-picryl hydrazyl) using vitamin C as reference. Concentrations of samples used were 75, 100, 115, 125 and 135 ppm. The antioxidant activity was measured by visible spectrophotometry at three wavelengths of 498, 518 and 538 nm. The result showed that the n-hexane fraction gave the highest antioxidant activity with IC50 of 52.5 ppm, 15 times lower than that of vitamin C (IC50 of 3.33 ppm). Phytochemical screening of the Plectranthus scutellaroides leaves indicated the presence of flavonoids, polyphenolic, monoterpenoids, sesquiterpenoids, steroids and triterpenoids. Penelitian ini bertujuan untuk mengetahui daya antioksidan dari daun Plectranthus scutellarioides (L.) R.Br., secara in vitro dengan metode spektrofotometri menggunakan pereaksi 1,1-difenil 2-pikrilhidrazil (DPPH) dengan vitamin C sebagai pembanding. Daun diekstrak menggunakan etanol lalu difraksinasi dengan n-heksana, etil asetat dan air. Variasi konsentrasi sampel uji yang digunakan pada pengujian ini adalah 75, 100, 115, 125 dan 135 ppm. Aktivitas antioksidan diukur secara spektrofotometri pada tiga panjang gelombang yaitu 498, 518 dan 538 nm. Hasil penelitian menunjukkan bahwa fraksi n -heksana pada konsentrasi tersebut memberikan aktivitas antioksidan paling kuat dengan nilai IC sebesar 52.5 ppm, 15 kali lebih lemah dibandingkan dengan vitamin C (IC50 = 3.33 ppm). Hasil penapisan fitokimia terhadap daun Plectranthus scutellaroides menunjukkan adanya senyawa golongan flavonoid, polifenolat, monoterpenoid, sesquiterpenoid, steroid dan tritertenoid. Bulletin of Health Research, 2012Co-Authors: Yun Astuti NugrohoAbstract:Malaria is a major public health problem in the world and developing countries in particular, causing an estimated 1-2 million deaths per year, an annual incidence of 300-500 million clinical cases and more than 2 billion people were at risk of infection from it. But it is also becoming more difficult to treat malaria due to the increasing drug resistance. Therefore, the need for alternative drugs is acute. This study aims at investigating the in vivo antiplasmodial activity of Piper betle L., fruit (buah sirih), Plectranthus scutellarioides (L.) R. BR., (daun miyana), honey and egg yolk combination. Methods: A rodent malaria parasite, Plasmodium berghei, was inoculated into Swiss albino mice. The mice were infected with Ixl0 5 parasites intraperitoneally. The Piper betle L., fruit (buah sirih), Plectranthus scutellarioides (L.) R. BR., (daun miyana), honey and egg yolk combination were combined and administered by an intra gastric tube daily for seven days starting from the day of parasite inoculation. The control groups received the same amount of solvent (vehicle) used to suspend each dose of the herbal drug. Chloroquine was used as a standard drug, administered through the same route. Results: Combination of Piper betle L., fruit (buah sirih), Plectranthus scutellarioides (L.) R. BR., (daun miyana), honey and egg yolk were observed to inhibit Plasomodium berghei parasitaemia in the Swiss albino mice 100 % on the sixth day. Conclusion: The study could partly confirm the claim in East Sulawesi traditional medicine that the Piper betle L., fruit (buah sirih), Plectranthus scutellarioides (L.) R. BR., (daun miyana), honey and egg yolk combination has therapeutic values in human malaria. There was, thus, the need to initiate further in-depth investigation by using different experimental models.

BR, madu dan kuning telur dimanfaatkan oleh masyarakat Sulawesi Utara untuk mengobati malaria. Diharapkan kombinasi buah sirih ( Piper betle L), daun miyana ( Plectranthus scutellarioides (L.) R. BR, madu dan kuning telur juga memiliki potensi untuk merangsang respon imun. Penelitian ini dilakukan untuk mengetahui pengaruh pemberian kombinasi buah sirih (Piper betle L), daun miyana (Plectranthus scutellarioides (L.) R. BR, madu dan kuning telur terhadap aktivitas dan kapasitas sel makrofag. Metode, tiga puluh ekor mencit Swiss dibagi ke dalam 5 grup. Grup I mendapatkan air suling, grup II mendapatkan 0,55 mg/20g BB Stimuno. Sementara kelompok perlakuan: grup III sampai V masing-masing mendapatkan 0,07 mL; 0,21 mL; 0,6 mL/20 g BB. Bahan uji diberikan secara oral pada hari pertama sampai dengan ketujuh. Pada hari kedelapan, kepada masing-masing mencit disuntik secara intraperitoneal bakteri Staphylococcus aureus (SA). Aktivitas dan kapasitas sel makrofag dihitung dari sediaan apus cairan peritoneum dengan menghitung persentase fagosit yang melakukan fagositosis dari 100 fagosit. Kapasitas fagositosis ditetapkan berdasarkan jumlah SA yang difagositosis oleh 50 fagosit aktif. Hasil, aktivitas dan kapasitas sel makrofag meningkat seiring dengan peningkatan dosis kombinasi buah sirih ( Piper betle L), daun miyana ( Plectranthus scutellarioides (L.) R. BR, madu dan kuning telur dibandingkan dengan kontrol negatif. Kata kunci: Buah sirih, Piper betle L, Makrofag, Fagositosis

2018Co-Authors: Adriani WizeAbstract:Tumbuhan miana (Plectranthus scutellarioides (L.) R. Br) adalah satu tumbuhan hias yang tergolong kedalam famili Lamiaceae. Daun tumbuhan ini banyak digunakan sebagai obat tradisional seperti mengobati asma, bronchitis, batuk, melancarkan haid, bisul, diare, cacingan, wasir, dan kencing manis. Pada penelitian ini telah dilakukan penentuan kandungan fenolik total, uji aktivitas antioksidan, aktivitas antimikroba, dan sitotoksik dari fraksi metanol daun miana. Ekstraksi daun miana dilakukan dengan metode maserasi secara bertahap menggunakan pelarut heksana, etil asetat dan metanol. Pada penelitian ini penentuan kandungan fenolik total dilakukan dengan metode Folin Ciocalteu, uji aktivitas antioksidan dengan metode DPPH (1,1 diphenyl-2-picrylhydrazyl), aktivitas antimikroba (antibakteri dan antijamur) dilakukan dengan metode difusi cakram melalui penentuan zona bening terhadap bakteri Escherichia coli bakteri Staphylococcus aureus serta jamur Candida albicans. Uji sitotoksik dilakukan dengan metode BSLT (Brine Shrimp Lethality Test). Kandungan fenolik total fraksi metanol diperoleh 3,03 mg/L, dan menunjukkan aktivitas antioksidan yang sangat kuat dengan nilai IC50 yaitu 11,254 mg/L. Hasil uji aktivitas antimikroba terhadap bakteri Escherichia coli, bakteri Staphylococcus aureus serta jamur Candida albicans menunjukkan nilai zona bening masing-masing (1,6 mm), (1,8 mm), dan (1,1 mm). Fraksi metanol daun miana tidak menunjukkan aktivitas sitotoksik. Kata Kunci : Tumbuhan Miana, (Plectranthus scutellarioides (L.) R. Br), Total Fenolik, Antioksidan, Antimikroba dan Aktivitas Sitotoksik.

In vivo diagnostics of early abiotic plant stress response via Raman spectroscopy.

ScullyAbstract:Development of a phenotyping platform capable of noninvasive biochemical sensing could offer researchers, breeders, and producers a tool for precise response detection. In particular, the ability to measure plant stress in vivo responses is becoming increasingly important. In this work, a Raman spectroscopic technique is developed for high-throughput stress phenotyping of plants. We show the early (within 48 h) in vivo detection of plant stress responses. Coleus (Plectranthus scutellarioides) plants were subjected to four common abiotic stress conditions individually: high soil salinity, drought, chilling exposure, and light saturation. Plants were examined poststress induction in vivo, and changes in the concentration levels of the reactive oxygen-scavenging pigments were observed by Raman microscopic and remote spectroscopic systems. The molecular concentration changes were further validated by commonly accepted chemical extraction (destructive) methods. Raman spectroscopy also allows simultaneous interrogation of various pigments in plants. For example, we found a unique negative correlation in concentration levels of anthocyanins and carotenoids, which clearly indicates that plant stress response is fine-tuned to protect against stress-induced damages. This precision spectroscopic technique holds promise for the future development of high-throughput screening for plant phenotyping and the quantification of biologically or commercially relevant molecules, such as antioxidants and pigments.
