Traditional medicinal systems are based on plant as a source of medicine for the treatment of chronic debilitating and life threatening disease and disorders. Annona reticulate Linn. is widely used in the traditional medicine for the treatment of various disease conditions. Methanolic leaves extract of Annona reticulata Linn. Was investigated for anticancer potential using sulforhodamine B (SRB) cytotoxicity assay against colon cancer (HCT15), Human lung cancer (Hop65) and Human hepatoma (HEPG2) cell lines. Adriamycin was used as a standard to compare the results. The extract exhibited anticancer effect against all the cell lines. The median growth inhibition (GI50) concentration for extract was <10 µg/ml against Human lung cancer and hepatoma cell line, showing moderate anticancer efficacy of methanolic extract against these cell lines. In conclusion, methanolic leaves extract of Annona reticulata Linn. Possess moderate anticancer activities in vitro which could be attributed to the presence of anticancerous phytochemicals like acetogenins.

Leaves of A. reticulata were screened for phytochemicals and in vitro antioxidant, antibacterial and antifungal activity. The shade dried leaves were extracted with methanol and aqueous methanolic extract was partitioned successively with n-butanol, chloroform and acetone solvents. Methanolic extract was subjected to antioxidant screening using DPPH free radical scavenging activity and H 2 O 2 scavenging activity. Antibacterial and antifungal activity of extract and fractions were analyzed on eight different clinical bacterial and fungal strains using agar well diffusion method and broth dilution method (MIC and MMC determination). The antioxidant activity showed that the extracts exhibited scavenging effect in concentration-dependent manner. The extract showed potent inhibitory effect against Bacillus subtilis and Escherichia coli bacterial strains while in case of fungal strains the maximum effect was observed against Candida blanki . The maximum zone of inhibition of n-butanol, chloroform and acetone fractions was observed against B. subtilis , and E. coli respectively while all fractions exhibited potent inhibitory effect against C. blanki . MIC and MBC values were determined for active samples, methanol extract and chloroform fraction against Staphylococcus aureus , B. subtilis , E. coli and Pseudomonas aeruginosa which revealed lower MIC and MBC values. The fungal strains Candida albicans , Saccharomyces cerevisiae and C. blanki were used to calculate MIC and MFC values for methanol extract and acetone fraction which demonstrated lower MIC and MFC values. The results provided evidence that the plant is richly supplied with numerous phytoconstituents that might indeed be potential sources of natural antioxidant, antimicrobial agents and supplementary food.

ChandakAbstract:The present study was envisaged to evaluate the ethno-medical claims of Annona reticulata L. roots for anthelmintic efficacy in vitro in comparison with the standard chemotherapeutic agent, albendazole on adult Indian earthworms ( Pheretima posthuma ). The efficacy was evaluated by monitoring gross visual motility, paralysis and mortality time and new software based tool, PASS (Prediction of Activity Spectra for Substances). Literature confirmed that roots of the plant are rich in aporphine alkaloids and acetogenins. Therefore, study includes investigation and correlation of the biological activity spectrum of the aporphine alkaloids and acetogenins using PASS with in vitro anthelmintic activity. In vitro anthelmintic trials of extract was conducted at 20, 40, 60 mg/mL. The extract was found to be effective at all concentrations but a more significant result was at concentration 60 mg/mL. This could be due to synergistic effect of both the phytoconstituents. However, exact mechanism of action need to be studied.

Phytochemical and different products obtained from plant are used as medicines, pharmaceuticals, cosmetics and food supplements. Annona reticulata Linn. (牛心果 niu xīn guǒ; Bullock's heart) is a versatile tree and its fruits are edible. Parts of A. reticulata are used as source of medicine and also for industrial products. It possesses several medicinal properties such as anthelmintic, analgesic, anti-inflammatory, antipyretic, wound healing and cytotoxic effects. It is widely distributed with phytochemicals like tannins, alkaloids, phenols, glycosides, flavonoids and steroids. Present article is an attempt to highlight over taxonomy, morphology, geographical distribution, phytoconstituents and pharmacological activities of A. reticulata reported so far.

Antioxidant, antimicrobial activity and in silico PASS prediction of Annona reticulata Linn. root extract

ChandakAbstract:Abstract Microbial infections and diseases are frequently associated with several pathogenic strains of bacteria and fungi. Plants of the reticulata genus are a notable source of new therapeutic agents including antioxidant and antimicrobial. This study reports the antioxidant and antimicrobial activities of methanolic root extract of Annona reticulata Linn. The antioxidant property of extract was evaluated using DPPH free radical scavenging and hydrogen peroxide assay. Antibacterial tests were performed using the agar cup method whereas Poison plate method was used to assess sensitivity of fungal strains. The biological potential of major phytoconstituents as antimicrobial agent was screened by new software based tool, PASS. The dose dependent scavenging was observed at concentrations 20, 40, 60, 80 and 100 μg/ml which were compared to ascorbic acid. The probable activity (Pa) of neoannonin using PASS was found to be 0.541. The extract was significantly active against all strains of bacteria but the largest zone of inhibition was found against B. cereus. Predominant growth reduction was observed in fungi Tricoderma viride and Candida albicans. The results indicate that the extract show potential as a source of new antimicrobial drug and may impart health benefits by its antioxidant property.

Larvicidal Efficacy of Root and Stem Bark Extracts of the Plant, Annona reticulata against Filarial Vector, Culex quinquefasciatus

(Annonaceae) against early 3 rd instar larvae of Culex quinquefasciatus Say, 1823 ( Cx. quinquefasciatus ) up to 72 hours. Among all the solvent extracts used, chloroform: methanol (v/v 1:1) root and stem bark extracts of the plant showed highest larvicidal activity. Further, the effect of chloroform: methanol (v/v 1:1) root and stem bark extracts of the plant was investigated on early 1 st – 4 th instars larvae of Cx. quinquefasciatus . Early 1 st - 3 rd instars larvae of Cx. quinquefasciatus showed 100% mortality with remarkably very low concentrations of chloroform: methanol (v/v 1:1) root and stem bark extracts. Non- target organisms were non responsive to chloroform: methanol (v/v 1:1) solvent root and stem bark extracts. So, chloroform: methanol (v/v 1:1) root as well as stem bark extracts of A. reticulata can effectively be used as potential sources to control Cx. quinquefasciatus population.

Evaluation of Larvicidal Efficacy of Acetone Leaf Extracts of Annona reticulata Linn. Against Aedes aegypti, Anopheles stephensi and Culex quinquefasciatus (Diptera: Culicidae)

( A. reticulata ), (family: Annonaceae) against 1 st to 4 th instars larvae of three dreadful mosquitoes namely Aedes aegypti L., 1762 ( Ae. aegypti ), Anopheles stephensi Liston, 1901 ( An. stephensi ) and Culex quinquefasciatus Say, 1823 ( Cx. quinquefasciatus ). Cent percent mortality was observed at 12 ppm concentration in 1 st , 2 nd and 3 rd instars larvae of Ae. Aegypti , where as 9 ppm concentration showed 100% mortality in 1 st , 2 nd and 3 rd instars larvae of An. stephensi after 72 h of exposure. At 6 ppm concentration, 100% mortality was observed in 1 st instar larvae after 24 h of exposure and in 2 nd and 3 rd instars larvae of Cx. quinquefasciatus after 48 h of exposure. In the 72 h bioassay experiment, LC 50 and LC 90 values progressively decreased with the increase in exposure period, having the lowest values at 72 h of exposure. Among the three mosquitoes, Cx. quinquefasciatus proved to be most susceptible to acetone leaf extract of A. reticulata . Some non target organisms were found to be non-responsive to the bioactive extract when exposed. So, acetone leaf extract of A. reticulata was proved to be an efficient larvicidal agent against 3 important vectors like Ae. aegypti, An. stephensi and Cx. quinquefasciatus .

Larvicidal Potentiality of Root Extracts of Annona reticulata Linn. Against the Filarial Vector Culex quinquefasciatus Say (Diptera: Culicidae)

( A. reticulata ), family Annonaceae against filarial vector Culex quinquefasciatus Say, 1823 ( Cx. quinquefasciatus ). Petroleum ether, hexane, and ethyl acetate root extracts of the plant were examined to established their larvicidal potentiality against 3 rd instar larvae of Cx. quinquefasciatus mosquito species. Crude and only ethyl acetate root extracts of the plant at different concentrations were examined on 1 st - 4 th instars larvae of Cx. quinquefasciatus . LC 50 value of ethyl acetate extract after 24 h of exposure against 3 rd instar larvae of Cx. quinquefasciatus were also examined on some non target organisms. Phytochemical analyses of the root extracts of A, reticulata were also investigated. 100% mortality were noticed at very low concentrations of petroleum ether, hexane, and ethyl acetate root extracts of A. reticulata against 3 rd instar larvae of Cx. quinquefasciatus . LC 50 and LC 90 values of ethyl acetate root extract was lower than petroleum ether and hexane root extracts after 24, 48, and 72 h of exposure. Crude and ethyl acetate root extracts showed excellent larvicidal activity against Cx. quinquefasciatus mosquito species. 100% mortality was observed at very low concentration for crude and ethyl acetate root extracts. LC 50 and LC 50 values gradually decreased with time of exposure for crude and ethyl acetate root extracts of A. reticulata . No mortality and sluggishness were observed in control experiments as well as on examined non target organisms. Phytochemical analyses of root extracts of the plant revealed the presence of different secondary metabolites. So crude and ethyl acetate root extracts of A. reticulata can effectively be used for larval mortality against Cx. quinquefasciatus mosquito species at a very low concentration.

Cumulative Cytotoxicity Assay of the Aqueous and Ethanolic Extracts of the Selected Medicinal Plants using Crown Gall Tumor Disc Bioassay

Gorde, Ajay KharcheAbstract:The present study was conducted to test for in vivo crown gall tumor disc bioassay using potato discs of the aqueous and ethanolic extracts of Annona reticulata with Allium sativum, Allium fistolisum Brassica oleraceae and correlate cytotoxicity results with known pharmacological activities of the plants. Cytotoxicity was evaluated in terms of Crown Gall Tumor Disc Bioassay. The use of highly specific, quantitative bioassays which require only a short period of time to obtain results are available for studying crown gall tumor formation. Results showed that the extracts of Annona reticulata with Allium sativum, Allium fistolisum and Brassica oleraceae were potent against the Crown Gall Tumor Disc Bioassay when compared alone with combined extracts. It indicated that bioactive components are present in these plants that could be accounted for its pharmacological effects. Thus, the results support the uses of these plant species in traditional medicine.

Brine Shrimp Lethality Assay of the Aqueous and Ethanolic Extracts of the Selected Species of Medicinal Plants

PatilAbstract:The present study was conducted to test for in vivo Brine Shrimp Lethality Assay (BSLA) of the Aqueous and ethanolic extracts of Annona reticulata with Allium fistolisum and Brassica oleraceae and correlate cytotoxicity results with known pharmacological activities of the plants. Cytotoxicity was evaluated in terms of LC50 (lethality concentration). Ten nauplii were added into three replicates of each concentration of the plant extract. After 24 hours the surviving brine shrimp larvae were counted and LC50 was assessed. Results showed that the extracts of Annona reticulata with Allium fistolisum and Brassica oleraceae were potent against the brine shrimp when compared alone with combined extracts. It indicated that bioactive components are present in these plants that could be accounted for its pharmacological effects. Thus, the results support the uses of these plant species in traditional medicine.

Microwave Assisted Extraction, Fractionation, Total Phenolic and Flavanoid Estimation and Antioxidant Potential of Annona reticulate Leaves

The extract obtained was subjected to column chromatography and the fractionation was done to get different fractions. Preliminary phytochemical screening was done and the fractions were subjected to total phenolic and flavanoid content estimation. The crude extract contained 502 mg GAE/100 g of phenolic content and among the fractions AM8 contained the maximum amount of phenolic content that is 360 mg GAE/100 g. The flavanoid content present in the crude extract of the Annona reticulata was found to be 127 mg QE/100 g. Among all the tested fractions, the fraction AM8 showed more flavanoids i.e., 112 mg QE/100 g and 118 AN/100 g. The antioxidant activity has been studied in-vitro by using DPPH, hydrogen peroxide assay. DPPH, H2O2 and Nitric-oxide scavenging effect of Annona reticulata L. leaf extracts were found to be dose dependant with maximum inhibition at highest concentration. Methanolic extract of the leaves of Annona reticulata Linn. is found to have most potent anti-oxidant activity in DPPH, H2O2 Radical scavenging methods. IC50 value DPPH and H2O2 inhibition of methanolic extract at highest concentration (400 μg/mL) are 62.58 ± 1.15, 68.27 ± 1.05 and 64.01 ± 1.02. The results suggest that all the tested extracts are having antioxidant property, but the methanol is having significantly higher flavonoid and phenol content. Due to presence of higher flavonoid and phenol content in methanol, it may be considered as the fraction with better pharmacological property in comparison to other tested extracts.

(Annonaceae) an interesting plant for investigating its anti-hyperglycemic potential. Objective: Different fractions prepared from hydro-alcoholic extract of A. reticulata leave were investigated for their blood glucose lowering effect on Streptozotocin (STZ) induced hyperglycemic rats. Methods: Ethyl acetate, methanol, and residual fractions (at dose level of 100 mg/kg by oral route) prepared from the hydro-alcoholic extract of A. reticulata leave were administered for 14 consecutive days to STZ induced hyperglycemic rats for evaluation of their anti-hyperglycemic potential. Anti-hyperglycemic potential was assessed by observation of a decrease in fasting blood glucose level. Results: The studies revealed that ethyl acetate fraction decreased the blood glucose level of hyperglycemic rats from 447.67 to 234.17 mg/dL and is significant (p<0.001) when compared with diabetic control group. The residual fraction and methanolic fraction decreased blood glucose level from 417.83 to 402.50 mg/dL and 432.33 to 371.67 mg/dL respectively but not significant when compared with the diabetic control group. Standard drug metformin (dose 300 mg/kg) reduced the blood glucose level from 447.33 to 219.50 mg/dL. Discussion: Ethyl acetate fraction at tested dose level was capable not only to control the elevated blood glucose level but also able to attenuate certain secondary parameters associated with STZ induced hyperglycemia. Conclusion: This study suggested that the ethyl acetate fraction prepared from hydro-alcoholic extract of A. reticulata leave exhibit potential antihyperglycemic property in the tested experimental models and should be investigated further. 2013Co-Authors: Soumya P Rout, Durga Madhab Kar, Santosh B Mohapatra, Sharada P SwainAbstract:Context: Annona reticulata Linn (Annonaceae) commonly known as bullock's heart is widely distributed all over India and leaves are used as insecticides, anthelmintic, styptic and are also used externally as suppurant. Objective: The present study has been designed to determine the effectiveness of Hydro-Alcoholic extract of leaves of Annona reticulata L. (HAAR) for potential hypoglycemic activity against streptozotocin (STZ) induced hyperglycemic rats. Materials and methods: Hyperglycemia was induced in rats by administration of multiple low doses (40 mg/kg) of Streptozotocin (STZ) to the overnight fasted rats for five consecutive days intraperitoneally. After 12days of STZ administration, the fasting blood glucose levels (FBG) were measured and the rats with FBG level >250 mg/dL were considered to be diabetic and were used in the study. The study was undertaken by using the plant extract at dose levels of 200 mg/kg and 400 mg/kg (as test), p.o. and standard drug as Metformin (300 mg/kg) in both normoglycemic and streptozotocin induced hyperglycemic animals. Hypoglycemic potential was assessed by the measurement of fasting blood glucose, Lipid Profile (total cholesterol, triglycerides), physical parameters (body weight, food and water intake) and Oral Glucose tolerance test. Results and discussion: Results of the studies showed that, the fasting blood glucose level in hyperglycemic and in oral glucose tolerance test, showed a significant (p<0.05) decrease at defined time points, while the observed biochemical and physical parameters showed a good agreement with hypoglycemic property of the extract. Conclusion: The present study suggests that Annona reticulata leaves possess potent glucose lowering effect.
