    Abstract:

    Effects of dietary administration of Bacillus subtilis FPTB13 and chitin, single or combined, on the systemic immunity, mucosal immunity and resistance of Catla (Catla Catla) against Edwardsiella tarda infection were investigated. The probiotic attributes of B. subtilis was tested by conducting antagonism study, safety in Catla, in vitro immunomodulation and dietary immunomodulation. Results of these studies indicated the probiotic potential of the strain. From the preliminary dietary immunomodulation study, a dose of 10(9) B. subtilis cells g(-1) was selected for inclusion into diets for subsequent experiments. Experimental diets were prepared by adding B. subtilis (10(9) cells g(-1)), chitin (2%) and their combination to the basal diet. Different systemic and mucosal immunological parameters viz. oxygen radical production, myeloperoxidase content, lysozyme activity, total protein content and alkaline phosphatase activity showed significant enhancement (p<0.05) after 2 weeks of feeding with the combined diet. B. subtilis and chitin alone also significantly elevated most of the immune responses. All the diets significantly increased the resistance of Catla against E. tarda challenge. The highest post-challenge survival was observed in combined group (i.e. 63.33%). In conclusion, B. subtilis and chitin, alone or combined, had a health ameliorating effect in Catla. The results also collectively suggest the usefulness of applying a combined probiotic and immunostimulant supplemented diet to achieve greater benefits.

    Abstract:

    Immunostimulants increase the immunocompetency and disease resistance of fish by enhancing immune responses. This study determined the in vitro and dietary effects of chitin on immune responses of Catla, Catla Catla. Catla head-kidney leukocytes were cultured in complete RPMI-1640 medium containing 0.01, 0.1, and 1.0 mg/mL of chitin. Leucocyte viability and different cellular immune responses, namely, respiratory burst, nitric oxide production, and leucocyte peroxidase content were analyzed in vitro. In the dietary experiment, chitin-supplemented diets (0.5, 1.0, and 2.0% of feed) were fed to Catla for 2 wk, and respiratory burst, serum lysozyme, and total protein contents were measured after 1 and 2 wk of feeding. Leucocytes incubated in vitro with chitin for 72 h maintained their viability. Chitin significantly (P < 0.05) enhanced all the immune responses in vitro. The lowest dose showed the best response. Dietary chitin also significantly stimulated all the immune parameters in a dose-dependent manner with the highest dose showing the maximum responses. All the responses were higher after 1 wk of feeding and declined thereafter. The results suggest that chitin stimulates Catla cellular and humoral immune responses in vitro and in vivo.

The laboratory maintained B. amyloliquefaciens was used to study antagonistic activity against fish pathogenic bacteria by agar well diffusion assay. Healthy Catla were challenged by this bacterium for determination of its safety. For preparation of probiotic supplemented diet, the bacteria were added to the basal diet (control) at three different inclusion levels i.e., 1 × 10 9 , 1 × 10 8 and 1 × 10 7  CFU/g diet. Fish (weight 25–30 g) were fed with these diets and various immune parameters and disease resistance study were conducted at 4 weeks and 8 weeks post-feeding. The bacterial antagonism study showed inhibition zone against E . tarda, Aeromonas hydrophila, Vibrio parahaemolyticus and V. harveyi. B. amyloliquefaciens was harmless to Catla as neither mortalities nor morbidities were observed after the challenge. Study of different systemic and mucosal immunological parameters viz. superoxide anion production and nitric oxide production, myeloperoxidase content, lysozyme activity and total protein content showed significant enhancement ( p 8 and 10 9  CFU/g B. amyloliquefaciens at both time points with the highest values observed in case of 10 9  CFU/g.     Abstract Dynamic changes in immunoreactivity (innate and specific) of Catla ( Catla Catla ) against the bacterial pathogen Edwardsiella tarda was evaluated in the present study. Healthy Catla were injected intraperitoneally with the LD 50 dose of 10 5.5  CFU/ml to induce edwardsiellosis. Different innate and specific immune responses were measured for the injected and control group on 0th, 2nd, 4th, 7th, 10th, 14th, 20th and 27th day post-injection (p.i.). The clinical and gross signs were first visible at 1 day p.i. and the infected fish showed typical signs of hemorrhagic septicemia. Study of innate immunological parameters viz. superoxide anion production and nitric oxide production showed initial decrease at 2nd and 4th day p.i. followed by increase at 7th and 10th day p.i., respectively. However, the lysozyme activity showed a different trend with no decrement at initial stage. Anti- Edwardsiella antibody production was measured by indirect ELISA that showed an increasing trend with the highest observed on 20th day p.i. Another specific immune parameter i.e. lymphoproliferation showed almost similar trend like antibody production with the highest response observed on 20th day p.i. The results suggest that the protective immunity of Catla against E. tarda essentially depends on the co-operation of both innate and specific immune responses.

    Abstract A cell line, CTE, derived from Catla ( Catla Catla ) thymus has been established by explant method and subcultured for more than 70 passages over a period of 400 days. The cell line has been maintained in L-15 (Leibovitz) medium supplemented with 10% fetal bovine serum. CTE cell line consists of homogeneous population of epithelial-like cells and grows optimally at 28 °C. Karyotype analysis revealed that the modal chromosome number of CTE cells was 50. Partial amplification, sequencing and alignment of fragments of two mitochondrial genes 16S rRNA and COI confirmed that CTE cell line originated from Catla. Significant green fluorescent signals were observed when the cell line was transfected with phrGFP II-N mammalian expression vector, indicating its potential utility for transgenic and genetic manipulation studies. The CTE cells showed strong positivity for cytokeratin, indicating that cell line was epithelial in nature. The flow cytometric analysis of cell line revealed a higher number of cells in S-phase at 48 h, suggesting a high growth rate. The extracellular products of Vibrio cholerae MTCC 3904 were toxic to the CTE cells. This cell line was not susceptible to fish betanodavirus, the causative agent of viral nervous necrosis in a large variety of marine fish.

    Abstract:

    Complete nucleotide sequence of mitochondrial genome (mitogenome) of the Catla Catla (Ostariophysi: Cypriniformes: Cyprinidae) was determined in the present study. Its length is 16,594 bp and contains 13 protein coding genes, 22 transfer RNAs, two ribosomal RNAs and one non-coding control region. Most of the genes were encoded on the H-strand, while the ND6 and eight tRNA (Gln, Ala, Asn, Cys, Tyr, Ser (UCN), Glu and Pro) genes were encoded on the L-strand. The reading frames of two pair of genes overlapped: ATPase 8 with 6 and ND4L with ND4 by seven nucleotides each. The main non-coding region was 929 bp, with three conserved sequence blocks (CSB-I, CSB-II, and CSB-III) and an unusual simple sequence repeat, (TA)(7). Phylogenetic analyses based on complete mitochondrial genome sequences were in favor of the traditional taxonomy of family Cyprinidae. In conclusion present mitogenome of Catla Catla adds more information to our understanding of diversity and evolution of mitogenome in fishes.

    Abstract The compatibility of olive barb, Puntius sarana (Hamilton) with major carps was studied in grow-out carp polyculture system for one year in a set of nine earthen ponds of 0.08 ha each. Three different species combinations evaluated were Control: Catla (Catla Catla Ham.), silver carp (Hypophthalmichthys molitrix Valenciennes), rohu (Labeo rohita Hamilton) and mrigal (Cirrhinus mrigala Hamilton) at 0.5:0.5:1:1; T1: Catla, silver carp, rohu and olive barb at 0.5:0.5:1:1 and T2: Catla, silver carp, mrigal and olive barb at 0.5:0.5:1:1 at combined density of 7500 fingerlings/ha. While survival levels of the carps did not differ significantly in treatments (P > 0.05), silver carp recorded highest survival levels (94–96%) followed by olive barb (87–90%), mrigal (72–74%), rohu (72–73%) and Catla (67–69%). The specific growth rate (SGR) and average harvested body weight (ABW) of Catla and silver carp did not differ significantly among the treatments revealing their competition with mrigal or olive barb to be minimum. In absence of rohu in T2, both mrigal and olive barb showed higher SGR and ABW revealing minimal competition between these two species, while their lower performance in presence of rohu in Control and T1 indicated inter-specific competition with the latter. Such olive barb–rohu inter-specific competition, however, failed to yield significant effect on growth of rohu as revealed from its non-significant SGR difference in presence and absence of olive barb. The lower FCR (2.54 ± 0.06) and higher treatment biomass production (3418.4 ± 95.0 kg ha− 1 year− 1) in T1 with rohu–olive barb combination compared to T2 with mrigal–olive barb (2.84 ± 0.11; 3155.1 ± 104.7 kg ha− 1 year− 1) indicated feasibility and advantage of culturing rohu with olive barb rather than mrigal in carp polyculture. Further, similar biomass production in Control and T1 also indicated feasibility of replacing mrigal with olive barb in the grow-out carp polyculture system.

    The compatibility and growth performance of silver barb Puntius gonionotus (Barbonymous gonionotus) with the three Indian major carps, i.e., Catla Catla, Labeo rohita and Cirrhinus mrigala, were assessed in a 10-month carp polyculture trial. Treatments T-1, T-2, T-3 and T-4 were stocked with three of the above four carp species, with an absence of silver barb, mrigal, rohu and Catla, respectively, while all four species were stocked in treatment T-5. The treatments were stocked at 6000 fingerlings ha−1, with an equal species ratio maintained in each treatment. Incorporation of silver barb into the polyculture system neither affected the survival of any carp irrespective of species combination nor yielded significant changes in biomass production among treatments, except for the one without Catla, where it was significantly low. The study revealed a higher extent of competition between silver barb and rohu, perceptible from the lower growth of one in the presence of the other. Although a certain level of competition of silver barb with mrigal was evident, competition with Catla was not perceptible. Irrespective of species combination with silver barb as a component species, similar total biomass production in treatments revealed the feasibility of its incorporation into the Indian major carp-based polyculture practice without affecting the total yield.

    The effect of water pH on certain haematological parameters was studied in the three species of Indian major carps, Catla (Catla Catla), rohu (Labeo rohita) and mrigal (Cirrhinus mrigala) for 21 days by subjecting fingerlings of these fishes to acidic (pH 5.5, 6.5) and alkaline (pH 8.0, 8.5 and 9.0) water, control groups were maintained at neutral pH. A change in water pH either to acidic or alkaline conditions exerted stress in fish characterized by swelling of erythrocytes, production of immature erythrocytes, and reductions in the total erythrocyte counts, haemoglobin and serum protein content. There were also increases in total leukocyte counts and blood glucose. However, the degree of these responses varied among the three species. Rohu was found to be least affected followed by mrigal to the stress of altered water pH, while Catla was the most vulnerable to pH changes.

    UV-B radiation is a potential stressor to the aquacultural species. Catla Catla, Catla larvae (1.08 ± 0.065 g) were exposed to different doses of UV-B radiation, 0 (control), 504, 1008, 1512 and 2016 mJ/cm2 at a mean radiant energy of 80 μW/cm2 for 21 days. The dose of UV-B radiation was selected on the basis of the field study conducted in Lake Naini, Delhi, India (Latitude: 28°41′26″N and Longitude: 77°12″37″E). Significantly (P < 0.05) lower survival, average weight and specific growth rate were found in UV-B irradiated larvae compared to the control one. Food conversion ratio was 1.5-4-fold higher in UV-B treated larvae compared to the control one. The carbonyl protein (CP), thiobarbituric acid reactive substances (TBARS), superoxide dismutase (SOD) levels were significantly (P < 0.05) higher in UV-B irradiated larvae compared to the control group. Among the treated larvae, CP and SOD were significantly (P < 0.05) higher in larvae exposed at 1512 mJ/cm2 UV-B. A correlation was found between the CP and SOD (R2 = 0.834). Highest TBARS level was found in 2016 mJ/cm2 UV-B exposed Catla. Nitric oxide synthase level was significantly (P < 0.05) lower in UV-B exposed larvae compared to the control one. A 3-fold increased Hsp 70 level was recorded in UV-B irradiated Catla compared to the control larvae. Comet assay analysis indicated that UV-B irradiation enhanced DNA fragmentation. Tail extent moment and the olive tail moment were significantly (P < 0.05) higher in 2016 mJ/cm2 UV-B exposed Catla compared to others. The tail length was significantly (P < 0.05) higher in 1512 and 2016 mJ/cm2 UV-B exposed larvae compared to the other doses. The present study suggests that the Catla is a useful species for the biomonitoring of stress in the aquatic environment.

    Evaluation of functional mechanism of immunostimulant in fish is essential for realizing their therapeutic potential. The impact of dietary supplementation of seeds of Achyranthes aspera on the immune system of Catla Catla was studied. Catla (37.73 ± 6.7 g) were fed with test diets containing 0.5 (D1) and 1.0% (D2) seeds of A. aspera and control diet (D3) for 50 days. Then fish were immunized with heat-killed Aeromonas hydrophila. Blood and tissue samples were collected after 7 days of immunization. Three fish were sacrificed for each parameter. Respiratory burst activity, bactericidal activity and antibody response were significantly (P < 0.05) higher in test diets fed Catla compared to the control one. Nitric oxide synthase and serum lysozyme were significantly (P < 0.05) higher in fish fed with diet D2 compared to the other treatments. There were up-regulations of TNFα, lysozyme c and g gene expressions in kidney and IL-10, lysozyme c and g expressions in hepatopancreas of test diets fed fish compared to the control one. The expression of IL-10 was down-regulated in the kidney of D2 diet fed fish compared to others. In the gill, significantly (P < 0.05) higher expressions of TNFα was found in D1 diet feed fish and lysozyme c and in D2 diet fed fish compared to the other groups. This study showed tissue-specific gene expression pattern in Catla. An inverse relationship was found between the expression of TNFα and IL-10 in kidney of Catla.

    Characteristics and functional efficacy of digestive proteases of Catla Catla, Catla, Labeo rohita, rohu and Hypophthalmichthys molitrix, silver carp were studied. Total protease activity was significantly (P < 0.05) higher in rohu (1.219 ± 0.059 U mg protein )1 min )1 ) followed by silver carp (1.084 ± 0.061 U mg protein )1 min )1 ), and Catla (0.193 ± 0.006 U mg protein )1 min )1 ). Trypsin activity of silver carp and rohu was 89–91% higher than Catla. Chymotrypsin activity was significantly (P < 0.05) higher in silver carp compared with rohu and Catla. The protease activity of rohu and silver carp displayed bell-shaped curves with maximum activity at pH 9; whereas in Catla, maximum activity was found between pH 8 and 11. Inhibition of protease activity with soybean trypsin inhibitor (SBTI), phenylmethylsulfonyl fluoride (PMSF) revealed the presence of serine proteases and inhibition of activity with N-a-p-tosyl-L-lysine-chloromethyl ketone (TLCK) and N-tosyl-L-phenylalanychloromethane (TPCK) indicated the presence of trypsin-like and chymotrypsin-like enzymes in all these three carps. SDS-PAGE showed the presence of several protein bands ranging from 15.3 to 121.9 kDa in enzyme extracts of Catla, rohu and silver carp. The substrate SDS-PAGE evidenced the presence of various protease activity bands ranging from 21.6–93.7, 21.6–63.8 and 26.7–98.5 kDa for Catla, rohu and silver carp respectively. In pH-stat hydrolysis of Chilean fishmeal showed significantly (P < 0.05) higher degree of hydrolysis compared with soybean meal, silver cup (a commercial fish feed of Mexico) and wheat flour, with enzyme preparations of three fishes. The rate of hydrolysis was significantly (P < 0.05) higher in silver carp compared with others.

    Achyranthes aspera, an Indian medicinal plant (family: Amaranthaceae) was incorporated in artificial fish diet, and fed to Catla Catla Catla. After 4 weeks of feeding, fish were immunized with bovine serum albumin (BSA), spleen and blood were sampled on weekly intervals for four times after immunization. Antigen-specific antibody level in serum was determined by ELISA. Antigen clearance was determined in spleen by immuno-electron microscopy. Achyranthes has significantly (P<0.05) enhanced the BSA-specific antibody titers than the untreated control group throughout the study period. The efficiency of antigen clearance was also enhanced in C. Catla treated with Achyranthes.

The main non-coding region was 929 bp, with three conserved sequence blocks (CSB-I, CSB-II, and CSB-III) and an unusual simple sequence repeat, (TA)(7). Phylogenetic analyses based on complete mitochondrial genome sequences were in favor of the traditional taxonomy of family Cyprinidae. In conclusion present mitogenome of Catla Catla adds more information to our understanding of diversity and evolution of mitogenome in fishes.

    Abstract The compatibility of olive barb, Puntius sarana (Hamilton) with major carps was studied in grow-out carp polyculture system for one year in a set of nine earthen ponds of 0.08 ha each. Three different species combinations evaluated were Control: Catla (Catla Catla Ham.), silver carp (Hypophthalmichthys molitrix Valenciennes), rohu (Labeo rohita Hamilton) and mrigal (Cirrhinus mrigala Hamilton) at 0.5:0.5:1:1; T1: Catla, silver carp, rohu and olive barb at 0.5:0.5:1:1 and T2: Catla, silver carp, mrigal and olive barb at 0.5:0.5:1:1 at combined density of 7500 fingerlings/ha. While survival levels of the carps did not differ significantly in treatments (P > 0.05), silver carp recorded highest survival levels (94–96%) followed by olive barb (87–90%), mrigal (72–74%), rohu (72–73%) and Catla (67–69%). The specific growth rate (SGR) and average harvested body weight (ABW) of Catla and silver carp did not differ significantly among the treatments revealing their competition with mrigal or olive barb to be minimum. In absence of rohu in T2, both mrigal and olive barb showed higher SGR and ABW revealing minimal competition between these two species, while their lower performance in presence of rohu in Control and T1 indicated inter-specific competition with the latter. Such olive barb–rohu inter-specific competition, however, failed to yield significant effect on growth of rohu as revealed from its non-significant SGR difference in presence and absence of olive barb. The lower FCR (2.54 ± 0.06) and higher treatment biomass production (3418.4 ± 95.0 kg ha− 1 year− 1) in T1 with rohu–olive barb combination compared to T2 with mrigal–olive barb (2.84 ± 0.11; 3155.1 ± 104.7 kg ha− 1 year− 1) indicated feasibility and advantage of culturing rohu with olive barb rather than mrigal in carp polyculture. Further, similar biomass production in Control and T1 also indicated feasibility of replacing mrigal with olive barb in the grow-out carp polyculture system.

    The compatibility and growth performance of silver barb Puntius gonionotus (Barbonymous gonionotus) with the three Indian major carps, i.e., Catla Catla, Labeo rohita and Cirrhinus mrigala, were assessed in a 10-month carp polyculture trial. Treatments T-1, T-2, T-3 and T-4 were stocked with three of the above four carp species, with an absence of silver barb, mrigal, rohu and Catla, respectively, while all four species were stocked in treatment T-5. The treatments were stocked at 6000 fingerlings ha−1, with an equal species ratio maintained in each treatment. Incorporation of silver barb into the polyculture system neither affected the survival of any carp irrespective of species combination nor yielded significant changes in biomass production among treatments, except for the one without Catla, where it was significantly low. The study revealed a higher extent of competition between silver barb and rohu, perceptible from the lower growth of one in the presence of the other. Although a certain level of competition of silver barb with mrigal was evident, competition with Catla was not perceptible. Irrespective of species combination with silver barb as a component species, similar total biomass production in treatments revealed the feasibility of its incorporation into the Indian major carp-based polyculture practice without affecting the total yield.

    The effect of water pH on certain haematological parameters was studied in the three species of Indian major carps, Catla (Catla Catla), rohu (Labeo rohita) and mrigal (Cirrhinus mrigala) for 21 days by subjecting fingerlings of these fishes to acidic (pH 5.5, 6.5) and alkaline (pH 8.0, 8.5 and 9.0) water, control groups were maintained at neutral pH. A change in water pH either to acidic or alkaline conditions exerted stress in fish characterized by swelling of erythrocytes, production of immature erythrocytes, and reductions in the total erythrocyte counts, haemoglobin and serum protein content. There were also increases in total leukocyte counts and blood glucose. However, the degree of these responses varied among the three species. Rohu was found to be least affected followed by mrigal to the stress of altered water pH, while Catla was the most vulnerable to pH changes.

    Abstract Three different antigenic preparations from the epizootic ulcerative syndrome (EUS) pathogen Aphanomyces invadans were evaluated as vaccine candidate in Catla (Catla Catla). Anti-Catla enzyme immunoconjugate was prepared after isolating Catla immunoglobulin and raising hyperimmune sera against it, in rabbit. Three antigens namely, fungal extract (FE), fungal extract mixed with Freund’s incomplete adjuvant (FIA) in a 1:1 (v/v) ratio (FE + A) and extra cellular product (ECP) were prepared and three groups of Catla were vaccinated intramuscularly with all these antigens (200 μg/fish). Different cellular and humoral immune responses were measured for the entire vaccinated and control group on 0th, 5th, 15th and 25th day post vaccination. Thirty days after the vaccination, the fish were challenged with an A. invadans zoospore dose of 1 × 105 ml−1 and mortality and relative percent of survival (RPS) were recorded. Study of cellular immunological parameters including antigen-specific leukocyte proliferation, antigen-specific nitric oxide production and superoxide anion production showed significantly higher (p   0.05). Increased immune responses and protection have important implications with regard to the control of EUS by vaccination.
